The Effects of a Culture & Arts Treatment Program for Elderly Women for Regional Community Consciousness (문화예술치유프로그램이 여성 노인의 지역공동체의식에 미치는 영향)

  • This research demonstrates the effects of a culture & arts treatment program for elderly women's regional community consciousness. Study subjects of this research are 18 women whose ages are 65 years old and older. (8 women participated in a culture & arts program and 10 women participated in an elderly center program.) The culture & arts program held 16 sessions where the subjects wrote letters or drew pictures. After this program, we measured the degree of regional community consciousness. The results of the study were, first, that the cultural and arts treatment program improved the regional community consciousness among the elderly woman. Second, the treatment program has improved the sub-region of the regional community consciousness among the woman's elderly. This result shows the culture & arts treatment program can be used as an effective way to enhance the consciousness of the regional community on elderly women

Effect of a co-culture of scenedesmus dimorphus and nitrifiers on advanced wastewater treatment capacity (Scenedesmus dimorphus와 질산화 박테리아의 공배양이 하수고도처리능에 미치는 영향)

  • This study investigated the effect of a co-culture of Scenedesmus dimorphus and nitrifiers using artificial wastewater on the removal of ammonium, nitrate and phosphate in the advanced treatment. To test the synergistic effect of the co-culture, we compared the co-culture treatment with the cultures using S. dimorphus-only and nitrifiers-only treatment as controls. After 6 days of incubation, nitrate was removed only in the co-culture treatment and total amount of N removal was 1.3 times and 1.6 times higher in the co-culture treatment compared to those in the S. dimorphus- and nitrifiers-only treatments, respectively. In case of total amount of P, co-culture treatment removed 1.2 times and 12 times more P than the S. dimorphus -and nitrifiers-only conditions, respectively. This indicates that the co-culture improved removal rates for ammonium, nitrate, and phosphate. This further implies that there was no need for denitrification of nitrate and luxury uptake of P processes because nitrate and phosphate can be removed from the uptake by S. dimorphus. In addition, co-culture condition maintained high DO above 7 mg/L without artificial aeration, which is enough for nitrification, implying that co-culture has a potential to decrease or remove aeration cost in the wastewater treatment plants.

Effect of chlorella culture solution using animal liquid manure on improving seed germination in perennial ryegrass

  • This experiments were conducted to evaluate the influence of Chlorella culture solution on seed germination of perennial ryegrass seeds. Chlorella are known to contain different bioactive compounds. In present research work, Chlorella culture solution using liquid manure as medium have been used to study their effects on germination and root length. The study conducted a germination experiment in petri-dishes. Four treatments were compared: non-treated control treated with distilled water, Chlorella culture solution and Chlorella culture filtrate, and liquid manure. The germination percentage of perennial ryegrass seeds was highest in the Chlorella culture solution treatment. Days required for 50, 70% seed germination was the fast in Chlorella culture solution and the Chlorella culture filtrate treatment. Root length of perennial ryegrass seeds was long by 1~2cm in the Chlorella culture solution compared with no treated control. The germination index of perennial ryegrass seeds was high by 180~202% in the Chlorella culture solution treatment compared to no treatment. Chlorella culture solution and the Chlorella culture filtrate have shown stimulatory effects in germination and development of root. Use of Chlorella culture solution and the Chlorella culture filtrate can be recommended to farmers as a ecofriendly practice for better germination and growth. Present research work reveals that Chlorella contain certain growth promoting substances which enhances seed germination.

Effects of Heating on Hydroxyl Radical-Generated Toxicity in Mouse Forebrain Tissue Culture

  • This experiment was carrid out to know the effects of heating and serum on hydroxyl radicals in embryonic mouse forebrain (cerebrum) culture. The heating to mouse embryonic cerebrum cells in culture was done in a water bath at 43${\circ}C$ for 60min. After that, two supernatants were prepared at 20 hrs and 48 hrs respectively after heat treatment to the brain cells. To find out the heating effects on neuron cells, mouse cerebrum cells (13 embryonic day) were cultured in hydroxyl radical generation system composed of 20mU/ml glucose oxidase (GO system), using condition of normal culture media (MEM, 5% serum, 5% $CO_2$or supernatant prepared after heating at 43${\circ}C$ for 60 min in a water bath. Supernatant prepared at 20 hrs after heat treatment had a greater protective effects against hydroxyl radical than supernatant prepared at 48 hrs after heat treatment . Otherwise, the protective effect of serum against hydroxyl radicals in the cultured brain cells is higher than that in the heat treatment. These results indicated that serum in culture media reduced cytotoxicity of hydroxyl radicals in mouse forebrain culture, also that heat treatment showed the protective effects against hydroxyl radicals generated with 20mU/ml GO system in mouse forebrain culture.

