In the fungal genus Cordyceps, the type species Cordyceps militaris produces bioactive ingredients and exhibits medicinal effects as a Traditional Chinese Medicine (TCM), The fruiting bodies of C.militaris have now been mass-produced artificially and used as functional food and medicine in China. The unstable variation in forming fruiting body is however a key restrictive factor in industrial production. The genetic study on in vitro stromata formation of C. militaris has rarely been carried out. Here, we report the effects of genetic variation including the mating system on perithecial stromata formation of C. militaris. Monoconidial isolates which have both MAT1-1-1 and MAT1-2-1(genotype MAT1-1/2) could produce stromata. While the isolates only have either MAT1-1-1 or MAT1-2-1 (genotype MAT1-1 or MAT1-2) failed to produce stromata. Despite obvious heterothallism, homothallism was occasionally observed in a few isolates of C. militaris. High genetic variation was observed amongst the different monoconidial isolates of C. militaris. The unstable variation or lose of fruiting body formation was found to be caused by the inner-species high genetic variation of C. militaris. These results also indicated that C. militaris sexually behaved as both heterothallic and homothallic and required two mating type compatible in the same culture in order to produce regular clubshaped perithecial stromata.
Cordyceps militaris, known as Dong-Chong-Xia-Cao, produces the most cordycepin among Cordyceps species and can be cultured artificially. For these reasons, C. militaris is widely used as herb or functional food in the East Asia. In this study, we developed a new strain of C. militaris that produces higher cordycepin content than parent strains through mating-based sexual reproduction. Twenty parent strains were collected and identified as C. militaris based on internal trasncrived spacer and rDNA sequences. Seven single spores of MAT 1-1 idiomorph and five single spores of MAT 1-2 idiomorph were isolated from 12 parent strains. When 35 combinations were mated on the brown rice medium with the isolated single spores, eight combinations formed a stroma with a normal perithecia and confirmed mated strains. High pressure liquid chromatography analysis showed that mated strain KSP8 produced the most cordycepin in all the media among all the tested strains. This result showed due to genetic recombination occurring during the sexual reproduction of C. militaris. The development of C. militaris strain with increased cordycepin content by this approach can help not only to generate new C. militaris strains, but also to contribute to the health food or medicine industry.

The effect of media composition and culture condition on mycelial growth and cordycepin (3'-deoxyadenosine) production was determined using Cordyceps spp. Among the strains of C. militaris and C. sinensis tested, C. militaris KCTC 6862, C. militaris DGUM 32003 and C. militaris KCTC 16932 were excellent for the production of cordycepin. The optimal temperature and pH for production of cordycepin were $24^{\circ}C$ and pH ranged from 6.0 to 10, respectively. Among various sources of carbon and nitrogen tested, glucose and tryptone were very excellent for the production of cordycepin, respectively. After 5days cultivation with 1% of tryptone with nitrogen source, 39mg/l of cordycepin was produced. However, addition of phosphorus sources reduced the production of cordycepin.

The aim of this study is to evaluate the efficacy of cordyceps militaris extracts for the improvement of cognitive dysfunction in PC12 and BV2 cells. Cordyceps militaris extracts was prepared by extracting with distilled water. Cell viability was assessed by MTT assay using PC12 cells and BV2 cells. Confirmed effects of L-glutamate induced cytotoxicity test, Acetylcoline (ACh) concentration, and Acetylcolinestase (AChE) activity in PC12 cells. Anti-inflammatory activities of cordyceps militaris extracts was measured through changes in the levels of nitric oxide (NO), and prostaglandin E2 ($PGE_2$) on lipopolysaccharide(LPS)-induced BV2 cell. In addition, we measured the expression of $NF-{\kappa}B$, p38, JNK, and caspase-3 in western blot analysis. Cordyceps militaris extracts showed no cytotoxicity at the concentrations of 1, 10, and $100{\mu}g/m{\ell}$ except for the concentration of $200{\mu}g/m{\ell}$. Cordyceps militaris extracts protected the cell and exhibited significant increases in the ACh concentration and a significant decrease in the AChE activity in L-glutamate induced PC12 cells. Moreover, cordyceps militaris extracts inhibited the productions NO, and PGE2 level and the protein expression of $NF-{\kappa}B$, p38, JNK, caspase-3 in LPS-induced BV2 cells. These results indicate that cordyceps militaris extracts possible prevented and improved cognitive dysfuction symptoms. Thus, cordyceps militaris extracts may be a novel natural material option for the improvement of cognitive dysfunction.
We previously isolated cordycepin, guanosine and tryptophan from Cordyceps militaris as antiadipogenic constituents. For the quality control of C. militaris for anti-adipogenic activity, simultaneous analytical method using high-performance liquid chromatography (HPLC)-diode array detection (DAD) was developed and validated. Quantitation of these compounds in various Cordyceps samples from different sources and various extraction methods were conducted using developed method. Our study shows that natural Cordyceps and host insect possess higher content than cultured ones and fruiting bodies, respectively. The content of cordycepin showed great difference in different C. militaris samples whereas trytophan content was similar in tested samples. Addition of water to extraction solvent greatly increased the yield of guanosine and tryptophan. High temperature and longer extraction time increased yield of guanosine, whereas the content of trytophan was decreased in high temperature during extraction with water. Extraction using ultrasonic apparatus slightly increased extraction efficiency. Cordycepin, however, has little variation in different extraction method tested. Strong anti-adipogenic activity was observed in the samples that contain all the three constituents. Taken together, quantitation of these compounds using developed analytical method might provide basic requirement for the anti-adipogenic activity of C. militaris.

