번데기동충하초(Cordyceps militaris) 에탄을 추출물의 항산화성 및 항돌연변이원성 효과 (Antioxidative and Antimutagenic Effects of the Ethanol Elrtract from Cordyceps militaris)

  • 번데기동충하초(Cordyceps militaris(L. ex fr.) Link)의 수소전자공여능 및 항돌연변이원성을 살펴본 결과 부탄을 분획물과 에틸 아세테이트 분획물에서 수소전자공여능이 다른 시료에 비해 2배 이상 높게 나타났다. 항돌연변이원성 실험결과에서는 직접변이원인 MNNG, 4NQO 그리고 간접변이원인 B($\alpha$)P, Trp-P-1에 대해서 유의성 있는 돌연변이 억제효과를 보였다. MNNG(0.4 $\mu\textrm{g}$/plate)의 경우 S. typhimurium TA100 균주에서는 시료농도 200 $\mu\textrm{g}$/plate에서 에탄을 추출물이 78.1%인데 반해 부탄을, 물 분획물에서는 각각 90.4%와 87.6%로 매우 높은 억제효과가 나타났다. 4NQO(0.15 $\mu\textrm{g}$/plate)에 대한 S. typhymurium TA98 균주의 경우 에틸 아세테이트 분획물의 경우 80.4%였고, TA100 균주에서는 부탄을 분획물이 88.6%의 억제효과를 보였다. 또한 간접변이원의 경우 B($\alpha$)P(10 $\mu\textrm{g}$/plate)을 사용한 경우 에틸 아세테이트와 부탄을 분획물은 시료농도 200 $\mu\textrm{g}$/plate에서 85% 이상의 높은 억제율을 나타내었다. Trp-P-1(0.15 $\mu\textrm{g}$/plate)에서는 TA98 균주의 경우 200 $\mu\textrm{g}$/plate 농도에서 핵산 분획물과 부탄을 분획물이 각각 90.2%, 87.6%로 높은 억제율을 보여주었고, TA100 균주에 대해서는 에틸 아세테이트 분획물이 90.1%로 억제효과가 가장 높게 나타났다. 또한 같은 농도에서 부탄올과 물 분획물에서 각각 87.0%와 75.4%의 억제율을 나타내었다. 번데기동충하초 에탄을 추출물 및 분획물은 시료 농도증가에 따라 TA98과 TA100 균주 모두에서 각 변이원에 대한 돌연변이 억제효과도 증가하였다.

건강한 성인을 대상으로 면역증강 및 상기도 감염 예방에 대한 동충하초주정추출물의 유효성 평가 : 이중맹검 플라세보 대조 연구 (Effects of Cordyceps militaris supplementation on the immune response and upper respiratory infection in healthy adults: a randomized, double-blind, placebo-controlled study)

  • 본 연구는 한국인 건강한 성인에게서 12주간 동충하초 주정추출물의 섭취가 면역기능 증진 및 상기도감염 예방효과에 대해 확인하였다. 면역관련 지표인 NK-cell activity는 시험식품 섭취군이 플라세보군에 비해 유의적으로 증가하여 (p = 0.047) 면역증강 효과에 대해 확인하였다. 또한, 점막 면역에 주요 역할을 하는 항체 IgA 변화량은 시험식품 섭취군에서 플라세보군에 비해 통계적으로 유의하게 증가하였다 (p = 0.035). 본 연구대상자의 상기도감염(URI) 발생률은 시험식품 섭취군과 플라세보군, 두 군 간 유의한 차이는 관찰할 수 없었다. 안전성 평가인 진단검사의학 검사와 활력진후 결과에서도 섭취군 내 및 섭취군 간 의미 있는 변화 또는 차이는 관찰되지 않았다. 본 연구에서 동충하초주정추출물의 섭취가 URI 발생률에 대한 유의한 변화는 관찰할 수는 없었으나 면역지표인 NK-cell 활성 증가와 혈청 IgA 농도 증가를 통해 선천성 면역과 후천성 면역을 증진시키는 작용이 있어, 잠재적인 상기도감염 예방 효과에 관련성 있을 것으로 사료되며, 항생제의 약물 의존도를 낮추며 삶의 질 개선 및 국민보건 향상에 기여할 것으로 사료된다.

