Antioxidant Activities of the Mycelium and Culture Broth of Phellinus igniarius and Agrocybe cylindracea (말똥진흙버섯 및 버들송이버섯의 균사체 및 배양액의 항산화활성)

  • The fractions obtained from the mycelia and their culture broth of Agrocybe cylindracea and Phellinus igniarius were tested for the antioxidation activities. In electron donating ability test, strong activities more than 70% were observed in ethyl acetate and butanol fractions of A. cylindracea mycelium, butanol fraction of A. cylindracea culture broth, diethyl ether fraction of P. igniarius mycelium, and diethyl ether and ethyl acetate fractions of P. igniarius culture broth. Lipid peroxidation inhibition was observed in diethyl ether, butanol and water fractions of A. cylindracea mycelium, and water fraction of A. cylindracea induced the decrease in uric acid production. Activities of $50{\sim}60\;%$ of nitrite scavenging ability were observed in diethyl ether fraction of A. cylindracea mycelium, ethyl acetate and water fractions of A. cylindracea culture broth, diethyl ether fraction of P. igniarius mycelium, and diethyl ether, chloroform, butanol and water fractions of P. igniarius culture broth. In melanin biosynthesis inhibition activity measurement, water fraction of A. cylindracea mycelium and butanol fraction of P. igniarius mycelium showed twice higher activities than control. From these results, the fungi of P. igniarius and A. cylindracea had strong antioxidant activities.

Hydrogen Production from Wastewater in Takju Manufacturing Factory by Microbial Consortium (탁주제조공장 폐수로부터 혼합균주에 의한 수소생산)

  • Culture conditions for biological hydrogen production were investigated in wastewater of Takju manufacturing factory. Rhodobacter spaeroides KCTC1425, photosynthesis bacteria, and Enterobacter cloacae YJ-1, anaerobic bacteria were used. The hydrogen production were $195.3m{\ell}{\cdot}H_2/{\ell}$ broth for Rhodobacter spaeroides KCTC1425 and $271.8m{\ell}{\cdot}H_2/{\ell}$ broth for Enterobacter cloacae YJ-1 during 36 h. The hydrogen production increased with light intensity, and were highest over 12000Lux. In mixed culture of Rhodobacter spaeroides KCTC1425 and Enterobacter cloacae Y J-1, the optimum mixing ratio of hydrogen production was 20 and 80. Adding volume of yeast extract for maximum hydrogen production was 15 $g/{\ell}$, but there was no effect over that. $Na_2MoO_4$ was most effective among the inorganic salts, and the optimum volume was 0.4 $g/{\ell}$. In semi-continuous culture, total hydrogen production was $13086m{\ell}{\cdot}H_2/{\ell}$ broth for 144 h with operating period of 24 h.

Extraction and Purification of an Antifungal Antibiotic Saccharide from Bacillus sp.

  • An antifungal antibiotic was extracted three times using n-butanol from the culture broth of Bacillus sp. Bioassayguided column chromatography with silica gel and Sephadex LH-20 yielded 62 mg of the original active compound from 1 L of culture broth. The minimal inhibitory concentration values were 25 and $50{\mu}g/ml$ against Pyricularia oryzae and Pellicularia filamentosa, respectively. Based on results obtained from the analysis of the structure of the antibiotic using MS, NMR, and IR spectroscopy, the antifungal antibiotic was shown to consist of only six of fructose.

  • Rhodospirillum rubrum KCTC 1372 produced hydrogen from glucose for first 48hrs culture under the anaerobic photosynthetic conditions, and after 48hrs culture the hydrogen production was decreased by the accumulation of producing organic acids in broth. Only 41% of glucose was consumed and 143mL/day/L hydrogen were produced after 96hrs culture. However the hydrogen production and glucose consumption were substantially increased when the pH of the culture broth were controlled to 6.8-7.2. After 96hrs culture, 450mL/day/L hydrogen were produced, and about 80% glucose was consumed. Specific hydrogen production rate was 48.33mL/hr/g cells under pH not controlled, but 45.42mL/hr/g cells under pH controlled.

THE EFFECT OF LEUCONOSTOC LACTIS 51 AGAINST THE PLAQUE FORMATION OF STREPTOCOCCUS MUTANS (Streptococcus mutans의 치태형성에 대한 Leuconostoc lactis 51의 영향)

  • Dental caries is a bacterial disease of the dental hard tisssus, characterized by a localized, progressive, molecular disintegration of tooth structure. The action of Leuconostoc lactis 51 about plaque formation and replication by Streptococcus mutans was studied as follows. 1. Lower amount of plaque was produced at the mixed culture of S. mutans and L. lactis 51 than S. mutans alone on the wires in the beaker. 2. Fewer cells of S. mutans were replicated at the mixed culture of S. mutans and L. lactis 51 than S. mutans alone. 3. In M17Y broth, viable cells of S. mutans and L. lactis 51 increased for 12 hours, and decreased for 24 hours. In M17YS broth, viable cells of S. mutans showed time-dependent decrease at mixed culture of S. mutans and L. lactis 51. 4. The culture supernatant of L. lactis 51 didn't inhibit the replication of S. mutans and the formation of artificial plaque. 5. Sucrose and frutose were extracted from the culture supernatant of L. lactis 51 in M17YS broth. These results suggest that L. lactis 51 isolated from the oral cavity inhibits the replication of S. mutans and the formation of artificial plaque.

