Corn-fiber의 희석된 황산 가수분해에 의한 L-arabinose의 생산 (L-Arabinose Production from Diluted Sulfuric Acid Hydrolysis of Corn-fiber)

  • 농산 폐자원 및 견과류 껍질의 산 가수분해를 통하여 L-arabinose를 생산하기 위한 최적조건의 결정에 대한 연구를 수행하였다. 다양한 원료들을 분말로 제조하여 희석된 황산으로 가수분해 시킨 결과 corn fiber에서 arabinose가 가장 많이 생성되어, 이를 원료로 선정하였다. Corn fiber로부터 arabinose를 생성하기 위한 산 가수분해 반응의 최적조건은 0.4%의 황산으로 130$^{\circ}C$에서 60분간 처리하는 것이며, arabinose의 생성 수율은 기질의 농도가 낮을수록 증가하였으나 생성농도는 기질의 농도가 높을수록 증가하였다. 최종적으로 90 g/L의 corn fiber를 최적조건에서 산 가수분해 시킨 결과 20.1 g/L의 glucose, 10.1 g/L의 xylose, 7.8 g/L의 arabinose 및 1.8 g/L의 galactose가 생성되었다. 산 가수분해 용액을 암모니아수로 pH를 5.5로 조정하고 C. tropicalis를 접종하여 배양한 결과 7.6 g/L의 arabinose, 0.6 g/L의 xylose, 0.5 g/L의 xylitol 및 0.5 g/L의 galactose만이 존재하였다. 이는 효모의 배양에 의하여 당 혼합물로부터 L-arabinose의 비율을 증가시킬 수 있다는 것을 보여주었다. 막 여과에 의하여 고형물을 제거시킨 배양액에 activate carbon을 처리하여 organic contaminants와 색소를 제거하고 양이온과 음이온 교환 수지를 통과시켜 이온물질들을 제거시킨 후에 농축시켜 3.1 g의 L-arabinose의 분말을 얻었다.

Nutrient composition and in vitro fermentability of corn grain and stover harvested at different periods in Goesan, a mountainous area

  • With South Korea's limited capability of feed production because of its relatively small cultivable area, the country is pushed to depend on foreign feed imports despite the immensely fluctuating price of corn. Hence, intensive efforts to increase the total cultivable area in Korea like extending of farming to mountainous area is being practiced. Corn was planted in Goesan County, a mountainous area in the country. Grain and stover were harvested separately in three harvest periods: early-harvest (Aug 8), mid-harvest (Aug 18), and late-harvest (Aug 28). The nutrient composition such as dry matter (DM), crude protein (CP), crude fat (EE), organic matter (OM), neutral detergent fiber (NDF), acid detergent fiber (ADF), and non-fibrous carbohydrates (NFC) was determined after harvest. Effective degradability (ED) of the major nutrients (DM, NDF, ADF, and CP) were measured through in vitro fermentation of rumen fluid from Hanwoo (Korean cattle). pH, ammonia-N concentration, volatile fatty acid (VFA) concentration, and gas production were periodically measured at 0, 3, 6, 12, 24, 48, and 72 h. Corn grain showed higher nutrient content and ED than stover. It also had higher gas production but its pH, ammonia-N, and total VFA concentration were lower than corn stover. The best nutrient composition of corn grain was observed in early-harvest (high CP, EE, NDF, OM, NFC, and low ADF). Early-harvest of corn grain also had high effective degradability of dry matter (EDDM), effective degradability of neutral detergent fiber (EDNDF), effective degradability of acid detergent fiber (EDADF), and total VFA concentration. On the other hand, the best nutrient composition of stover was observed in mid-harvest (high DM, CP, NDF, and low ADF). EDDM, EDNDF, and EDADF were pronounced in early-harvest and mid-harvest of stover but the latter showed high total VFA concentration. Hence, early and mid-harvested corn stover and grain in a mountainous area preserved their nutrients, which led to the effective degradation of major nutrients and high VFA production.

COMPARISON OF UTILIZATION OF CELLULOSE AND CORN DIETARY FIBER AS AN ENERGY SOURCE IN CHICKS

  • An experiment was conducted to investigate effects of fiber source on growth performance, N and neutral detergent fiber (NDF) digestibility, and utilization of energy in chicks fed an isocaloric low-energy diet from 7 to 21 days of age. Two fiber sources, cellulose and corn dietary fiber (CDF), were included in a diet at 10, 20 and 30% at the expense of kaolin, an inert diluent. The CDF contained 76.5% NDF consisting mainly of hemicellulose. The results showed that growth performance, N and NDF digestibility, dietary DE and ME values, energy deposition, and NE for production in birds fed CDF were inferior to those in birds fed cellulose. It can be concluded, from the present study, that chicks can utilize cellulose more efficiently than CDF up to a level of 30%.

