Human activities greatly affect the environment causing its degradation. Urban development and road networks construction cause main impacts on ecosystems and particularly on vegetation cover: road constructions induce complete degradation of the vegetation cover and often leaves a hare land, sometimes without even a soil cover. Reconstitution of vegetation cover is necessary to limit superficial erosion and land slipping on the road, towards a reintegration of the site in the neighbouring landscape. Many approaches have been studied over the last 30 years aiming at this reconstitution of vegetation cover. At frost, the main purpose of land reclamation was to create a new ecosystem. At this time, the environment created was rather a "garden" with a new soil adapted to the plantation of "decorative" species. Then, in early 90′s many studies on the restoration ecology concept rather focused on adapting the vegetation to the existing conditions on the site, as in a side road embankment for example. Nowadays, we notice a large tendency towards the use of such adapted native species instead of industrially produced seeds. In southern France, our team have led research on the potentials of those local species for their use in revegetation processes with hydro-seeding. We therefore developed an approach combining the use of different types of species: Industrially produced, native and wild cultivated species. This method integrates the benefits of using available low costing seeds that are already used on large scale projects with better adapted species, issued form the cultivation of native species and seed production for their use on smaller scale and more costly but more effective results. The use of wild cultivated species seeds was developed in order to limit the cost and reduce harsh natural seed withdrawal in the natural environment In the case of the use of native species. Besides, the use of such seeds allowed a larger geographical scale of use than with local native seeds. In addition, our team began two years ago a research project in Lebanon aiming at the Introduction and development of the revegetation techniques in Lebanon. In fact, this country bared since 20 years the consequences of urban pressure on its environment especially by the development of quarries and road networks. Therefore, pioneer work is necessary to aim at the adaptation of these techniques to the local environment.

Recently, we reported the robust in vitro three-dimensional (3D) expansion of intestinal organoids derived from adult bovine (> 24 months) samples. The present study aimed to establish an in vitro 3D system for the cultivation of intestinal organoids derived from growing cattle (12 months old) for practical use as a potential alternative to in vivo systems for various purposes. However, very few studies on the functional characterization and 3D expansion of adult stem cells from livestock species compared to those from other species are available. In this study, intestinal crypts, including intestinal stem cells, from the small intestines (ileum and jejunum) of growing cattle were isolated and long-term 3D cultures were successfully established using a scaffold-based method. Furthermore, we generated an apical-out intestinal organoid derived from growing cattle. Interestingly, intestinal organoids derived from the ileum, but not the jejunum, could be expanded without losing the ability to recapitulate crypts, and these organoids specifically expressed several specific markers of intestinal stem cells and the intestinal epithelium. Furthermore, these organoids exhibited key functionality with regard to high permeability for compounds up to 4 kDa in size (e.g., fluorescein isothiocyanate [FITC]-dextran), indicating that apical-out intestinal organoids are better than other models. Collectively, these results indicate the establishment of growing cattle-derived intestinal organoids and subsequent generation of apical-out intestinal organoids. These organoids may be valuable tools and potential alternatives to in vivo systems for examining host-pathogen interactions involving epithelial cells, such as enteric virus infection and nutrient absorption, and may be used for various purposes.

