Dongchunghacho is traditionally believed to be effective against various diseaseas. It includes many different genera such as Cordyceps, Paecilomyces, Torrubiella and Podonectria. The effects of liquid culture conditions and nutrient sources on the formation of protein-binding polysaccharide (PS) in Cordyceps militaris were examined. The formation amount of PS was increased in proportion to the growth rate of mycelium, in case of higher aeration or lower acidity. The optimum growth temperature of the mycelia was 25$^{\circ}C$ for the formation of PS. The optimum carbon source and nitrogen source were glucose and peptone, respectively. The ratio of C/N was optimal with 3% glucose to 0.5 % peptone. The sugar composition in the PS was greatly changed according to the carbon sources. The mycelium of Cordyceps militaris by liquid culture showed a higher electron donating ability than that by solid culture.
Cordyceps militaris (CM) has been used as a tonics in the traditional medicine. To investigate the anti-diabetic principle of CM, activity guided fractionation was conducted. Hot water extract of CM was fractionated into 3 parts: above 100,000(A), $100,000{\sim}20,000(B)$, below 20,000(C) in molecular weight using in membrane filter system. All fractions showed mild hypoglycemic activity in streptozotocin (STZ)-induced diabetic rats by oral administration (300 mg/kg). The fraction C which was most active among them was fractionated again into two parts, C-1 and C-2 by Sephadex LH 20 column chromatography. The fraction C-1 showed hypoglycemic activity but C-2 did not show activity compared with control in STZ mice. In glucose-fed hyperglycemic mice, fraction C, C-1 and C-2 also showed significant glucose lowering activity. Their decreasing rates of plasma glucose level after 1 hours administrations of fraction C, C-1 and C-2 were 24.5%, 29.3% and 22.0%, respectively (Tolbutamide: 48.4%). These results suggested that CM has both insulin like and insulin release promoting activity and could be developed as an antidiabetic agent.
Background: Cordyceps militaris has been used in traditional medicine to treat numerous diseases and has been reported to possess both antitumor and immunomodulatory activities in vitro and in vivo. However, the pharmacological and biochemical mechanisms of Cordyceps militaris extract (CME) on macrophages have not been clearly elucidated. In the present study, we examined how CME induces the production of proinflammatory cytokines, transcription factor, and the expression of co-stimulatory molecules. Methods: We confirmed the mRNA and protein levels of proinflammatory cytokines through RT-PCR and western blot analysis, followed by a FACS analysis for surface molecules. Results: CME dose dependently increased the production of NO and proinflammatory cytokines such as IL-$1{\beta}$, IL-6, TNF-${\alpha}$, and $PGE_2$, and it induced the protein levels of iNOS, COX-2, and proinflammatory cytokines in a concentrationdependent manner, as determined by western blot and RT-PCR analysis, respectively. The expression of co-stimulatory molecules such as ICAM-1, B7-1, and B7-2 was also enhanced by CME. Furthermore, the activation of the nuclear transcription factor, NF-${\kappa}B$ in macrophages was stimulated by CME. Conclusion: Based on these observations, CME increased proinflammatory cytokines through the activation of NF-${\kappa}B$, further suggesting that CME may prove useful as an immune-enhancing agent in the treatment of immunological disease.
This study was carried out to investigate the conditions of liquid culture and fruiting body formation of Cordyceps militaris using grain mediacontained the brown rice and black purple rice. Liquid mycelial growth and density of Cordyceps militaris were the highest in the medium of malt extract 0.7% and pH 5.0~5.5. The optimal condition of formation the fruiting bodies were shown to be temperature of $17^{\circ}C$, light intensity of 600 lux, $C0_2$ of 500ppm and humidity intensity of 95% produced highest amount of fruit bodies.

Light is an important factor for cordycepin production in Cordyceps militaris. We investigated the effects of different light-emitting diode (LED) conditions including various LED wavelengths and their combinations on cordycepin production in Cordyceps militaris cultivated in submerged culture. The results of our study showed that the combinations of LED wavelengths were more beneficial than single LED sources for cordycepin production. Among the three tested wavelength combinations, the greatest effects for cordycepin production were observed for the red:blue light combination at the wavelength ratio of 5:5 or 3:7. The optimal culture conditions were 19.2278 h/day of illumination time; 9.19497 g/50 mL of glucose content in the media; and 53.112 h of cultivation time. Our model predicted a maximum yield of 2860.01 ㎍/mL cordycepin. Finally, to verify the calculated maximum, we performed experiments in the culture media representing the obtained optimum combination and the cordycepin yield of 2412.5 ㎍/mL.

