Development of Bulgogi Sauce Added with Concentrated Curcuma longa L. (울금 농축액을 첨가한 불고기 소스의 개발)

  • This study was conducted the physicochemical and sensory characteristics of Bulgogi sauce added with concentrated Curcuma longa L.. Increasing the amount of concentrated Curcuma longa L. in the Bulgogi sauce tended to increase the pH values, lightness(L), redness(a) and yellowness(b) values. It also tended to increase DPPH radical activity, but total polyphenol contents of Bulgogi sauce added with concentrated Curcuma longa L. were not significantly different among all the samples. In sensory evaluation, Bulgogi sauce added with 10% concentrated Curcuma longa L. had an excellent score in sensory taste, but Bulgogi sauce added with 20% concentrated Curcuma longa L. had an excellent score in overall preference. Seasoned roasted beef addition with 20% concentrated Curcuma longa L. had an excellent score in sensory preference. Therefore, this results suggest that 20% concentrated Curcuma longa L. should be applied to Bulgogi sauce for high sensory preference.

Curcuma Longa L. Extract Controls Cancer Cell (Sarcoma 180) Growth (강황(薑黃) 추출물의 암세포 성장 억제 효과)

  • Objectives : The anticancer response of three different types of water extracts of Zingiberaceae Curcuma longa L. tested for sarcoma 180. Only few studies carried out to investigate the effects of other contents of Curcuma longa L. in anticancer activities, therefore, in this study we have investigated the effects of other component then curcumin in Curcuma longa L. for anticancer a activities. Methods : Three different types of water extracts of Curcuma longa L. were prepared as follows. The sarcoma cells (S180) were maintained in Dulbecco's modified Eagle's medium (DMEM) and were seeded on 24-well cell culture cluster flat bottom with lid tissue culture treated non-pyrogenic polystyrene. The growth of sarcoma 180 was monitored for 1, 2 and 5 days. The sarcoma cells were pictured using inverted microscope and cell density was counted using hemocytometry. Results : After 5 days in the culture medium the results showed high growth of sarcoma 180 for control condition and the surface of CCP plates were fully covered with the cells. In case of medium in which the 10% of filtered water extract of Curcuma longa L. was added a very limited growth of sarcoma 180 was observed. The results were showed only small difference in cell density for two different concentrations of unfiltered water extracts of Curcuma longa L. whereasin case of filtered water extracts the control of sarcoma growth shows better result. Conclusion : The filtered water extracts showed the best result relatively to the unfiltered water extracts for two different concentrations. This indicates that the water extracts of Curcuma longa L. can have anticancer activities possibly without curcumin.

Enhancement of Antioxidant Activities of Curcuma longa Leaves by Ultra High Pressure Extraction (초고압 공정을 이용한 강황 잎 추출물의 항산화 활성 증진)

  • This study was designed to evaluate antioxidant activity of Curcuma longa L. leaves treated by ultra high pressure extraction. Curcuma longa L. leaves was subjected to 5,000 bar for 5 and 15 min at $25^{\circ}C$ The highest phenolics and flavonoids content was observed in the treatment of high pressure extraction for 15 min ($308.28{\mu}g/mg$, $124.33{\mu}g/mg$). The DPPH scavenging activity was 82.34% at $1.0mg/m{\ell}$ of Curcuma longa L. leaves treated by ultra high pressure process for 15 min. The highest SOD-like acitivity of Curcuma longa L. leaves ($1.0mg/m{\ell}$) was observed at ultra high pressure extraction for 15 min (67.54%). The high pressure extraction significantly increased the contents of phenolics and flavonoids and also enhanced the antioxidant activity. These results provide useful information for enhancing biological properties of Curcuma longa L. leaves.

Effects of Curcuma longa L.on Human Stomach Cancer Cells (울금(鬱金)이 위암세포(胃癌細胞)에 미치는 영향(影響))

  • Objective: We are aimed to identify anti-tumor effects of Curcuma longa L. on the stomach cancer cells through molecular biologic methods. Material & Methods: We used AGS as human stomach cancer cells obtained from American Type Culture Collection. The boiled extract of Curcuma longa L. $5{\mu}l$ (Sample I), $10{\mu}l$ (Sample II) was treated to cultural media(ml) for 0, 6, 12, 24, 48 hours. We measured the killing effect on stomach cancer cells through Trypan blue exclusion test and the suppressive effect on viability of stomach cancer cells via MTT assay. For identification of its anticancer mechanism, the revelation of Bcl-2, Bcl-XL, and Bax which are genes related to apoptosis using the quantitative RT-PCR, change of mitochondria membrane permeability and membrane potential via flow cytometry, the cycle of cell mitosis, caspase cleavage and annexin V staining were examined. Results: 1. showed significant killing effect on stomach cancer cell than the control group with a time(6 hours later) and density dependent manner, which was statistical significance. 2. Extract of Curcuma longa L. showed suppressive effect on viability of stomach cancer cells that each test groups had more suppressive effects on viability of stomach cancer cells than the control group with a time(6 hours later), which was statistical significance.(p<0.05) 3. In the test about the revelation of genes related to apoptosis, the revelation of Bcl-2 and Bcl-XL decreased with a density manner which was statistical significance. but the revelation of Bax was not changed with statistical significance. 4. Extract of Curcuma longa L. caused apoptosis by decreasing the absorbance of mitochondria with statistical significance, and also induced apoptosis by decreasing the membrane potential of mitochondria. 5. Extract of Curcuma longa L. destructed the cell cycle of cell mitosos. 6. Cell apoptosis was induced by extract of Curcuma longa L. certificated by method of caspase cleavage and annexin V staining. Conclusion: This experiment showed that Curcuma longa L. has anti-tumor effect with statistical significance. This is in vitro experiment and basic experiment on Curcuma longa L.. We hope more progressive research on Curcuma longa L. will go on and its anti-tumor effects will be more practically identified.