옥수수 섬유질로부터 검과 수용성 아라비노자일란의 수율향상을 위한 압출성형 조건의 최적화 (Optimization of Extrusion Process Conditions to Increase the Corn Fiber Gum and Soluble Arabinoxylan Yield from Corn Fiber)

  • 본 연구는 압출성형 된 옥수수 섬유질로부터 화학적 추출을 통해 SAX와 CFG 수율을 향상시키기 위해 반응표면분석법을 이용하여 최적의 압출성형조건을 확립하였다. 압출성형공정 조건 중 수분함량($X_1$), 스크루 회전속도($X_2$), 배럴온도($X_3$)를 독립 변수로 하고 옥수수 섬유질의 residual solid, final solution, 그리고 CFG의 수율과 각각의 SAX값을 종속 변수로 하여 화학적으로 추출 하였다. 그 결과, 수율의 경우 final solution은 EDCF가 DCF보다 낮은 경향을 보였고, residual solid와 CFG의 경우, EDCF가 DCF보다 높은 경향을 나타내었다. SAX값은 모든 조건의 EDCF가 DCF보다 높은 경향을 나타냈다. 각각의 결정된 최적의 압출성형 조건에서 분석한 결과 모든 조건에서 예측값과 결과값이 유사하여 반응표면 분석의 결과가 양호한 것으로 나타났다. 이상의 연구결과에서 반응표면 분석법이 압출성형 공정에 의한 CFG의 수율과 총 SAX값의 최적화를 위한 적절한 방법 중 하나라는 것을 보여주었다.

Effects of Feeding Corn-lablab Bean Mixture Silages on Nutrient Apparent Digestibility and Performance of Dairy Cows

  • This study estimated the fermentation characteristics and nutrient value of corn-lablab bean mixture silages relative to corn silages. The effects of feeding corn-lablab bean mixture silages on nutrient apparent digestibility and milk production of dairy cows in northern China were also investigated. Three ruminally cannulated Holstein cows were used to determine the ruminal digestion kinetics and ruminal nutrient degradability of corn silage and corn-lablab bean mixture silages. Sixty lactating Holstein cows were randomly divided into two groups of 30 cows each. Two diets were formulated with a 59:41 forage: concentrate ratio. Corn silage and corn-lablab bean mixture silages constituted 39.3% of the forage in each diet, with Chinese wildrye hay constituting the remaining 60.7%. Corn-lablab bean mixture silages had higher lactic acid, acetic acid, dry matter (DM), crude protein (CP), ash, Ca, ether extract concentrations and ruminal nutrient degradability than monoculture corn silage (p<0.05). Neutral detergent fiber (NDF) and acid detergent fiber (ADF) concentrations of corn-lablab bean mixture silages were lower than those of corn silage (p<0.05). The digestibility of DM, CP, NDF, and ADF for cows fed corn-lablab bean mixture silages was higher than for those fed corn silage (p<0.05). Feeding corn-lablab bean mixture silages increased milk yield and milk protein of dairy cows when compared with feeding corn silage (p<0.05). The economic benefit for cow fed corn-lablab bean mixture silages was 8.43 yuan/day/cow higher than that for that fed corn silage. In conclusion, corn-lablab bean mixture improved the fermentation characteristics and nutrient value of silage compared with monoculture corn. In this study, feeding corn-lablab bean mixture silages increased milk yield, milk protein and nutrient apparent digestibility of dairy cows compared with corn silage in northern China.

