This study describes the effect of the growth regulators such as 2,L-D and BA, on the structural abnormalities of somatic embryos derived from leaf explants of Angelica gigas Nakai, Also, the relationship between the cotyledon number of a somatic embryo and its germinability is explored. Embryogenic calli were selected from calli formed on explants cultured on MS solid basal medium supplemented with 0.5mg/L 2,4-D, 1mg/L 2,4-D, 1mg/L plus 0.1mg/L BA, and 1 mg/L 2,4-D plus 0.5mg/L BA. Cotyledonary abnormalities were observed in somatic embryos which were developed from embryogenic calli cultured on MS medium containing 1mg/L 2,4-D for 8 weeks and then subcultured on 2,4-D free MS medium for 3 weeks. The frequency of abnormalities was as follows: 22.8% one cotyledon, 42.5% two cotyledons, 16.8% three cotyledons, 7.8% four cotyledons, 1.8% five cotyledons, and 8.2% jar shaped cotyledon. In addition, ABA treatment indicated an improvement of the somatic embryo with normal cotyledon (65.3%). ABA was important role to the high production of normal somatic embryos. Two cotyledon embryos showed germinability 77.8%. However the germinability of somatic embryos with anomalous cotyledons was prominently low: One cotyledon, 62.5%; three cotyledons, 43.3%; four cotyledons, 60%; five cotyledons, 50% and jar shaped cotyledon, zero%. Thus, germinability was essentially, inversely proportional to cotyledon number.

A successful, efficient system for multiple shoot induction and plant regeneration of soybean (Glycine max) was established. Four soybean genotypes were compared for organogenic responses on various media cultured under light conditions. The adventitious shoots (98%, 2.6 shoots/cotyledon) directly from one-day-old cotyledon after germination induced by the hormone treatment and its efficiency was higher than any other conditions. The optimal medium for the induction of multiple shoots from cotyledon in Pungsannamulkong(shoot formation rate, 98%), Lx 16 (83%) and IIpumgeomjeongkong(63%) was MS medium supplemented with 2 mg/L BAP, but for Alchankong(75%), MS medium supplemented with 1mg/L zeatin and 1mg/L IAA, 3% sucrose, 4% Phytagel. Higher root induction (88%) was observed from the shoots placed on rooting medium (hormone-free MS basal). Plantlets were transferred onto the same medium supplemented with 1% activated charcoal for further development. With this treatment, regenerated plantlets were obtained within 7-8 weeks (shoot induction for 4 weeks, rooting and shoot elongation for 3-4 weeks).
An efficient plant regeneration system has been developed for figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), which is exclusively used as a rootstock for cucumber. The protocol is based on results obtained from a series of culture experiments involving different parts of the cotyledons and various media. The culture of cotyledon explants was critical for the enhancement of shoot regeneration frequency. The lower parts of the cotyledon excised at the plumule base were found to display a markedly enhanced production of adventitious shoots compared to other cotyledon regions. Culture in silver nitrate-supplemented Murashige and Skoog (MS) medium was not beneficial for shoot regeneration and suppressed root regeneration. Efficient shoot regeneration was obtained on MS medium containing 1.0 $mg\;l^{-1}$ zeatin and 0.1 $mg\;l^{-1}$ indole-3-acetic acid. Regenerated shoots successfully elongated and rooted in medium containing 0.1 $mg\;l^{-1}$ 1-naphthalene-acetic acid after 10-15 days of subculturing. The plantlets were satisfactorily acclimatized in a greenhouse and grew into normal plants without any morphological alterations.
Cotyledon of Panax ginseng was cultured in the growth regulator-free Knudson C medium comprising only several kinds of mineral salts and sucrose. Shoot primordium or callus developed profusely from the cotyledonary tissue and finally plantlets were produced directly from the shoot primordium or indirectly through callus. Microscopic examination revealed that the epidermal cell as well as the mesophyll cell of the cotyledon became meristematic and divided, changing into multinucleate cell or multicellular body, eventually developing into either a shoot primordium or callus.
This study was conducted to determine changes in th contents and composition of dietary fiber during the growth of soybean sprout. Soybean was soaked in water at $25^{\circ}C$ for 2hrs and cultivated at 2$0^{\circ}C$ for 7 days under dark condition. The soybean sprouts were divided into cotyledon and axis and sampled every 24hrs. The analysis methodlogies used were Van Soest's NDF, AOAC's ADF and lignin and Prosky's IDF, SDF, TDF. The weight of 100 sprouts increased gradually from 20.26g to 90.12g during the growth periods. The weight increased to 344.9% of the original weight. The germination rate was 100% after soaking at $25^{\circ}C$ for 2hrs. Root length increased gradualy from 0.6cm at 1st day to 17.2cm at 7th day. The crude ash and crude fat contents showed no significant change in the cotyledon and axis. The crude protein contents increased in the cotyledon and axis, whereas the total carbohydrate content didn't have general tendency. The insoluble dietary fiber(IDF), soluble dietary fiber(SDF) and total dietary fiber(TDF) contents of cotyledon were no significantly different from 20.01%, 1.45%, 21.46% at 1st day to 22.75%, 2.07%, 24.82% at 7th day on dry basis. In axis those contents increased from 23.19%, 1.97%, 25.16% at 1st day to 32.78%, 3.02%, 35.80% at 7th day, respectively. The neutral detergent fiber(NDF) contents of cotyledon and axis increased from 4.35% to 6.39% and from 6.44% to 26.60% respectively on dry basis. The acid detergent fiber (ADF) contents of cotyledon and axis increased from 2.84% to 4.91% and from 2.5% to 4.7%, but there were no significantly different in the hemicellulose and lignin contents on dry basis. The hemicellulose and lignin contents of axis increased with culture periods from 1.70% to 4.41% and from 0.20% to 2.11%, respectively. The cellulose contents increased from 4.54% to 20.35% on dry basis.
