Metabolic profiles of Wolfiporia cocos mycelia cultivated under light and dark conditions

  • Wolfiporia cocos is an edible fungus commercially cultivated in Asia. To investigate metabolic changes of W. cocos mycelia under both light and dark culture conditions, gas chromatography mass spectrometry (GC-MS) and high-performance liquid chromatography (HPLC) analyses were performed. In terms of the total amount of sugars, alcohols, amino acids, organic acids, fatty acids, and purines, there no significant differences between the W. cocos mycelia cultivated under light (L) or dark (D) conditions (p < 0.05). However, there were some differences with respect to the production of particular sugars and proteins. The levels of trehalose (L: 17.2 ± 0.3% vs. D: 13.9 ± 1.6%), maltose (L: 0.9 ± 0.1% vs. D: 0.3 ± 0.1%), turanose (L: 0.7 ± 0.2% vs. D: 0.1 ± 0.1%), glutamine (L: 1.6 ± 0.3% vs. D: 0.7 ± 0.2%), and proline (L: 0.3 ± 0% vs. D: 0.1 ± 0%) were all significantly higher under light condition (p < 0.05). In contrast, the levels of galactose (L: 13.7 ± 1.2% vs. D: 17.6 ± 2.0%), aspartic acid (L: 0.6 ± 0.1 % vs. D: 0.9 ± 0.1%), cystathionine (L: 0.6 ± 0.1% vs. D: 0.8 ± 0 %), and malic acid (L: 0.7 ± 0.1% vs. D: 1.2 ± 0.1%) were higher under the dark condition. It is worth noting that the amount of pachymic acid, a pharmaceutically active compound of W. cocos, was 1.68 times greater under the light condition (p < 0.05).

농산물 생산이력제 도입 농가실태와 소비자 인지도 (The Adoption of Traceability Systems by Farmers and Its Consumers' Recognition)

  • The main goal of this study is to survey the actual condition of farm household adopt traceability of agricultural products and the consumers' recognition of the traceability. Thirty six farm household adopted traceability of agricultural products and one hundred twenty three consumers were surveyed for this study. A total of the cultivated area of surveyed farm household was 39.6ha-owned land(21.2ha) and Rented land(18.4ha)-and the cultivated area of crops with the traceability of agricultural product was 15.7ha, consisting of 39.6% of the whole cultivated area. 22.2% of agricultural traceability products was cultivated bychemical method and 77.8% of the them by environment-friendly agricultural methods-organic cultivation accounts for 2.8%, no-chemicals cultivation 47.2% and low-chemicals cultivation 27.8%. As a result of the consumer survey, 75.6% of respondents recognized agricultural product traceability and only 29.0% of them had experience to purchase traceability products. But 61.0% of surveyed people had intention to purchase traceability products in the future. It was found that consumers wanted to know about production traceability information of farm products in order to identify quality certification including environment-friendly certification, product quality such as taste, weight, grade, and the use of insecticides and fertilizers regarding use frequency and kinds of chemicals.

Establishment of Sesame Cultivation Practices as Succeeding Cropping System of Watermelon in the Greenhouse Condition

  • This experiment was conducted to develop optimum sesame cultivation practices as succeeding cropping system of watermelon in the greenhouse condition. We also compared major components in sesame seed cultivated in the greenhouse and open field condition. The adaptable varieties under the greenhouse condition were white sesame 'Pyoungan' and black sesame 'Jinki' which showed higher yield and disease resistance. We also conducted several experiments to determine optimum sowing date, planting density and pinching time. Grain yields were statistically different according to the several sowing dates. In general, sesame sowing on June 30th showed higher grain yields than July 10th, July 20th and July 30th in the greenhouse. We also found out sesame cultivation practice with the row spacing of 40 cm and interplant spacing of 30 cm (a few branch type) or 40 cm (many branch type) showed higher yield than other treatments. Optimum pinching time was 25 days after first flowering in main stem when thousand seed weight and grain yield were highest. In the comparison of major components of sesame at the different cultivation conditions, sesame seeds cultivated in the greenhouse contained 4% much more oil content than open field condition. The lignan compounds, sesamin and sesamolin were also higher by 6% in the greenhouse than open field condition. It was concluded that sesame cultivation practices as succeeding cropping system of watermelon in the greenhouse condition guaranteed higher grain yield with less labor input which is now emerging alternative farming practice system in present aging rural society and will also give sesame cultivation farmers to increase net income in Korea.

