Effects of extrusion conditions (barrel temperature and moisture content) and fermentation time on the antioxidant properties of root hair of tissue cultured raw mountain ginseng (MG) were investigated. The barrel temperature/ moisture combinations were: $110^{\circ}C$/25% (MG1), $140^{\circ}C$/25% (MG2), $110^{\circ}C$/35% (MG3) and $140^{\circ}C$/35% (MG4). Red ginseng (RG) was also investigated. The contents of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and polyphenolic increased after fermentation in RG and even more in MG, while extruded ginseng samples exhibited little change. The increases noted with MG and RG occurred during the first 4 days of fermentation. DPPH radical scavenging activity decreased after extrusion and was significantly higher in MG (20.93%) than RG (1.63%) on the first day of fermentation. DPPH radical scavenging activity in the barrel temperature/moisture combinations were 19.01% (MG1), 14.45% (MG2), 20.37% (MG3) and 15.78% (MG4). The content of polyphenolic compounds in ginseng samples displayed a similar trend. Acidic polysaccharide in RG and MG1${\sim}$MG4 were higher than MG, but decreased during fermentation. Crude saponin in RG and MG1${\sim}$MG4 decreased after 15 days of fermentation, while increasing in MG.

To develop new potent antitumor substancces with low toxicity from natural products, especially from higher fungi of Korea, the carpophores of some wild basidiomycetes were collected and the antitimor test for their polysaccharides was done. The dried carpophores were extracted with hot water, and from the extracts, crude polysaccharides were obtained by alcohol precipitation. The alcohol precipitated crude polysaccharides were partially purified by dialysis and then used as the samples for antitumor tests. Among tested, the polysaccharide of Laccaria laccata, Trametes sanguineus, Armlliariella mellea, Clitocybe in fundibuliformis, and Xeromphalina campanella respectively showed the inhibition ratio of 75%, 72%, 94% 55%, and 47% when administered i. p. once dailly for ten days at the dose level of 20mg/kg/day into the mice implanted with $10^{6}$ cells of sarcoma 180/mouse. However, those of Craterellus cornucopiodes, Daedaleopsis confrogosa, and Coriolus sp. showed almost no activities.
Polysaccharides of marine algae exhibit different structural characteristics and interesting biological functions. In this study, crude polysaccharides (CP) of eleven Sri Lankan marine algae obtained through hot water extraction and ethanol precipitation were investigated for DPPH, alkyl, and hydroxyl radical scavenging activities using electron spin resonance spectrometry and for intracellular reactive oxygen species scavenging activity in the Chang liver cell line. Characterization of CPs was done by Fourier transform infrared (FTIR) spectroscopy and by analysis of the monosaccharide composition. Time-dependent density functional theory quantum-chemical calculations at the RB3LYP/6-31G(d,p) level for constructed dimeric units of the corresponding polysaccharides were used to resolve the FTIR spectra. CPs from Chnoospora minima showed the highest DPPH and alkyl radical scavenging activities and higher intracellular reactive oxygen species scavenging effects for both AAPH and $H_2O_2$ induced ROS production in "Chang" cells. The major polysaccharide constituent in C. minima CP was identified as fucoidan and it displayed a higher sulfate content. The degree of sulfation of these polysaccharides suggests a positive correlation with the observed antioxidant properties.
η$_{evap}$분자량에 따라 다양한 생리활성을 나타내는 아가리쿠스버섯에서 분리한 조단백다당류를 고부가가치 기능성 소재로 개발하기 위하여 막분리 및 분무건조 공정에 따른 특성을 조사하였다. 아가리쿠스버섯 조단백다당류를 분자량 크기에 따라 여과하는 동안 공정온도에 따른 투과플럭스의 변화는 10 kDa와 130 kDa 모두에서 유사하였는데, 막분리 온도 및 압력이 증가함에 따라 비례적으로 증가하였으며, 4$0^{\circ}C$에서의 투과플럭스가 가장 높았다. 시간에 따른 투과플럭스의 변화는 막분리 30분 경과시 급속히 감소하였으며, pore size가 10 kDa와 150 kDa일 때 공정압력이 각각 1.2kgf/$\textrm{cm}^2$와 2kgf/$\textrm{cm}^2$에서 가장 높았다. 한외여과 후 아가리쿠스버섯 조단백다당류의 점도는 여과 전 13.9cP에서 여과후 10kDa이 하와 10∼150 kDa구간은 각각 10.8, 11.9cP로 점도가 저하되었으나, 150 kDa이상은 20.1cP로 점도가 증가되었다. 분자량 크기가 다른 3가지 분획물에 대한 분무건조 공정에서의 열효율성은 가열공기온도, 시료공급속도 및 농도가 증가할수록 열효율성이 증가함을 알 수 있었으며, 시료공급속도가 가장 중요한 영향인자임을 확인하였다.다.
