수중 중금속의 연속적인 제거에 있어 여러 흡착제의 제거능 비교에 관한 연구

  • Crab shell 충진 칼럼을 이용한 수중 중금속의 연속적 제거에 있어서 다른 흡착제 (CER, GAC, Zeolite)와 비교 실험을 통하여 crab shell의 중금속 제거능을 알아보았다. 수중 $Pb^2+/{2+}$ 의 column을 이용한 연속적제거에 있어서 미세침전부분이 외부로 배출됨으로 칼럼 외부에 여과시스템의 도입함으로써 중금속 제거능을 높일 수 있을 것으로 판단된다. 중금속의 연속적 제거에 있어서 CER의 1,000 BVs까지의 전체 제거량은 0.35mmol/g, GAC는 0.17mmo1/g, zeolite는 0.16 mmol/g으로 나타났고 crab shell에 의한 중금속 제거량은 0.61 mmol/g으로 다른 흡착제보다 높은 제거효율을 보였다. 수중 중금속의 연속적 제거에 있어서 여러 흡착제의 제거 순서는 crab shell > CER > GAC > zeolite 로 나타났다. Crab shell의 경우는 파과점이 2,000 BVs에서 나타나므로 일반적인 폐수처리의 화학적 처리 후 잔존하는 저농도의 중금속을 제거를 위해 crab shell 충진 칼럼이 유용 할 것으로 판단된다. Crab shell의 재사용 가능성을 알아보기 위하여 crab shell을 이용한 중금속 제거공정에서 연속적인 중금속제거와 탈착과정을 통하여 보았을 때 1회에서 $Pb^2+/{2+}$제거된 량은 0.71 mmol/g 제거되었고 2회에서는 0.27 mmol/g, 3회에서는 0.02 mmol/g으로 급격하게 crab shell의 제거능이 떨어지는 것을 관찰 할 수 있다. 이는 crab shell이 재사용이 불가능 할 것으로 판단되나 다른 흡착제보다 월등한 제거효율을 보이므로 수중 중금속의 연속적 제거 흡착제 적합할 것으로 판단된다.

  • Crab shell 충진 칼럼을 이용한 수중 중금속의 연속적 제거에 있어서 여러 중금속 ($Pb^{2+}$ , $Cd^{2+}$ , $Cu^{2+}$ , $Cr^{3+}$ )의 제거능 비교와 중금속 제거 전후의 crab shell의 표면 변화를 관찰해 보았다. Crab shell 충진 칼럼의 중금속 제거능을 알아보기 위해, 가장 변화가 많은 1,000 BVs까지의 crab shell g 당 각각 중금속의 제거량을 비교해 보면 $Pb^{2+}$(0.61 mmol/g) > $Cu^{2+}$(0.43 mmol/g) > $Cd^{2+}$(0.38 mmol/g) > $Cr^{3+}$(0.30 mmol/g) 순으로 나타났다. $Cd^{2+}$의 경우는 미세 침전보다는 내부의 화학적 침전이나 물리적 침전에 의한 제거가 더 많이 일어나고 $Pb^{2+}$의 경우는 유출되는 미세 침전량이 전체 제거량 중 26.6 %를 차지 하고 칼럼내 crab shell 표면에서도 다른 중금속들보다 월등히 많은 미세 침전물이 관찰 됨으로 $Pb^{2+}$의 경우는 중금속 제거는 화학적 침전이나 물리적 침전에 따른 미세침전에 의해 많이 이루어지고 있는 것으로 판단된다. $Cu^{2+}$$Cr^{3+}$의 경우, 중금속의 고유 색깔인 청색이 중금슥 제거가 끝난 후 crab shell 표면에 착색된 것을 관찰할 수 있었다. Crab shell 충진 칼럼을 이용한 수중 중금속 제거 후 중금속을 고농도로 수거하기 위한 탈착에 있어서는 초기 20 BVs 내에서 대부분의 중금속이 탈착되어 유출되는 것을 관찰 할 수 있었다.

Preparation and Physicochemical Properties of Chitosan from Red Crab Waste-Shell (붉은대게 폐각으로부터 키토산의 제조 및 물리화학적 특성)

  • Chitosans were prepared from red crab chitin under various alkali treatment conditions(different alkali concentrations, reaction times and temperatures) and theirphysicochemical properties were investigated. The nitrogen content and deacetylation degree of red crab chitin were 6.15% and 22.17A%, respectively. By the IR spectra, red crab and reference chitin showed the sharp bands at 1650 $cm^{-1}$ / and 1550 $cm^{-1}$ /, which are characteristic of chitin. The nitrogen contents of prepared chitosans ranged from 6.19~7.48%. Thedeacetylation degree was increased from 63~76% and 48~78% with increasing reaction time and temperature, whereas viscosity was decreased. The nitrogen content and yield of red crab chitosan perpared from chitin with 50% NaOH, 1:25(w/v) for 3.0 hr at 120$cm^{-1}$ / were 7.26% and 85.0%, respectively. and viscosity, deacetylation degree and molecular weight, 67.0 mPa.s, 75.0% and 6.5$\times$10$^{5}$ Dalton, respectively. From the IR spectra, the amide absorption bands of red crab and reference chitosan became very weak, similarly. And at solid state $^{13}$ C-NMR spectra, C=O(carbonyl carbon) signals absent, whereas $CH_3$(methyl carbon) was residues. Chemical shift of $^{13}$ C-NMR spectra of red crab and reference chitosans were in good agreement with slight experimental deviation.

