Influence of Cell Stage of Donor Nucleus on Nuclear Injection, Electrofusion and In Vitro Development in Nuclear Transplant Rabbit Embryos (토기에서 공핵란의 발달단계가 할구주입, 전기융합 및 핵이식 수정란의 체외발달에 미치는 영향)

  • This study evaluated the influence of cell stage of donor nucleus on nuclear injection, electrofusion and in vitro development in the rabbit to improve the efficiency of nuclear transplantation in the rabbit. The embryos of 8-, 16- and 32-cell stage were collected from the mated does by flushing viducts with Dulbecco's phosphate buffered saline(D-PBS) containing 10% fetal calf serum(FGS) at 44, 54 and 60 hours after hCG injection. The blastorneres separated from these embryos were used as donor nucleus. The ovulated oocytes collected at 14 hours after hCG injection were used as recipient cytoplasm following removing the nucleus and the first polar body. The separated blastomeres were injected into the enucleated oocytes by micromanipulation and were electrofused in 0.28 M mannitol solution at 1.5 kV /cm, 60 $\mu$sec for three times. The fused oocytes were cocultured with a monolayer of rabbit oviductal epithelial cells in M-199 solution containing 10% FGS for 72~120 hours at 39$^{\circ}C$ in a 5% $CO_2$ incubator. The cultured nuclear transplant embryos were stained with Hoechst 33342 solution and the number of cells were counted by fluorescence microscopy. The successful injection rate of 8-, 16- and 32-cell-stageblastomeres into enucleated oocytes was 86.7, 91.0 and 93.9%, respectively. The electrofusion rate of 8-, 16- and 32-cell-stage blastomeres with enucleated oocytes was 93.3,89.3 and 79.0%, respectively. Development of blastomeres to blastocyst was similar with 8-,16- and 32-cell-stage donor nuclei(26.2, 25.8 and 26.6%, respectively, P<0.05). The mean number of cell cycle per day during in vitro culture in nuclear transplant embryos which received 8-, 16- and 32-cell- stage nuclei was 1.87, 1.81 and 1.43, respectively.

OPTICAL AND NEAR-INFRARED IMAGING OF THE IRAS 1-JY SAMPLE OF ULTRALUMINOUS INFRARED GALAXIES

  • Optical (R) and near-infrared (K') images of the IRAS 1-Jy sample of 118 ultraluminous infrared galaxies have been studied. All but one object in the 1-Jy sample show signs of strong tidal interaction/merger. Most of them harbor a single disturbed nucleus and are therefore in the later stages of a merger event. Single-nucleus ULIGs show a broad distribution in host magnitudes with significant overlap with those of quasars. The same statement applies to R - K' colors in ULIG and quasar hosts. An analysis of the surface brightness profiles of the host galaxies in single-nucleus sources reveals that about $35\%$ of the Rand K' surface brightness profiles are well fit by an elliptical-like $R^{1/4}$-law, while only $2\%$ are well fit by an exponential disk. Another $38\%$ of the single-nucleus systems are fit equally well with an exponential or de Vaucouleurs profile. Elliptical-like hosts are most common among merger remnants with Seyfert 1 nuclei ($83\%$) and Seyfert 2 optical characteristics ($69\%$). The mean effective radius of these ULIGs is 4.80 $\pm$ 1.37 kpc at Rand 3.48 $\pm$ 1.39 kpc at K'. These values are in excellent agreement with recent quasar measurements obtained at H with HST. The hosts of elliptical-like 1-Jy systems follow with some scatter the same ${\mu}e - r_e$ relation, giving credence to the idea that some of these objects may eventually become elliptical galaxies if they get rid of their excess gas or transform this gas into stars.

A case of Hydrocephalus in S/D Rat with the Closure of Mesencephalic Aqueduct (중뇌수도관 폐쇄에 의한 S/D계통 랫드 수뇌증 1예)