Plant regeneration has been optimized increasingly by organogenesis and somatic embryogenesis using a range of explants with tissue culture improvements focusing on factors, such as the age of the explant, genotype, media supplements and $Agrobacterium$ co-cultivation. The production of haploids and doubled haploids using microspores has accelerated the production of homozygous lines in Brassicaceae crop plants. Somatic cell fusion has facilitated the development of interspecific and intergeneric hybrids in sexually incompatible species of $Brassica$. Crop improvement using somaclonal variation has also been achieved. Transformation technologies are being exploited routinely to elucidate the gene function and contribute to the development of novel enhanced crops. The $Agrobacterium$-mediated transformation is the most widely used approach for the introduction of transgenes into Brassicaceae, and $in$$vitro$ regeneration is a key factor in developing an efficient transformation method in plants. Although many other Brassicaceae are used as model species for improving plant regeneration and transformation systems, this paper focuses on the recent technologies used to regenerate the most important Brassicaceae crop plants.
Among non-timber forest products, Tricholoma matsutake is a high value added item. Many countries, including Korea, China, and Japan, are doing research and technology development to increase artificial cultivation and productivity. However, the production of T. matsutake is on the decline due to global warming, abnormal temperatures and pine tree pest problems. Therefore, it is necessary to identify trends in domestic and foreign research on T. matsutake, respond to preemptive research and development to preserve the genetic resources of T. matsutake and increase its productivity. Based on the correlation between keywords in the high frequency keywords, it was observed that microbial clusters of T. matsutake are mainly found in Korea. The main focus in China has been the pharmacology studies on the ingredients of T. matsutake. The main focus in Japan has been on preserving the genetic diversity and species of T. matsutake. Thus, future domestic studies of T. matsutake will require pharmacological studies on the ingredients of T. matsutake and on its genetic diversity and species conservation. In addition, unlike China and Japan, genetic keywords did not appear in Korea at high frequency. Therefore, Korea will have to proceed with research using modern molecular biology techniques.

King oyster mushroom(Pleurotus eryngii) is one of the most commercially important mushrooms in Korea. Development of fruit body and disease occurrence are sensitive to environmental conditions such as temperature, humidity, carbon dioxide(CO2) concentration. The purpose of this study was to investigate the changes in the growth environment of king oyster mushroom by installing Airocide, an air purifier for the purpose of improving mushroom cultivation environment. The results of the environment conditions, identification of pathogenic organisms and pathogenesis during the cultivation were as follows. Airocide operation increased the CO2 concentration of the cultivation room by more than 400 ppm on average, but the increase of CO2 concentration at this level had little effect on the quality and growth of fruit body. Operation of the Airocide tended to reduce the air humidity of the cultivation room and required more humidification. In humidifying conditions, the Airocide has the effect of lowering the species and density of bacteria and reducing bacterial symptoms and abnormal fruiting body of mushroom. Pseudomonas sp., the mushroom pathogen, was isolated from the cultivation room without Airocide, resulting in serious disease and loss of yields, so that only about 83% of substrate could harvest normal fruiting bodies. No disease symptom caused by bacteria and fungi in the cultivation room with Airocide. Trichoderma sp., Penicillium sp. and Cladosporium sp. were isolated from all experimental conditions, but did not inhibit fruit growth or caused diseased.
Leaf-rolling moths were collected from soybean fields and identified as Matsumuraeses falcana and Matsumuraeses phaseoli by comparison with laboratory-reared species based on the nucleotide sequence (658 bp) of the mitochondrial cytochrome c oxidase 1 subunit gene (COX1). Ten haplotypes with 0.15-0.46% genetic distance from each other in COX1 were found in 47 samples of M. falcana, in which haplotype A was dominant (approximately 70%). Only one type of COX1 was found in 30 samples of M. phaseoli, and its sequence showed 4.11-4.61% genetic distance from those of M. falcana. Amino acid sequences translated from COX1 were identical in all samples of both species, and they showed synonymous substitutions. Larvae of both species caused damage to soybean leaves and pods and co-occurred simultaneously in the field. The average density of M. falcana was 1.5 times higher than that of M. phaseoli. The results clearly indicate that soybean was the host plant for both species. In addition, Elodia flavipalpis (Diptera: Tachinidae) was found to be a larval parasitoid of Matsumuraeses sp. through identification of the COX1 gene.