재배조건, 수확시기 및 열처리에 따른 갯기름나물의 영양성분 변화 (Nutritional quality of Peucedanum japonicum Thunb. leaves in relation to ripening time, growing condition and blanching)

  • 본 연구에서는 재배조건(하우스재배, 노지재배), 수확시기(어린잎, 성잎) 열처리에 따른 영양성분 및 항산화활성의 변화를 비교 분석하여 국가표준성분표 발간을 위한 기초자료로 활용하고자 하였다. 일반성분 4종(수분, 지질, 회분, 단백질), 무기질 5종(칼슘, 철, 마그네슘, 칼륨, 나트륨), 비타민 3종(비타민 C, 엽산, 베타카로틴)의 영양성분을 분석하였고 폴리페놀 화합물과 DPPH 라디칼 제거능을 각각 측정하였다. 연구결과 일반성분의 경우 시료 간에 유의적인 차이를 나타내지 않았으나 무기질의 경우 Ca, Mg, Na은 하우스 재배가 노지재배 보다 유의적으로 높은 함량을 나타내었다. 비타민함량과 폴리페놀 함량 및 항산화 활성의 경우 수확시기보다는 재배방법과 열처리에 의해 함량변화가 큰 것으로 나타났다. 하우스보다 노지 재배된 갯기름나물의 비타민 C, 엽산, 폴리페놀 화합물과 항산화활성이 증가하였으며 열처리할 경우 비타민 C, 베타카로틴, 폴리페놀 화합물, 항산화활성이 각각 증가하였다. 비타민 C의 기존 보고된 이론과 상이한 결과가 도출되어 다양한 열처리온도와 시간을 적용한 성분 변화 연구가 필요할 것으로 생각된다. 베타카로틴과 폴리페놀 화합물의 열처리에 의한 증가는 산화가수분해 효소의 불활성화와 세포조직의 견고성 저하로 인한 활성물질의 추출 효율증가로 설명될 수 있다. 따라서 나물을 데쳐서 섭취할 경우 미생물 오염방지 등 안전성을 확보할 수 있을 뿐만 아니라 나물 세포조직이 연해져 베타카로틴과 같은 우리 몸에 유익한 성분의 소화흡수율이 증가될 것으로 생각된다.

한국, 일본에서 수집한 들깨와 차조기의 재배형 및 잡초형들의 종자발아 변이 (Variation of Seed Germination Among Cultivated and Weedy Types of Perilla Crop in Korea and Japan)

  • 본 연구는 우리나라와 일본에서 재배 및 이용하고 있는 들깨, 차조기 그리고 이들 잡초형 69 계통들에 대하여 종자 특성 및 종자발아 변이에 대한 분석결과는 다음과 같다. 1. 재배형 들깨에서 측정된 100립중은 0.177 g에서부터 0.402 g까지의 범위였고, 종자경도는 대부분의 계통들이 잘 부스러지는 부드러운 특성을 지니고 있었으나, 일부 계통들은 잘 부스러지지 않는 딱딱한 특성을 지니고 있었다. 반면에 잡초형 들깨에서 측정된 100립중은 0.045 g에서부터 0.172 g까지의 범위였고, 종자경도는 모든 계통들이 잘 부스러지지 않는 딱딱한 특성을 지니고 있었다. 2. 재배형 차조기에서 측정된 100립중은 0.054 g에서부터 0.101 g까지의 범위였고, 잡초형 차조기에서 측정된 100립중은 0.059 g에서부터 0.135 g까지의 범위였으며, 그리고 이들의 종자경도는 모든 계통들이 잘 부스러지지 않는 딱딱한 특성을 지니고 있었다. 3. 실온 및 $28^{\circ}C$ 항온조건의 발아율 조사에서 수확 후 1개월째에서는 들깨의 잡초형 그리고 차조기의 재배형 및 잡초형 계통들은 대부분이 50% 이하의 낮은 발아율을 나타내었으나, 재배형 들깨의 일부 계통들이 50% 이상의 발아율을 나타내었다. 그리고 2개월 및 3개월이 경과한 상태에서 들깨의 재배형 및 잡초형 그리고 차조기의 재배형 및 잡초형 계통들은 50% 이상의 발아율을 나타내는 계통들이 많이 나타났다. 4. 들깨와 차조기 작물의 재배형 및 잡초형 계통들에서의 평균발아율 및 평균발아세는 수확 후 1개월째에서 가장 낮고 수확 후 3개월째에서 가장 높은 것으로 나타났으며, 또한 실온 및 $28^{\circ}C$ 항온조건에서의 발아율과 발아세는 $28^{\circ}C$ 항온조건 보다는 실온조건에서 비교적 높은 것으로 나타났다.

