Comparison of Oral versus Rectal Administration of Processed-Scutellaria baicalensis on Colonic Inflammation in Mice

  • We previously showed that the water extracts of rice wine-baked Scutellaria baicalensis Georgi (RWBS) ameliorated colonic inflammation more than crude Scutellaria baicalensis (CS) after oral administration. The aim of this study is to compare the effect of rectal and oral administration of RWBS in the experimental colitis. Experimental colitis was induced in mice by daily treatment with 5% dextran sulfate sodium (DSS) in the drinking water for 7 days. Water was used as vehicle of oral administration, while Carbopol/PEG mucoadhesive gel was used as vehicle of rectal administration. RWBS and RWBS gel (RWBSG) were administered once per day for 7 days. RWBS and RWBSG significantly attenuated the disease activity index (DAI) calculated as the sum of scores of body weight loss, stool consistency and rectal bleeding. Furthermore, RWBS and RWBSG reduced the mucosal myeloperoxidase activity and COX-2 (cyclooxygenase-2) expression in colon tissue. Anti-inflammatory effect of CS on colonic inflammation was increased by baking with rice wine in both oral and rectal administration. Moreover, anti-inflammatory effects of oral administration on colonic inflammation was greater than those of rectal administration. Further study would be required for the development of intra-rectal formulation.

Isolation and Characterization of Acidic Polysaccharides Activating Complement System from the Hot Water Extracts of Pteridium aquilinum var. latiusculum (고사리 열수 추출물로부터 보체계 활성화 산성 다당의 분리 및 특성)

  • From the hot water extract of bracken(Pteridium aquilinum var. latiusculum), a Korean win edible plant, anti-complementary acidic polysaccharides were Isolated. Crude polysaccharide fraction(HPA-1) was obtain ed by methanol reflux, ethanol precipitation, dialysis, and lyophilization. HPA-1 contained 81.80% of total sugar, 30.40% of uronic acid, and 15.60cA of protein. HPA 1 was purified consecutively by cetavlon fractionation and chromatography including ion exchange nth DEAE-Sepharose CL 6B and gel permeation with Sephadex G-100 and Sepharose CL-6B. HPA-2- IVa and HPA-Va-2 were nearly homogeneous on HPLC and had 500,000 and 560,000 daltons of molecular weights, respectively. HPA-2-Wa consisted of fucose, galacturonic acid, and glucuronic acid at the molar ratio of 1.40 : 0.97 : 1.88. HPA-2-Va 2 was composed of rhamnose, galactose, and galacturonic acid at the molar ratio of 1.00 : 1.38 : 1.39. The polysaccharides were found to activate the C3 component both In the presence and In the absence of Ca2+ through the crossed-immunoelectrophoresis suggesting that those Involved in both classical and alternative complement pathway.

  • With the progress of chemical researches in ginseng studies, efforts to elucidate the pharmacological actions of ginseng have been greatly increased. The majority of ginseng reaserches in the past has been performed with crude extracts from ginseng roots to verify scientifically the empirical application of ginseng in men and animals recently. Ginseng reaserches have been done mostly with pure ginsenosides and there has been a shift in focus to the various biochemical pathways. It was demonstvated that ginseng had diverse effects by modulating the second-messenger system, such as cyclic nucleotides. calcium The demonstration in 1987 of the formation of nitric oxide(NO, endothelium-derlled rectating factor) by an enzyme in vascular endothelial cells opened up a new area of biological reaserches of ginseng. It was shown that vascular relaxations induced by glnsenosides are mediated by release of nitric oxide from endothelial cells. According to the literature search from'hledline". There have been 737 original and review articles during the last 30 years. In these review articles, an attempt has been made to summalize some results from some of these published papers. Ginseng has a wide range of phal.macologtcal and therapeutical actions. It acts on the centralral nervous system and cardiovascular system, promotes immune function and metabolism. Possesses anti-stress. Anti-cancer and anti-ageing activities, and so on.o on.

