Effect of Electron-beam Irradiation on Polymethoxylated Flavones Content of Citrus unshiu Pomaces

  • To determine the effect of electron-beam irradiation on the contents of polymethoxylated flavones (PMFs) extracts from citrus pomaces (CP), CP was irradiated at 0, 1, 2, or 5 kGy. Methanol extract of the irradiated CP were prepared and the PMF (nobiletin, sinensetin, and tangeretin) content of the extract was determined. Nobiletin and sinensetin of CP extract significantly increased with irradiation dose-dependent. However, electron-beam irradiation decreased the amount of tangeretin in the CP extract. These data suggest that irradiation can liberate phenolic compounds such as nobiletin or sinensetin, but tangeretin might have different pathway of conversion by irradiation. Therefore, irradiation can be a tool to change the composition of PMFs in CP.

Apoptosis-inducing Effect of Herba Patriniae Extract in the Prostate Cancer LNCaP Cells (전립선 암세포에서 패장 추출물의 세포고사 유도 효과)

  • Herba Patriniae(HP) has been known to exert anti-inflammation and -tumoral activity in Korea. However, its molecular mechanism of action is not understood. In this study, we found that HP extract induced apoptosis in androgen-dependent prostate cancer LNCaP cells as evidenced by DNA fragmentation. Our data demonstrated that HP extract-induced apoptotic cell death was accompanied by inhibition of NF- κB activation, lowering effects of intracellular prostate specific antigen(PSA) and androgen reoeptor(AR) expression in a time dependent manner. Taken together, HP extract may inhibit the proliferation of prostate cancer LNCaP cell associated with inhibition of NF- κB activation, PSA and AR expression and that of apoptosis.

In vitro and in vivo antibacterial activities of root extract of tissue cultured Pluchea indica (L.) Less.

  • The methanolic root extract of tissue cultured Pluchea indica (L.) Less. was tested for its antibacterial potentiality against 102 different strains of bacteria belonging to both Gram positive and Gram negative groups. The bacteria could be arranged according to their decreasing order of sensitivity as follows: Staphylococcus aureus, Vibrio cholerae, Bacillus spp. Vibrio parahaemolyticus, shigellae, Salmonella spp., and Escherichia coli. The extract was found to be bacteriostatic in nature against Salmonella typhimurium NCTC 74. When administered to Swiss strain of white mice at the doses of 0.5 and 1.0 mg/kg body weight, the extract could significantly protect the animals challenged with 50 MLD of S. typhimurium NCTC 74. According to the chi-square test, the in vivo data is highly significant (P < 0.001).

  • The effects of Artemisia princeps var orientalis(mugwort), Circium japonicum var ussuriense(Unggungqui) on serum lipid componets in rats was evaluated. Thirty rats divided into 5 experimental groups were fed with the diet containing 1% cholesterol, 0.25% sodium cholate, 10% coconut oil and 5% lard(control diet), supplemetned with 5% plant powder or its water soluble extract(experimental diet) for 4 weeks. Proximate analysis data and compositions of ash, dietary fiber and fatty acids in plants were presented. The concentrations of the total cholesterol was significantly lower in Ungungqui powder, Ungungqui extract and mugwort powder diet groups than the control. Serum HDL-cholesterol level was significantly higher in Unggunqui powder and Unggunqui extract diet group than the control. The concentration of LDL was significantly lower in Unggungqui power or extract diet group compared to the control. The values of VLDL and chylomicron were comparatively lower in Unggungqui power and mugwort powder diet group than in the other diet groups. The concentration of seum triglyceride and phospholipid was lower in Unggungqui powder diet group than in the control.