계피나무의 어린가지인 계지의 열수추출물에서 높은 보체계 활성효과(항보체 활성)를 발견하여 대량으로 열수추출한 추출물(CC-O)에 대하여 메탄올 환류, 에탄올 침전, 투석, 동결건조를 실시하여 메탄올과 에탄올에 비가용성인 고분자 획분(CC-1)에서 증가된 활성을 보였다. 60.3%의 당과 32.8%의 단백질로 구성되어 있는 CC-1 획분에서 보체계 활성화 본체를 파악하기 위하여 pronase처리에 의한 단백질 분해 및 periodate를 이용한 다당부위를 선택적으로 산화시킨 후, 각각의 활성을 조사한 결과 pronase 처리한 CC-1에서는 보체계활성 효과를 그대로 유지한 반면, CC-1의 periodate 산화물은 CC-1에 비하여 활성이 50% 이하로 감소한 사실로부터 보체계 활성화 물질이 다당임을 확인하였다. CC-1을 양이온 계면 활성제인 cetavlon으로 처리 후 CC-2, CC-3, CC-4 및 CC-5의 4획분으로 분리하였으며 수율과 활성이 가장 높은 CC-2분획을 음이온 교환 수지인 DEAE-Toyopearl 650C column에 흡착시켜 비흡착획분(CC-2-1)과 7개의 흡착획분$(CC-2-II{\rightarrow}CC-2-VIII)$으로 분획하였다. 이 중 0.2 M NaCl로 용출된 CC-2-III 획분을 Sephadex G-100 및 Sepharose CL-6B 겔여과 크로마토그래피를 행하여 주요 활성다당체인 CC-2-IIIa-3를 최종적으로 정제하였다. HPLC상에서 거의 순수한 단일 peak로 확인된 CC-2-IIIa-3는 41.1%의 산성당을 함유하는 산성다당체로서 분자량은 240,000으로 확인되었다. CC-2-IIIa-3의 항보체 활성$(ITCH_{50})$은 1 ㎎/ml의 농도에서 대조군의 94%를 나타내었으며 구성당 조성은 arabinose, xylose, glucose, galactose, galacturonic acid 및 glucuronic acid가 5.56 : 3.77 : 1.87 : 1.00: 5.12 : 3.13의 비율로 존재하였다.
A bacterial strain capable of producing a novel bioflocculant was isolated from a biofilm sample and identified as Bacillus megaterium G31. The highest biopolymer yield was achieved when the organism was cultivated in a medium containing acetate as the sole carbon source and ($NH_4)_2HPO_4$as the nitrogen source. In kaolin suspension, the flocculating activity was highest at 170 mg I$^{-1}$ and decreased at the higher bioflocculant concentrations. The crude bioflocculant produced by the organism was purified by ethanol precipitation and gel permeation chromatography. The FTIR spectrum of the purified bioflocculant revealed that the bioflocculant might be a heterogeneous polysaccharide composed of hexosamines and neutral sugars. The analysis of sugar components of the bioflocculant using high performance anion-exchange chromatography showed that the sugar constituents of the bioflocculant were glucosamine, fucose, galactosamine, galactose, glucose, mannose in approximate molar ratio of 4 : 1 : 6 : 3 : 8 : 19. Its flocculating activity was stimulated by various cations. The bioflocculant was thermo-stable and retained 64% of its original activity after heating at $100^{\circ}C$ for 50 min.