  • Hydrocepbalus may be an acquired or a congenital condition. We have studied the macroscopic and microscopic changes in the hydrocephalus of an inbred Sprague-Dawley rat at postnatal week 8. The animal suspected with the hydrocephalus showed clinical syndromes such as depression, severe ataxia, eye abnormalities, dome-shaped head, and persistent fontanelle. With the postmortem examination, the suspected animal was clearly revealed as a severe internal hydrocephalus. In this animal, severe ventriculomegaly was limited to the third and lateral ventricles, and cortical thining was most apparent in the parieto-occipital region. With the routine histological examination, brain tissue showed aqueductal obstruction, thinning of the cerebral cortex, severe ependymal damage, subependymal edema, damage of choroid plexus of fourth ventricle, enlarged cortical vessels, and expanded ventricles. Aqueductal obstruction was observed with the appearance of simple stenosis at the level of rostral colliculus. Subsequently, the other structures of brain such as septal nucleus, caudate nucleus, and hippocampus etc. were abnormally reconstructed by hydrocephalus. This study suggests that the hydrocephalus can be taken place by primary aqueductal obstruction and this type of hydrocephalus is classified as uncommunicating type. Though the mechanism of aqueductal obstruction is not clear, the morphological studies of this case may be helpful for the further study of hydrocephalus.

The Antinociceptive Effect of Sigma-1 Receptor Antagonist, BD1047, in a Capsaicin Induced Headache Model in Rats

  • Intracranial headaches, including migraines, are mediated by nociceptive activation of the trigeminal nucleus caudalis (TNC), but the precise mechanisms are poorly understood. We previously demonstrated that selective blockage of spinal sigma-1 receptors (Sig-1R) produces a prominent antinociceptive effect in several types of pain models. This study evaluates whether the Sig-1R antagonist (BD1047) has an antinociceptive effect on capsaicin (a potent C-fiber activator) induced headache models in rats. Intracisternal infusion of capsaicin evoked pain behavior (face grooming), which was significantly attenuated by BD1047 pretreatment. BD1047 consistently reduced capsaicin-induced Fos-like immunoreactivity (Fos-LI), a neuronal activator, in the TNC in a dose-dependent manner. Moreover, capsaicininduced phosphorylation of N-methyl-D-aspartate receptor subunit 1 was reversed by BD1047 pretreatment in the TNC. These results indicate that the Sig-1R antagonist has an inhibitory effect on nociceptive activation of the TNC in the capsaicin-induced headache animal model.

Expression of Neuropeptide Y(NPY) and NADPH-diaphorase Neurons in the Hypothalamus and Cerebral Cortex of Fasting and Anorexia Mutant Mice(anx/anx). (절식시킨 생쥐와 식욕부진 돌연변이 생쥐의 시상하부와 대뇌겉질에서 Neuropeptide Y와 NADPH-diaphorase의 이중면역조직화학법에 의한 발현)

  • Food intake is regulated by both central and peripheral mechanisms. In the central nervous, the hypothalamus acts for autonomic and endocrine homeostasis. The paraventricular nucleus(PVN) of hypothalamus is an imprtant site of interaction in central feeding pathways. Neuroepetide Y(NPY)is one of the most powerful neurochemical stimulants of food intake known. Also brain nitric oxide(NO), known as neurotransmitter, is involved in the mechanisms that regulate food intake. In this experiment, 24h fasting mice and anorexia mutant mice have been to examine the expression of NPY, which is the major neuropeptide increasing food intake. Double staining with NPY and nicotinamide-adenine-dinucleotide-phosphate diaphorase(NADPH-d), followed by immunohistochemical method and image analysis, have been used to observe coexisting neurons and the level of expression of each neurons. The results were as follows. 1) NPY-immunoreactivitys reduced immune response of the hypothalamus, particularly paraventricular nucleus(PVN), in anorexia mutant mice. Decreased level of NPY is assumed to be a major pathological factor in anorexia mutant mice. On the other hand, PVN in hypothalamus of fasting mice showed increased immunoreactivity which is in agreement of other researchers. 2) NPY and NADPH-d double staining revealed coexisting neurons in the cerebral cortex. Fasting mice had a tendency to have increased level of coexisting neurons compared to the control group. Compared to the control group, fasting mice express is not increase level of NPY-immunoreactivity, while anorexia mutant mice tended to have a decreased level.

  • Light and electron microscopical characteristics of Perkinsus sp. parasitizing in Manila clam, Ruditapes philippinarum, in Korea were investigated. Trophozoite within the tissue was spherical or ovoid and ranged $2.5-10.5\mu m$ $(mean = 6.2\mu m)$ in diameter. Trophozoite had a nucleus with a prominent nucleolus and a large cytoplasmic vacuole within the cytoplasm. Single trophozoite was phagocytozed by host hemocyte and cluster cells were encapusulated by hemocytes aggregation within the host tissues. Hypnospores incubated in thioglycollate medium (FTM) for 1 to 15 days were also spherical or ovoid and ranged $10-132\mu m$ $(mean\pm S.D.\;:\;44.25\pm 7.91\mu m)$ in diameter. Zoospores were spherical or ovoid, had a nucleus and two flagella. Zoospores contained apical complex, which consisted of conoid, subpellicular microtubules, rhoptries and rectilinear micronemes.

