Cultural Characteristics of Veiled Lady Mushroom, Dictyophora spp.

  • This study was carried out to obtain the basic data for artificial culture of veiled lady mushroom (Dictyophora spp). The optimal conditions for the mycelial growth were $25^{\circ}C$ and pH 5.0 for all isolates except the optimal temperature of $30^{\circ}C$ for D. echinovolvata ASI 32002 and Phallus rugulosus. The optimal medium for Dictyophora spp. was PBA (potato bamboo sawdust extract agar) medium. The strain ASI 32002, D. echinovolvata, grew faster than. D. indusiata ASI 32003 and Phallus rugulosus ASI 25007 on the medium. Carbon sources such as glucose, maltose and inuline were favorable for stimulating a mycelial growth of the two strains of ASI 32002 and ASI 32003. Asparagine and glutamine appeared to be favorable to the strain ASI 32002 and ASI 32003, where as alanine, one of nitrogen source also favorable to the strain ASI 32002. The optimum C/N ratio of the two isolates of ASI 32002 and ASI 32003 was about 25 : 1 when 2% glucose as carbon source was mixed with the basal medium. While, in the case of 4% as carbon source, the optimum C/N ratio was about 30 : 1.

Stabilization and Antifungal Activity of Isolated Symbiotic Bacteria from Entomopathogenic Nematodes (곤충병원성 선충에서 분리한 공생세균의 안정화 및 항진균활성)

  • In order to use the symbiotic bacteria from ethomophatogenic nematodes as a biological control agent for agriculture, the cultural condition for maintaining phase I and antifungal activity was investigated. Symbiotic bacteria (SB) 1 stain from nematodes were selected from the three strains isolated from entomopathogenic nematodes. The growth of the SB 1 strain in NB, TSB, TY and YS medium was higher than that of the SB 2 and SB 3 strain. The packed cell volume of the SB 1 strain was reduced in NB medium which showed radical pH change. Phase I of the SB 1 strain was maintained in TSB medium after being stored for 2 weeks at $4^{\circ}C$. Culture broth with the SB 1 strain in TSB medium for 6 days and 7 days showed antifungal activities against Rhizoctonia solani KACC 40142, Botrytis cinerea Pers. KACC 40854, and Botrytis cinerea Pers. KACC 41008. Culture broth with the SB 1 strain in TSB medium containing 100 mM L-proline for 5 days showed antifungal activities against Rhizoctonia solani KACC 40142, and Botrytis cinerea Pers. KACC 40854.

A Study on the Cultural Characteristics of Pholiota nameko Mycelium (맛버섯 균사체의 배양 특성에 관한 연구)

  • This Study was carried out to investigate the optimal mycelial growth of Pholiota nameko. The optimal medium for the mycelial growth was ME medium. The optimal temperature and pH were $25^{\circ}C$$\pm$1 and 5.5, respectively. The modified optimal medium compositions were glucose 3% (w/v), malt extract 0.25% (w/v), yeast extract 0.25% (w/v), $KH_2PO_4$ 0.046% (w/v), $K_2HPO_4$ 0.1% (w/v), $MgSO_4$$7H_2O$ 0.05% (w/v). From the result of experiments on the optimal temperature, pH and nutritional requirements, the mycelial growth of modified optimal medium was higher than that of ME medium.

Nutritional Conditions of Xylanase Production from Xylose Fermenting Yeast (Xylose 발효효모의 Xylanase 생성)

  • Cultural conditions for the formation of extracellular xylanase by Candida sp. X-6-41 were investigated. The xylanase was not produced in culture medium containing polypeptone or yeast extract as a nitrogen source, respectively, whereas the enzyme w8s produced in chemically defined medium containing (NH$_4$)$_2$SO$_4$as a sole nitrogen source. The xylanase production was affected by the amino acids such as isoleucine and tryptophan. The enzyme production of the strain was completely inhibited by the addition of isoleucine in the culture medium, but enhanced by tryptophan below the concentration of 25$\mu$g/$m\ell$.

Growth Stimulation of In Vitro Shoots by the Post-supplying of Liquid Medium in Anthurium andreanum ′Atlanta′ (액체배지 첨가에 의한 Anthurium andreanum ′Atlanta′의 기내생육 촉진)

  • In order to enhance shoot elongation and rooting of Anthurium andreanum 'Atlanta' in vitro, 15mL of liquid media containing various concentrations of activated charcoal, sucrose and MS salts were added in same vessels after small shoots were induced from the calli on mudium supplemented with 10.0mg/L BA and 0.1mg/L 2.4-D. The post-supplying of 15mL liquid medium containing MS macro and micro elements, 30g/L sucrose and 5.0∼10.0g/L activated charcoal was significantly stimulated the shoot elongation and rooting of regenerated shoots from calli. The medium addition was also resulted in the enhanced soil survival, elongation and rooting of plantlets in cultural soil mixed with perlite and vermiculite(1 : 1)

Screening of Salicylic acid Producing Rhizobacteria Isolated from Plant Roots and Rhizosphere (식물의 뿌리와 근권으로부터 Salicylic acid를 생성하는 근권세균의 검색)

