Effects of Auxins and Cytokinins on Organogenesis of Soybean Glycine max L.

  • To select the section with shoot formation ability, the calli and shoot formation from three sections (first leaf including cotyledonary node, hypocotyl and cotyledon explants) of 5-days-seedlings of soybean were induced on MS medium supplemented with 1.0 mg/L BAP, 3% sucrose, and 0.3% gelrite for one month. The first leaf section exhibited the highest shoot formation rate (51%), followed the hypocotyl section (10%) and the cotyledon section (0%). The shoot formation rates and shoot number of the four excised sections (whole first leaf, a half of the first leaf, a third of the first leaf and only node) of the first leaf were also investigated on the same medium. A half of the first leaf explant and the third of the first leaf explant had higher shoot formation rates (76-80%) and numbers (3-4 / explants) than those in other two explants. Effects of six cytokinins (kinetin, zeatin, BAP, 2iP, PBA, and TDZ) on shoot formation were determined, using the half of the first leaf explants. Zeatin (1.0 mg/L) exhibited the highest in shoot formation rate (94%) and numbers (8 / explant). In addition, the combined effects of three cytokinins (zeatin, BAP, and TDZ; 0.5, 1.0, 2.0 mg/L, respectively) and an auxin (IAA; 0.0, 0.5, 1.0, 2.0 mg/L) were determined. The combination (1:1, v/v) of zeatin (1.0 mg/L) and IAA (1.0 mg/L) exhibited the highest in shoot formation rate (96%) and numbers (16 / explant), twice more than zeatin (1.0 mg/L) alone. The shoot cuttings were transferred and cultivated on the rooting media supplemented with only auxin, IBA at various concentrations. The highest root formation (8 / shoot) was achieved on the medium supplemented with 1.5 mg/L. After 4 weeks of cultivation, the plantlets with an extensive root system were transplanted in pots with a soil mixture of vermiculite and fine sand. Transferred to field, about 75% of the plantlets survived.

  • To elucidate the relationship between in vitro flowering and the structure of cytokinins in ginseng (Panax ginseng C.A. Meyer), zygotic embryos, seedlings, and cotyledonary nodes were cultured on MS medium supplemented with 5 $\mu$M of various cytokinins (BA, kinetin, 2-iP, and zeatin) with or without GA$_3$ (5 $\mu$M). The frequency of in vitro flowering was the highest when explants were cultured on the medium containing BA regardless of the kinds of explants, followed by kinetin, 2-iP, and zeatin. Flowering frequency of cotyledonary node explants was significantly increased by the combined treatment of cytokinin and GA$_3$. Flowering frequency was highly correlated with the logP of cytokinins, indicating that the lipophilicity of each cytokinin may involved in the in vitro flowering of ginseng.

Effect of Genotypes and Cytokinins on Shoot Multiplication of Cut Roses by Nodal Culture (절화 장미의 마디 배양시 품종과 사이토키닌류가 신초의 대량증식에 미치는 영향)

  • This experiment was carried out to determine the effects of genotyes (cultivars) and kinds of cytokinins and concentrations on shoot multiplication of roses (Rosa hybrida) by nodal culture. The results from this study were summarized as follows. Shoot multiplication was significantly affected by genotypes. Most shoots were produced from 'Carl Red 'rather than two other cultivars, 'Golden Emblem' and 'Tineke'. Among 3 cytokinins, BA was more significantly effective for multiplication than two other cytokinins, 2iP and kinetin. However subsequent shoot growth after multiplication was more enhanced by 2iP and kinetin rather than BA. An optimal concentration of cytokinin for shoot multiplication was 1 mg/L in this study but BA at 1-5 mg/L was considered optimal for multiplication ratio itself. Subsequent shoot growth was significantly inhibited by increases in cytokinin concentrations, regardless of kinds of cytokinin.

  • Cytokinin is one of major growth regulators in plants. In this study, the gene isopentenyl transferase (jpt) which encodes a key enzyme involved in the biosynthesis of the growth regulator cytokinin isolated from Agrobacterium tumefaciens was introduced ito tobacco plant via Agrobacterium-mediated transformation. The jpt gene was modulated using the proteinase inhibitor II (PI-IIK) promotor. In general, this promoterlipt gene fusion resulted in overproduction of cytokinins throughout the transgenic plants. The overproduction of cytokinin caused dramatic changes in morphology of the plant, including stem thickness and reduced root development. The studies reported in this paper were initiated to examine the consequences of overproduction of cytokinin in plant.

  • The activities of auxins and cytokinins have been examined in the growth of callus tissue derived from Phaseolus vulgaris L. cv. Damyang. The synthetic auxin, picloram was the most effective in promoting callus growth and the range of effective concentrations (0.1$\mu{M}$ to 32$\mu{M}$) was broad. 2, 4-D also enhanced callus growth at the optimal concentration of 3.2$\mu{M}$. NAA promoted callus growth at relatively higher concentrations than other auxins tested. IAA was less effective in supporting callus growth. Cytokinin bearing saturated side chain ($N^6$-isopentyladenine) was approximately 30 times more active than the corresponding unsaturated compound, $N^6$-($\D^2$-isopentenyl) adenine. The abilities of cytokinin-autonomous growth were also examined. Callus tissues previously grown on concentrations lower and/or higher than optimal concentrations of cytokinins were better habituated in the subsequent passage. It was suggested that the development of cytokinin autonomy may be related to dosage-concentrations of cytokinin in the previous passage.