Effects of Supplemention of Antibiotic, Probiotic and Yeast Culture of Performance and Meat Quality in Broiler Chicks (항생제, 생균제 및 효모제 첨가가 육계의 성장과 육질에 미치는 영향)

  • The present study was conducted to investigate the effects of dietary supplementions of 0.1% probiotic and 0.1% yeast culture on the growth performance and meat quality of broiler chicks. A total of 160 Arbor Acre broiler chicks were randomly allotted to 16 pens ; four pens per treatment and 10 birds per pen. Feeding trial lasted for 6 weeks. The results obtained are summerized as follows : The body weight gain and feed intake was slightly higher in group of broiler chick supplemented antibiotic than those of other treatment groups, but were not significantly different among treatments. The feed conversion were slightly lower in groups of broiler chick supplemented antibiotic and probiotic than those of other treatment groups, but were not significantly different among treatments. The pH of thigh musle was slightly higher in group of broiler chick supplemented probiotic than those of other treatment groups. Crude protein, crude fat and crude ash of thigh muscle were lower in groups of broiler chick supplemented probiltic and yeast culture than those of other treatment groups, but were not significantly different among treatments. The saturated fatty acid content of the thigh muscle was tende to be higher in group of broiler chick supplemented yeast culture, but the unsaturated fatty acid content was tende to be lower in group of broiler chick supplemented yeast culture than those of other treatment groups.

CYTOTOXICITY OF D-GALACTOSAMINE ON PRIMARY CULTURES OF ADULT RAT HEPATOCYTES

  • Primary cultures of adult rat hepatocytes were used to study the cytotoxicity of D-galactosamine. Hepatocytes were isolated by a collagenase perfusion technique and maintained as monolayers in serum-free medium on collagen-coated culture dishes. Treatment of galactosamine to the culture markedly inhibited the uptake of ${\alpha}$-aminoisobutyric acid (AIB) inducible with glucagon and dexamethasone. At0.1 mM of galactosamine, AIB uptake was inhibited significantly when treated for 12 hr. At higher doses (0.25, 0.5 and 1.0mM), a significant inhibition was noticed after 1 hr exposure. Generally the magnitude of the inhibition was related to the dose and treatment time of galactosamine. Treatment of galactosamine also produced a dose- and treatment time-related suppression of the tyrosine aminotransferase (TAT) induction caused by dexamethasone. Meanwhile, uptake of ouabain was not affected by the treatment of galactosamine. The viability of the hepatocytes was decreased only slightly by the treatment of galactosamine; more than 87% of the cells excluded tryphane blue when treated 1 mM galactosamine for 12 hr. Galactosamine induced depressions of AIB uptake and TAT activity were prevented by the simultaneous addition of uridine to the culture. Effect of Fluorescent Light Treatment during Imbibition and Culture on Growth of Soybean Sprout

  • The lateral root formation in soybean sprout culture declines its quality. This study was done to measure the effect of fluorescent light treatment during 24 hour imbibition and 6-day culture on seed germination and growth of soybean sprout. After 6 day culture, the sprouts were sorted as normal (>4cm), abnormal (<4cm) and non-germination by their hypocotyl lengths, and lateral roots, fresh and dry weights were measured. Lateral roots were less formed in the fluorescent light treatment lasted during the whole period of the imbibition than in the treatment for 50 minutes a day during the culture. The fluorescent light treatment during the imbibition mainly affected the germination and growth compared to the treatment done during the culture. Compared to the dark imbibition, the light treatment during the imbibition resulted in more normal sprouts, thicker diameters of hypocotyl and hook, and more fresh weights in cotyledon, hypocotyl, whole sprout, and economic yield. However, these results were reverse in lengths of hypocotyl and root, and fresh and dry weights of roots. It is concluded that the fluorescent lamp mainly irradiating red and blue lights can be used for the sprout production as an alternative light replacing blue and red lights treated during the imbibition because it blocked the lateral root appearance and stimulated growth of the sprout.

