The present study has been undertaken to characterize availability of citrus as a medicinal plant with antineoplastic property. The crude extracts from 40 species of fruits with 12 species of the local Citrus in Cheju island were evaluated on their potential activities against mouse P388 lymphocytic leukaemia in vitro. The percent cytotoxicity varied from 25.40 to 97.94% at a concentration of $100{\mu}g/mL$. Among 40 spp., 8 species showed high toxicity more than 90% against P388 cells and Cheongkyool(C. nippokoreana) exhibited the most cytotoxicity as 97.94%($IC_{50}=20.2{\mu}g/mL$). Nine varieties of C. junos were showed insiginicant cytotoxicity. In trifoliate orange, immature fruit was stronger than mature and peel extract showed higher cytotoxicity($IC_{50}=18{\mu}g/mL$) than the other tissues. Hexane fraction from methanol(MeOH) extract of trifoliate orange showed highly significant inhibition of cell growth($IC_{50}=3.9{\mu}g/mL$). In addition, its cytotoxicity increased remarkably from 3.95 to $0.40{\mu}g/mL$ as exposure time legthened. Cytotoxic activities of crude extracts were decreased considerably during a six months storage period. It was apparent that there is considerable variation in cytotoxicity, depending upon species, maturity and storage time of extracts. There was no meaningful cytotoxic difference between archicitrus and metacitrus in the genus Citrus.
To evaluate anti-aggregatory activity of traditional prescriptions and medicines for treatment of Ohyul symptom, 70% methanol extracts were examined using collagen stimulated in vitro platelet aggregation by impedance method in rat whole blood. The crude extracts from DoHaekSeungKiTang, BoYangHwanOhTang, Caesalpinia sappan, Rhus verniciflua, Rheum palmatum, Polygonum cuspidatum, Salvia miltiorrhiza were found to inhibit platelet aggregation. The effective crude extracts of traditional medicine were fractionated to dichloromethane, ethyl acetate, butanol and aqueous layer. Polygonum cuspidatum, Caesalpinia sappan aud Rhus verniciflua ethyl acetate fractions concentration-dependently $(250-50{\mu}m/ml)$ inhibited the aggregation of platelet in whole blood induced by collagen. These results suggested that ethyl acetate fractions of Polygonum cuspidatum, Caesalpinia sappan and Rhus verniciflua have potent anti-aggregatory activity.
In this study, the inhibitory effect of Atriplex gmelinii C. A. Mey. against the activity of MMP-2 and MMP-9 secreted from phorbol-12-myristate-13-acetate (PMA)-stimulated HT-1080 cells was evaluated by gelatin zymography and enzyme-linked immunosorbent assay (ELISA), reverse transcription polymerase-chain reaction (RT-PCR), and Western blot assay. Specimens of the halophyte A. gmelinii were extracted twice for 24 hr with methylene chloride ($CH_2Cl_2$), and then twice with methanol (MeOH), in turn. Each extract significantly inhibited the enzymatic activities in gelatin zymography and MMP ELISA kit, and expression of MMP-2 and 9 in mRNA and protein levels. Two crude extracts were combined and then the combined crude extracts were fractionated into n-hexane, 85% aqueous methanol (85% aq.MeOH), n-butanol (n-BuOH), and water ($H_2O$) fractions, according to solvent polarity. Among solvent-partitioned fractions, the 85% aq.MeOH fraction showed the strongest inhibitory effect against MMP-2 and -9 in gelatin zymography and MMP ELISA kit. In RT-PCR, all solvent-partitioned fractions significantly suppressed mRNA expression of MMP-2 and -9. On the other hand, in Western blot assay, all solvent-partitioned fractions except $H_2O$ significantly reduced expression levels of protein. HT 1080 cell migration was most significantly inhibited by the n-BuOH fraction followed by the 85% aq.MeOH and $H_2O$ fractions. These results suggest that A. gmelinii could be used as a potential source to inhibit tumor cell metastasis.