  • The effects of various plant growth regulators, both auxins and cytokinins, on cell growth and berberine production were investigated in cell suspension cultures of Thafictrum rugosum. Indole-%-acetic acid (IAA) was found to be the best for berberine production among five examined plant growth regulators and the optimum concentration of IAA was 1 $\mu \textrm M$. The enhancement compared to control 2, 4-dichlorophenoxyacetic acid (2, 4-D) was more than 60%. Simultaneous addition of cytokinins such as kinetin and 6-benzylamiroyurine (BA) was inhibitory.

Effects of Auxins end Cytokinins on Callus Induction from Leaf Blade, Petiole, and Stem Segments of in Vitro-grown 'Sheridan' Grape Shoots

  • To establish an the mass production system of grape anthocyanin pigments through callus and cell suspension culture, the effects of various combinations of auxins and cytokinins on friable callus production were studied. for friable callus production, 2,4-D was superior to other regulators. IAA at 2 mg/L induced callus from stem and petiole while NAA resulted in rooting. Callus induction rate increased with the 2,4-D level, and stem segments were superior to leaf blade or petiole, showing nearly 100% with 1 and 2 mg/L 2,4-D from petiole and stem. Combined treatments of 2,4-D + kinetin and NAA + BA also yielded friable callus from stem segments. In treatments with 1 mg/L 2,4-D + 1 mg/L kinetin and 1 mg/L NAA + 1 mg/L BA, callus induction rate was nearly 100%. The combination effect of 2,4-D and BA on anthocyanin production was not significant.

Effect of Cytokinins on Nodal Cultures of Citrus Species (감귤류의 마디배양에서 싸이토키닌류가 기내 대량증식에 미치는 영향)

  • This study was conducted to determine the effects of 3 cytokinins (BA,2iP and kinetin) and their concentrations (0, 0.1, 0.5, 1.0, and 2.0 mg/L) on multiple shoot production of Citrus spp. 'Sambokam' and 'Byungkyool' by nodal culture. Nodal explants were obtained from in vitro germinated seedlings of both cultivars. 'Sambokam' produced more multiple shoots than did 'Byungkyool' by nodal culture. Among the 3 cytokinins tested in this study BA supplemented in semi-solid MS basal medium was the most effective stimulator for multiple shoot production, and an optimal concentration was determined to be 1.0 mg/L. Shoot elongation and root formation were inhibited by increasing cytokinin concentration, regardless of cytokinin types. BA at 1.0 mg/L produced the most multiple shoots and the highest number of leaves in 'Sambokam', whereas any cytokinin and concentration studied in this experiment did not affect any scored variables such as shoot and leaf numbers, etc. in 'Byungkyool'.

Effects of Cytokinins on Secondary Embryogenesis and Plant Regeneration from Somatic Embryos of Aralia cordata Thunb. (땅두릅의 체세포배로부터 2차배 발생과 식물체 재생에 미치는 싸이토카이닌의 영향)

  • Embryogenic suspension cultures were initiated using embryogenic callus from immature inflorescence explants (Aralia cordata Thunb.) cultured on solid MS medium containing 1 mg/L 2,4-D for 8 weeks and then the embryogenic callus was proliferated in liquid MS medium containing 1 mg/L 2,4-D. After sieving the suspensions (pore size 270$\mu$m), embryogenic cells were cultured in liquid MS medium with cytokinins (kinetin, BA, zeatin) for two weeks. When the embryogenic cells were transferred to liquid MS basal medium, primary somatic embryos were developed after 5 weeks of culture. Secondary embryos were developed directly from the primary torpedo and cotyledonary embryos cultured in solid MS basal medium. Frequency of secondary embryogenesis was higher on medium containing 2 mg/L kinetin than the other cytokinins. Plant regeneration was highly recorded by placing secondary cotyledonary embryos induced from primary cotyledonary embryos in MS medium containing 2 mg/L kinetin or 2 mg/L zeatin (25.4% and 28.6%, respectively). The plant regeneration from secordary embryos was prohibited by tertiary embryogenesis.

Effect of Cytokinins on the Number of Cotyledons of Sometic Embryos from Immature Zygotic Embryo Culture of Heracleum moellendorffii Hance (어수리(Heracleum moellendorffii Hance.)의 미숙배로부터 형성된 체세포배의 자엽수에 미치는 Cytokinin의 영향)

  • In order to investigate the effect of cytokinins on the cotyledonary variability of somatic embryos in Heracleum moellendorffii Hance., somatic embryos were induced from the immature zygotic embryo on MS medium containing 1.0mg/l 2, 4-D, and then transferred to 2,4-D-free and cytokinin-added medium for somatic embryogenesis after 4weeks culture. Polycotyledonary embryos were formed most abundantly(84.9%) on the 0.2mg/l BA medium as compared with the 0.2mg/l 2ip(42.6%) and kinetin(32.9%) media, and it was proportional to BA concentrations(0.01~0.1mg/l). The rate of polycotyledonary embryo formation increased proportional to the period of BA treatment and also increased more at the globular stage than at the heart, torpedo-shaped and cotyledonary stages.
