Studies on the Media Development of Pleurotus ostreatus by Waste Cotton Stuff (느타리버섯 배지개발을 위한 폐면포 이용에 관한 연구)

  • Media development of Pleurotus ostreatus were analyzed to recognize the compositional differences depending on rice straw and waste cotton stuff media. Yields of add the rice bran 20% to the waste cotton stuff were increment 11% as compared with rice straw media. Crude protein of products to the waste cotton stuff media on Pleurotus ostreatus was a much and crude ash was less than of rice straw media. Glutamic acid was most and cystine was lowest among the amino acids of Pleurotus ostreatus. Total amino acids and essential amino acids of waste cotton stuff were much than of rice straw media.

  • Amino acids of Pleurotus ostreatus in various cultural media(rice straw, sawdust and cotton waste) were analyzed to recognize the compositional differences depending on pileus size and portions(pileus and stipe) Total amino acids of rice straw were a little less than that of sawdust and cotton waste. Seventeen amino acids were identified and quantified. Among the amino acids glutamic acid was higest and cystine was lowest. The amino acid contents depending on the pileus size was higest in $3{\sim}7\;cm$ of rice straw, under 3 cm of sawdust, over 7 cm of cotton waste, respectively. The content of them was found to be higher in pileus than stipe.

  • The results of examining cultivation of tlyster mushroom using NSM(Non Sterilized Media) for determining the condition of artificially culturing oyster mushroom(Pleurotus ostreatus) are as follows. Mycelial growth an[1 density of oyster mushroom. were the highest in the medium of waste cotton(spinning) : corn cob(80 : 20, V/V) followed by the order of rice bran, beet pulp. Expecially, mycelial growth and density of oyster mushroom is the lowest at the mixture rate of 80% waste cotton(spinning) : 10% beet pulp. Mycelial growth and density of oyster mushroom. were the highest in the medium of cotton seed hull and beet pulp mixture followed by the order of rice bran, corn cob. Expecially, mycelial growth and density of oyster mushroom is the lowest at the mixture rate of 80% cotton seed hull : above 20% rice bran. Mycelial growth and density of oyster mushroom were the highest in the medium rate of 70% waste cotton(spinning), 10% corn cob and 10% beet pulp(V/V). Mycelial growth and density of oyster mushroom were the highest in the medium rate of 70% cotton seed hull , 10% corn cob and 10% beet pulp(V/V). Optimal concentration of NSM for the mycelial growth and density of oyster mushroom were shown to be 500 times concentration. Optimal water contents for the mycelial growth and density of NSM was 70%.

Additive effect of tobacco waste in substrates on yield and mycelial growth of oyster mushroom, Pleurotus spp. and Trichoderma disease (느타리버섯 배지에 담배가루의 처리가 버섯균과 푸른 곰팡이병원균의 균사생장과 수량에 미치는 영향)

  • This study was carried out to investigate effect of adding tobacco waste to promote oyster mushroom, Pleurotus ostreatus, mycelial growth and control of Trichoderma disease. Tobacco waste was treated on the surface of rice straw inoculated with oyster mushroom strains. The mycelial growth treated with 0.1g tobacco waste was somewhat faster than non-treatment. In the case of mixed media with 1.0g tobacco waste, mycelial growth was increased compare to control, but adding 2.0 g tobacco waste decreased the mycelial growth. We also treated the tobacco waste on Trichoderma disease. All of the tobacco waste treatment force down the growth of Trichoderma disease. We treated tobacco waste on the surface of rice straw substrates. ASI 2072 mycelial growth was decreased regardless of amount of tobacco waster and method of treatment. Mycelial growth of ASI 2180 was somewhat decreased with treatment of 0.1g and 0.2g In the case of media mixed, mycelial growth of ASI 2072 was decreased in the overall treatment. Mycelial growth of other strains were decreased as only 2.0g treatment. Field test of mycelial growth was conducted using rice straw and waste cotton with substrates. Rice straw culture with treated 3% tobacco waste and cotton waste culture with treated 6% tobacco waste showed the highest yield. There was no effect to Trichoderma disease.

Characteristics of microorganism isolated from Cotton Waste Media for the Oyster Mushroom Cultivation (느타리버섯 균상재배 중 배지에서 분리한 미생물의 특성)

  • This study was carried out to investigate interaction between mushroom mycelium and microorganisms in cotton waste media for the shelf cultivation of oyster mushroom. Two oyster mushroom farms was selected for this experiment. One was good mushroom farm (farmhouse I) and the other failed mushroom farm (farmhouse II). In farmhouse I, the inhibition microorganisms were higher toward the end of growth stage than the early stage, but the result of farmhouse II was opposite. Effects of the mycelium growth on plate culture showed same results on mushrooms as the earlier one. And the mycelium growth was influenced by secretory materials of microorganisms. Among of the isolates, Only few microorganism had inhibitory effects on either P. tolaasii or T. harzianum causing the disease of oyster mushrooms. But more microorganisms had inhibition effects on P. agarici.

  • Main materials used as media for oyster mushroom cultivation are pine sawdust, beet pulp, cotton seed pomace. Increases in the price and the unbalance of demand and supply of cotton seed pomace was often damage to oyster mushroom cultivation farm, so we investigated agricultural by-product to replace the cotton seed pomace for bottle cultivation of oyster mushroom. Treatment of coconut oil meal or coconut pomace delayed incubation period about 3 days compared with cotton seed waste treatment(control), but yield and income index showed similar to each other in three treatment. Consequently coconut oil meal and coconut pomace could select for cotton seed pomace substitute.