We investigated the chemical components of red onion powder dried using the low temperature vacuum method and the inhibitory effects of solvent extracts of the dried red onion powder on the growth of HT-1080 human fibrosarcoma and HT-29 human colon cancer cells and $H_2O_2$-induced oxidative stress. The moisture content of the dried red onion powder was 17.95%, while the vitamin C content was 96 mg/100 g and the total phenols content was 39.1 mg/mL. The inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts of the red onion powder on the growth of HT-1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was greater on the growth of HT-29 cells, while the A+M extracts had a higher inhibitory effect than the MeOH extracts. Treatment with the hexane, 85% aq. methanol, butanol and water fractions of the extract led to significant inhibition of the growth of both cancer cell lines (p<0.05). Among the fractions, the hexane and 85% aq. methanol fractions showed a greater inhibitory effect. To determine the protective effect on $H_2O_2$-induced oxidative stress, a DCFH-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including the crude extracts of dried red onion, appeared to lead to a significant reduction in the levels of intracellular reactive oxygen species (ROS), and these reductions occurred in a dose dependent fashion (p<0.05). Among the fractions, the 85% methanol fraction showed the greatest protective effect on the production of lipid peroxides.

In this study, antioxidant activities of acetone/dichloromethane and methanol extracts of Arctic seaweeds were investigated. The antioxidant properties of both extracts of arctic seaweed were evaluated using two different peroxynitrite tests, including scavenging power on authentic peroxynitrite and inhibitory activities on peroxynitrite generation from 3-morpholinosydnonimine (SIN-1) producing superoxide anion and nitric oxide simultaneously in vitro. At concentration of $10\;{\mu}g/ml$, the acetone/dichloromethane and methanol extracts of Odonthalia dentata exhibited 54.6 and 64.2% inhibition against peroxynitrite generation from SIN-1 while they exhibited 24.3 and 23.0% scavenging activities on authentic peroxynitrite, respectively. On the other hand, the acetone/dichloromethane extract of Polysiphonia stricta exhibited 61.8% inhibition only against the induced peroxynitrite from SIN-1. Furthermore, the crude extracts of Odonthalia dentata and Polysiphonia stricta were fractionated into n-hexane, 85% aq. MeOH, n-BuOH, and $H_2O$ fractions, successively, and only 85% aq. MeOH fraction exhibited the best inhibition.
생열귀나무의 잎에 대한 일반성분과 tocopherol 동족체를 분석하였으며, 생리활성 기능을 검색하기 위해 메탄을 추출물의 항산화력을 보다 더 정제하기 위해 헥산, 클로르포름, 에틸아세테이트, 부탄을 및 물층으로 분획하여 얻어진 추출물과 분획물을 이용하여 자외선 (UV 308, 350 nm) 차단효과와 DPPH 자유라디칼 소거법에 의한 항산화성 효과를 검토하여 다음과 같은 결론을 얻었다. 일반성분의 분석 결과 수분이 67.5%로 가장 높게 나타났으며, 탄수화물 18.9%, 조단백질 6.8%, 회분 6.1% 그리고 조지방 0.7% 순이었다. 무기질 함량으로 가장 높은 것은 칼륨으로 1,637.2 mg%로 나타났다. 메탄올 추출물을 물로 현탁시킨 후 헥산, 클로르포름, 에틸아세테이트, 부탄을 순서로 용매를 분획하여 각각의 분획물을 얻은 결과 물(34.5%)이 가장 높게 분획되었으며, 메탄을 추출물을 각각 n-헥산, 클로르포름, 에틸아세테이트, n-부탄올 및 물 분획물을 얻어 자외선 차단효과를 측정한 결과 장파장대 (UVA 320∼400 nm)의 경우 에틸아세테이트 분획물(151.4)에서 가장 높게 측정되었다. 한편, 메탄을 분획물의 항산화 활성을 검토한 결과 에틸아세테이트 분획물의 항산화 활성 ($IC_{50}$/ : 6.0 $\mu\textrm{g}$)이 가장 높았으며, 그 다음이 부탄올 분획물($IC_{50}$/ : 14.0 $\mu\textrm{g}$)로 나타났다.