  • High levels of extracellular phospholipase $A_2$ (Plase $A_2$) associated with inflammatory process in man and animal models have been extensively reported elsew here. Thus, a logical approach to the treatment of inflammatory diseases should involve the inhibitors of Plase $A_2$. To develop new Plase $A_2$ inhibitors from natural products, two hundred crude drugs were screened using group II PLA$A_2$ inhibitory activity. Among them, methanol extract of 5 medicinal plants such as, Raphani Semen, Moutan cortex radicis, Arecae semen, Caryophylli Cortex and Betulae Cortex inhibited more than 90% of PLase $A_2$ activity at a concentration 2.5${\mu}g/ml$. Then, 10 methanol extracts sample were transferred into organic solvents, PLase $A_2$ inhibitory effects were found mainly in CHCl3 and EtoAc fractions.

  • The adrenergic blocking activity and refractory period of cardiac muscle on isolated rabbit atria were measured after administration of Scutellaria. In rabbits and cats the antiarrhythmic action of Scutellaria on atrial and ventricular arrhythmias produced by epinephrine or ouabain was examined and also compared with that of propranolol and quinidine. The alcoholic extract of Scutellaria produced a marked decrease in heart rate and contractile amplitude of the isolated rabbit atria. Pretreatment with Scutellaria rendered the atria to fail to respond to epinephrine, indicating that this crude drug possesses an adrenergic blocking activity. The extract produced a marked prolongation of the refractory period of atrial muscle. The extract effectively abolished the spontaneous arrhythmia occurring in the isolated rabbit atria. As propranolol and quinidine it also suppressed the atrial arrhythmia induced by ouabain. The extract prevented, as propranolol and quinidine, the induction of ventricular arrhythmia arising from excessive dose of epinephrine in anesthetized rabbits and cats. With regard to the ventricular arrhythmia induced by a continuous infusion of ouabain, the alcoholic extract of Scutellaria exerted some suppressive effect in anesthetized rabbits but no effect on cats. From the above results, it may be concluded that Scutellaria is effective against atrial and ventricular arrhythmias. The antiarrhythmic effects of this drug may be the result of adrenergic beta receptor blocking and cardiac depressive activities including prolongation of the refractory period of cardiac muscle.

Biological Effects of Vinca minor extract; Tyrosinase inhibition, stimulation of ROS generation and increasement of cell migration activity in keratinocytes

  • Vinca alkaloids from plant Vinca minor have been investigated for their effects of tyrosinase inhibition, stimulation of ROS generation and increasement of cell migration activity. The methanolic crude extract and the water-soluble fraction exhibited $IC_{50}$ value of 3.1 mg/mL and 2.1 mg/mL. Vinca minor extract treatment significantly increased ROS levels in HaCaT cells, in a concentration-dependent manner. Treatments of Vinca minor extract led to increase wound closure when compared with non-treatment. Low dose (0.1% or 0.3%) of extracts have not significantly affected, compared with that in controls. By contrast, 0.5% extract have dramatic effect on wound healing activity of keratinocytes. Effects of Vinca minor extract in a filter-based cell mobility assay appear similar to that of wound closure assay, which suggests that the Vinca minor extract have wound healing effects on skin.

The dynamics of proteinase activity at different stages of toxocariasis in mice (개회충에 감염된 생쥐에 있어서 시기별 단백분해효소 활성의 변동)

  • Toxocariasis is a parasitic zoonosis caused by infestation of humans with larvae of Toxocara canis, the common roundworm in dogs. Two syndromes have been identified : visceral larva migrans and ocular toxocariasis. In this study we were characterized proteinase activity in crude extracts from liver, lung, kidney and heart of mice infected with Toxocara canis and the dynamics of their changes in different stages of disease. The optimal pH was 5.5. In liver of mice infected with Toxocara canis, the maximun activity of proteinase was observed in 5 day post infection. In lung, the activity reached its maximun on 5th day in A group (infected with 100 embryonated eggs), and on 5th week in B group (infected with 50 embryonated eggs). In kidney, the maximum activity was shown at 6th week in A group, and in B group was shown at 10th day. In early infection, the activity reached its maximun in heart of mice infected with Toxocara canis. As we could see, the dynamics of the changes of proteinase activity in mice is similar in the case of the disease with other biochemical and immunological indices observed in toxocariasis.