  • Dental personnels faced risks of infection in the clinic. For infection control, recognition and practice of dental personnels are important factor. This study was performed to investigate the recognition and practice of dental hygiene for infection control and infection waste control. A stratified convenience sample of dental hygienists in dental health-care settings. The major finding of the present study are as follows: 1. The existence of education about Standard Precaution and low of infection waste storage was higher dental hospital than dental clinic. 2. The degree of practice in the storage of dental wastes was low in absorbent cotton and body tissue exclude damage waste. And the degree of practice in the disposal of dental wastes was high in all three. 3. Practice in the storage of dental waste was higher dental hospital than dental clinic. 4. At the conclusion of this investigation, systematic refresher training of infection control should be prepared by campaign an various media, Dental health care workers should be encouraged to practice those action items from training. For successful implementation of infection control in every dental health-care settings, it is highly demanded as well that development of effective safe-guard tools, stategic support, and standardized action items against infection problems.

Changes and Distributional Pattern of Microflora in Cotton Waste Media of Oyster Mushroom Cultivation (느타리버섯 균상재배 중 배지내 미생물상의 변화 및 분포양상)

  • The diversity of microflora according to growth stage of Pleurotus ostreatus and the correlation between microbe and medium fermentation were investigated. In farmhouse I, the aerobic bacteria and fungi as longer of growing period were increased. And, thermophilic bacteria and fluorescent Pseudomonas sp. showed high density at the early stage of spawn inoculation. The thermophilic actinomycetes were distributed evenly during all the growing period, but mesophilic actinomycetes were not observed. In farmhouse II, thermophilic actinomycetes were not observed in fermented medium and density of fungi were suddenly increased at 60 days after spawn inoculation. And also, mushrooms can hardly be harvested due to Penicillium spp. After medium fermentation, density of aerobic bacteria, thermophilic bacteria, and fluorescent Pseudomonas sp. was higher at farmhouse I than those of farmhouse II. In farmhouse I, Bacillus sp. and Pseudomonas sp. dominated at early stage of mushroom growth but as time goes by, density of Bacillus sp. was higher than the others. And also, the kind of microbe showed a few at early stage of mushroom growth but increased as time goes by. In farmhouse II, Bacillus sp. was dominated at early stage of mushroom growth. And the growth of Bacillus sp. and Pseudomonas sp. showed intersect aspect each other in the farmhouse I but Bacillus sp. dominated during all growth periods in the farmhouse II.

A Study on the Treatment of Radioactive Liquid Wastes using Synthetic textile by Air Intake System (공기유입시스템에서의 섬유매체에 의한 방사성액체폐기물 처리에 관한 연구)

  • In this study based on the mass transfer theory, experiments for the evaporation rates depending on various conditions were carried out through the operation of the existing Natural Evaporation Facility in KAERI. Evaporation media were made of the cotton and polyester. Air circulation in the facility was forced by exhausting fans. The evaporation rate and the decontamination factor were calculated by the result of experiment. The evaporation rate increased as the flow rate of air supply, the feed rate of liquid waste, and the temperature of supplied air increased. As for the humidity of supplied air, the evaporation rate was getting higher as the humidity was getting lower. As the result of this study, operation conditions of the Natural Evaporation Facility are optimized as follows : The air temperature above $8^{\circ}C$, the air humidity below 70%, the air flow rate 1.14-1.47 m/sec, and the liquid waste feed rate $4.6{\ell}/hr\cdotm^2$. The decontamination factor and the radioactivity are $5.1{\times}10^3$and $4.7{\times}10^{-13}{\mu}Ci/\textrm{m}{\ell}$ respectively, at the above mentioned optimum operation conditions. The air factor in the Dalton's equation for evaporation was determined from results of experiment on the temperature, the humidity, and the flow rate of supplied air as following : $[\textit{Eh}=(0.018 + 0.0141\textitv) {\delta}textitH]$

Biological Control of Cucumber Powdery Mildew by Bacillus amyloliquefaciens M27 (Bacillus amyloliquefaciens M27에 의한 오이 흰가루병의 생물적 방제)

  • Bacillus amyloliquefaciens M27 was isolated from the cotton-waste compost for cultivation of oyster mushroom (Pleurotus ostreatus). B. amyloliquefaciens M27 is a biocontrol agent with antagonistic activities against a wide range of fungal pathogens. The aim of this work was to evaluate the possibility of exploiting antagonistic bacteria, B. amyloliquefaciens M27, in the biological control of the cucumber powdery mildew fungus, Podosphaera fusca. In greenhouse tests, the isolate was found to be very effective to control powdery mildew on cucumber leaves showing 4.0% diseased area, whereas diseased area in the control was 80.5%. The filtrate of the isolate cultured on MH and LB media were more effective for control of the disease than those cultured on TSB, NB, and KB media. When two, five, ten, 20, 50 and 100-fold diluted culture broth of isolate on LB media were treated, disease areas were 0%, 0%, 0%, 1.3%, 3.1% and 5.0%, respectively, whereas diseased area in the control was 60.0%. The filtrate of the isolate cultured on LB media was treated to cucumber plants on July, October and December just before the outbreak of the powdery mildew occurred. When 10-fold diluted filtrate of the isolate was treated, control efficacy was 88.9~98.9% in the treated seasons. The results showed that the culture filtrate of B. amyloliquefaciens M27 was very effective to control powdery mildew of cucumber.