A Preliminary Study on Hypocholesterolemic and Hypoglycemic Activities of Some Medicinal Plants

  • Total cholesterol level in mice with hypercholesterolemia was determined after intraperitoneal administration of the methanolic extract of some medicinal plants. From the data obtained, it was suggested that the methanolic extract of Elaeagnus crispa, Ixeris dentata, Prunus davidiana, Eriobotrya japonice, Aralia elata and Phragmites communis produced a significant hypocholesterotemic effect. In the case of the extract of Saussurea diamantiaca, on the other band, the total cholesterol level was markedly increased. The methanolic extract of Ixeris dentata, Prunus davidiana and Phragmites communes also decreased the level of blood glucose in alloxan-diabetic male albino mice while that of Eriobotrya japonica, Allium tuberosum, Houttuynia cordata and Eucommia japonica did not produce this effect.

Anti-diabetic activity of Thespesia lampas Dalz & Gibs on alloxan induced rats

  • Anti-diabetic effect was observed with Thespesia lampas Dalz & Gibs (Family: Malvaceae) when given as a root extract in normal as well as alloxan induced diabetic rats. The effects, however, were more pronounced in diabetic animals in which administration of plant extract for 15 days after alloxan induced diabetes, significantly reduced blood glucose levels. After alloxan induced diabetes it was observed that both standard drug (glibenclamide) and aqueous extract of Thespesia lampas were significantly superior to control in reducing blood sugar on long term treatment (15 days). The aqueous extract of T. lampas (300 and 600 mg/kg) reduced the blood glucose levels from $349.2{\pm}7.2$ to $120.7{\pm}4.6$ and $346.3{\pm}3.4$ to $101.8{\pm}6.3$, respectively. The data suggested that T. lampas could be of beneficial in diabetes mellitus in controlling blood sugar. The present investigation established pharmacological evidence to support the folklore claim as an anti-diabetic.

Studies on Cytotoxic Constituents of Korean Forsythia Fruits (한국산개나리 Forsythia viridissima의 세포 독성 성분에 관하는 연구)

  • In the preliminary antitumor screening tests of Crude Drugs and collected plants, the methanol ic extract of Forsythia Fruits in Korean marcket showed signif icant cytotoxic activity against Chinese hamster V-79 cells, but that in Japanese market did not. The former was identifid as Forsythia viridissima Lindley and latter as F.suspensa Vahl on the basis of the morphological observation. When an aqueous solution of the extract prepared from the fruits of F. viridissima (Oleaceae) was partitioned successively with n-hexane, methylene chloride, n-butanol, the cytoyoxic activitywas concetrated in the methylene chloride extract. Fractionation of the extract was made with the guidance of bioassay against V-79 cells to give cytotoxic lignans, matariresiol (1) and arctigenin (2). Their ICso values of compounds 1 and 2 were respectively $7.8{\;}\mu\textrm{g}/ml$ 1 and $1.65{\;}\mu\textrm{g}/ml$. Also, their structures were confirmed by comparison of physical and spectral data in the literature.

  • Glutamate acts as an important neurotransmitter in brain. However, high concentration of glutamate showed an excitatory neurotoxicity and resulted to neuronal cell death. Neuronal cell death is known for one of the reason of Alzheimer's disease, a neurodegenerative disease. We tried to find neuroprotective medicinal plants by neuroprotection activity against glutamate injured HT22 cells as a model system. In the course of bioscreening of various medicinal plants, Taraxacum platycarpum extract showed significant neuroprotective activity. We tried to elucidate mechanisms of neuroprotective activity. T. platycarpum extract reduced ROS and intracellular $Ca^{2+}$ concentration increased by glutamate induced neurotoxicity. In addition, mitochondrial membrane potential was restored to the control level. Also, glutathione level, glutathione reductase and glutathione peroxidase activity were increased by T. platycarpum extract treatment. These data suggested that T. platycarpum showed neuroprotective activity via antioxidative activity.

