한국산 사과식초에서 분리한 다당의 면역 및 항전이 활성 (Immunomodulatory and anti-metastatic activities of a crude polysaccharide isolated from Korean apple vinegar)

  • 전통발효식초에 속하는 사과식초로부터 에탄올 및 증류수를 이용, 다당체를 분리하여 새로운 면역활성 물질을 확인하고자 하였다. 사과식초를 농축하고, 80% 에탄올을 가하여 침전물을 얻었으며 소량에 물에 용해하여 투석 및 동결건조를 거쳐 조다당 KAV-0를 얻었다. 구성당 분석 결과, KAV-0는 주로 mannose 38.2%, galactose 19.1% 및 glucose 14.3%로 구성되어있음을 확인하였고, 정상세포주(RAW 264.7 세포 및 대식세포) 및 종양세포주(B16-BL6 melanoma 세포)에 대해 세포독성 측정 결과, 직접적인 세포독성을 나타내지 않았다. 대식세포 유래 세포주인 RAW 264.7 세포 자극에 대한 사이토카인 및 산화질소 생산능을 측정한 결과, IL-6, $TNF-{\alpha}$ 및 산화질소 모두 농도 의존적으로 분비능이 증가하고 저농도에서도 우수한 활성을 나타냈으며, 마우스 복강유래 대식세포 자극에 대한 사이토카인 및 산화질소 생산능에서도 동일한 양상의 결과를 보여 사과식초 유래 조다당 KAV-0가 높은 대식세포 활성능을 소유함을 확인할 수 있었다. 한편, KAV-0의 정맥투여에 따른 NK 세포 활성화를 측정한 결과(ex vivo), 시료에 의해 활성화된 NK 세포는 Yac-1 종양 세포주에 대한 치사활성을 농도 의존적으로 증진시켰으며, $1,000{\mu}g/mouse$에서 월등히 우수한 NK 세포 활성능을 유도하였다. 또한, B16-BL6 melanoma를 이용한 폐암전이모델에서 KAV-0를 정맥투여한 뒤 항전이 활성을 측정한 결과, 농도 의존적으로 암 저해 효과가 나타났으며, 특히 $1,000{\mu}g/mouse$에서는 약 53%의 우수한 암전이 억제 활성을 나타냈다. 이상의 결과로부터 사과식초 유래 조다당 KAV-0는 인체의 면역계를 자극하여 우수한 면역활성 및 항전이 효과를 나타낼 수 있음이 확인되었고 건강 기능성 소재로의 활용 가능성이 충분할 것으로 최종 판단되었다.

Monitoring of Chemical Changes in Explosively Puffed Ginsengvand the Optimization of Puffing Conditions

  • Response surface methodology was used to predict the optimum conditions of explosive puffing process for ginseng. A central composite design was used to monitor the effect of moisture content and puffing pressure on dependent variables such as functional compounds (extract yield, crude saponin, acidic polysaccharide, and total phenolic content) and sensory properties. Correlation coefficients $(R^2)$ of models for crude saponin, acidic polysaccharide, and total phenolic content were 0.9176 (p<0.05), 0.9494 (p<0.05), and 0.9878 (p<0.001), respectively. Functional compounds increased with decreasing moisture content and increasing puffing pressure. Overall palatability was high at 15-20% moisture content and 98-294 kPa of puffing pressure. On the basis of superimposed contour maps for functional compounds and overall palatability of puffed ginseng, the optimum ranges of puffing conditions were 10-17% moisture content and 294-392 kPa puffing pressure.

세포주를 이용한 군소 다당류와 Glycosaminoglycan의 기능성 검색 (Screen of Functional Activity of Polysaccharide and Glycosaminoglycan from Sea Hare (Aplysia kurodai) by Cell Line)

  • 군소에서 정제한 GAG는 대식세포주인 RAW 264.7에서 세포독성은 보이지 않았으며 NO 생성량, TNF-$\alpha$, IL-6 생성량을 증가시켜 면역조절 효과가 있음을 확인하였다. 그리고 $100{\mu}g$/mL 농도에서 배양 72시간 후 약 40%에 가까운 위암세포(AGS) 성장 억제 효과가 나타났고, 정상세포 IEC-6에는 독성을 보이지 않았다. 이 같은 결과는 면역조절을 위한 기능성식품 혹은 위암 치료제로서 군소의 당 추출물과 GAG의 활용 가능성을 제시한다.

  • The purpose of this study was to investigate extract from mixed culture with Trichloloma matsutake mycelium in oriental medicine and cereal medium(OCM) to develop new material for pharmaceutical products and medicinal food. To evaluate of physiological activity of OCM extracts, we examined ${\beta}$-glucan contents, SOD-like activity, nitric oxide production and cytotoxicity by MTT assay. ${\beta}$-glucan contents was found to be 52.85% for crude polysaccharide of hot water extracts of OCM(HEE) and 49.74% for crude polysaccharide of ultra sonic waves, micro waves, and micro bubble extracts of OCM (UEE). SOD like activity was showed UE 74.66%, HE 67.16%, UEE 31.34%, HEE 26.10%, respectively. NO production of UEE and HEE, at LPS 1 ug/mL, 1 mg/mL UEE showed 66.62 uM and HEE 45.68 uM. at LPS 10 ug/mL, 1 mg/mL UEE showed 63.91 uM and HEE 51.74 uM. The inhibitory effect against HT1080 was increased dose-dependently in UEE.