Effects of Curcuma longa L. on Some Kinds of Cancer Cells (강황이 수종의 암세포에 미치는 영향)

  • Objectives : The Purpose of this study was to identify anti-tumor effects of Curcuma longa L. on some kinds of cancer cells through molecular biologic methods. Materials & Methods : We used 4 kinds of cancer cell lines such as glioma cells(A172), cervical cancer cells(HeLa), Prostate cancer cells(PC3), lung cancer cells(A549). We injected the boiled extract of Curcuma longa L. $5{\mu}g,\;10{\mu}g$ to culture media(ml) for 24 hours. We measured the cytotoxic effect on 4 kinds of cancer cells through trypan blue exclusion test and the suppressive effect on viability of 4 kinds of cancer cells via MTT assay. We measured the change of mitochondria membrane potential via flow cytometry. The quantitative RT-PCR was used to examine the effect on the revelation of Bcl-2 and Bax which genes are related to apoptosis. We examined the effect on the revelation of Bcl-2 protein and Bax protein by western blot analysis. Results: 1. Extract of Curcuma longa L. showed significant cytotoxic effect on A172, HeLa, PC3 compared to the control group with density dependent manner. 2. Extract of Curcuma longs L. showed significant suppressive effect on viability of A172, HeLa, PC3 compared to the control group with density dependent manner. 3. Curcuma longs L. induced apoptosis by decreasing the membrane potential of mitochondria in A172, HeLa, PC3. 4. In the test about the revelation of genes related to apoptosis, the revelation of Bcl-2 decreased and the revelation of Bax increased in A172. HeLa, PC3 treated with Curcuma longa L. with density dependent manner. 5. In the test about the revelation of protein related to apoptosis, the protein levels of Bcl-2 decreased and the protein levels of Bax increased in A172, HeLa, PC3 treated with Curcuma longa L. Conclusions: This experiment shewed that Curcuma longs L. has anti-tumor effect on glioma, cervical, Prostate cancer cells except on lung cancer. We hope that anti-tumor effects of Curcuma longa L. will be more Practically identified.

Dyeability and Antimicrobial Activity of Silks Dyed Singly with Sophora japonica L., Curcuma longa L., Combination-Dyed with Polygonum tinctorium L. and Sophora japonica L., and with Polygonum tinctorium L. and Curcuma longa L. (회화 및 울금에 의한 단독 염색 견직물 그리고 쪽과 회화 및 쪽과 울금에 의한 복합 염색 견직물의 염색성 및 항균성)

  • We examined the dyeability and antimicrobial activity of silks dyed singly with Sophora japonica L., Curcuma longa L., combination-dyed with Polygonum tinctorium L. and Sophora japonica L., and with Polygonum tinctorium L. and Curcuma longa L.. The color of silk dyed singly with Sophora japonica L., Curcuma longa L. was Yellow with Hue of 5.0Y and 0.6Y, respectively. Two types of combination-dyed silk were Blue Green with 2.6BG and 1.5BG, respectively. In the case of dry Cleaning fastness and rub fastness, all four types of dyed silk was good, achieving grade 4~5. The antimicrobial activity of Staphylococcus aureus of silk dyed with Sophora japonica L. extract showed excellent antibacterial activity of 99.5% and the other three types of dyed silk of 99.9%. In addition, in antibacterial activity against Klebsiella pneumonia, silk dyed with Sophora japonica L. extract showed 99.6%, and the other three types of dyed silk exhibited excellent antibacterial properties of 99.9%.