Anti-stress Effects of Ethanol Extract of Ziziphus jujuba Against Corticosterone-Induced Apoptosis in PC12 Cells

  • The coronavirus disease 2019 (COVID-19) pandemic may be stressful for people. Public health actions, such as social distancing, can make people feel isolated and lonely and can increase stress and anxiety. As a result, there is a growing interest towards various materials to relieve stress. Thus, the present study aimed to investigate the anti-stress effects of ethanol extract of Ziziphus jujuba in PC12 cells treated with corticosterone and its underling mechanisms. Furthermore, the viability of the cells, the apoptosis of the cells, the level of phosphorylation of extracellular signal-regulated kinases (p-ERKs) expression were measured by MTT assay, LDH assay, Hoechst staining assay and western blotting. Our results showed that the extract of Ziziphus jujuba reversed corticosterone-induced damage in PC12 cells, which increased cell viability, decreased LDH release, and attenuated corticosterone-induced apoptosis as compared with the corticosterone-treated group. Therefore, these data suggest that the extract of Ziziphus jujuba could be a good candidate for development as a functional food supplement in the improve the anti-stress effect.

The Effects of Alcohol-Steamed Rhei Rhizoma and Row Rhei Rhizoma on varied Extraction Time in Blood Stasis Model (전탕시간(煎湯時間)에 따른 생대황(生大黃) 및 주대황(酒大黃)이 어혈병태모형(瘀血病態模型)에 미치는 영향(影響))

  • In order to investigate the effects of Alcohol-Steamed Rhei Rhizoma and Row Rhei Rhizoma on varied extract time in both endotoxin-induced blood stasis model(hereafter Endotoxin Model) and hydrocortisone acetate-induced blood stasis model (hereafter HA Model), Half of rats were treated with endotoxin(0.4mg/kg, single Ⅳ, into caudal vein) for Endotoxin Model. Thereafter, they were orally administrated water extract of Alcohol-Steamed Rhei Rhizoma or Row Rhei Rhizoma, which were boiled during 30, 60, 120 minute, respectively. Finally, the number of platelet, fibrinogen, prothrombin time, hematocrit, the number of RBC and WBC were measured after sacrifice. The remainder rats were treated with hydrocortisone acetate(10mg/kg, daily IM for 7 days into the muscular rump) for HA significantly decreased. Together, they were orally administrated for 7 days water extract of Alcohol-Steamed Rhei Rhizoma or Row Rhei Rhizoma that were boiled above methods Finally, the number of platelet, fibrinogen, prothrombin time, hematocrit, the number of RBC and WBC were measured after sacrifice. The results were summarized as follows : 1. The number of platelet was significantly increased in boiled water extract for 30 min of Row Rhei Rhizoma group as compared with that of control group in Endotoxin Model. 2. Fibrinogen level was significantly increased in all administration groups as compared with that of control group in Endotoxin Model. It was significantly increased in all administration groups except boiled water extract for 30 min of Alcohol-Steamed Rhei Rhizoma group as compared with that of control group in HA Model. 3. Prothrombin time was significantly shortened in boiled water extract for 60 min of Alcohol-Steamed Rhei Rhizoma group and boiled water extract for 120 min of Alcohol-Steamed Rhei Rhizoma group as compared with that of control group in Endotoxin Model It was significantly shortened all administration groups as compared with that of control group in HA Model. 4 Hematocrit was significantly increased in all administered groups except boiled water extract for 60 min of Alcohol-Steamed Rhei Rhizoma group and boiled water extract for 30 min of Row Rhei Rhizoma group as compared with that of control group in Endotoxin Model. It was significantly increased in all administration groups as compared with that of control group in HA Model. 5. The number of RBC was significantly decreased in boiled water extract for 60 min of Alcohol-Steamed Rhei Rhizoma group, boiled water extract for 120 min of Alcohol -Steamed Rhei Rhizoma group and boiled water extract for 30 min of Row Rhei Rhizoma administered group in Endotoxin Model. It was significantly increased boiled water extract for 30 min of Row Rhei Rhizoma group and boiled water extract for 60 min of Row Rhei Rhizoma group in HA Model as compared with data of control group. 6 The number of WBC was significantly decreased in all administered groups except boiled water extract for 30 min of Alcohol-Steamed Rhei Rhizoma group and boiled water extract for 60 min of Alcohol-Steamed Rhei Rhizoma group as compared with that of control group in HA Model.