  • Traditionally, Cordyceps species have been used as a part of herbal medicine in Oriental countries, including Korea a for intemal health, vigorosity and to cure different diseases related to heart, lung etc. In the recent years, research on artificial fruiting of different Cordyceps species including C. militaris has been carried out in the world because of their medicinal value. Variationas well as instability occur during artificial fruiting. Instability observed in the in vitro fruiting of C. militaris is reported in the present study.

  • Cordyceps militaris (CM) has been used as antidiabetics, anticancer, endocrine and sexual functions enhancement in the traditional medicine. Water soluble fractions of CM showed cytotoxic activities on the three kinds of human cancer cell lines, stomachic adenocarcinoma(SNU-1), colorectal adenocarcinoma (SNU-C4), and hepatocellular carcinoma(SNH-354). Cytotoxic activity guided isolation and identification of active fractions afforded cordycepin as an active component.

  • Extract of water-soluble polysaccharide (CFWx), showing inhibiting activity on ${\alpha}-glucosidase$, was prepared from the fruiting bodies of Cordyceps militaris by hot-water extraction, and ethanol precipitation. Chemical characteristics of CFWx were as follows: carbohydrate content 30% including 16% of uronic acid; 51% protein content; monosaccharide composition, Man:Glu:Gal (30:43:27); molecular weight $3-5{\times}10^4$. CFWx was further purified by ion-exchange, gel-permeation, and affinity chromatography and $CFWx-AH-{\alpha}$ fraction was isolated. Fundamental structure of $CFWx-AH-{\alpha}$ was deduced as ${\alpha}-(1{\to}4$)-D-glucan with ${\alpha}-(1{\to}3$)- and/or ${\alpha}-(1{\to}6$)-D-glycosidic side chains based on methylation analysis.

  • Cordyceps militaris (CM) is one of the most important medicinal mushrooms known to possess various biological activities. Cordycepin (CP) is a bioactive compound present in the fruiting bodies of CM and is known to have anti-tumor, anti-metastatic immunomodulatory and anti-inflammatory activities. In this study, we aim to analyze CP quantitatively under various CM extraction conditions. CP was measured using high-performance liquid chromatography, quantified using a reversed phase column using a gradient elution system of water and acetonitrile, and detected with a UV absorbance wavelength of 260 nm. The CP content of CM was the highest in 100% ethanol extract of the fruiting bodies and 60% ethanol extract of the mycelium. This study provides an efficient analysis method to determine the optimal extraction conditions for CP that can be used as a basis for developing functional foods and pharmaceutical products derived from CM.

동충하초가 사염화탄소로 유발된 간 손상 및 간암세포증식에 미치는 영향 (Effects of Cordyceps militaris on $CCl_4$ - Induced Liver Damage and Cancer Cell (HepG2 Cell) Growth)

  • Cordyceps militaris has been known as a Chinese traditional medicine for the treatment of tuberculosis, asthma, kidney disease, debility and fatigue etc. This study was attempted to investigate the therapeutic effect of C. militaris extract on the cytotoxic activity of HepG2, human hepatocellular carcinoma cells and the liver damage induced by carbon tetrachloride in SD rats. C. militaris extracts inhibited significantly the proliferation of HepG2 cells in vitro. Carbon tetrachloride(CCl₄) caused a significant an increase in liver weight, serum aspartate aminotransferase(AST) and alanine aminotransferase(ALT) activity, alkaline phosphatase(ALP), serum thiobarbituric acid reactive substances (TBARS), microsomal TBARS, and decrease in microsomal detoxification enzymes (cytochrome P-450, P-450 reductase, cytochrome b5, b5 reductase). TBARS and ALP in serum pretreated with C. militaris extracts (300mg/kg/day, 600mg/kg/day) was significantly reduced compared to control group(CCl₄). Cytochrome b5 and b5 reductase activities were significantly increased in CM300 (300 mg/kg/day) and CM600 group(600 mg/kg/day), and cytochrome P-450 reductase was significantly increased in CM300 group. Pretreatment (100, 300, and 600 mg/kg/day for 7 days) of C. militaris with CCl₄ was significantly inhibited the accumulation microsomal TBARS and the significantly increased in the cytochrome P-450 activity. These results suggested that C. militaris (300mg/kg/day for 7 days) has appreciable therapeutic effect on CCl₄ induced hepatotoxicity.