  • Rice hull was used as a additive in order to find the effect for incresing of mushroom growth and yield in Chungnam Provincial techinical institution. 1 Treatment of 80% rice hull in small Neutaribeosut mycelial grow duration is shorter about 11 days and yield increased about 7% than conventional culture. 2. In case of Chongpung Neutaribeosut bottle culture, mycelial growth duration is shorter about two to three days in additive of 30 to 80% rice hull compared to conventional but yield similar to conventional. 3. Treatment of 30% rice hull in Agrocybe aegerita bottle culture, mycelial growth and yield increased 6days and 6% than convrntional, respectively 4 Treatment additived of 30% to 40% rice hull in Ganoderma lucidum bottle culture, similar to 454ays demand in mycelial grow duration and 38g yield/bottle in conventioal culture methods.

Maintenance of Proliferation and Adipogenic Differentiation by Fibroblast Growth Factor-2 and Dexamethasone Through Expression of Hepatocyte Growth Factor in Bone Marrow-derived Mesenchymal Stem Cells

  • Several studies have investigated the various effects of dexamethasone (Dex) on the proliferation and differentiation of mesenchymal stem cells (MSCs). Previously, we reported that co-treatment with L-ascorbic acid 2-phosphate and fibroblast growth factor (FGF)-2 maintained differentiation potential in MSCs through expression of hepatocyte growth factor (HGF). In this study, we investigated the effects of co-treatment with FGF-2 and Dex on the proliferation and differentiation potential of MSCs during a 2-month culture period. Co-treatment with FGF-2 and Dex increased approximately a 4.7-fold higher accumulation rate of MSC numbers than that by FGF-2 single treatment during a 2-month culture period. Interestingly, co-treatment with FGF-2 and Dex increased expression of HGF and maintained adipogenic differentiation potential during this culture period. These results suggest that co-treatment with FGF-2 and Dex preserves the proliferation and differentiation potential during long-term culture.

Snake Venom synergized Cytotoxic Effect of Natural Killer Cells on NCI H358 Human Lung Cancer Cell Growth through Induction of Apoptosis

  • Objectives : I investigated whether snake venom can synergistically strengthen the cytotoxic effects of NK-92 cells, and enhance the inhibition of the growth of lung cancer cells including NCI-H358 through the induction of death receptor dependent extrinsic apoptosis. Methods : Snake venom toxin inhibited cell growth of NCI-H358 Cells and exerted non influence on NK-92 cell viability. Moreover, when they were co-cultured with NK cells and concomitantly treated with $4{\mu}g/m{\ell}$ of snake venom toxin, more influence was exerted on the inhibition of growth of NCI-H358 cells than BV or NK cell co-culture alone. Results : The expression of Fas, TNFR2 and DR3 and in NCI-H358 lung cancer cells was significantly increased by co-culture of NK-92 cells and treatment of $4{\mu}g/m{\ell}$ of snake venom toxin, compared to co-culture of NK-92 cells alone. Coincidentally, Bax, caspase-3 and caspase-8 - expressions of pro-apoptotic proteins in the extrinsic apoptosis pathway, demonstrated significant increase. However, in anti-apoptotic NF-${\kappa}B$ activities, activity of the signal molecule was significantly decreased by co-culture of NK-92 cells and treatment of $4{\mu}g/m{\ell}$ of snake venom toxin, compared to co-culture of NK-92 cells or snake venom toxin treated by NCIH358 alone. Meanwhile, in terms of NO generation, there is a significant increase, in co-culture of NK-92 cells with NCI-H358 cells as well as the co-culture of NK-92 cells and concomitant treatment of $4{\mu}g/m{\ell}$ of snake venom toxin. However, no synergistic increase of NO generation was shown in co-culture of NK-92 cells and treatment of $4{\mu}g/m{\ell}$ of snake venom toxin, compared to co-culture of NK-92 cells with NCI-H358 cells. Conclusion : Consequently, this data provides that snake venom toxin could be useful candidate compounds to suppress lung cancer growth along with the cytotoxic effect of NK-92 cells through extrinsic apoptosis.
