The morphological changes in normal and weathering hair shafts of the human scalp were investigated by using the transmission and scanning electron microscopes. The hair shaft composed of cuticular layer, cortex and medula. The surface of normal hairs are smooth and covered by imbricated cuticular scales. The cuticular layer consists of five to seven cuticle cells. These cells, which are flat and thin, measuring about $100{\mu}m$ long and $0.4{\mu}m$ thick, appears intercellular membrane complex in diameter 25 nm. The cortex composed of melanin granules and cornified cells, which multicomponent concentric microfibrils in diameter about 8 nm give rise to macrofibrils in diameter $0.5{\mu}m$ to $0.8{\mu}m$ encased in limiting membrane. The melanin granules are spherical shaped about $0.5{\mu}m$ in size and scattered between macrofibrils. The medulla in the normal hairs are $16{\mu}m$ in diameter centrally region of cortex. Normal hair shafts undergo progressive degenerative changes due to a variety of environmental insults. In the initial weathering process of hair, the cuticular scales became irregularly raised and broken, and then cuticle cells formed cytoplasmic vacuolation, following dissociated intercellular membrane complex, ultimately entirely lost and nuded cortex. Occasionally, transverse fissures were seen at hair shafts indicating that the hairs were deteriorated. Complete removal of the cuticular layer in the heavily damaged cortex portions appeared splitting of the cortical cell into its macrofibrils and scattering of melanin granules.

Objective: Zona pellucida (ZP) has been thought to be the barrier of egg to sperm penetration before and after fertilization. The phenomenon of ZP hardening has been considered as a post-fertilization event until now, and it is generally accepted that it is caused by the secretory products of cortical granules released during the cortical reaction. Hardening of ZP could occur "spontaneously" in mammalian oocytes in standard culture conditions, and that it is probably not a consequence of cortical reaction. The purpose of our study was to investigate the effect of human amniotic fluid (HAF) on nuclear maturation (NM) and fertilization ability of mouse immature oocytes. Methods: HAF was obtained from patients undergoing amniocentesis at $16{\sim}20$ weeks of gestation. HAF from five to ten patients was centrifuged and the supernatants was pooled. Cumulusenclosed mouse immature oocytes were incubated in the medium containing HAF, and examined to confirm NM and fertilization. Female ICR mice (about 3 weeks old) were stimulated with 7.5 IU PMSG. Immature oocytes were isolated at $48{\sim}52$ hrs post PMSG injection and cultured in TCM-199 supplemented with 20% HAF for 18 hrs. FBS was used as a control for the examination. Matured oocytes (MII) were fertilized with sperms collected from the epididymis of male mice (over 10 weeks old). Fertilization was in conducted T6 medium containing 15 mg/ml BSA, and confirmed at 6 hrs post-insemination. Fertilization rate was assessed in zona-intact or zona-free oocytes (denuded by trypsin). Evaluation of NM and fertilization was carried out by rapid staining method. ZP hardening was evaluated by incubating cumulus cell-free mature oocytes in 0.001% chymotrypsin at $37^{\circ}C$ for 10 min. Results: There was no significant difference between the effects of HAF (86.6%) and FBS (87.7%) supplements on NM of immature oocytes. When maturation medium was supplemented with HAF, total fertilization rates (7%) were significantly lower (p<0.01) than that of FBS (85.1%). In HAF group, fertilization rate was increased (p<0.01) in zona-free oocytes (7% versus 100%). The resistance of mouse oocyte ZP to digestion by chymotrypsin after maturation in vitro was significantly higher (p<0.01) in HAF group (86.7%) than in FBS (6.7%). To culture oocytes in FBS were very effective in preventing ZP hardening. However cultured oocytes in HAF showed high rate of ZP hardening (p<0.01). Conclusions: These results suggest that HAF can be used as a supplement for the NM of mouse immature oocytes in vitro. However, HAF induces spontaneous hardening of ZP of mouse immaure oocytes during maturation in vitro.