시판 인스턴트 커피에서 추출한 다당류의 화학적 특성 및 면역활성 (Chemical Characteristics and Immunostimulating Activity of Crude Polysaccharide Isolated from Commercial Instant Coffee)

  • 국내 시판중인 인스턴트 커피에서 다당류를 추출하여 새로운 면역 활성 물질을 확인하고자 하였다. 시판중인 인스턴트 커피를 구입하여 냉수추출 후, 에탄올 침전과 투석을 통하여 약 24%의 높은 수율로 다당류(ICP-0)를 얻을 수 있었다. ICP-0은 중성당 89.9%, 산성당 10.1%로 이루어진 다당체로써, 구성 성분으로는 갈락토스 55.5%, 만노스 25.7%와 소량의 아라비노스, 람노스 등으로 이루어진 것을 확인하였으며, 갈락토만난(galactomannan)이나 팩틴 다당류(pectic polysaccharide)가 혼합된 구조일 것이라고 추정되었다. 인스턴트 커피 유래 ICP-0에 대하여 대식세포 자극에 대한 세포증식능을 확인한 결과, NC 대비 유의적으로 높은 증식활성을 보였으며, IL-6, IL-12 및 TNF-${\alpha}$의 경우, $10{\mu}g/mL$에서도 NC 대비 우수한 사이토카인 분비 유도능을 보임이 확인되었다. 또한 NO, ROS를 분석한 결과에서도 NO의 경우 $10{\mu}g/mL$ 이상에서 NC 대비 양호한 활성을 보여 주었다. ICP-0에 대하여 2차 면역기관 중 하나인 비장세포상의 증식능을 확인한 결과, NC 대비 115% 정도 증식하는 것이 확인되어 면역세포를 직접 증진시키는 마이토겐 활성이 있음을 나타내었다. 인스턴트 커피 유래 ICP-0가 장관 내 분포되어 있는 Peyer's patch를 경유하여 장관면역에 공헌할 수 있는지 여부를 확인한 결과, NC 대비 유의적으로 골수세포의 증식능을 확인하였으며 GM-CSF와 같은 관련 사이토카인의 분비 유도가 확인되었다. 이상의 결과는 시판 인스턴트 커피로부터 분리한 다당류가 대식세포의 기능과 장관면역계를 증진시킬 수 있는 가능성을 제시한다.

참취에서 분리한 다당의 면역자극 활성 (Immunostimulating Activites of Polysaccharide Fractions isolated from Aster scaber Thunb.)

  • ASW0 is a polysaccharide derived from the perennial herb Aster scaber Thunberg. We isolated ASW0, a fraction of crude polysaccharide, by means of ethanol precipitation and dialysis after hot water extraction to investigate its physicochemical properties and immunostimulatory effects. ASW0 contains neutral sugar (45.7%), acidic sugar (51.6%), protein (2.3%), and 2-keto-3-deoxy-D-manno-octonate (KDO) (0.4%). The neutral sugar in ASW0 (in mole percentage) was mainly composed of arabinose (34.5 mol%), glucose (31.1 mol%), galactose (14.9 mol%), and rhamnose (8.1 mol%), which are characteristic of pectic polysaccharides. ASW0 also contained small amounts of xylose, mannose, and fucose. The anti-complementary activity of ASW-0 was similar to that of polysaccharide K (used as positive control). ASW0 exhibited no cytotoxicity in RAW 264.7 macrophages and dramatically increased nitric oxide (NO) production in a dose dependent manner ($0.3{\sim}30{\mu}g/mL$). Also, macrophages stimulated with ASW0 showed enhanced production of immunostimulatory cytokines such as interleukin-6 (IL-6) and tumor necrosis factor alpha ($TNF-{\alpha}$) in a dose dependent manner. These results suggest that the ASW0 have a potent immunostimulatory effect and can be used as a natural immune health ingredient.