This study was conducted to investigate the effects of the Cordyceps militaris extract on the improvement of the glucide metabolism in serum of streptozotocin (STZ, 50 mg/kg BW, IP injection)-induced diabetic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of creatinine, lipid peroxide and NEFA in sera were significantly higher in the STZ-induced diabetic group (group DMG) and STZ+Cordyceps militaris extract administration group (group DMS) than those in the control group (group BD). However, the concentrations of creatinine, lipid peroxide and NEFA in sera were reduced in the group DMS than those in the group DMG. The activity of creatine phosphokinase (CPK) in serum was lower in the group DMS than in the group DMG. The activity of LCAT in serum was increased in the group DMS (Cordyceps militaris extract administration) than in the group DMG. The results indicate that Cordyceps militaris extract were effective in the improvement of the glucide metabolism in the sera of STZ-induced diabetic rats.

Background: Cordyceps militaris have been reported to modify the immune and inflammatory responses both in vivo and in vitro. Macrophages play important roles in the innate immunity through the phagocytosis of antigens. This study examined the effects of Cordyceps militaris on the activation of murine macrophage RAW 264.7 cells and primary macrophages. Methods: The components contained in culture broth of Cordyceps militaris were purified by propyl alcohol extraction and HP 20 column chromatography to CMDB, CMDBW, CMDB5P, and CMDB25P. The amounts of nitric oxide (NO) were determined by using ELISA, Griess reagent respectively. The amounts of some cytokines were determined by using ELISA, western blot, and RT-PCR The expression levels of cell surface molecules (ICAM-1, B7-1 and B7-2) were measured by flow cytometric analysis. Results: All the components of Cordyceps militaris produced significant amounts of NO. In particular, CMDB produced much more NO in RAW 264.7 cells and primary macrophages than other fractions of Cordyceps militaris. CMDB increased significantly the production of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-1${\beta}$, and IL-6 dose-dependently in RAW 264.7 cells. Examination of the gene expression level also showed that the enhanced production of cytokines was correlated with the up-regulation of i-NOS expression, cycloxygenase (COX)-2 expression, IL-1${\beta}$ and IL-6 expression, and TNF-${\alpha}$ expression on the expression of mRNAs by semi-quantitative RT-PCR Western blot analysis also confirmed that CMDB enhances the expression level of these cytokines. Conclusion: These results show that CMDB stimulates the production of NO and pro-inflammatory cytokines and can also up-regulate the gene expression levels in macrophages.
This study aimed to verify the whitening effect of Cordyceps militaris, which is distributed in several countries worldwide, including Korea, Japan, and China, and has various medical effects. To screen the efficacy of C. militaris, the inhibitory activity of tyrosinase, which was 66% at a concentration of 1 mg/mL, was measured. Thereafter, the survival rate of melanoma cells was measured, and cell experiments were conducted at a concentration of 90% or more in which C. militaris was not toxic to cells. After measuring the inhibitory effect of TRP-1, TRP-2, tyrosinase protein, and mRNA expression, which are factors influencing melanin synthesis, C. militaris was found to decrease in all factors, with an expression level that was significantly lower compared to quercetin. This confirmed that C. militaris stimulated with LED has excellent whitening activity and can be used as a functional whitening cosmetics material.
Many Cordyceps rpp were collected for excellent strains selection at Mts. Moak, Younsuk and Daea-ree herbarium near Wanju-kun from mid May to October,1999. They were identified as the resulting 2 genera and 5 species, 10 strains were got through 5 species culture. Among them 4 species used for induction of artifitial fruiting body and 6 strains of them were selected to excellent strains: Cordyceps militaris (99-0007), Isaria japonica (99-0001, 99-0002, 99-0003), Cordyceps soborifera (99-0008), Cordyceps kanzashiana(99-0009).
The comparative effects of the fibrinolytic action, antioxidative activity, and tyrosinase inhibition of Cordyceps militaris powder and fermented Cordyceps militaris powders were investigated using several microscopic organisms. The nutritional components such as phenolic compounds, flavonoids, and minerals were also measured. The total phenolic compounds and flavonoid concentrations were highest in the Cordyceps militaris powder fermented by Aspergillus oryzae. Major minerals were K, Ca, Mg, and Zn. Native polyacrylamide gel electrophoresis (native-PAGE) analysis of the total protein patterns of Cordyceps militaris powder and fermented Cordyceps militaris powders revealed slight varietal differences. Fibrinolytic activity was highest in the Cordyceps militaris powder fermented by Bacillus subtilis and Aspergillus kawachii. The DPPH radical scavenging activity was slightly stronger in the powder fermented by Monascus purpureus; however, these samples all exhibited a relatively low activity when compared with butylated hydroxytoluene (BHT). Tyrosinase inhibition activity was stronger in the powder fermented by Aspergillus oryzae than in unfermented powder. These results may provide basic data for understanding the biological activities and chemical characteristics of Cordyceps militaris powder fermented by several microscopic organisms for the development of functional foods.
