THE STUDY ON TISSUE CULTURED WILD MOUNTAIN GINSENG(Panax Ginseng C.A. Meyer) ADVENTITIOUS ROOTS EXTRACT AS A COSMETIC INGREDIENT

  • Korean ginseng(Panax Ginseng C.A. Meyer) known as a oriental miracle drug is an important medicinal plant. Ginseng has been used for geriatric, tonic, stomachic, and aphrodisiac treatments for thousands years. Also, it is an antibiotic and has therapeutic properties against stress and cancer. Ginseng is widely distributed all over the world. Among them, Korean mountain ginseng has the most valuable effect on pharmaceuticals. The roots of mountain ginseng contained several kinds of ginsenosides that have many active functions for the human body. However, the study of mountain ginseng has a limit because the mountain ginseng is very expensive and rare. So, we artificially cultured mountain ginseng adventitious roots using the bioreactor culture system. We induced callus from original mountain ginseng, directly dug up in mountain and aged about one hundred ten years. Separated adventitious roots were precultured in 500ml conical flasks and then, transferred in 20L bioreactors. The adventitious roots of mountain ginseng were harvested after culturing for 40days, dried and then, extracted with several solvents. In this study, we investigated the whitening effect, anti-wrinkle effect and the safety of tissue cultured adventitious roots extract of mountain ginseng in order to identify the merit as a cosmetic ingredient. Particularly, extract of mountain ginseng adventitious roots showed whitening and anti-wrinkle effects. The inhibitory effect of this extract on the melanogenesis was examined using B-16 melanoma cell. When B-16 melanoma cells were cultured with adventitious root extract, there was a dramatically decrease in melanin contents of 8-16 melanoma cell. And we identified this extract inhibited Dopa auto-oxidation significantly. Also, when transformed mouse fibroblast L929 cells were treated with this extract, there was a significant increase in collagen synthesis. The results show significant inhibited melanization and wrinkle without inhibiting cell viability.

동종 종양 세포 용해액을 이용한 수지상 세포 항암 백신의 흑색종 폐암 전이 모델에서의 효과 연구 (Effect of Dendritic Cell Based Cancer Vaccine Using Allogeneic Tumor Cell Lysate in Melanoma Pulmonary Metastasis Model)

  • Background: To perform the successful dendritic cell-based cancer immunotherapy one of the main issues to be solved is the source of antigen for DC pulsing. Limitations occur by using auto-tumor lysate due to the difficulties obtaining enough tumor tissue(s) quantitatively as well as qualitatively. In this study the possibility of allogeneic tumor cell lysate as a DC pulsing antigen has been tested in mouse melanoma pulmonary me tastasis model. Methods: B16F10 melanoma cells $(1{\timeS}10^5/mouse)$ were inoculated intra venously into the C57BL/6 mouse. Therapeutic DCs were cultured from the bone marrow myeloid lineage cells with GM-CSF and IL-4 (1,000 U/ml each) for 7 days and pulsed with lysate of either autologous B16F10 (B-DC), allogeneic K1735 (C3H/He origin; K-DC) or CloneM3 (DBA2 origin; C-DC) melanoma cells for 18 hrs. Pulsed-DCs $(1{\times}10^6/mouse)_{[CGP1]}$ were injected i.p. twice with one week interval starting from the day 1 after tumor cell inoculation. Results: Without observable toxicity, allogeneic tumor cell lysate pulsed-DC induced the significantly better anti-tumor response (tumor scale: $2.7{\pm}0.3,\;0.7{\pm}0.3\;and\;0.3{\pm}0.2$ for saline, B-DC and C-DC treated group, respectively). Along with increased tumor specific lymphocyte proliferations, induction of IFN-${\gamma}$ secretion against both auto- and allo-tumor cell lysates was observed from the DC treated mice. (w/B16F10-lysate: $44.97{\pm}10.31,\;1787.94{\pm}131.18,\;1257.15{\pm}48.27$, w/CloneM3 lysate: 0, $1591.13{\pm}1.83,\;1460.47{\pm}86.05pg/ml$ for saline, B-DC and C-DC treated group, respectively) Natural killer cell activity was also increased in the mice treated with tumor cell lysate pulsed-DC ($8.9{\pm}_{[CGP2]}0.1,\;11.6{\pm}0.8\;and\;12.6{\pm}0.7%$ specific NK activity for saline, B-DC and C-DC treated group, respectively). Conclusion: Conclusively, promising data were obtained that allogeneic-tumor cell lysate can be used as a tumor antigen for DC-based cancer immunotherapy.