Study of Macrophage Activation and Structural Characteristics of Purified Polysaccharide from the Fruiting Body of Cordyceps militaris

  • Cordyceps militaris, an entomopathogenic fungus belonging to the class Ascomycetes, has been reported to have beneficial biological activities such as hypoglycemic, anti-inflammatory, antitumor, antimetastatic, hypolipidemic, immunomodulatory, and antioxidant effects. In this study, the crude water-soluble polysaccharide CMP, which was obtained from the fruiting body of C. militaris by hot water extraction and ethanol precipitation, was fractionated by DEAE-cellulose and Sepharose CL-6B column chromatographies. This process resulted in three polysaccharide fractions, termed CMP Fr I, CMP Fr II, and CMP Fr III. Of these fractions, CMP Fr II, with an average molecular mass of 127 kDa, was able to upregulate effectively the phenotypic functions of macrophages such as NO production and cytokine expression. The chemical property of the stimulatory polysaccharide, CMP Fr II, was determined based on its monosaccharide composition, which consisted of glucose (56.4%), galactose (26.4%), and mannose (17.2%). Its structural characteristics were investigated by a combination of chemical and instrumental analyses, including methylation, reductive cleavage, acetylation, Fourier transform infrared spectroscopy (FTIR), and gas chromatography-mass spectrometry (GCMS). Results indicated that CMP Fr II consisted of the (1${\rightarrow}$4) or (1${\rightarrow}$2) linked glucopyranosyl or galactopyranosyl residue with a (1${\rightarrow}$2) or (1${\rightarrow}$6) linked mannopyranosyl, glucopyranosyl, or galactopyranosyl residue as a side chain. The configuration of the ${\beta}$-linkage and random coil conformation of CMP Fr II were confirmed using a Fungi-Fluor kit and Congo red reagent, respectively.

Comparison of Physicochemical Properties of Extruded Ginseng Samples

  • This study compared the physicochemical properties of root hair of white ginseng (WG), root hair of tissue cultured mountain ginseng (MG), root hair of red ginseng (RG) and extruded ginseng samples. The comparison of crude ash and total sugar resulted insignificant differences between extruded and raw samples. MG had a higher content of crude ash, crude protein, amino acids and polyphenolic compound than WG and RG; the total sugar and reducing sugar were highest in RG. Crude fat and acidic polysaccharide in RG and WG were similar to and higher than MG. Crude saponin of treated samples WG1 (moisture content 25%, barrel temperature $110^{\circ}C$) and WG3 (moisture content 35%, barrel temperature $110^{\circ}C$) were 9.80% and 9.73%, respectively, which were the highest among ginseng samples. In conclusion, the extrusion process can be applied to red ginseng manufacturing, and some characteristics of MG were higher than in RG and WG.

Physicochemical Properties and Intestinal Bacterial Growth-Promoting Effect of Cell-Wall Polysaccharides from Cucumber Peel

  • Physicochemical properties, intestinal microbial growth, and inhibitory effects of alcohol-insoluble polysaccharide (AIP) extracted from cucumber peel were investigated. AIP was composed of 14.54% crude protein, 1.04% crude lipid, 13.74 % crude ash, 9.1% soluble dietary fiber, and 41.2% insoluble dietary fiber. AIP showed low bulk density (0.18 g/mL) and water-holding capacity (6.39 g/g), and high oil-holding capacity (3.96 g/g). Pectic substance fractions [water-soluble pectic substance (WSP), ethylenediaminetetraacetic acid-soluble pectic substance (ESP), and alkali-soluble pectic substances (ASP)] and hemicellulose fractions [1 M KOH-soluble hemicellulose (KHP1) and 4 M KOH-soluble hemicellulose (KHP4)] were obtained from sequential chemical fractionation of AIP. WSP showed higher total sugar contents than total uronic acid contents, whereas opposite results were observed in ESP and ASP. Molecular weight distributions of three pectic substance fractions were in order of ASP>ESP>WSP. Ion exchange chromatogram pattern of WSP was different from those of ESP and ASP. Major component of WSP was fraction eluted by 0.05 M ammonium acetate buffer, whereas that of ESP and ASP was fraction eluted by 0.2 M NaOH. WSP and ASP showed growth-promoting activities against Lactobacillus brevis, Bifidobacterium bifidum, and B. longum, whereas B. bifidum and B. longum for ESP. KHP1 and KHP4 fractions had significant growth-promoting activities against B. bifidum.

Comparison of Protein Binding Polysaccharide from Agaricus blazei Murill Prepared by Ultrafiltration and Spray-Drying Process

  • Chemical properties of spray-dried powders separated based on molecular weight from crude protein binding polysaccharide (CP-SD) of Agraricus blazei were examined. Contents of ${\beta}$-glucan in SD-1, SD-2 and SD-3 were 18.67%, 48.24%, and 37.15% respectively, and SD-2 (10-150 kDa) showed the highest molecular weight. Obtained ${\beta}$-glucans were not pure glucan, but was determined to be an acidic proteo-heteroglycan with a large amount of glucose (74.46-80.05%), galactose (8.91-15.2%), and mannose (4.9-5.46%). Composition of their amino acids was mainly aspartic and glutamic acids. FT-IR spectrum revealed SD-1, SD-2 and SD-3 were structures of ${\beta}$-1,3-glucans and ${\alpha}$-1,6-glucans at 890 and 930 $cm^{-1}$, respectively, signals of ${\alpha}$-1,6-glucans for CP-SD was not found. Useful CP-SD was recovered from A. blazei for preparation of three powder types as food materials.