A marine ciliate Amphisiella milnei (Kahl, 1932) $Horv{\acute{a}}th$, 1950 was discovered from the tidal pool of Baekdo Island, South Korea. The existence of extra cirri between leftmost frontal cirrus and buccal cirrus discriminates this species from its congeners. Its morphological features are described as follows: body size in vivo $110-130{\times}35-45{\mu}m$; elongate rectangular to elliptical in shape; two large and several small ring-shaped structures; yellowish cortical granules arranged irregularly on ventral side but longitudinally along dorsal kineties on dorsal side; 34-40 adoral membranelles, 3 frontal cirri, 1 buccal cirrus, 1 parabuccal cirrus, usually 2 extra cirri behind leftmost frontal cirrus, and 3 frontoventral cirri; amphisiellid median cirral row composed of 25-31 cirri with 27-36 left and 27-44 right marginal cirri; usually 5 transverse cirri and 2 pretransverse cirri with 7 dorsal kineties; two macronuclear nodules. In addition to, 18S rDNA sequence of A. milnei was analyzed to understand its phylogenetic relationship.
The ultrastructure of damaged hair shaft from blow-drying at typical temperature $40{\sim}170^{\circ}C$ for daily beautiful face have been investigated by using transmission electron microscope and scanning electron microscope. When we used to hair dryer for a long time in our everyday life, the following morphological alternations were found in hair. First, the partial of scales in outer cuticle were detached simultaneously with separation of intercellular membrane complex of cuticle cells. Then hair broke cuticle off and exposed to cortex. Secondly, the cortical cell in the cortex was fissured into its macrofibril. The melanin granules were scattered between macrofibrils. As a result, I confirmed that blow-drying removed the hair's bonded water and made hard on hair which lost elasticity. After all, hair showed irregular, rough surface and vanished its luster.
Two heterotrichid ciliates, Climacostomum virens (Ehrenberg, 1838) Stein, 1859 from brackish water and freshwater, and Fabrea salina Henneguy, 1890 from a solar saltern, were collected in Korea. They are novelly investigated in Korea by means of live observation, protargol staining and nuclear small subunit (SSU) rRNA gene sequencing. Climacostomum virens is characterized by pouch-like body shape, body length of $200-370{\mu}m$ in vivo, conspicuous cytopharyngeal tube, macronuclei ribbon-like shape, and one to four in number, with or without symbiont algae in cytoplasm, 34-66 somatic kineties, 67-113 adoral zone of membranelles, 8-42 peristomial kineties, 24-37 apical membranelles. SSU rDNA sequence size is 1,591 bp and GC contents 48.52%. Fabrea salina is also characterized by scoop-like body shape with proboscis, body length of $190-240{\mu}m$ in vivo, one to two rod-shaped macronuclei, oval micronuclei, grayish green cortical granules, 104-186 somatic kineties, 4-8 preoral kineties, 7-19 peristomial kineties and fragmented paroral membrane. SSU rDNA sequence size is 1,598 bp and GC contents 47.50%.
Two Spirostomum species collected from freshwater in Korea were identified as S. caudatum (Muller, 1786) and S. teres (Claparede and Lachmann, 1858). They are recorded for the first time in Korea. The description was based on the observation of living specimens and protargol impregnated specimens. Diagnostics of these species are as follows. Spirostomum caudatum: body size $400-450{\times}20-30\;{\mu}m$ in vivo, shaped long and slender with a tapered posterior part, highly contractile; macronucleus ellipsoid; adoral zone of membranelles occupied 30% of body length; somatic kineties 14-22 in number. Spirostomum teres: body size $240-460{\times}25-40\;{\mu}m$ in vivo, shaped long and slender with a flattened posterior end, highly contractile; cortical granules arranged in 2-3 rows; adoral zone of membranelles occupied 50% of body length; somatic kineties 20-30 in number; macronucleus ellipsoid; micronuclei 2-3 in number. Spirostomum caudatum and S. teres are the most similar congeners, but they are different in the posterior part of body (tail-like posterior part vs. flattened posterior end), length of adoral zone of membranelles in body length (1/3 vs. 1/2), and the number of somatic kineties (14-22 vs. 20-30). These populations match with European populations in morphological characters.