반하 추출물이 B-16 마우스 흑색종 세포의 멜라닌 생성과 타이로시네이즈 mRNA 양에 미치는 영향 (Effects of Banha Extract on the Melanin Biosynthesis and Tyrosinase mRNA Level in Bl6 Mouse Melanoma Cells)

  • 피부 내에서의 멜라닌 색소의 생성은 태양광에 존재하는 유해한 자외선으로부터 생체를 보호하는 중요한 방어 수단으로 여겨지고 있다. 타이로시네이즈라는 효소가 이러한 멜라닌의 합성 과정에 있어서 매우 중요한 역할을 수행한다. 이러한 이유로 피부의 색소생성을 조절하는 많은 연구의 대부분이 타이로시네이즈의 효소적 조절에 초점이 맞추어져 왔다. 본 연구자들은 전통 한약재인 반하의 물 추출물이 비록 타이로시제이즈에 대한 저해 작용은 없으나 배양된 B-16 마우스 흑색종 세포의 멜라닌 생성을 억제한다는 것을 발견하였다. 본 연구자들은 또한 반하 추출물이 배양된 세포의 타이로시네이즈 효소 활성을 낮춘다는 사실도 발견하였다. 이러한 반하 추출물의 작용 기작을 밝혀내기 위하여 반하 추출물이 B-16 마우스 흑색종 세포의 타이로시네이즈 mRNA양에 미치는 영향에 대하여 reverse transscription-polymerase chain reaction 기법을 이용, 연구를 수행 하였다. 연구 결과 반하 추출물은 타이로시네이즈 mRNA의 양을 용량 의존적인 경향으로 감소시킴이 밝혀졌다. 즉, 반하 추출물을 2$\mu\textrm{g}$/$m\ell$ 및 5$\mu\textrm{g}$/$m\ell$ 처리시 각각 20%와 44%의 mRNA 양의 감소가 관찰 되었다. 이러한 결과는 반하 추출물이 타이로시네이즈 mRNA의 전사 조절을 통하여 그 미백효과를 발휘한다는 것을 의미한다.

백부자산(白附子散)이 자외선 조사된 피부 손상과 색소침착에 미치는 영향 (The Effects of Baickbujasan Extract on the Skin Damage and Pigmendation Induced by Ultraviolet Irradiation)

  • Objective : The purpose of this study is to examine the effects of Baickbujasan(BB) on the skin damage and depigmentation. Method : The inhibition of tyrosinase activity, melanogenesis and cell viability in cultured B16 melanoma cells were measured. In order to test effects of reduction of melanogenesis, B16 F-10 mouse melanoma stem line was employed to extract melanin from cultured cell, where BB was added or not, and was dissolved in alkali for colorimetric analysis. Also, in order to test skin alteration in C57BL/6 after UV irradiation, the animals were grouped into a UV urradiation group and UV irradiation after BB application group. Dopa oxidase tissue staining was excuted to invesitage the change in the distribution of active melanin cell. The distribution of active melanin cell in inner skin of iNOS after damage from UVB irradiation and the manifestation condition of P53 which takes part in natural death of keratinocyte were examined. Result : The results indicate that BB has significant effects on tyrosinase activity, and melanogenesis in vivo test. BB seems to reduce C57BL/6, external dermatological damage, for instance, erythematous papule, eczema, loss of keratinocyte, reduction in pus, and relieves dermatological damages. Conclusion : BB can be applied externally for UV protection and depigmentation.

The Effects of Bamboo Extract on Human Melanocytes and B16 Melanoma Cells In vitro

  • To identify inhibitory effect of Bamboo extract on melanogenesis, the effect was compared with arbutin. ascorbic acid, hydroquinone, and kojic acid on the melanin biosynthesis in Bl6 mouse melailoma cells and cultured hunlan melanocytes. The cell viability of the agent was tested on cultured human melanocytes. We also examined. its free radical scavenging activity on 1,1-dipheny1-2-picrylhydrazyl (DPPH). Bamboo extract showed considerable effect against melanin production and did not reduce cell viability at the concentration tested. It also showed potent free radical scavenging activity.

  • Four compounds were isolated from the culture broth of Volvariella bombycina and they were identified as ergosta-4,6,8(14),22-tetraene-3-one (1), ergosterol peroxide (2), indole-3-carboxaldehyde (3), and indazole (4) by interpretation of spectroscopic data. Among them, compound 2 exhibited melanogenesis inhibitory effect in cultured B16 mouse melanoma cells.