3D epigenomics and 3D epigenopathies

  • Mammalian genomes are intricately compacted to form sophisticated 3-dimensional structures within the tiny nucleus, so called 3D genome folding. Despite their shapes reminiscent of an entangled yarn, the rapid development of molecular and next-generation sequencing technologies (NGS) has revealed that mammalian genomes are highly organized in a hierarchical order that delicately affects transcription activities. An increasing amount of evidence suggests that 3D genome folding is implicated in diseases, giving us a clue on how to identify novel therapeutic approaches. In this review, we will study what 3D genome folding means in epigenetics, what types of 3D genome structures there are, how they are formed, and how the technologies have developed to explore them. We will also discuss the pathological implications of 3D genome folding. Finally, we will discuss how to leverage 3D genome folding and engineering for future studies.

Comparative Study on the Salivary Gland between Two Species (Achatina fulica and Incilaria fruhstorferi) of the Snails in Stylommatophora ( Mollusca, Gastropoda ) (병안목 달팽이류 두 종간 (Achatina fulica and Incilaria fruhstorferi)의 타액선에 관한 비교 연구)

  • Histochemical experiment was carry out respectively to confirm the properties of the salis (Achatina fulica and Incilaria fruhstorferi). SDS-PAGE was carried out to compare and invertigate the distribution aspects of protein patterns between the two species. Five types(A, B, F, H and I)of gland cells with four neutral mucopolysaccharide cells and one acid mucopolysaccharide cells and one acid mucopolysaccharide cell were observed in acinous of Achatina fulica, while six types were observed in acinous of Incilaria fruhstorferi: ond acid mucopolysaccharide cell(type-A) and four neutral mucopolysaccharide cells(type-B, C, D and F) and one cell that acid mucopolysaccharide is only mimbrane that surrounded granule(type-E). The results are follows:The thpe-A fland cell is commonly observed between the two species. The type-A gland cell in Achatina fulica possesses a nucleus with a developed heterdchromatin, and the cytoplasm was filled with round granules. The granules were surrounded with an uncertain boundary mimbrane and confirmed with neutral mucopolysaccharides, but is confirmed acid mucopolysaccharide in Incilaria fruhstorferi.The type-B gland cell is obwerved in the two species, too. The type-B gland cell in Achatina fulica was round shaped, and included an evenly alrge nucleus. The uncleoplasm included granules that were confirmed in the neutral mucopolysaccharides of the two species. The type-C and D gland cells exist only in Incilaria fruhstorferi, nucleoplasm was well developed heterochromatins. The type-E gland cell appears in the acinous surrounded the salivary gland of Incilaria fruhstorferi. Thdse granules appear irregular irregular shape and size and the cytoplasm is formed in alveolar. The type-F gland cells are commonly observed in the salivary glands of the two species. They are similar with the type-B gland cell, but the granular shape is comparatively small and irregular, and possess the neutral mucos granules. The type-H gland cells are mainly seen in only Achatina, and in nucleus is a well developed heterochromatin. The cytoplasm is filled with round small granules with acid mucopolysaccharide for alcianophilia observed. The type-I cell was small cell with an irregular shape and only observed in the gland cells of Achatina fulica. The heterochromatins were developed in the nucleus and the granules are not observed in cytoplasm.Secretory ducts of saliva are composed of the interlobular duct and interlobar secretory duct. In Achatina fulica the interlobular duct consists of a simple cuboidal epithelium, while the endothelium of intralobar secretory duct of Incilaria fruhstorferi consists of a simple squamous epithelium and in the cytoplasm is filled with granules(type-G secretory cell). A SDS-PAGE was carried out to confirm that the protein band pattern consist of salivary gland. In conclusions, five more bands in Achatina fulica and three bands in Incilaria fruhstorferi were confirmed in MW<29 kDa. one main band coincides comparatively with both and is between 29-45 kDa. There are four main bands in Achatina fulica and two main bands in Incilaria fruhstorferi between 45-66.5 kDa respectively. The bands in Achatina fulica seem more complex than in incilaria fruhstorferi.