Antioxidant and ACE Inhibitory Activity of Cultivated and Wild Angelica gigas Nakai Extracts Prepared Using Different Extraction Conditions

  • The purpose of this study was to investigate the biological activities of cultivated Angelica gigas Nakai (CAG) and wild Angelica gigas Nakai (WAG) extracts prepared by extraction with water, 30% ethanol, 60% ethanol, or 90% ethanol. The electron donating ability of the WAG extracts was higher than that of the CAG extracts and 0.1% and 1.0% solutions of the comparative substance, L-ascorbic acid. The superoxide dismutase-like activity of the CAG extracts was higher than that of WAG extracts. Superoxide dismutase-like activity was highest (33.95%) in the CAG water extract. The total polyphenol content was highest in the 60% ethanol extracts of WAG. The nitrite scavenging ability of the CAG and WAG extracts was highest at a pH of 1.2. The tyrosinase inhibitory effect was highest (43.72%) in the water extract of WAG. The angiotensin converting enzyme inhibitory activity was highest (83.84%) in the 60% ethanol extract of WAG. The results of the present study will be useful for understanding the antioxidant and angiotensin-converting enzyme inhibitory activities of Angelica gigas Nakai extracts.

HPLC-CAD에 의한 산양삼의 극성 및 비극성 ginsenoside 동시 분석 (Simultaneous Analysis Method for Polar and Non-polar Ginsenosides in Cultivated Wild Ginseng by Reversed-phase HPLC-CAD)

  • 산양삼은 식품 또는 약초로써 많이 사용되고 있다. 이번 연구의 목표는 high performance liquid chromatography (HPLC) - charged aerosol detection (CAD)에 의해 산양삼의 추출물에 사포닌들의 분석을 최적화 하는 것이다. CAD는 물리적인 특성은 물론이고 입자크기, 스펙트럼 특성과는 무관하게 비휘발성 물질도 분석할 수 있다. 산양삼 추출물 중 Gensenoside Rb1, Rd, Rg1, Rf, Re 및 Rh1을 분석하였다. HPLC-CAD분석을 위한 사포닌 추출물은 건조 산양삼 10 g에 95% 에탄올 100 ml을 넣어 80℃에서 24시간 동안 추출하여 사포닌을 얻었다. 6개의 주요한 사포닌을 정량화하였을 경우 값은 다음과 같다. 사포닌들의 함량은 Rb1 (5.48±0.12 mg/g), Rd (5.33±0.14 mg/g), Rg1 (12.80±0.05 mg/g), Rf (19.08±0.68 mg/g), Re (19.87± 0.05 mg/g), 및 Rh1 (16.47±0.16 mg/g)로 확인하였다. 대체적으로 산양삼 추출물의 사포닌은 protopanaxatriol (Rg1, Rf, Re, Rh1)의 값이 protopanaxadiol (Rb1, Rd)보다 높게 확인 되었다. 이번 연구에서는 산양삼 사포닌의 분석조건을 HPLC-CAD를 이용하여 최적화하였고 제품화를 위해 약리활성에 관한 연구도 매우 중요할 것이다.

Effect of continuous maize cultivation on soil condition and yield in Northern Laos