Antimicrobial effect of tea extracts from green tea(steamed, roasted), oolong tea and black tea was investigated. Minimal inhibitory concentration(MIC) of tea extraxcts against 9 well known strains of foodborne pathogenic bacteria such as the Gram positive and Gram negative bacteria was determined at 37$^{\circ}C$. Significant antimicrobial activity was observed in the steamed green tea and the roasted green tea of the water-soluble fraciton, and the steamed green tea of the methanol-soluble fraction, and the steamed green tea, roasted green tea and the oolong tea of the crude catechin fraction. The MIC of these extracts against B. subtillis were 700$\mu\textrm{g}$/ml, 500$\mu\textrm{g}$/ml and 120 $\mu\textrm{g}$/ml, respectively. The crude catechin fraction possessed greater antimicrobial activity than did the other fractions. Among tea extracts, extracts of steamed green tea, roasted green tea and oolong tea showed higher antimicrobial activity than them of black tea. The MIC of the crude catechin fraction obtained from tea extracts against Gram-positive bacteria such as M. Iuteus, B. subtillis and S. mutans were 30~50$\mu\textrm{g}$/ml, 120~240$\mu\textrm{g}$/ml and 120~180$\mu\textrm{g}$/ml, and against Gram-negative bacteri such as e. aerogenes and V. parahaemolyticus were 50~60$\mu\textrm{g}$/ml and 60~70$\mu\textrm{g}$/ml in the broth medium, respectively. Especially, the MIC to Streptococcus mutans which has known as a causative bacteria of a decayed tooth were 120$\mu\textrm{g}$/ml, 140$\mu\textrm{g}$/ml, 180$\mu\textrm{g}$/ml and above 1,000$\mu\textrm{g}$/ml in steamed green tea, roasted green tea, oolong tea and black tea, respectively. Tea extracts had strong growth inhibition activity against foodborne pathogenic and dental bacteria.
White, red, and extruded ginsengs were studied against 8 strains of food-borne pathogens and/or food spoilage microorganisms. The ginseng powders were extracted with different extractants and screened for antimicrobial activity using the disc diffusion and broth dilution techniques. The results showed that the yield of extraction was higher with increase of aqueous solution content and temperature. Preliminary screening revealed that the red ginseng extracts were most active, that has been found to be highly effective against all tested microbe except Listeria monocytogenes. Moreover, Bacillus subtilis has shown highly susceptible, which the diameters of inhibition zone values of 28 extracts were between 7 and 14 mm. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) recorded for the different crude ginseng extracts against microorganism using ranged from 6.25 to 100 mg/mL, indicated that the methanol extract of ginseng were more effective than ethanol and water extracts. The 60% methanol extract of red ginseng had the greatest effects against B. subtilis with MIC and MBC values at 6.25 mg/mL.

This study was carried out to examine the constituents of Amorpha fruticosa (Leguminosae), a shrub originated from North Africa. Dried and ground fruit of A. fruticosa were extracted with methanol and then concentrated to give the crude extracts. The crude extracts was successively fractioned with organic sovents, such as n-hexane, CH$_2$Cl$_2$ and EtOAc. Seven compounds were isolated from the fruits of A. fruticosa. On the basis of spectrosopic data, the structures of these compounds were determined as: kaempferol 7-O-${\alpha}$-L-rhamnopyranoside (I), methyl 3,4,5- trihydroxybenzoate (methyl gallate, II), tephrosin (III), dalbinol (IV), gallic acid (V), 2",4",5",7-tetramethoxyisofavone (Ⅵ) and Dalbinol 2"-O-${\beta}$-D-Glucopyranoside (Ⅶ) respectively. Brine shrimp toxicity of water extracts of twenty three crude drugs was tested by the brine shrimp bioassay. Eight crude drugs were shown to have 100% mortality in $1000{\;}{\mu}g/ml$. They were fractionated by Amberlite XAD-2 column chromatography and were tested by the brine shrimp bioassay. Methanol fraction of Rhei Rhizoma $(LC_{50}=49.60{\;}{\mu}g/ml)$ and Cantharis $(LC_{50}=54.08{\;}{\mu}g/ml)$ were shown to have potent brine shrimp toxicity in this test.
