Effects of Culturing Parameters on the Production of Microbial Biosurfactant from Candida bombiocola (Candida bombiocola로 부터 미생물 계면활성제 생산시 관여 인자에 관한 연구)

  • Effects of nitrogen souses and C/N ratio were investigated on the production of extracellular microbial surfaclant, sophorolipid, from C. bombiocola. Organic nitrogen sources, such as urea, peptone and yeast extract was found to be more effective for sophorolipid production, than inorganic nitrogen sources. Depending on the nitrogen sources, sophorolipid production pattern varied by increasing C/N ratio. Increased production of sophrolipid could be obtained up to 90g/L by feeding carbon source again 2 days after cultivation.

Induced Change in DNA Methylation of Fusarium oxysporum f. sp. niveum due to Successive Transfer

  • Changes in pathogenicity of old and successively-cultured isolates of Fusarium oxysporum f. sp. niveum have been observed and the concept that such cultures will become attenuated is generally accepted. However, the genetic basis for this phenomenon has not been studied. In an effort to identify a DNA marker closely linked to variations, DNA methylation was investigated both before and after the successive transfers of F. o. f. sp. niveum isolates on artificial media. A sector of mycelium in F. o. f. sp. niveum race 2 isolate (TXXID) which showed variation in pigmentation and colonial morphology occurred after 18 successive weekly transfers on potato dextrose agar (PDA). The sector characteristics were stable and did not change after more successive transfers. It was shown that DNA methylation preexists in ribosomal RNA gene (rDNA) of F. o. f. sp. niveum and that additional changes in DNA methylation occurred during successive culturing.

  • The condition necessary for maximal production of pectin trans-eliminase by culturing Aspergillus D-11 on a moistened wheat bran medium was studied. The maximum production of pectin trans-eliminase was obtained after cultivation for two, days at $29^{\circ}C.$ One tenth milligram of crude pectin trans-eliminase of Aspergillus D-11 having 0.15 units of activity could clarify 15-20 ml of apple juice within 3 hours at $40^{\circ}C.$ Optimal pH and temperature for clarifying apple juice by the crude-enzyme were found to be pH 4.5~5.0 and $45^{\circ}C$ respectively.

  • An experimental study was conducted to Indentify the dlrpct denitrification of ammonium nitrogen In culture water by ozone. During the experimnet period, pH was 7.8-8.8. pH was grdually lower after ammonium nitrogen was reacted with ozone under Br . In addition, it can be tmown that the culturing water was improved greatly form the inverstigation of T-N by biofilm and ozonation. As the results of a variation of recirculation rate, denitrification of ammonium nitrogen was in increased in proportion to the recirculation rate. But Nitrification of microorganism was opposite to the recirculation rate. With the increasing injected ozone in ozonation tank uner 21 clrculation/day(6.71 /min), dinitrification of ammonium nitrogen was Increased lineraly in propotion to the Increasing of injected ozone concentration.

  • Human cytochrome P450 (CYP or P450) 3A4 (CYP3A4) is the most abundant among P450s in human liver. We previously reported that the expression of CYP3A4 in membranes prepared from E. coli coexpressed the bicistronic construct of CYP3A4 and NADPH-P450 reductase with cytochrome b$_{*}$ (b5) was showed 20-60% higher than that in membranes from E. coli expressed only the bicistronic construct with culturing longer times (48-72h).(omitted)

  • The heavy chain monoclonal antibody (HC MAb) was produced in suspension cultures of genetically modified Nicotiana tabacum. The HC MAb secreted to the medium was unstable due to unfavorable interactions in the plant cell medium. The addition of gelatin (5g/l) stabilized the extracellular HC MAb and increased its production 10-fold. A kinetic model was developed describing the interaction between the secretedprotein and the stabilizer. The model accounted for the inactivation of the protein by simple aggregation and general instability. It was assumed that the secreted protein and the stabilizer form a stable complex. Culturing the cells semicontinuously could further increase the productivity of HC MAb.

  • Telomere erosion is the earliest chromatin modification in cells entering the apoptotic regime. The purpose of this investigation was to demonstrate that loss of telomeric DNA was involved in the induction of mitotic catastrophe and cell death of Chinese hamster Don cells that were grown continuously without subculturing for several days. Don, which is a diploid male Chinese hamster-derived cell line, was grown without sub-culturing for one to four days at 37℃ and then harvested for cytological preparations. (omitted)

  • The main objective of modern reproductive technologies in pig reproduction is to increase reproductive efficiency and rates of genetic improvement. They also offer potential for greatly extending the multiplication and transport of genetic materials and the conservation of unique genetic resources in reasonably available forms for possible future use. The development and refinement of these technologies is concentrating on gamete and embryo collection, sorting and preservation, in vitro production of embryos, culturing, manipulation of embryos (splitting, nuclear transfer, production of chimeras, establishment embryo stem cells, and gene transfer) and embryo transfer. Also, the development of these novel technologies is facilitated by modern equipment for ultrasonography, microscopy, cryopreservation, endoscopy, and flow cytometry, microinjectiors, micromanipulators and centrifugation. The real impact on herd productivity will come from combining new reproductive techniques with powerful DNA technologies. The new reproductive techniques will allow a rapid turnover of generations, whereas the DNA technology can provide selection, which does not need phenotypic information when the selection decisions are made.

Enumeration of Vibrio vulnificus in Natural Samples by Colony Blot Hybridization

  • Colony blot hybridization using a VVHP DNA probe derived from the sequence of the hemolysin gene, vvhA, was specific in identifying all V. vulnificus strains, thereby, eliminating the need for any additional phenotypic identification. The colony blot hybridization procedure revealed a sensitivity and broad applicability sufficient for the direct enumeration of V. vulnificus in various natural samples, without the use of enrichment or culturing on selective medis. V. vulnificus was detected in all natural samples collected during August and May at concentrations ranging from $2.1{\times}10^1\;to\;4.0{\times}10^3$ organisms per ml. However, during November and February, when the mean temperatures of the seawater were $12^{\circ}C$ and $5^{\circ}C$, respectively, V. vulnificus was not detected in any natural samples.

  • Intracytoplasmic photosynthetic membranes of Rhodocyclus gelatinosus and Rhodocyclus tenuis are known to be of weakly developed type of in the Rhodospirillaceae. We compared the membrane invagination of Rhodocyclus gelatinosus KS-117 (isolated in lur laboratory) with that of Rhodocyclus tenuid (ATCC 25093) after culturing under several different light intensities. We observed the significant membrane invagination in Rhodocyclus gelatinosus, in particular under 3,000 Lux, while none was observed in Rhodocyclus tenuis.