Identification and Bioassay of Bioactive Compounds Isolated from Phytolacca americana (미국자리공 (Phytolacca americana)에서 추출한 생물활성 물질의 동정 및 생물검정에 관 하여)

  • This study was carried out for the identification and bioassay of bioactive compounds which isolated from Phyutolacca americana L. obtained results are summarized as follows: 1. Two biological active compounds were found from crude extracts of P. americana roots, using the systematic solvent fractionations, such as ethyl acetate fraction and butanol fraction. One biological compound was isolated from the ethyl acetate fraction with silicagel column chromatography, which was identified as a ${\alpha}-spinasterol$ by spectral analysis of IR, H-NMR, C-NMR and MS. The other one is isolated from butanol fraction which was identified as phytolaccoside E by spectral analysis of IR, H-NMR, C-NMR and MS. 2. The ${\alpha}-spinasterol$ and phytolaccoside E induced necrosis of primary root and resulted in death of the tested plant. These two compounds strongly inhibited to the growth of Mucor racemous and Phytophthora infestants but did not inhibited the growth of Colletotricum lagenarium and Fusarium oxisporum. 3. Cytotoxicity of the two biological active compound was exammined to the two different animal cancer cell lines (L1210, K562). The phytolaccoside E has some cytotoxical activity to the growth of cancer cell lines (L1210, K562) but $\alpha$-spinasterol has not cytotoxical effect.

Cloning and Characterization of Actinorhodin Biosynthetic Gene Clusters from Streptomyces lividans TK24

  • Actinorhodin antibiotics produced by Streptomyces lividans TK24 are blue pigments with a weak antibiotic activity, derived from one acetyl-CoA and 15 malonyl-CoA units via a typical ployketide pathway. In an attempt to clone polyketide biosynthetic genes of S. lividans TK24, hybridizing fragments in the genomic DNA of S. lividans TK24 were detected by use of acn and act III polyketide synthase gene probes. Since typical aromatic polyketide bio-synthetic gene clusters are roughly 22-34 Kb long, we constructed in E. coli XL-Blue MR using the Streptomyces-E. coli bifunctional shuttle cosmid vector (pojn46). Then, about 5,000 individual E. coii colonies were thor-oughly screened with acrl-ORFI and actIII probes. From these cosmid libra-ries, 12 positive clones were identified. Restriction analysis and southern hybridization showed two polyketide biosynthetic gene clusters in this organism. These cosmid clones can be transformed into Streptomyces parvulus 12434 for expression test that identify product of actinorhodin biosynthetic genes by heterologous expression. Thus, heterologous expres-sion of a derivative compound of a actinorhodin biosynthetic intermediate was obtained in pKE2430. Expression of these compounds by the trans-formants was detected by photodiode array HPLC analysis of crude extracts.

Anti-allergic Action of Some Medicinal Plants (수종 생약의 항알레르기 작용)

  • Anti-allergic action of each water extracts of some crude drugs was investigated in mice and rats. The activity of hyaluronidase which was used in the screening test of anti-allergic action was inhibited significantly by Amomi Semen, Asiasari Radix, Cimicifugae Rhizoma, Cinnamomi Ramulus, Glycyrrhizae Radix and Scutellariae Radix. The 48-hour homologous passive cutaneous anaphylaxis(48-hr PCA) in mouse ear was inhibited significantly by intraperitoneal(i.p.) injection of Amomi Semen, Cimicifugae Rhizoma, and ketotifen, a comparative drug of an anti-allergic action. The increase of vascular permeability induced by histamine or serotonin was inhibited significantly by i.p. injection of Amomi Semen, Cimicifugae Rhizoma, Cinnamomi Ramulus and ketotifen. In rat dorsal skin, the increase of vascular permeability which was induced by histamine, serotonin or compound 48/80 was inhibited significantly by i.p. injection of Amomi Semen, Asiasari Radix, Cimicifugae Rhizoma, Scutellariae Radix and ketotifen. Armeniacae Semen and Liriopis Tuber which had not inhibited hyaluronidase activity did not inhibit 48-hr PCA and the increase of histamine, serotonin or compound 48/80-induced vascular permeability in mice and rats. These results suggest that each water extract of Amomi Semen and Cimicifugae Rhizoma has anti-allergic action.