Fusarium root rot is an increasingly severe problem in soybean cultivation. Although several Fusarium species have been reported to infect soybean roots in Heilongjiang province, their frequency and aggressiveness have not been systematically quantified in the region. This study aimed to investigate the diversity and distribution of Fusarium species that cause soybean root rot in Heilongjiang province over two years. A total of 485 isolates belonging to nine Fusarium species were identified, with F. oxysporum and F. solani being the most prevalent. Pot experiments were conducted to examine the relative aggressiveness of different Fusarium species on soybean roots, revealing that F. oxysporum and F. solani were the most aggressive pathogens, causing the most severe root rot symptoms. The study also assessed the susceptibility of different soybean cultivars to Fusarium root rot caused by F. oxysporum and F. solani. The results indicated that the soybean cultivar DN51 exhibited the most resistance to both pathogens, indicating that it may possess genetic traits that make it less susceptible to Fusarium root rot. These findings provide valuable insights into the diversity and distribution of Fusarium species that cause soybean root rot and could facilitate the development of effective management strategies for this disease.
Interest in commercial cultivation and product development of Cordyceps species has shown a recent increase. Due to its biochemical and pharmacological effects, Cordyceps militaris, commonly known as orange caterpillar fungus, is being investigated with great interest. Cultivation of C. militaris has been practiced on a large scale in order to fulfill a demand for scientific investigation and product development. Isolates of C. militaris can be easily established from both spores and tissue. For isolation of spores, ascospores released from mature stromata are trapped in sterile medium. Multi-ascospore isolates, as well as combinations of single ascospore strains, are used for production of fruiting bodies. Progeny ascospore strains can be isolated from artificial fruiting bodies, thus, the cycle of fruiting body production can be continued for a long period of time. In this study, we examined fruiting body production from multi-ascospore isolates and their progeny strains for three generations. $F_1$ progeny strains generally produced a larger number of fruiting bodies, compared with their mother multi-ascospore isolates; however, $F_2$ and $F_3$ progeny strains produced fewer fruiting bodies. Optimum preservation conditions could help to increase the vitality of the progeny strains. In order to retain the fruiting ability of the strains, further testing of various methods of preservation and different methods for isolation should be performed.

Since 2000, severe rots on aerial and underground parts of paprika( Capsicum annum L.) has occurred in most cultivation glasshouses throughout the country. Totally 169 isolates of a fungus were consistently isolated from the diseased plant tissues of fruits, stems, branches, and roots collected from 19 farms in six provinces. Anamorph stage of the fungus was identified as Fusarium solani based on morphological characteristics. However, the fungus readily produced sexual structure of perithecia on infected plant tissues and on agar medium. Since the fungus formed abundant perithecia by single isolate, it was considered as a homothallic strain of Nectria hematococn, the teleomorph of F. solani. Irregularly globose perithecia with orange to red color formed sparsely to gregariously on dead tissues of fruits and basal stems at the 3ate infection stage, which is a diagnostic sign for the disease. Abundancy of perithecium varied among isolates and they sized 125-220$\mu\textrm{m}$ in diam. Asci enveloping eight ascospores were cylindrical and measured 60-80x8-12$\mu\textrm{m}$. Ellipsoid to obovate ascospores are two-celled and measured 11-l8${\times}$4-7$\mu\textrm{m}$. Ascospores are hyaline, slightly constricted at the central septum, and revealed longitudinal striations that is a typical trait of the species. This fungus that has never been reported in Korea previously became a threat to paprika cultivation because of its strong pathogenicity and nationwide distribution.
Environmental risk assessment of living modified (LM) crops is essential for their cultivation. In this study, we cultivated β-carotene enhanced transgenic soybean (LM soybean) and non-LM soybean (Gwangan) in living modified organism (LMO) isolated fields, and investigated changes in the insect fauna using three types of collection methods for three years. In total, 331,483 individual insects and arachnids, representing 82 families in 14 orders, were captured during the study. Totals of 166,518 and 164,965 individual insects and arachnids were collected from LM soybean and Gwangan, respectively. Throughout the study, although there were differences between the investigation year, region, and methods, there were no significant differences between the population densities of insect pests, natural enemies, and other insects on LM soybean and non-LM soybean. Also, there were no statistically significant differences between varieties in the results of the species diversity analysis. The data on insect species population densities were subjected to multidimensional scaling (MDS), which did not distinguish between the two varieties, LM soybean and the non-LM soybean, in all cultivated fields. However, the results of the MDS analysis were completely divided into six groups based on the yearly survey areas. These results provided the insect diversity for an environmental risk assessment of LM soybean and suggested that the guideline could be useful to detect LM crops.