Effects of Curcuma longa L. Extracts on Natural Killer Cells and T Cells (울금 주정 추출물이 자연살해세포와 T 면역세포에 미치는 영향)

  • The present study investigated the immunomodulatory effects of Curcuma longa L. ethanol extracts on natural killer (NK) cells and T cells. We treated Curcuma longa L. ethanol extracts at concentrations of 20, 50, 100, and $150{\mu}g/mL$ to murine NK cells co-incubated with YAC-1 cells. Curcuma longa L. ethanol extracts resulted in increased NK cell activity compared to the control group at all concentrations. In the groups treated with Curcuma longa L. ethanol extracts, CD69 and IFN-${\gamma}$ expression levels significantly increased compared to the control group at 100 and $150{\mu}g/mL$. In addition, Curcuma longa L. ethanol extracts induced significant elevation of CD8+ T cell numbers in a dose-dependent manner. However, Curcuma longa L. ethanol extracts also led to reduction of CD4+ T cell and MHCII numbers. The findings of this study suggest that Curcuma longa L. ethanol extracts could enhance the immune response through activation of NK and cytotoxic T cells due to a proliferative shift of antigen presentation from MHCII to MHCI, presumably.

Quality Characteristics of Fish Paste Containing Curcuma longa L. Powder (울금 분말을 첨가한 어묵의 품질특성)

  • This study was conducted to investigate rheological and sensory characteristics of fish paste prepared with Curcuma longa L. powder (0, 1, 3, 5, 7%). The moisture content of the fish paste ranged from 71.35 to 72.97%. Increasing the amount of Curcuma longa L. powder in the fish paste tended to decrease the pH value and the folding test good score in all samples. Increasing the amount of Curcuma longa L. powder in the fish paste also tended to decrease the lightness (L) in the hunter color value, but to increase the redness (a) and yellowness (b). For the textural characteristics, the addition of Curcuma longa L. powder increased the strength and decreased the springiness. In a sensory evaluation, the addition of 3% Curcuma longa L. powder had the best score in taste and overall preference. Therefore, this results suggest that adding 3% Curcuma longa L. powder would be feasible for making Curcuma longa L. fish paste.

The Quality Characteristics of Curcuma longa L powder Sulgitteok (강황분말 설기떡의 품질 특성)

  • This study aims to provide preliminary data to popularize Sulgitteok with optimum mix proportion of Curcuma longa L. powder by conducting moisture content, color values, texture and quantitative descriptive evaluations on Sulgitteok with 0 to 2.4% of Curcuma longa L. powder. The chemical composition of Curcuma longa L. powder was $12.6{\pm}0.21%$ of moisture, $1.8{\pm}0.12%$ of protein, $1.0{\pm}0.00%$ of fat, $1.3{\pm}0.01%$ of ash, and that of rice flour was $11.7{\pm}0.17%$ of moisture, $7.6{\pm}0.32%$ of protein, $2.1{\pm}0.01%$ of fat, $1.6{\pm}0.01%$ of ash. As the content of Curcuma longa L. powder increased, L-values significantly decreased while a-value and b-value significantly increased. The hardness of the texture characteristics significantly increased with increasing the amount of Curcuma longa L. powder(p<0.05). As for gumminess, cohesiveness, and chewiness, there was no significant difference(p<0.05) depending on the amount of Curcuma longa L. powder. The quantitative descriptive analysis of the color, flavor, moistness and chewiness of Sulgitteok showed that one with 0.8% of Curcuma longa L. powder was evaluated the highest.

Immunomodulatory Effects of Curcuma longa L. Extract in LP-BM5 Murine Leukemia Viruses-induced Murine Acquired Immune Deficiency Syndrome (면역결핍 동물모델에서 울금 주정 추출물의 면역조절 효과)

  • The immune system protects the body against harmful substances and infectious agents. Normally, the body can maintain a state of immune homeostasis. However, failure of immune homeostasis results in severe diseases when the immune system is defective. We investigated the immunomodulatory effect of Curcuma longa L. extract in LP-BM5 MuLV (murine leukemia viruses)-induced murine AIDS (acquired immune deficiency syndrome). Mice were divided into six groups: normal control, infected control (LP-BM5 MuLV infection), positive control (LP-BM5 MuLV infection+dietary supplement of red ginseng 200 mg/kg), CL50 (LP-BM5 MuLV infection+dietary supplement of Curcuma longa L. 20% alcohol extract 50 mg/kg), CL200 (LP-BM5 MuLV infection+dietary supplement of Curcuma longa L. 20% alcohol extract 200 mg/kg), and CL500 (LP-BM5 MuLV infection+dietary supplement of Curcuma longa L. 20% alcohol extract 500 mg/kg). We found that dietary supplementation with Curcuma longa L. 20% alcohol extract inhibited elevation of spleen, lymph node, and liver weights as well as reduction of T- and B-cell proliferation and natural killer cell activity induced by LP-BM5 MuLV infection. Moreover, Curcuma longa L. 20% alcohol extract inhibited Th1 (IL-2, IFN-${\gamma}$)/Th2 (IL-4, IL-10) cytokine imbalance and pro-inflammatory cytokine production. In conclusion, these data suggest that Curcuma longa L. has immunomodulatory effects in LP-BM5 MuLV-induced murine AIDS.
