Extraction and Purification of Antitumor Protein-bound Polysaccharides from Mycelia of Lentinus edodes (표고버섯 균사체로부터 항암 단백다당체의 추출 및 정제)

  • Korean Lentinus edodes SR-1 was cultured to multiply the mycelia in the complete broth medium (C/N=13.1) for mushroom, and protein-bound polysaccharides were extracted from the cultured broth containing mycelia (The whole cultured broth was used to increase the yields: 80% of protein-bound polysaccharides were existed at the cell wall of mycelia and 20% of those were secreted extracellularily in this culture). Protein-bound polysaccharides in the cultured broth containing mycelia were extracted by using three different methods: 1) Extraction with hot water, 2) Disintegration of cell wall by glass bead mill treatment before extraction with hot water, and 3) Cellulase treatment before extraction with hot water. The highest yield was obtained (930 mg polysaccharides/100 mL culture broth) when protein-bound polysaccharides were extracted with 2) method. The extracted crude protein-bound polysaccharides were purified using protease, DEAE-cellulose and Sephadex G-100. The growth inhibition activity for $P_{388}$, mouse leukemic cell, increased (53.7, 62.2, 93.7% and 97.4%) as the purification level increased. Protein-bound polysaccharides contained 46.1% of polysaccharides, 7.3% of protein, and trace amounts of minerals. Polysaccharides contained glucose, galactose, xylose and mannose. The content of proline and glycine were high, however, methionine and leucine were not found. The major minerals were Na, K, Zn, and Ca.

Effect of Ganoderma lucidum Mycelial Fractions on the Functional Recovery of Primary Cultured Hepatocytes. (영지 균사체 분획이 일차배양 간세포 기능회복에 미치는 영향)

  • The cultured mycelia of Ganoderma lucidum were extracted and the extract was separated into six fractions by organic solvent fractionation. The antihepatotoxic activity of all the fractions was evaluated by measuring activities of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT). Among the fractions tested, the high-polarity fractions such as aqueous and n-butanol fractions significantly reduced activities of GPT and GOT in CCl4- and galactosamine-intoxicated rat primary hepatocytes. When intracellular synthetic activities were measured by pulsing the rate primary cultured hepatocytes with [3H]-uridine and [3H]-leucine, activities of DNA, RNA and protein. When direct toxicities of the fractions were measured against human hepatoma(SK-Hep-1), the non-polarity fractions such as n-butanol and ethyl acetate fractions showed potent direct cytotoxicities even at the concentration of 1 $\mu\textrm{g}$/ml. These data showed that Ganoderma lucidum has hepatoprotective and hepatotoxic recovery principles in its mycelia.

Anti-Diabetic Studies of Mass Cultured Mycelia from Ganoderma applanatum in db/db Mice and Human (잔나비걸상버섯(Ganoderma applanatum) 균사체의 db/db Mice 및 인체에서의 항당뇨 효능연구)

  • Anti-diabetic activities of cultured mycelia from Ganoderma applanatum are being evaluated in this study. The OGTT and 4-weeks of repeated oral efficacy tests are conducted in mice at the doses of 0 (vehicle treatment), 25, 75 and 225 mg/kg/day, respectively. In human study, the test article was administered orally every day for 8-week at a dose of 1,500 mg/kg, tid and placebo group. The blood glucose levels (BGL) at 0.5 hour after treatment are significant decreased in all treatment groups of OGTT test. In the 4-week test, BGL of 75 and 225 mg/kg/day group is continuously decreased during all treatment periods and the BGL of 25 mg/kg/day group show decreasing trends at the final week, the pancreas weight of all treatment groups are being increased, and the Langerhans-islet numbers were increased at all treatment groups with a dose-response manner. There are no test article-related abnormal signs and the fasted blood glucose (FBG), postprandial blood glucose (PPG) and HbA1c are decreased significantly after 8-week treatments. These results that the cultured mycelia from Ganoderma applanatum could decrease BGL by protecting the degeneration of Langerhans islets.

  • Seven compounds were isolated from stationary cultured mycelia of Inonotus obliquus, of which several of them showed certain antitumor activities. Seven compounds were identified as lanosterol(1), inotodiol(2), trametenolic acid(3), 3β,22,25-trihydroxy-lanosta-8-ene(6), 3β,22-dihydroxy-lanosta-8,24-diene(A), 3β-hydroxy- lanosta-8,24-dien-21-al(B) and methyl trametenolate(C), respectively. The precursor compound of sclerotium obtained from shaking-cultured and stationary-cultured mycelia of Inonotus obliquus is lanosterol. Ergosterol and ergosterol peroxide were obtained by shaking-culture, and the substituted compounds of C-21 and C-22 of lanosterol were obtained by stationary culture.