Characteristics of Lead Removal in a Fixed-Bed Column Packed with Crab Shell Particles (게껍질 충진 칼럼에 의한 납의 제거 특성)

  • Crab shell particles (Protunus trituberculatus) and activated carbon (Norit 0.8 SUPRA) were used as packing material in a fixed-bed column. When 1 g crab shell was added in the column packed with 10 g activated carbon. breakthrough to influent 10 mg Pb/L occurred at 1520 bed volumes. as compared to 380 bed volumes for 10 g activated carbon only. Addition of crab shell particles into activated carbon column resulted in increased uptake of lead. The dramatic improvement might be attributed to an increase in $co_3{^{2-}}$ and $OH^-$ available for the binding of lead. From the result of analyses, the major mechanism of lead removal was based on dissolution of $CaCO_3$ in the crab shell followed by precipitation of $Pb_3(CO_3)_2(OH)_{2(s)}$ on the surface of activated carbon. The lead uptake increased two fold as the influent lead concentration was increased from 10 to 50 mg/L. However, it decreased by 40% as the influent pH was lowered from 5.0 to 3.0.

Antioxidant and Angiotensin I Converting Enzyme Inhibitory Activities of Red Snow Crab Chionoecetes japonicas Shell Hydrolysate by Enzymatic Hydrolysis

  • We investigated the antioxidant and angiotensin I converting enzyme (ACE) inhibitory activities of red snow crab Chionoecetes japonicas shell (RSCS) hydrolysate by enzymatic hydrolysis and its molecular weight cut-off fractions. The RSCS hydrolysate was fractionated through two ultrafiltration membranes of 3 and 10 kDa cut-offs. Three fractions (<3 kDa, 3-10 kDa, and >10 kDa) were evaluated for total amino acid composition, antioxidant activities using 2'-azino-bis[3-ethylbenzthiazoline-6-sulfonic acid] ($ABTS^+$) radical scavenging and superoxide dismutase (SOD)-like activities and reducing power assays, and ACE inhibitory activity using Hou's method. Although all fractions showed activity, the <3 kDa fraction of RSCS hydrolysate exhibited the greatest $ABTS^+$ radical scavenging, SOD-like and ACE inhibitory activities. However, these fractions exhibited low reducing power. These results suggest that the low-molecular-weight enzymatic hydrolysate of RSCS could be used as a functional ingredient to control oxidative stress and ACE activity.

Mesh selectivity of gill net for swimming crab Potunus trituberculatus in the western coastal waters of Korea (서해 꽃게 자망의 망목 선택성 연구)

  • A series of fishing experiments were carried out in the western coastal waters of Korea from May, 2006 to September, 2007, using gill nets of different mesh sizes (m=101, 121.8, 152.5, 176.8, and 191 mm) to determine the mesh selectivity of gill net for swimming crab, Potunus trituberculatus. The catch species was composed of swimming crab (87%), Thomas's rapa whelk Rapana venosa (5%), arthritic neptune Neptunea cumingi (2%), Japanese swimming crab Charybdis japonica (2%), marbled sole Pleuronectes yokohamae (1%), pen shell Atrina pectinata, ocellate spot skate Okamejei kenojei, and so on. The carapace length (CL) of each swimming crab caught in the experimental nets was measured. Sponge crab (egg-bearing female crab) was appeared from the end of June to August in the coastal waters. The master selection curve was estimated by applying the extended Kitahara's method. The selection curve showed that the gill nets of larger mesh size allowed more crabs of small carapace size to escape. The optimum value (CL/m) was 0.645 and the CL/m was estimated to be 0.301, 0.354, 0.391, 0.422, and 0.450 when the retention probabilities were 0.1, 0.2, 0.3, 0.4 and 0.5, respectively.

Hydroponic Culture of Leaf Lettuce Using Mixtures of Fish Meal, Bone Meal, Crab Shell and the Pig Slurry Leachate of Woodchip Trickling Filter (목편살수여상 침출액비와 어분, 골분, 게껍질 혼합액을 이용한 상추의 수경재배)

