Prediction of the Pollutant Loading into Estuary Lake according to Non-cultivation and Cultivation conditions of Reclaimed Tidal Land (담수호 유입 오염부하량의 간척농지 영농 전.후 변화 예측)

  • Estimation of current and future loading from watershed is necessary for the sound management of water quality of an estuary lake. Pollution sources of point and non-point source pollution were surveyed and Identified for the Koheung watershed. Unit factor method was used to estimate potential pollutant load from the watershed of current conditions. Flow rate and water qualify of base flow and storm-runoff were monitored in the main streams of the watershed. Estimation of runoff pollutant loading from the watershed into the lake in current conditions was conducted by GWLF model after calibration using observed data. Prospective pollutant loading from the reclaimed paddy fields under cultivation conditions was estimated using the modified CREAMS model. As a result, changes of pollutant loading into estuary lake according to non-cultivation and cultivation conditions of reclaimed tidal land were estimated.

Optimum Condition for Pigment Production and Antioxidative Activity of the Products by Bacillus subtilis DC-2 with Response Surface Methodology (반응표면 분석에 의한 Bacillus subtilis DC-2의 색소생성 및 그 생성물의 항산화성에 대한 최적조건)

  • The conditions for color intensity and electron donating ability to $\alpha$,$\alpha$-diphenyl-$\beta$- picryl-hydrazyl (DPPH) of Bacillus subtilis DC-2 were investigated. Temperature, pH and cultivation time were chosen as three factors, and the optimal conditions of color intensity and DPPH was determined with response surface methodology. Color intensity was affected by cultivation temperature(p<0.1). DPPH was influenced by cultivation temperature(p<0.05) and pH(p<0.1). But cultivation time was affected neither color in- tensity nor DPPH. Optimal conditions of color intensity with Bacillus subtilis DC-2 were appeared at cultivation temperature of 39.$25^{\circ}C$, pH 8.83 and cultivation time of 84.41hrs. Optimal conditions of DPPH with Bacillus subtilis DC-2 were revealed at cultivation temperature of 39.19$^{\circ}C$, pH 8.84 and cultivation time of 82.21hrs.

Comparison of Cultivation, Mushroom Yield, and Fruiting Body Characteristics of Lentinula edodes Strains according to the Inoculation Method

  • The cultivation in sawdust media, mushroom productivity, and fruiting body characteristics of Lentinula edodes strains NIFoS 2778 and NIFoS 3363 were compared according to the inoculation conditions. The cultivation period was 5% shorter when liquid spawn was used. Fruiting bodies were induced after 113 days of incubation on media inoculated with liquid spawn, and the cultivation period was 119 days on media inoculated with solid spawn. Mushroom productivity of NIFoS 2778 was the highest (661.4 g) when 36 mL of liquid spawn was used. For NIFoS 3363, mushroom production was higher under liquid inoculation conditions when the same amount of liquid and solid spawns were used. The mushroom characteristics of the two strains were not significantly different, except for gill width and stipe diameter.

Comparison of Plant Growth and Glucosinolates of Chinese Cabbage and Kale Crops under Three Cultivation Conditions

  • Purpose: The objective of this study is to evaluate the effect of cultivation conditions on the growth and glucosinolate content of Chinese cabbage and kale. Methods: Chinese cabbage and kale were grown in three different cultivation conditions, including a plant factory, greenhouse, and open field. Samples were collected at two harvesting times (10 d and 20 d after transplanting the seedlings). Nine growth parameters (plant height, plant width, number of leaves, petiole diameter, SPAD readout, leaf length, leaf width, stem diameter, and plant weight) were measured immediately after harvesting, and the samples were freeze-dried and stored until the glucosinolate content was analyzed. Mean values of the growth parameters and glucosinolate contents were evaluated using Duncan's multiple range tests. Results: The results indicated that the plant parameters of the Chinese cabbage and kale were greater for plants grown in the plant factory and greenhouse. The plant height, width, and weight showed significant differences in the Duncan's multiple range tests at a 5% level. The plant factory also produced greater contents of most of the glucosinolates. Conclusions: Three different cultivation conditions significantly affected the growth and glucosinolate contents of Chinese cabbage and kale. Further study is necessary to investigate other functional components and different vegetable varieties.

Optimization of Cultivation and Extraction Conditions of Pupae-Cordyceps for Cordycepin Production

  • Cordycepin is a characteristic bioactive compound of Cordyceps militaris with various beneficial effects. Cordyceps grows on both grains and insects, and the content of cordycepin varies depending on the cultivation conditions. In this study, the effect of culture conditions on the cordycepin content was analyzed and the extraction conditions were optimized. Analysis of cordycepin content in Pupae-Cordyceps found that it was highly affected by temperature in culture conditions. In the case of mycelium, it grows well at 20 and 25 ℃, but not at 30 ℃. However, the content of cordycepin was highest at 30℃ and less at 20 ℃. The fruiting body also showed a similar tendency: growth was 20 ℃ > 25 ℃ > 30 ℃, but the cordycepin content was 30 ℃ > 25 ℃ > 20 ℃. The content of cordycepin decreased after the fruiting bodies were produced. Next, extraction conditions such as solvent and time were optimized for maximum cordycepin content using response surface methodology (RSM). There was a large difference in the content of cordycepin according to the content of ethanol and the extraction temperature. Through RSM, it was confirmed that the optimum condition for extraction of cordycepin was 48.9 ℃ using 49.0% ethanol, and 160.9 mg/g extract could be obtained under this condition. In conclusion, this study suggested the optimized conditions for the cultivation and extraction of Pupae-Cordyceps for maximizing the content of cordycepin, and this may be applied to the discovery of materials using cordycepin.