  • In Northern Laos, maize is cultivated in continuous cropping without fertilizer, fallowing nor any other soil conservation practice. It is expected that this inadequate management in maize cultivation will degrade soil and decrease yield. However, there is limited information about the change of soil condition and yield under continuous maize cultivation. We tried to evaluate the change of soil condition and yield under continuous maize cultivation in Northern Laos. Our study was conducted in farmer's flat and slope fields in Sainyabuli province where maize cultivation had been introduced earlier than the other provinces of Northern Laos. Our study was conducted in 21 fields in 2014, and in 19 fields in 2015. We analyzed grain yield and soil characteristic (total carbon (TC), total nitrogen (TN), available phosphorus (Av-P), exchangeable cation, pH, soil texture) at 3 places in each field. The 3 places were set at different elevation level (lower position, middle position, upper position) in slope fields. Further, the period of continuous maize cultivation and crop management practice were investigated. Then, by evaluating the relation between the period of continuous maize cultivation and yield and the soil characteristics, the effect of maize cultivation was estimated. Crop management practices were similar among the investigated fields. Maize was cultivated in rain season. Grain seed and cob were harvested on September or October, but shoot was left on the fields. No crop was cultivated during dry season. Fertilization and fallowing has never been conducted under continuous maize cultivation. On the other hand, the period of maize cultivation was different among the fields, and ranged from 2 years to 30 years. In the slope fields, as the period of continuous maize cultivation was longer, the contents of TC and TN were lower at all 3 positions, Av-P content was lower at the upper position, exchangeable potassium (Ex-K) content was lower at the middle and the upper positions. The other soil characteristics weren't related with the period of maize cultivation in the slope fields. In contrast, soil characteristics weren't related with the period of maize cultivation in the flat fields. Yield was lower as the period of maize cultivation was longer at the upper position of the slope fields. At middle position of slope fields, yield tended to be low with increase of the period of maize cultivation. In contrast, yield wasn't related with the period of continuous maize cultivation in flat fields. From the results about crop management, it was presumed that the period of maize cultivation was one of the main factors causing the difference of yield and soil characteristics among the fields. Therefore, from the result of yield and soil condition, it was considered that the continuous maize cultivation decreased soil productivity in the slope fields with decline of TC, TN, Av-P, Ex-K and yield at upper position of slope fields, and decline of TC and TN in the other positions in Sainyabuli province.

고추냉이 밭재배시 함황비료 처리에 따른 생육 및 Allylisothiocyanate 함량 (Effects of Sulfuric Fertilizers on Growth and Allylisothiocyanate Contents of Wasabia japonica $M_{ATSUM}$ Cultivated in Heating Condition)

  • 고추냉이 동계가온 밭재배시 allylNCS함량을 증대시키기 위하여 allylNCS함량과 밀접한 관계가 있는 것으로 알려져 있는 $SO_4^{2-}$을 함유한 함황비료를 토양과 엽면에 시비하여 생육특성, 식물체 총 황함량 및 allylNCS함량과의 관계를 조사한 결과는 다음과 같다. 생육 정체기인 월동기간동안의 가온재배로 평균기온 $14{\sim}16^{\circ}C$, 지온 $13{\sim}16^{\circ}C$로 적정 생육온도의 범위를 유지할 수 있었으며, 함황비료의 토양시용으로 토양중의 황함량은 $82.6{\sim}127.4%$까지 증가되었고 토양 pH는 황분말에서 최저 2.8까지 저하되었다. 함황비료의 처리가 지상부 및 지하부의 생육에 미치는 영향에서는 함황비료 처리로 본엽수는 감소되고 측엽수는 증가하는 경향이였으며, 근중은 황산가리 엽면시비구가 가장 양호하였고 근경중등에서는 처리간 차이가 없었다. 식물체내 총황함량은 엽면시비구 보다는 토양시용구에서 높은 경향이였고, allylNCS함량은 황분말 토양시용구에서 지상부 194%, 근경 23%의 증대효과가 있었다. 지상부의 allylNCS함량은 지상부, 근, 근경의 총 황함량과 유의한 정의 상관을 보였으며, 근경의 allylNCS함량은 지상부의 총 황함량, allylNCS함량 및 근경의 총 황함량과 유의한 정의 상관을 보였으나 분주수와는 유의한 부의 상관을 나타냈다.

Mycoplasma pneumoniae의 성장과 형태에 미치는 환경요소적 영향 (Effects of environmental factors on growth and morphology of mycoplasma pneumoniae)

  • Mycoplasma pneumoniae was examined for growth characteristics and morphology when cultivated in several media supplemented with a variety of sera and under different atmospheric conditions. different formula of the medium as well as different sources of lot numbers of the serum in the same medium exhibited varying effects on growth rate and adherence. When the organisms were cultivated in SSR-2 medium in a normal atmospheric environment or under a facultative anaerobic condition provided with carbon dioxide, they developed filamentous cells with heavy growth, whereas mainly round-shaped cells were produced under strict anaerobic conditions of hydrogen and carbon dioxide. Both morphologies of the organism were transformed by switching the incubation environments. An inverted phasecontrast microscopy using modified petri dishes was excellent to observe single cells and useful to follow the development of the cells. Growth, turbidity, and pneumoniae colonies developed on a solid medium and produced clear when overlaid with sheep blood agar.
