Crataegus pinnatifida Bunge root extract induces apoptosis of murine lung carcinoma cells in vitro

  • This study sought to evaluate the anticancer effects of Crataegus pinnatifida Bunge root extract (CPE) on murine Lewis lung carcinoma cells (LLC1) in vitro. CPE treatment (2.5, 5, 10 ㎍/mL, 24 h) of LLC cells led to a dose-dependent decrease in cell viability, while CPE treatment did not have a cytotoxic effect on non-cancer cells (NIH/3T3). CPE affects LLC by flipping the plasma membrane and making the membrane more permeable; by flow cytometry, CPE-induced annexin V and propidium iodide positivity, indicating induction of apoptosis in LLC cells. In addition, CPE enhanced the expression of apoptotic proteins caspase-3 and poly (ADP-ribose) polymerase 1 (PARP-1). CPE upregulated the proapoptotic protein BCL-2-associated X while downregulating the anti-apoptotic protein B-cell lymphoma 2 (BCL-2), suggesting that CPE induces apoptosis via the mitochondrial pathway. Furthermore, CPE upregulated the phosphorylation of the mitogen activated protein kinase p38. In conclusion, the results suggest that CPE has an anticancer effect in LLC cells by inducing apoptosis via p38.

Taxonomy and Phylogeny of Neosiphonia japonica (Rhodomelaceae,Rhodophyta) Based on rbcL and cpeA/B Gene Sequences

  • Neosiphonia japonica is a rhodomelacean red alga that occurs in Korea, Japan, China, far-east Russia, northwest America, and New Zealand. Although it is distinguished by a bush-like habit having four pericental cells with cortication and numerous branches on axes, the taxonomy of N. japonica is still problematic. To investigate the taxonomy and phylogeny of the species, we analyzed rbcL and phycoerythrin (cpeA/B) genes from 19 samples of N. japonica and putative relatives. Phylogenetic trees from both genes show that N. japonica from Korea, Japan, New Zealand, and USA is clearly separated from N. decumbens, N. harlandii, and N. flavimarina from the Pacific Ocean. Instead, N. harveyi from the Atlantic Ocean was more related to N. flavimarina than to N. japonica. This result supports morphological and distributional differences between N. japonica and N. harveyi. However, the close relationship between these species suggests that they might have a recent most common ancestor. This is the second report to use the cpeA/B gene for evaluating species diversity in the Rhodophytes.

Highly-sensitive Detection of Salvianolic Acid B using Alumina Microfibers-modified Electrode

  • Alumina microfibers with porous structures were prepared through hydrothermal reaction, and then used to modify the surface of carbon paste electrode (CPE). After modification with alumina microfibers, the electrochemical activity of CPE was found to be greatly improved. On the surface of alumina microfibers-modified CPE, the oxidation peak current of salvianolic acid B, a main bioactive compound in Danshen with anti-oxidative and anti-inflammatory effects, was remarkably increased compared with that on the bare CPE surface. The influences of pH value, amount of alumina microfibers and accumulation time were studied. Based on the strong signal amplification effects of alumina microfibers, a novel electrochemical method was developed for the detection of salvianolic acid B. The linear range was from 5 ${\mu}gL^{-1}$ to 0.3 mg $L^{-1}$, and the detection limit was 2 ${\mu}gL^{-1}$ (2.78 nM) after 1-min accumulation. The new method was successfully used to detect salvianolic acid B in ShuangDan oral liquid samples, and the recovery was over the range from 97.4% to 102.9%.

Carboxypeptidase E Is a Novel Modulator of RANKL-Induced Osteoclast Differentiation

  • Osteoclasts are large polykaryons that have the unique capacity to degrade bone and are generated by the differentiation of myeloid lineage progenitors. To identify the genes involved in osteoclast development, we performed microarray analysis, and we found that carboxypeptidase E (CPE), a prohormone processing enzyme, was highly upregulated in osteoclasts compared with their precursors, bone marrow-derived macrophages (BMMs). Here, we demonstrate a novel role for CPE in receptor activator of NF-${\kappa}B$ ligand (RANKL)-induced osteoclast differentiation. The overexpression of CPE in BMMs increases the formation of tartrate-resistant acid phosphatase (TRAP)-positive multinuclear osteoclasts and the expression of c-Fos and nuclear factor of activated T cells c1 (NFATc1), which are key regulators in osteoclastogenesis. Furthermore, employing CPE knockout mice, we show that CPE deficiency attenuates osteoclast formation. Together, our data suggest that CPE might be an important modulator of RANKL-induced osteoclast differentiation.

Genetic diversity of Clostridium perfringens form food-poisoning outbreak in Gyeonggi-do, 2013-2014 (경기도 식중독에서 분리된 Clostridium perfringens의 유전적 특성 분석)

  • Clostridium perfringens is both a ubiquitous environmental bacterium and a major cause of human gastrointestinal disease, and C. perfringens food poisoning ranks among the most common gastrointestinal diseases in developed countries. 120 isolates of C. perfringens were obtained from food-poisoning outbreaks in 2013~2014, Gyeonggi-do. Using PCR, all 120 isolates were identified as C. perfringens type A. Of the tested isolates, 49 isolates carried the cpe gene, 71 isolates carried the cpb2 gene. The outbreak cases of cpb2 and cpe /cpb2 genes were 7 and 7, whereas the outbreak cases of cpe-gene were 2. The epidemiological relationship between C. perfringens isolates has previously been investigated chiefly by pulsed-field gel electrophoresis (PFGE) and multi-locus sequence typing (MLST). The genetics relatedness of the isolates raged from 53.5-100% and 75 district PFGE type were observed. The PFGE results revealed a wide genetic diversity among the 64 cpb2 carrying isolates (except 7 isolates), while 46 cpe-carrying isolates (except 3 isolates) showed a high genetic similarity. The MLST analysis revealed that 14 cpe isolates (cpe-chromosomal isolates) belong to a distinct cluster that is significantly distant from all the other cpb2 isolates (cpe-plasmid carrying and cpe-negative isolates). The isolates carrying a cpb2 appear to be rarely related, and are more variable than chromosomal cpe isolates. The results suggest that the cpe-positive outbreak isolates showed close genetic relation, whereas the cpb2-positive isolates revealed a wide genetic diversity.