Effects of Cordyceps militaris Mycelia on In vitro Rumen Microbial Fermentation

  • Effects of Cordyceps militaris mycelia on rumen microbial fermentation were determined by measuring in vitro gas production, cellulose digestion and VFA concentrations. C. militaris mycelia was added to buffered rumen fluid with final concentrations of 0.00, 0.10, 0.15, 0.20, 0.25 and 0.30 g/L and incubation times were for 3, 6, 9, 12, 24, 36, 48 and 72 h. At all incubation times, the gas production showed a quadratic increase with the supplementation of C. militaris mycelia; maximum responses were seen with 0.25 g/L supplementation. However, the gas production was significantly lower for the 0.30 g/L supplementation than for the 0.25 g/L supplementation from 9 h to 72 h incubation. The cellulose filter paper (FP) digestion showed a quadratic increase, as did the gas production except at 3 h incubation. The concentration of total VFA was significantly increased by the supplementation of C. militaris mycelia compared with the control treatment; the highest response was also seen with 0.25 g/L supplementation. This was true for responses in the concentration of acetic and propionic acids. As opposed to other responses, the responses of pH to the supplementation of C. militaris mycelia showed a quadratic decrease from 3 h to 36 h incubation. In conclusion, C. militaris mycelia alter the mixed rumen microbial fermentation with increases in the production of gas and VFA, and cellulose FP digestion.

Comparison of the metabolic profile of the mycelia and fruiting bodies of artificially cultured Cordyceps militaris

  • Cordyceps militaris, a well-known traditional Chinese medicine, has multiple health-promoting effects. It is used as a herbal remedy and health food in Asian countries. Cultured mycelia are often used as a substitute for natural C. militaris. In the present study, the mycelia and fruiting bodies of artificially cultured C. militaris were analyzed using a metabolomics approach. The protein and crude fat contents of the mycelia were substantially higher than those of the fruiting bodies. The top three abundant amino acids in the mycelia were proline (3.9 g/100 g), aspartic acid (2.9 g/100 g), and glutamic acid (2.7 g/100 g). The carbohydrate content was similar in the fruiting bodies and mycelia. Analysis revealed that both the fruiting bodies and mycelia are rich in phenolic compounds and exhibit antioxidant activity. Further, six metabolites were significantly different between the mycelia and fruiting bodies. The levels of Ca, glucose, Mg, and Se were higher in the mycelia than in the fruiting bodies. In contrast, mannitol and Zn were more abundant in the fruiting bodies. The current study provides a comprehensive metabolic profile of the mycelia and fruiting bodies of artificially cultured C. militaris. Such an exercise is potentially important for understanding the metabolism of C. militaris and facilitating the use of cultured mycelia as a supplement to C. militaris fruiting bodies in traditional Chinese medicine.

Effect of cordycepin-enriched Cordyceps militaris L. extract powder on alcohol degradation and hepatoprotection in mice alcohol model

  • Cordyceps militaris L. (C. militaris) has been traditionally used as tonic medicine for metabolic syndrome. Cordycepin, has been reported with immunomodulatory, antitumor, and hepatoprotective effect, is the main extract from C. militaris. This study was conducted to evaluate the alcohol degradation and hepatoprotective effect of cordycepin-enriched C. militaris extract (CM) powder in chronic and binge ethanol (ethanol Lieber-DeCarli diet)-fed male C57BL/6 Mice. Cordycepin-enriched C. militaris extract powder was orally administered once daily at dose levels of 0, 125, 250, and 500 mg·kg-1·day-1 for 16 days. For evaluating alcohol degradation, ethanol concentration and alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activity were measured in serum. Serum ethanol (EtOH) concentration was decreased at CM treated groups, and the activities of ADH and ALDH were increased dose-dependently at CM treated groups compare to EtOH model group. In clinical chemistry, the values of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were lower in CM treated groups than those in EtOH model group. Additionally, absolute and relative (to body weight) liver weights were statistically decreased in the CM treated groups compared to the EtOH model group. In conclusion, our study showed that cordycepin-enriched C. militaris extract powder exhibits hepatoprotective effect by upregulating the ADH and ALDH enzyme in an alcoholic liver disease model.