Effect of Air Cleaner on the Occurrence of Mushroom Disease During Cultivation of Pleurotus eryngii (공기정화장치가 큰느타리버섯 병 발생에 미치는 영향)

  • King oyster mushroom(Pleurotus eryngii) is one of the most commercially important mushrooms in Korea. Development of fruit body and disease occurrence are sensitive to environmental conditions such as temperature, humidity, carbon dioxide(CO2) concentration. The purpose of this study was to investigate the changes in the growth environment of king oyster mushroom by installing Airocide, an air purifier for the purpose of improving mushroom cultivation environment. The results of the environment conditions, identification of pathogenic organisms and pathogenesis during the cultivation were as follows. Airocide operation increased the CO2 concentration of the cultivation room by more than 400 ppm on average, but the increase of CO2 concentration at this level had little effect on the quality and growth of fruit body. Operation of the Airocide tended to reduce the air humidity of the cultivation room and required more humidification. In humidifying conditions, the Airocide has the effect of lowering the species and density of bacteria and reducing bacterial symptoms and abnormal fruiting body of mushroom. Pseudomonas sp., the mushroom pathogen, was isolated from the cultivation room without Airocide, resulting in serious disease and loss of yields, so that only about 83% of substrate could harvest normal fruiting bodies. No disease symptom caused by bacteria and fungi in the cultivation room with Airocide. Trichoderma sp., Penicillium sp. and Cladosporium sp. were isolated from all experimental conditions, but did not inhibit fruit growth or caused diseased.

Change of Major Functional Components of Gastrodia elata Blume with Cultivation Conditions and Harvest Times (재배조건과 수확시기에 따른 천마의 기능성 물질함량 변화)

  • Gastrodia elata Blume is a achlorophyllous orchid plant that has been used in traditional medicine to treat many human illnesses, such as vertigo, hemiplegia, and epilepsy. It contains three major pharmacological substances; gastrodin, ergothioneine, and vanillyl alcohol. Here we investigated the quantitative changes in those functional components by cultivation conditions and harvest times. The content of ergothioneine was different with cultivation areas, while gastrodin accumulated the higher levels than vanillyl alcohol regardless of cultivation areas. The smaller tubers contained the higher concentrations of gastrodin and the lower concentrations of ergothioneine than did the larger tubers. The cultivation under sun shade screen increased the concentrations of both gastrodin and vanillyl alcohol but decreased the levels of ergothioneine. We also determined the contents of functional substances depending on the harvest times. The concentration of gastrodin was higher in October than in April, while the level of ergothioneine was higher in April than in October. Interestingly, the use of sexually propagated seed tubers produced 1.9 times higher levels of gastrodin compared to vegetatively propagated seed tubers. In this study, we found that three major functional substances in G. elata tubers altered depending on various cultivation practices, which might be important to increase the economic values of G. elata tubers.

Effect of the Cultivation Technology on the Yield of Paddy Straw Mushroom (Volvariella volvacea)

  • The edible paddy straw mushroom Volvariella volvacea (Bull. ex F.) Singer has high nutritional and medicinal values. They are grown on a wide variety of agricultural by-products using different several methods. The result of this present study showed that type of bed and cultivation condition (outdoor/indoor) had effects on the yield of paddy straw mushrooms grown on the spent oyster mushroom sawdust. The treatment of circular compact bed under indoor cultivation condition (CYIC) had the best yield and biological efficiency (B.E.) with 2,119.2 g/bed and 14.5%, respectively, followed by the treatment of conventional bed under outdoor cultivation condition (COOC) with the yield and B.E. of 1,935.5 g/bed and 13.2%, respectively, but the lowest yield and B.E. were observed in the treatment of conventional bed under indoor cultivation condition (COIC) with 1,226.1 g/bed and 8.4%, respectively. Paddy straw mushrooms cultivated on spent oyster mushroom sawdust should be proper in both outdoor and indoor conditions by conventional and circular compact methods as well. However, in indoor conditions, paddy straw mushrooms should be grown in a circular compact bed to ensure the temperature of the bed is suitable for the mushroom growth.

  • Culture conductivity and on-line NADH fluorescence were used to measure cellular growth in plant cell suspension cultures of Podophyllum hexandrum. An inverse correlation between dry cell weight and medium conductivity was observed during shake flask cultivation. A linear relationship between dry cell weight and culture NADH fluorescence was obtained during the exponential phase of batch cultivation In a bioreactor under the pH stat (pH 6) conditions. It was observed that conductivity measurement were suitable for biomass characterisation under highly dynamic uncontrolled shake flask cultivation conditions. However, if the acid/alkali feeding is done for pH control the conductivity measurement could not be applied. On the other hand the NADH fluorescence measurement allowed online-in situ biomass monitoring of rather heterogenous plant cell suspension cultures in bioreactor even under the most desirable pH stat conditions.

  • The optimum conditions of the production of exo-polymer by Ganoderma lucidum WK-003 and it's hepatoprotective effect was studied. Optimum conditions for the production of exo-polymer (3.18 g/$\ell$) by using shaken flask culture of G. lucidum WK-003 were pH 4.5, 30$^{\circ}C$, 120 rpm for 18 days cultivation. Also exo-polymer productio (7.15 g/$\ell$) was optimized by 5$\ell$ jar fermenter cultivation with condition of pH 4.5, 30$^{\circ}C$, 200 rpm, 1.0 vvm for 6 days cultivation. Glutamic pyruvic transaminase(GPT) activities in serum of intoxicated Sprague-Dawley rat were decreased from 704 IU/L to 330 IU/L by oral administration of the exo-polymer (20mg/kg$.$day)f or 4 consecutive days.