  • Vibrio, Photobacterium, Alteromonas and Xenorhabdus species are capable of emitting light, called bioluminescence. They exist in marine, freshwater and terrestrial environments. Bacterial bioluminescent reaction is that reduced riboflavin phosphates and a long-chain aldehyde are oxidized in the presence of molecular oxygen and enzyme luciferase. This experiment aims to develop the proper culture media and to optimize the storage condition for the recovery of bioluminescent activity in Photobacterium phosphoreum. The Luria broth (LB) medium was modified for cultivation of Photobacterium phophoreum, called as modified LB(mLB) medium. The mLB medium is LB fortified with 3% glycerol and 1.5% NaCl. In mLB medium. bacterial growth and bioluminescent activity are 25% higher than those in a Nutrient broth medium. When the cell stocks were stored at $-20^{\circ}C$, $-70^{\circ}C$ and LN2 for 3 months, cell growth and bioluminescent activity of culture after stored at $-20^{\circ}C$ were better than those of other treatments. The highest bioluminescent activity obtained at the late exponential phase in all treatments. When the cell stock was freeze-dried with 5% adonitol as a cryoprotectant, the recovery of cell was better than those of control and freeze-dried cell stock without addition of cryoprotectant.

Evaluation of the Selective Enrichment Culture to Recover Clostridium difficile

  • To evaluate the recovery rates to increase toxigenic C. difficile, the selective enrichment broth culture methods were compared with commonly used cytotoxin assays and toxigenic culture. First, the enrichment culture, using the selective medium broth for 2 to 5 days, was performed and then, toxigenic C. difficile was confirmed by C. difficile toxin gene-specific PCR after being cultured on C. difficile selective agar. The sensitivity of C. difficile from the enrichment culture (100%) was higher than that of C. difficile selective agar culture (93.8%), while positive predictive values (PPV) were low; 72.7% (16/22) and 88.2% (15/17). PPV of the enrichment culture are not high. Recently, combinations of C. difficile selective agar culture, C. difficile A & B assays, glutamate dehydrogenase, and nucleic acid amplification method are widely used. The enrichment culture was disadvantageous in PPV, turn-around time, and cost. So, what we performed is not considered as a common method of diagnosis of C. difficile-associated diarrhea.

Isolation and Properties of Amino Acid Antimetabolite from Streptomyces sp. YS-943 (Streptomyces sp. YS-943균주가 생산하는 아미노산 대사길항물질의 정제와 성상)

  • A Streptomyces strain YS-943, which produced amino acid antimetabolite, was isolated from soil. During the course of screening for new amino acid antimetabolites from the culture broth of Actinomycetes, we found that the strain produced a substance active against Gram-positive bacteria and its activity was reversed by L-methionine and L-histidine on the synthetic minimal agar medium in the culture broth.The morphological and cultural characteristics serve to identify the producing organism strain YS-943 as the genus Streptomyces. Fermentation was carried out in the synthetic medium at 28$\CIRC$C for 48 hours. The fermentation yield reached about 12 mg per liter of the broth. The YS-943 substance was obtained as white powder, mp 194$\CIRC$C and has the molecular formular of C$_{4}$H$_{8}$N$_{2}$O$_{4}$. Its structure was determined to be o-carbamyl-D-serine by spectroscopic data. It is active against some Gram-positive bacteria and reversed by L-methionine and L-histidine.

  • Two novel compounds SSC1 and SSC2 were isolated from culture broth produced from the strain of Stereum hirsutum by using of YM broth. They were isolated through HP-20 column chromatography, silica gel column chromatography and preparative HPLC, successively. The molecular formulas of SSC1 and SSC2. were determined as C$\sub$15/H$\sub$22/O$_3$ by high resolution EI -mass. The chemical structures of SSC1 and SSC2 were determined as sesquiterpenoid compounds by spectroscopic analysis of UV, IR, $^1$H NMR, $\^$13/C NMR, DEPT, HMQC and HMBC spectrum.

Production of Red Pigment by Serratia sp. KH-95 and its Cultural Properties (Serratia sp. KH-95에 의한 적색 색소 생산 및 배양학적 특성)

  • Optimal media and cultural conditions for the production of prodigiosin-like pigment were established using Serratia sp. KH-95. Glucose and phosphate(K2PO4) stimulated the cell growth, but inhibited the production of pigment at concentration levels of above 10 g/L and 2.0 g/L, respectively. Addition of soy been oil or rice oil to the production medium accelerated cell growth up to more than 2-3 times, but the production of prodigiosin increased about 15-20% in spite of the good cell growth. The effect of pH on the production of pigment was investigated in a 5 liter-bioreactor. When the pH of culture broth was maintained below 8.0, most of pigment was attached to the surface of cells. When the pH of culture broth was above 8.5, however, about 70% of total pigment was suspended in the supernatant of the broth. The cell growth and production of pigment were inhibited at dissolved oxygen concentration of below 10% of air-saturation.