Assessment of the Nutritive Value of Whole Corn Stover and Its Morphological Fractions

  • This study investigated the chemical composition and ruminal degradability of corn stover in three maize-planting regions in Qiqihaer, Heilongjiang Province, China. The whole stover was separated into seven morphological fractions, i.e., leaf blade, leaf sheath, stem rind, stem pith, stem node, ear husk, and corn tassel. The assessment of nutritive value of corn stover and its fractions was performed based on laboratory assays of the morphological proportions, chemical composition, and in situ degradability of dry matter (DM), neutral detergent fiber (NDF), and acid detergent fiber (ADF). The chemical composition of corn stover was significantly different from plant top to bottom (p<0.05). Among the whole corn stover and seven morphological fractions, leaf blade had the highest crude protein (CP) content and the lowest NDF and ADF contents (p<0.05), whereas stem rind had the lowest CP content and the highest ADF and acid detergent lignin (ADL) contents (p<0.05). Ear husk had significantly higher NDF content and relatively lower ADL content than other corn stover fractions. Overall, the effective degradability of DM, NDF, and ADF in rumen was the highest in leaf blade and stem pith, followed by ear husk. The results indicate that leaf blade, ear husk, and stem pith potentially have higher nutritive values than the other fractions of corn stover. This study provides reference data for high-efficiency use of corn stover in feeding ruminants.

옥수수의 수확시기가 사초의 생산성과 품질에 미치는 영향 (Effect of Different Harvest Dates on Dry Matter Yield and Forage Quality of Corn ( Zea mays L. ))

  • The corn is one of the most important forage crop in Korea. The harvest time for silage affects dry matter (DM) yield and silage quality. This study was carried out to determine the effect of harvest time on the DM yield and nutritive value of corn forage at the Experimental Livestock Farm, College of Agriculture and Life Sciences, Seoul National University, Suweon in 1991. Forage DM yield and DM content of corn forage significantly increased mainly due to increase of ear. And also the ear to total DM ratio increased from 30% to 55% as the harvest was delayed. Chemical composition of the corn forage was improved by reduction in crude fihr, neutral detergent fiber and acid detergent fiber as the harvest was delayed. This results indicated that the harvest time of corn forage plays an important role to determine DM yield and DM contents for silage materials.

Determination of the energy contents and nutrient digestibility of corn, waxy corn and steam-flaked corn fed to growing pigs

  • Objective: The research was conducted to determine the digestible (DE) and metabolizable energy (ME) contents as well as the apparent total tract digestibility (ATTD) of nutrients in corn, waxy corn and steam-flaked corn fed to growing pigs. Methods: Eighteen growing pigs with initial body weight of $15.42{\pm}1.41kg$ were randomly allotted to three diets including a corn diet, a waxy corn diet and a steam-flaked corn diet in a completely randomized design. Each treatment contained six replicates. The experiment lasted for 12 days, which comprised 7-d adaptation to diets followed by a 5-d total collection of feces and urine. The energy contents and the nutrient digestibility in three ingredients were calculated using direct method. Results: Compared to normal corn, both the amylose and dietary fiber contents in waxy corn were numerically lower, but the starch gelatinization degree was numerically greater. Moreover, the DE and ME contents as well as the ATTD of neutral detergent fiber and acid detergent fiber (ADF) in waxy corn were significantly greater (p<0.05) than those in normal corn when fed to growing pigs. Furthermore, the steam-flaked corn had greater (p<0.05) DE and ME contents, and ATTD of ether extract and ADF compared to normal corn. Conclusion: Both variety and processing procedure have influence on chemical compositions, energy contents and nutrient digestibility of corn. The waxy corn and steam-flaked corn had greater degree of starch gelatinization and DE and ME contents compared to normal corn when fed to growing pigs.

Cassava Chips and Ground Corn as Sources of Total Non-Fiber Carbohydrates in Total Mixed Rations for Dairy Cows

  • Six Holstein$\times$indigenous multiparous dairy cows, $60{\pm}8$ days in milk, were used in a double $3{\times}3$ Latin square design to investigate the efficiency of milk production. The dairy cows were randomly allocated to total mixed rations (TMR) containing ground corn, ground corn plus cassava chips 50:50 and cassava chips as main sources of total non-fiber carbohydrates. Ether extract and calculated energy intakes for dairy cows fed TMR containing cassava chips were lower (p<0.05) than those fed TMR containing ground corn and ground corn plus cassava chips 50:50. There were no differences (p>0.05) in daily DM intake (3.51, 3.41 and 3.29% BW), in 4% fat corrected milk (19.66, 20.59 and 20.23%), in milk protein (3.37, 3.27 and 3.33%), and in solids-not-fat (9.03, 8.90 and 8.99%) but there were differences in cost of diets per kg of 4% fat corrected milk (40.75, 34.33 and 28.17%; p<0.01) for dairy cows fed TMR containing ground corn, ground corn plus cassava chips 50:50, and cassava chips. It can be concluded that the efficiency of milk production (4% fat corrected milk per dry matter intake) for dairy cows fed TMR containing cassava was greater than for those fed TMR containing corn.