  • Twenty two rhizobacteria were isolated from the roots and rhizosphere of radish, carnation, potato and tomato. There isolates produced a fluorescent pigment in King's B medium and identified as Pseudomonas spp. These isolates colonized roots and rhizosphere of the host plants. In the study of cultural characteristics of the bacteria, the pH of the culture broth was changed from neutral (7.0) to alkali (8.8∼9.41) and the numbers of cells were increased from 106 to 108 after 40 hr of incubation in basal standard succinate medium. The salicylic acid production identified by pink color reaction were observed in 7 bacteria. Out of these 7 salicylic acid producing bacteria, only 2 strains of bacteria such as Pseudomonas fluorescens RS006, and Pseudomonas sp. EN401 were confirmed as salicylic acid producers by optical density measurement. Therefore, for screening of salicylic acid producing bacteria from the roots and rhizosphere, color reaction of the culture medium should be done in the first step, and then optical density measurement of culture extract should be made for the confirmation of salicylic acid production.

A Study on the Purification and Characteristics of Branched-Chain Amino Acid Aminotransferase in Cultural Mycelia of Cordyceps militaris (번데기동충하초 균사 중의 Branched-Chain Amino Acid Aminotransferase의 분리정제 및 그 특성에 관한 연구)

  • The optimum conditions of Cordyceps militaris mycelial growth, purification and characteristics of branched-chain amino acid aminotransferase [BCAT(EC 2.6.1.42)] in this mycelium were studied. Optimum pH, temperature and medium of culture of mycelia were 5.5, $22.5^{\circ}C$ and Hamada medium (HM), respectively. BCAT in homogenate of this mycelia was precipitated by 20-40% saturated solution of ammonium sulfate and then purified by DEAE (diethylaminoethyl)-Sephadex A-50 column chromatography with linear concentration gradient and Sephadex G-200 gel filtration. A single band of purified enzyme was detected on SDS-PAGE (sodium dodecylsulfate-polyacrylamide gel electrophoresis). Optimum pH and temperature of BCAT were found to be 7.8 and $29^{\circ}C$, respectively. It showed activity toward L-leucine, L-isoleucine and L-valine as a substrate. The Km values of this enzyme for L-leucine were determined to be 5.88 mM for L-leucine.

Isolation and Characterization of Bacteria Able to Grow with Phenol at High Concentrations for Bioremediation (생물학적 환경정화를 위한 고농도 페놀에서 생육할 수 있는 세균의 분리 및 특성)

  • For the biological treatment of industrial wastewater containing high concentration of phenol, isolation and characterization of phenol - degrading bacterium were carried out. A bacterial strain P2 capable of degrading phenol was isolated from contaminated soils by enrichment culture technique and identified as the genus Rhodococcus by morphological, cultural, biochemical characteristics, and Biolog system. The optimal medium composition and cultural conditions for the growth and degradation of phenol by Rhodococcus sp. P2 were 0.1% of (NH$_4$)$_2$SO$_4$, 0.2% of KH$_2$PO$_4$, 0.25% of Na$_2$HPO$_4$ㆍ12$H_2O$, 0.2% of MgSO$_4$ㆍ7$H_2O$, and 0.008% of CaC1$_2$ㆍ2$H_2O$ along with initial pH 8.5 at 3$0^{\circ}C$. Rhodococcus sp. P2 could grow with phenol as the sole carbon source up to 1,800 ppm in batch cultures, but did not grow in medium containing above 2,000 ppm of phenol. When 800 ppm phenol was given in the optimal media, Rhodococcus sp. P2 completely degraded it within 24 h. Meanwhile, 1,800 ppm of phenol was degraded within 9 days. Rhodococcus sp. P2 could utilize toluene, n-hexane, xylene and benzene as sole carbon source .

  • Bacillus subtilis SH-1 have been isolated and identified from coastal sea, in Pusan, The optimal cultural characterization of Bacillus subtilis SH-1 for 속 production of bacteriolytic enzyme was determained. Bacillus subtilis SH-1 produced the bacteriolytic enzyme well in the medium consist of 1.0% glucose, 1.0% yeast extract, 1.0% NaCI, 0.02% $K_2HPO_4,\;0.002%\;MgSo_4{\cdot}7H_2O,\;0.001%\;MnSO_4{\cdot}5H_2O,\;0.0001%\;FeSO_4{\cdot}7H_2O$. The optimal medium pH, incubation temperature, and shaking tome for the highest production of the enzyme were 8.0, $30^{\circ}C$ and 28 hours respectively.

  • The taxonomical properties of strain J-275L isolated from soil as a microorganism which produces extracellular adenine deaminase and cultural conditions for the enxyme production were studied. The hyphae of strain J-275L is fragmented into rod-or coccus-like elements. The elements of fragmented aerkal hyphae has smooth surfaces. The cell wall of the organism contains LL-diaminopimelic acid. Mycolic acid are not produced. As a result of taxonomical studies, strain J-275L is designated as Nocardioides sp. J-275L. The optimum medium for the enzyme production from Nocardioides sp.J-275L wascomposed of 0.5% peptone, 0.5% dextrin, 1% yeast extract, and 0.2% $K_{2}HPO_{4}$. The optimum initial pH of the medium was pH 7.5.