This study was performed to investigate the influence of extracts from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium on hypoglycemia in streptozotocin-induced diabetic rats. Experimental animals were divided into 7 groups : normal control group(NC), diabetes control group (DC), positive control group(PC), non-fermented OCM(oriental medicine & cereal medium) extracts group (UM), crude polysaccharide of non-fermented OCM extracts group (UME), fermented OCM extracts group (UF), crude polysaccharide of fermented OCM extracts group (UFE). NC, DC groups were orally administered saline, PC group was orally administered acarbose. UM, UME, UF, UFE groups were orally administered each extract once a day for 14days. Blood glucose level was lower in the all administering OCM extract groups (UM, UME, UF, UFE) than in the diabetes group(p<0.05), and specially UF, UFE groups were similar to tendency of PC group. ALT, ALP activity in OCM groups were not significantly lowered than PC group(p<0.05). AST activity was not different with PC group. The results of this study show that extracts from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium may have a beneficial effect on the hypoglycemia.
Cordyceps militaris(CM) has been used as a traditional folk medicine to strengthen the immune system and regain energy for hundreds of years in Far East Asian countries. In the present studies, we determined that the stimulatory effect of Cordyceps militaris on testosterone production in male mouse rats. Results illustrated that changes of the body weight, food and water intake of the rats were not observed in this study but the concentration of testosterone in the serum of the rats was significantly increased by CM(p < 0.05). Therefore fruiting bodies of CM grown on the drone bee medium may be an integrative medicine for the treatment of reproductive problems caused by insufficient testosterone levels in human males.
The effects of environmental and nutritional requirement such as temperature, pH, different media, carbon, nitrogen, and carbon-to-nitrogen ratio on the mycelial growth of Cordyceps militaris strain 'Yedang 3' were studied. The optimum temperature and pH for the growth of mycelium were $20-25^{\circ}C$ and pH 6-7, respectively. Out of ten media tested, mushroom complete media (MCM) was the best medium for fast mycelial growth, and Sabouraud's dextrose agar yeast extract (SDAY), malt extract yeast extract agar (YMA) also were favored. The color and shape of colonies varied in different media. The best carbon sources for mycelial growth were fructose, mannitol, and sucrose, whereas the best nitrogen sources were tryptone and peptone. However, mycelia grew slowly in inorganic nitrogen compounds such as $NH_4Cl$, $(NH_4)_2SO_4$, $NH_4NO_3$, and $NaNO_3$. The optimum C:N ratio observed on the culture media was 30-40 range. These results provided basic information on cultural characteristics of vegetative growth and might be useful for spawn production in Cordyceps militaris.

Hot water-extracts prepared from Cordyceps militaris of silkworm pupa (CMP) or Cordyceps militaris of silkworm larva (CML) were tested for tumor growth inhibitory and immunomodulatory activities in ICR mice bearing sarcoma-180 cells solid tumor, and compared with those of the known compound, cordycepin, found in Cordyceps militaris. Mice were subcutaneously injected with sarcoma-180 cells, and i.p. injected with either saline (Control), 50 mg/kg or 100 mg/kg of CMP (CMP50 or CMP100, respectively), or CML (CML50 or CML100, respectively), or 1 or 2 mg/kg of cordycepin (C1 or C2, respectively) for 10 days. Mice injected with CMP50 or CMP100 showed a 47.3% or 57.6% inhibition in the solid tumor growth (P<0.05), while those injected with CML50 or CML100 exhibited a 35.5% or 37.1% reduction (p<0.05) in solid tumor size compared to the value for control mice treated with saline. Animals injected with corcycepin showed a 26∼30% inhibition in the solid tumor growth (P<0.05). Mice bearing solid tumor and injected with CMP or CML showed a significantly increased thymus weight (38∼44% increase), lymphocyte percentages of CD4+ T-cell, CD8+ T-cell, and NK-cell (63∼110% increase) in the spleen, and interleukin-2 excretion (33∼51% increase) by the isolated splenocytes compared to those in control mice (p<0.05). These results indicate that the anti-tumor activity of hot water extracts of Cordyceps militaris, raised on both silkworm pupa and silkworm larva, appears to be associated with their immunomodulatory activity, and these activities found in Cordyceps militaris are superior to those for the single compound, cordycepin.