Effect of Calamansi Pulp Ethanol Extracts on the Meat Quality and Biogenic Amine Formation of Pork Patty during Refrigerated Storage

  • This study evaluated the antibacterial and antioxidant activities of ethanol extract of calamansi pulp (CPE) and its effect on quality and biogenic amine (BAs) formation in pork patties during storage. The CPE were prepared in various conditions (ethanol concentrations of 50%, 70%, and 90% with extraction periods of 3 and 6 days). The extract with potent antibacterial and antioxidant activities (90%, 6 days) was selected for addition to pork patties. Three groups were tested: Control (without extract addition), CPE addition at 0.2% w/w (0.2PCPE), and 0.4% w/w (0.4PCPE). The addition of CPE inhibited the formation of BAs, mainly cadaverine, histamine, and tyramine, in pork patties during storage. The pH and bacterial count of pork patties decreased significantly in a concentration-dependent manner following the addition of CPE. The instrumental color (CIE L*, CIE a*, and CIE b*) tended to be higher in 0.4PCPE than in the control during storage. The thiobarbituric acid reactive substances and volatile basic nitrogen (VBN) values of pork patties were affected by CPE, showing a reduction toward lipid oxidation at any storage period, and maintaining the lowest VBN value in 0.4PCPE at the final storage day. Similarly, the reduction of total BAs in pork patties was observed ranged between 3.4%-38.1% under treatment with 0.2% CPE, whereas 18.4%-51.4% under 0.4% CPE addition, suggesting significant effect of CPE to improve meat quality. These novel findings demonstrate the efficacy of 0.4% CPE as a natural compound to preserve the quality and reduce BAs formation in pork patties during storage.

Investigation of Equivalent Circuit for PEMFC Assessment (고분자 전해질 FC 평가용 등가회로 검토)

  • Chemical reactions occurring in a PEMFC are dominated by the physical conditions and interface properties, and the reactions are expressed in terms of impedance. The performance of a PEMFC can be simply diagnosed by examining the impedance because impedance characteristics can be expressed by an equivalent electrical circuit. In this study, the characteristics of a PEMFC are assessed using the AC impedance and various equivalent circuits such as a simple equivalent circuit, equivalent circuit with a CPE, equivalent circuit with two RCs, and equivalent circuit with two CPEs. It was found in this study that the characteristics of a PEMFC could be assessed using impedance and an equivalent circuit, and the accuracy was highest for an equivalent circuit with two CPEs.

A Study on the EM Wave Absorber for the Electromagnetic Environment of Indoor Wireless LAN (실내 무선 LAN 전자파 환경 대책용 전파 흡수체에 관한 연구)

  • Recently, wireless LAN are often applied in home or office because of its various of convenience. Frequency rage of wireless LAN specified by IEEE 802.11a is at 5.2 GHz and IEEE 802.11b is 2.4 GHz. But in offices with wireless LAN devices, reflection of waves against walls, ceilings, floors and desks made of metal creates multipath problems that reduce communication speed and lose data. These problems can be solved by using EM wave absorber. In this paper, we designed and fabricated EM wave absorbers using MnZn-ferrite, sendust, carbon and CPE(Chlorinated Polyethylene). The EM wave absorber with the ratio of MnZn-Ferrite : sendust : CPE=64 : 16 : 20 wt.% has thickness of 3.7 mm and absorption ability more than 17 dB at 2.4 GHz and the EM wave absorber with the ratio of MnZn-ferrite : carbon : CPE=40 : 15 : 45 wt.% has thickness of 3.8 mm and absorption ability more than 23 dB at 5.2 GHz.

Development of EM Wave Absorber for Countermeasure against EM Wave Environment of 2.4 GHz Wireless LAN (2.4 GHz 무선LAN 전자파 환경대책용 전파흡수체 개발)

  • In this paper, the EM wave absorber was designed and fabricated for improvement of Wireless LAN environment at 2.4 GHz. We fabricated several samples in different composition ratios of Sendust and CPE(Chlorinated Polyethylene). Absorption abilities were simulated in accordance with different thicknesses of the prepared absorbers and changed complex relative permittivity and permeability due to composition ratio. The mixing ratio of Sendust and CPE was searched as 80: 20 wt.% by experiments and simulation Then the EM wave absorber was fabricated and tested using the simulated data. As a result, the EM wave absorber was fabricated based on simulated data. Simulated and measured results agreed well. As a result, the developed EM wave absorber with thickness of 3.25 mm has absorption ability of 19 dB at 2.4 GHz.

A Study on the EM Wave Absorber for Eliminating False Images in Collision-Avoidance Radar (충돌방지레이더의 허상방지용 전파흡수제에 관한 연구)

  • In this paper, we developed the EM wave absorber for eliminating false images in collision-avoidance radar. First of all, we fabricated some samples in different composition ratio of $TiO_2$ and CPE. And the relative permittivities of samples are calculated from S-parameter of samples by using 1-port method. We designed and fabricated the EM wave absorber by using the calculated relative permittivity. As a result, the EM wave absorber with composition of $TiO_2$:CPE=70:30 wt% has thickness of 1.85 mm and absorption ability higher than 20 dB in the frequency range 76-77 GHz.