Mycorrhizal Formations and Seedling Growth of Pinus desiflora by in vitro Synthesis with the Inoculation of Ectomycorrhizal Fungi

  • The mycelia were directly isolated from eight species of fungal basidiocarps, confirmed to the ectomycorrhiza in the roots from the fields(forestry); Suillus bovinus, Paxillus involutus, Lactarius hysginus, Russula fragilis, Lepista nuda, Lyophyllum shimeji, Tricholoma matsutake, and Russula integra. The mycelia were pure-cultured with several transferring in various agars, and inoculated to the roots of pine(Pinus densiflora) seedling by in vitro method. After ten months growth under artificially aseptic conditions, all pine seedlings inoculated were stimulated at the growth-height, whereas those not inoculated were nearly dead. Also, the ramifications of ectomycorrhizal pine roots formed in the synthetic in vitro systems and were various according to the different mycelia. Synthesis of ectomycorrhiza were clearly confirmed in ten months growth, but not distinguished at this moment. It was clearly proved that the mycelia isolated caused the ectomycorrhizae in the roots of pine seedlings.

Liquid Culture Enhances Protoplast Formation from the Auxotroph (Ser-) of lentinula edodes

  • The optimal conditions for the production and regeneration of the protoplasts from Lentinula edodes were studied. Protoplast formation from the mycelia of L. edodes which were cultured in liquid medium showed a significantly high yield compared with that of the mycelia which were cultured on cellophane covered agar media. A mixture of Novozyme 234 (15 mg/ml) and Cellulase Onozuka R10 (10 mg/ml) in 0.6 M mannitol (pH 4) was optimal lytic enzyme for the protoplast release. The optimal incubation time and mycelia age were 3.5-4 hours at $30^{\circ}C$ and 6-8 days, respectively. Regeneration frequency was 0.18% plated onto a medium containing 0.6 M sucrose, and 0.08% plated onto a medium containing mannitol. But hardly any regeneration was observed in the media containing NaCl, KCl, or $MgSO_{4}$ More than 90% of the protoplasts contained nuclei and the nucleus number per protoplast was 1.1. The DNA content per nucleus was 5.1 pg. The diameter of the protoplast was $3-5{\mu}m$ and it had a well defined cell structure.

  • It has been known that the novel diterphenoids, hericenone and erinacine isolated from the fruiting body and cultured mycelia of Hericium erinaceus showed potent stimulating activity of nerve growth factor (NGF)-synthesis. To investigate the biological activities of extracts from fruiting body, cultured mycelium and cell-free broth of H. erinaceus, the activity experiments of antioxidation and AChE inhibition were carried out. Sephadex G-10 gel filtration followed by HPLC on a ${\mu}Bondapak$ $C_{18}$ column of EtOAc extract from cultured mycelium showed a biological activity.

  • To investigate antitumor component of Lyophyllum decastes, the aqueous extract of its shake-cultured mycelia was subjected to antitumor test against sarcoma 180 cells implanted in ICR mice. The extract showed an inhibition ratio of 65.4% and was found to consist of a polysaccharide moiety and a protein moiety. After purification with DEAE-Sephadex A-50 ion exchange chromatography, Fraction D showed the highest inhibition ratio of 75.7%. The antitumor constituent was examined for immunoaccelerating activity and was found to increase macrophage accumulation in the peritoneal cavity and plaque forming cells of the spleen cells. It was named lyophyllan after the genus name.

  • A fraction (CMEx-AH-${\beta}$) of water-soluble polysaccharides, showing selective antitumor activities, was isolated from the mycelia of solid cultured Agaricus blazei Murill by hot-water extraction, ethanol precipitation, and series of chromatography. Chemical characteristics of CMEx-AH-${\beta}$, were as follows: carbohydrate content, 48%; monosaccharide composition, Man:Glu:Gal (2:93:5); molecular weight, $2{\times}10^5$; uronic acid content, 6.2%. Fundamental structure of CMEx-AH-${\beta}$, was deduced as ${\beta}-(1\;{\rightarrow}\;6)$-D-glucan with ${\beta}-(1\;{\rightarrow}\;3)$-D-glucosidic side chains based on results of methylation and $^{13}C$ NMR spectroscopic analyses.

  • Fraction (PMEx-AH-${\beta}$) of water-soluble polysaccharide, showing stimulating activity against macrophages, was isolated from mycelia of solid cultured Phellinus linteus by hot-water extraction, ethanol precipitation, and chromatography. Chemical characteristics of PMEx-AH-${\beta}$ were as follows: carbohydrate content, 71%; monosaccharide composition, Man:Glu:Gal (9:64:27); molecular weight, $1-7{\times}10^4$; uronic acid content, 6.8%. Fundamental structure of PMEx-AH-${\beta}$ is deduced as ${\beta}$-($1{$\rightarrow}6$)-D-glucan with ${\beta}$-($1{\rightarow}3$)-D-glucosidic side chains based on methylation analysis.