"Spontaneous" hardening of the zona pellucida of mouse oocytes during in vitro culture is most likely due to cortical granules exocytosis. Thus the purpose of the present study was to determine whether the exocytosis factor is involved in spontaneous zona pellucida hardening during in vitro culture of the mouse. The results obtained form these experiments were summarized as follows; 1. When a protein synthesis inhibitor(100${\mu}g$/ml puromycin) was added to the culture medium, it did not prevent spontaneous ZPH of mouse oocyte during in vitro culture. 2. Calmodulin antagonists (trifluoperazine and chlorpromazine) and calcium channel blocker (verapamil) had no inhibitory effect in spontaneous ZPH. 3. A microtubule assembly inhibitor, colcemid had some inhibitory effect on spontaneous ZPH. 4. Treatment with a microfillament formation blocker(cytochalasin-B) at 1${\mu}g$/ml concentration, resulted in the excellent inhibitory effect on spontaneous ZPH. However cytochalasin-B did not inhibit ethanol-induced ZPH.
Heterotrichous ciliates were isolated from marine and brackish habitats in Korea, and their morphology, infraciliature and morphometrical characters were investigated using silver impregnated methods. These three Condylostoma species were identified as Condylostoma curva Burkovsky, 1970, C. minutum Bullington, 1940 and C. spatiosum Ozaki and Yagiu in Yagiu, 1944. The species of Condylostoma based on morphology are highly variable in most of characters and distinguished as following. Condylostoma curva is oval to elongate ellipsoidal, size about $245{\times}100{\mu}m$ in vivo, and conspicuously dark greenish brown with cortical granules on their surface, buccal cavity relatively large about 40% of body length, 5-11 macronuclear nodules, 4-8 frontal cirri serially arranged and 22-38 somatic kineties. Condylostoma minutum is elongate ellipsoidal and the size about $310{\times}55{\mu}m$ in vivo, buccal field about 35% of body length, 12-20 macronuclear nodules, adoral zone consisted of 82-107 membranelles, large and long one frontal cirrus and 38-44 somatic kineties. Condylostoma spatiosum is large elongate ellipsoidal, size about $670{\times}105{\mu}m$ in vivo, buccal cavity about 25% of body length, 11-25 macronuclear nodules, 111-144 adoral membranelles, membrane-like 2 frontal cirri, 49-74 somatic kineties. These three Condylostoma species are described here for the first time in Korea.
Actin dynamics is critical for the formation and sustainment of the immunological synapse (IS) during T cell interaction with antigen-presenting cells (APC). Thus, many actin regulating proteins are involved in spatial and temporal actin remodeling at the IS. However, little is known whether or how actin stabilizing protein controls IS and the consequent T cell functions. TAGLN2 − an actin-binding protein predominantly expressed in T cells − displays a novel function to stabilize cortical F-actin, thereby augmenting F-actin contents at the IS, and acquiring leukocyte function-associated antigen-1 activation following T cell activation. TAGLN2 also competes with cofilin to protect F-actin in vitro and in vivo. During cytotoxic T cell interaction with cancer cells, the expression level of TAGLN2 at the IS correlates with the T cell adhesion to target cancer cells and production of lytic granules such as granzyme B and perforin, thus expressing cytotoxic T cell function. These findings identify a novel function for TAGLN2 as an actin stabilizing protein that is essential for stable immunological synapse formation, thereby regulating T cell immunity. Two poorly known and often confused pleurostomatid ciliates, Litonotus paracygnus Song, 1994 and L. pictus Gruber, 1884, were collected from the coastal waters of Yeonggeumjeong and Bongpo-port, Gangwondo in the East Sea and from the Iwon tide embankment near Ganwol-do, Chungcheongnam-do in the Yellow Sea, Korea. These species were described based on live observations, the protargol-impregnation and morphometrics of the species. Also provided are their diagnoses. The small subunit ribosomal DNA (SSU rDNA) sequences of these species were compared with previously known sequences of related species. The diagnostics of the two Litonotus species are as follows. L. paracygnus: 150-300 $\mu$m long in vivo, strongly contractile neck region, two ellipsoid macronuclei (Ma) and one micronucleus (Mi), 7 left (LSK) and 11-14 right somatic kineties (RSK), 2-4 contractile vacuoles (CV) located on the posterior end, extrusemes (Ex) distributed on the anterior region of the ventral margin only. L. pictus: about 200-600 $\mu$m long in vivo, extremely contractile, beautiful body color with rows of yellow to yellow-brownish cortical pigment granules, 12-21 Ma arranged in moniliform pattern, infrequently vermiform, 7-11 LSK and 18-26 RSK, several CV located on both margins, Ex distributed on the anterior region of the ventral margin only. In this study, this genus was firstly recorded in Korea.