  • The pig slurry leachate was dark brown-colored solution that leaches out of woodchip trickling filter. The purpose of this research was to investigate the effect of pig slurry leachate and byproduct on growth characteristics of leaf lettuce in hydroponics culture. The effects of addition of fish meal, bone meal and crab shell for the growth of leaf lettuce were investigated. Leaf lettuce were grown in each of six combination treatment solutions; slurry leachate, slurry leachate + fish meal, slurry leachate+bone meal, slurry leachate + crab shell and chemical hydroponic solution for lettuce based on EC content. The chemical nutrient solution was the solution of National Horticulture Research Station for the growth of lettuce. The all of nutrient solution was adjusted 1.5 mS/cm in EC in hydroponics culture. 1. The pH level of leachate of trickling filter was increased and EC decreased gradually during treatment. Pig slurry leachate was low in suspended solids (SS), phosphorus (P), but rich in potassium (K). 2. The plot of slurry leachate (SL) was lowest in the growth characteristics of lettuce. The leaf length and width of lettuce treated with mixture plot of slurry leachate and fish meal (SL + FM) was higher compared with plot in slurry leachate. The chlorophyll reading was reduced in plot treated with slurry leachate, but that in plot of SL+FM was similar compared with control plot. 3. The fresh weight of lettuce showed lowest in the plot treated with slurry leachate. The addition of fish meal increased the yield of comparing plot of slurry leachate, but plots of bone meal and crab shell addition were not significantly difference. The fresh weight of leaf lettuce in plot of SL+FM was 87% as 400.0g compared with control. In conclusion, the mixture solution of pig slurry leachate and fish meal could be used as a nutrition solution of organic lettuce hydroponics.

Physical Properties of Chitosan Film made from Crab Shell (꽃게 껍질에서 분리제조한 키틴산 필름의 물성에 관한 연구)

  • Chitin was isolated from the residue of enzymatically hydrolyzed crab, Portunus trituberculatus, and further deacetylated by alkaline boiling to make chitosan. The physical properties of chitosan solution and its film forming properties were examined. The functional characteristics of chitosan film were compared to those of cellophane, polyvinyl chloride (PVC) and polyethylene (PE) films. The proximate chemical composition of chitin obtained from crab residue was 6.95% nitrogen, 0.3% crude ash and 4.57% moisture and the product yield was 12.8% based on a dry material basis. The degree of deacetylation of chitosan was $79{\sim}92%$ and $70{\sim}86%$ as determined by IR spectroscopy, and $70{\sim}86%$ as determined by colloid titration method each respectively. The chitosan at 1% acetic acid solution showed distinct pseudoplastic flow behavior. The flow behavior index and consistency index were 0.8886, 0.2084 $MPa{\cdot}s^n$ for 0.4% solution and 0.8498, 0.6190 $MPa{\cdot}s^n$ for 0.8% solution, respectively. The chitosan film had the highest tensile strength $(888 kg/cm^2)$ and water permeability $(100\;g/m^2{\cdot}24\;hrs)$ among the tested films, but relatively low elongation property (49%). It showed the similar tear strength (90kg/cm) and light permeability (87.7%) to other films tested in spite of the relatively high haze value (12.5%). As the thickness of chitosan film increased from 0.025 to 0.050 mm, the tensile strength of film decreased distictively, and the degree of elongation, tear strength, and water permeability of film also decreased slightly. Whereas the light permeability of film did not change and the haziness of film slightly increased by the increase of film thickness.

Quality Characteristics of Kimchi Added with Blue Crab (꽃게를 첨가한 김치의 품질특성)

  • This study was carried out to investigated the quality characteristics of kimchi, made by adding blue crab. Kimchi is made into a fermented vegetable food by adding red pepper powder, garlic, ginger and fermented salted fish to the salted cabbage or radish. Chitin/chitosan, found in the shell of crab is the biopolymer. Chitin possesses many beneficially biological properties. Salt consumption impacts on human health problems such as hypertension and cardiovascular disease. In this study, we hypothesized that kimchi added with raw blue crab would reduce excessive salt consumption and increase protein supplementation. We analyzed lactic acid bacteria level, pH, acidity, salinity and free amino acid of kimchi added with blue crab during 1, 15 and 30 days storage period at $5^{\circ}C$. Lactic acid bacteria, pH, salinity increased significantly in kimchi added with blue crab compared to the control. Accordingly, this result suggested that kimchi should be manufactured by adding raw fish rather than fermented salted fish.

In vitro Study on the Functionality in Digestive Tract of Chitin and Chitosan from Crab Shell (게껍질 Chitin 및 Chitosan의 소화관내 기능성에 관한 in vitro 연구)

  • Chitin and chitosan samples prepared from crab's shells under different conditions were compared for their physicochemical properties; and functionality in gastrointestinal tract by in vitro test. Their bulk density was in the range of $127{\sim}208\;mg/ml$, and their viscosity was $80{\sim}581\;cP$ in 0.1 chitin and $80{\sim}3,670\;cP$ in 0.5% chitosan solution, showing a wide variation. The degree of deacetylation in chitosan samples as determined by IR spectral analysis was relatively high, showing $81{\sim}93%$. At the same alkali concentration and reaction temperature, a longer reaction period gave an increased degree of deacetylation and lower viscosity. The water holding capacity of chitic substance became greater at higher soaking temperature; chitosan D at $37^{\circ}C$ showed the greatest value. Chitic substance with lower bulk density showed the higher water holding capacity. The retardation effect toward glucose absorption was higher in critic substances of lower density and higher water holding capacity; chitosan D showed the highest value of 38%. The retardation index toward bile acid absorption after 1 hour dialysis was $15{\sim}34%$ in chitic substances, 39% in pectin and 9% in cellulose. The retarding effect showed the highest value of 34% in chitosan D at 3% concentration.