A simple method for the purification of porphyran from laver Porphyra yezoensis was developed to obtain information for the development of food materials with biological functionality. Crude porphyran (CP) was extracted from dried laver in boiling water for 3 h, and then fractionated using cetylpyridinium chloride into an acidic fraction (CP-F1) and a neutral fraction (CP-F2). CP-F1 was fractionated further by fractional ethanol precipitation. Fraction CP-F1-70, precipitated at an ethanol concentration of 61-70% was the major fraction containing 68.1% of the yield from the initial fraction CP-F1. The CP-F1-70 fraction displayed a single band on Sepharose CL-4B with a molecular mass of 550 kDa, indicating a homogeneous polysaccharide. The molar ratio of galactose, 3,6-anhydro-L-galactose, 6-0-methyl-D-galactose and ester sulfate of CP-F1-70 was 1:0.32:0.07:0.53. This method is very useful for rapid and large-scale preparation of purified porphyran because it is compatible with mass production.
Crude polysaccharides from rapeseed meal (PRM) were extracted with 0.3% NaOH aqueous solution, followed by further purifications and 2 fractions, namely PRM1 and PRM2, were separated with a DEAE-cellulose DE-52 column. Their primary compositions were analysed and antioxidant activity was determined, including scavenging activity toward superoxide anion radicals, hydroxyl radicals, and nitric oxide radicals, reducing power, and inhibitory effects against the microsomal lipid peroxidation, compared to that of L-ascorbic acid. The results indicated that PRM1 and PRM2 exhibited not only good reducing power and inhibitory effects on the microsomal lipid peroxidation, but also strong scavenging activity toward superoxide anion radicals, nitric oxide radicals, and hydroxyl radicals. In addition, positive correlations were also observed between the superoxide anion radical scavenging activity and the protein contents of the polysaccharides, and the reducing power and the sulfate contents. These findings thus clearly suggest the polysaccharides possess direct and potent antioxidant activity.
This study was designed to investigate the intracellular signaling pathways and immunoenhancing effect of macrophage activation by crude polysaccharides (CPP) extracted from citrus peels. CPP did not affect the cytotoxicity of RAW264.7 cells, but showed dose-dependent effects on cell viability. Also, CPP showed high production of chemokine (nitric oxide (NO)) and cytokines (interleukin (IL)-6 and tumor necrosis factor (TNF)-α). CPP increased IL-6, TNF-α, and inducible nitric oxide synthase (iNOS) mRNA expression dose-dependently. CPP also strongly induced the phosphorylation of the ERK, p38, and IκBα pathways in RAW 264.7 cells. In anti-pattern recognition receptors (PRRs) experiments, the effect of CPP on NO production was strongly suppressed by neutralizing toll-like receptor (TLR)2, TLR4, and Dectin1 antibodies, whereas IL-6 and TNF-α production by CPP was mainly suppressed by mannose receptor (MR). Therefore, these results suggest that CPP treatment-induced NO production was regulated by the ERK, p38, and NF-κB pathways through TLR2, TLR4, and Dectin1 receptors, whereas IL-6 and TNF-α production was primarily regulated by the ERK, p38, and NF-κB pathways through MR receptors.
백삼과 홍삼의 추출수율은 각각 33.56%와 48.79%로 홍삼의 추출수율이 높았고, 산성다당체의 경우 1~4차 추출단계까지는 홍삼군이 백삼군보다 높았으나, 5차 추출단계에서 백삼군이 6차 추출단계에서는 홍삼군이 높게 나타났다. 백삼과 홍삼의 추출단계별 농축액의 조사포닌 함량은 백삼(56.44 mg/g)에 비해 홍삼(53.15 mg/g)이 약 5.8 % 증가하는 것으로 나타났다. 이들을 혼합한 선택적 홍삼농축액 모델 4종의 기호도 평가 결과 1, 2, 6차 추출물이 혼합된 RGB-4가 가장 높은 기호도를 나타내었고 시판 홍삼 농축액 3종에 대해 비교한 결과에서도 RGB-4가 쓴맛, 떫은맛, 인삼향 강도가 약해지고 종합적 기호도에서 높은 평가를 받았다. 본 실험에서는 추출방법을 달리 함으로써 추출수율 및 조사포닌 함량을 감안하여 인삼 쓴맛과 향의 강도를 낮출 수 있다는 것을 확인하였다. 이 추출방법을 이용하여 쓴맛을 저감시키고 인삼향미의 강도를 저감시킨 제품을 개발하여 좀 더 편하고 자연스럽게 홍삼제품을 접할 수 있도록 더 많은 연구가 진행 되어야 할 것이다.