  • The bioassay-guided fractionation of the methylene chloride soluble portion of a methanol extract of Gastrodia elata tubers led to the isolation of a new furfural, 5-(4-hydroxy-benzyloxymethyl)-furan-2-carbaldehyde (2), together with four known compounds (1, 3-5), which exhibited potent inhibitory activity at the concentration of 25 $\mu\textrm{g}$/ml on melanin biosynthesis in cultured B-16 mouse melanoma cells.

감초 물추출물의 멜라닌 형성 억제효과 (Inhibitory Effect on Melanogenesis of Radix Glycyrrhizae Water Extract)

  • This study was conducted to evaluate the effects of Glycyrrhizae Radix water extract, known as depigmenting agent, on melanin biosynthesis in cellular level. The inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis was identified by mushroom tyrosinase assay, To determine whether Glycyrrhizae Radix water extract suppress melanin synthesis in cellular level, B16 mouse melanoma cells were cultured in the presence of different concentrations of Glycyrrhizae Radix water extract. The maximum concentration of Glycyrrhizae Radix water extract that was not inhibitory to growth of the cells was 2 mg/ml. At that concentration, melanin synthesis was significantly inhibited without cytotoxicity after 5 days, compared with untreated cells. The treatment with Glycyrrhizae Radix water extract reduced tyrosinase and DOPAchrome tautomerase activity in a dose-dependent manner. These results suggest that the inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis is due to the suppression of tyrosinase and DOPAchrome tautomerase activity.

In vitro studies of anti-inflammatory and anticancer activities of organic solvent extracts from cultured marine microalgae

  • Marine microalgae are a promising source of organisms that can be cultured and targeted to isolate the broad spectrum of functional metabolites. In this study, two species of cyanobacteria, Chlorella ovalis Butcher and Nannchloropsis oculata Droop, one species of bacillariophyta, Phaeoductylum tricornutum Bohlin, and one species of Dinophyceae, Amphidinium carterae (Hulburt) were cultured and biomasses used to evaluate the proximate comical compositions. Among the determined proximate chemical compositions of the cultured marine microalgae, the highest content of crude proteins and lipids were exhibited in P. tricornutum and A. carterae, respectively. Solvent-solvent partition chromatography was subjected to fractionate each of the cultured species and separated n-hexane, chloroform, ethyl acetate, and aqueous fractions. Nitric oxide production inhibitory level (%) and cytotoxicity effect on lipo-polysaccharide-induced RAW 264.7 macrophages were performed to determine the anti-inflammatory activity. N. oculata hexane and chloroform fractions showed significantly the strongest anti-inflammatory activity at $6.25{\mu}g\;mL^{-1}$ concentration. The cancer cell growth inhibition (%) was determined on three different cell lines including HL-60 (a human promyelocytic leukemia cell line), A549 (a human lung carcinoma cell line), and B16F10 (a mouse melanoma cell line), respectively. Among the extracts, C. ovalis ethyl acetate and A. carterae chloroform fractions suppressed the growth of HL-60 cells significantly at 25 and $50{\mu}g\;mL^{-1}$ concentrations. Thus, the cultured marine microalgae solvent extracts may have potentiality to isolate pharmacologically active metabolites further using advance chromatographic steps. Hence, the cultured marine microalgae can be described as a good candidate for the future therapeutic uses.

금선련 조직 배양체 추출물의 멜라닌 합성 및 지방축적 억제 효과 (Inhibitory Effect of Jewel Orchid (Anoectochilus Formosanus) Plantlet Extract against Melanogenesis and Lipid Droplet Accumulation)

  • 일반적으로 보석란으로 알려진 금선련은 대만에서 폐나 간의 질병 및 발열이나 두통 치료를 위한 전통식물약제로 사용되어 왔다. 본 연구에서는 생물반응장치를 이용하여 조직배양된 금선련 식물체에 대하여 화장품 성분으로써 응용 가치를 평가하였다. 이미 몇몇 보고 된 논문에서 금선련은 항암활성, 면역 활성, 간 보호 활성 및 지질대사의 약리학적 활성 등에 대한 연구가 되고 있지만 화장품 성분으로 효능들에 대한 연구는 알려져 있지 않다. 따라서 본 연구에서는 생물반응장치를 이용하여 조직배양된 금선련 추출물에 대하여 미백 및 항비만 관련한 효능 효과를 평가하였다. 실험 결과 조직배양된 금선련 추출물은 tyrosinase 활성 및 멜라닌 합성 억제 효과뿐만 아니라 지방 전구 세포의 지방세포로의 분화를 억제시킴으로써 세포 내 지질 축적을 억제하였다. 이러한 결과들은 피부보호를 위한 화장품 성분으로서 응용 가능성을 제공 할 수 있을 것으로 사료된다.