This experiment was conducted to determine seed storage protein pattern and structural character of differed peanut cultivars during germination. Soluble protein content in both Namdae and Daekwang cultivars remarkably decreased in cotyledon site at 2 or 3 days after incubation(DAI) and in embryonic axis site at 1 or 2 DAI, showing 28∼29% in cotyledon site and 10% in embryonic axis site at 5 DAI. Protein subunits such as 66, 43, 40 and 35.5kD bands in the cotyledon site of Namdae and Daekwang cultivars disappeared, but 21.5-23kD band disappeared slightly, but low polypeptide band such as 14-16kD increased gradually, and the same trend has been obserbed in embryonic axis site during 2 DAI. The amount of new protein formed during germination period was highest in cotyledon site at 3 DAI, and in embryonic axis site at 2 DAI. 16kD bend detected in cotyledon site of Daekwang cultivar during germination.
This investigation was to determine the changes in the trypsin inhibitor activity(TIA) and electrophoresis patterns of the soybean cotyledon and axis during germination. The TIA of the cotyledon decreased slightly and that of the axis decreased rapidly to 50% activivity after 7 day germination. At the 2nd, 3rd and 4th day's germination the TIA of the defatted dry axis was higher than that of cotyledon. However, the TIA of the fresh cotyledon was lower than that of the axis, due to its higher moisture content. Results from the electrophoretic studies showed that band 1 (polymer, 15S etc.), 2(11S), and 3(7S) whichare the major reserve proteins of soybean were decreased consid erably in cotyledon and axis and the fragments with Rm values between 0.5 and 1.0 were increased and band 5 showed up during germination. The band 4 of the cotyledon and band 6 of axis were not changed during germination. Generally speaking, the TIA and thereserve protein decreased as germination proceed.
Embryogenic callus was obtained from cotyledonary explants of Codonopsis lanceloata on Murashige & Skoog's medium supplemented with 1 mg/L 2,4-D. Suspension cultures of the embryogenic calli were grown on a shaker at 100 strokes/min, and then the calli were subcultured for 2 weeks in 2,4-D-free medium to produce somatic embryo. In somatic embryos at the globular stage, cotyledon initials began to differentiate themselves in the near distal end of the procambial strand. Dicotyledons, tricotyledon, tetracotyledon and fused cotyledon were differentiated from the distal ends of two, three, four and circular procambial strands, respectively. Nearly circular procambial strand in lower hypocotyls were independently differentiated into two, three, four procambial tissues at cotyledonary node and cotyledons to form somatic embryos with dicotyledon, tricotyledon, tetracotyledon. If the distal subepidermal cells of globular embryo exclusively became cotyledon initials, the torpedo or cotyledonary embryo was characterized by somatic embryos with fused cotyledon.
Immature embryos of Glycine max L. was cultured on Murashige and Skoog's (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D). After 6 to 8 weeks of culture, immature embryos produced somatic embryos. Of somatic embryos, two cotyledonary embryo (14%), one cotyledonary embryo (37%), fused cotyledonary embryo (43%), and stunted globular embryos (6%) were observed. The procambial strand of cotyledons originated from circular procambial tissues of lower hypocotyl. The circular procambial tissues were independently divided into one or two procambial strand at the edge of cotyledonary-node, and then connected to each cotyledon to form somatic embryos with one or two cotyledons. When cotyledon was a fused type, the circular procambial strand in lower hypocotyl was continuously connected to the cotyledon. Also, somatic embryos with two cotyledons developed a functional shoot apex with the tunica-corpus structure. In contrast, somatic embryos with one or fused cotyledon formed an abnormal shoot apex without the tunica-corpus structure or with non-dome shape in the inter-cotyledonary area. These results indicated that the variation of cotyledon in somatic embryos is closely related to procambial differentiation and shoot apical meristem development.
Low seedling growth rates of white clover (Trifolium repens L.) have been limited its good establishment to pastures. The experiment was done to determine the effect of removal of cotyledon and unifoliolate on the growth and morphological characters of contrasting white clover cultivars for 8 weeks after the treatment. Individual plants of cv. Regal (large leaf), Louisiana S-l (medium-large leaf), Grasslands Huia (medium-small leaf) and Aberystwyth S184 (small leaf) were grown in 10cm plastic pot containing a 2:1:1 soil:sand:peat moss mixture until the cotyledon or unifoliolate stage and then removed one (C1) or two cotyledons (C2) at cotyledon stage, and unifoliolate only (U), unifoliolate and one cotyledon (DC1) or unifoliolate and two cotyledons (DC2) at the unifoliolate stage. To measure the removal effect on biomass and morphological characters (leaf area, petiole and stolon lengths, growing tips and leaves), plants were sampled 4, 6 and 8 weeks after the treatment. Intact plants had greater biomass and morphological characters than removal-treated ones, Removal treatments at cotyledon stage, C1 and C2, were decreased more biomass and morphological characters than removal ones at unifoliolate stage while the severer cotyledon removal, the more reduction. Stolon length per plant and petiole length markedly inclined 6 weeks after the treatments although biomass and the other characters continuously did. Relatively large-leaved cultivar had more biomass, leaf area per plant and longer petiole than the other(s) but the reverse results were true in stolon length, growing tips and no. of leaves per plant. Biomass was linearly increased with increased leaf area but the earlier and severer removal, the less slope.
