Inhibitory Effects of Bioactive Fractions Containing Protoberberine Alkaloids from the Roots of Coptis japonica on Monoamine Oxidase Activity

  • The effects of bioactive fractions containing protoberberine alkaloids from Coptis japonica on monoamine oxidase (MAO) activity were investigated. The MAO was obtained from the mitochondrial fraction of mouse brain. The butanol fraction from Coptis japonica was fractionated into separate bioactive fraction (Fr I-IV) by silica gel column chromatography. MAO activity was strongly inhibited by Fr III and IV, which mainly contain protoberberine alkaloids such as berberine, palmatine and coptisine. These results indicated that the protoberberine alkaloids from Coptis japonica had an inhibitory effect on MAO activity.

Isolation and Determination of Alkaloids in Coptis Rhizome by Forming Complex (착체 생성에 의한 황련 알칼로이드의 분리 및 정량)

  • The Coptis Rhizome is known for containing a number of isoquinoline type alkaloids. Berberine, coptisine and palmatine are the major constituents of alkaloids. The alkaloids were isolated and determined by forming complex from Coptis japonica (Ranunculaceae). For the determination of these alkaloids, a new spectrophotometric method was developed with a simple and selective sample clean-up using thiocyanatocobaltate[II] complex ion. The absorbance of alkaloidal complex in l,2-dichloroethane solution was measured at 625 nm. A calibration curve for the alkaloids isolated from Coptis Rhizome was linear over the concentration range of 0.2-0.3 mg/mι. The method proved to be rapid, simple and reliable for the isolation and the determination of the alkaloids in Coptis Rhizome.

Studies on the Constituents of Berberis amurensis Ruprecht (매발톱나무의 성분에 관한 연구)

  • Berberis amurensis Ruprecht(Berberidaceae) is a medicinal plant indigenous to the middle and northern part of Korean peninsula. The woody parts of this plant have been used for the ocular, peptic and intestinal disorders. The stems of this plants were extracted with MeOH and the MeOH extract was partitioned between organic phases and water layer, successively to fractionated quarternary alkaloids. The acetone-soluble part of guarternary alkaloidal fraction had antibacterial activities and it contained four protoberberine alkaloids such as palmatine(I), Berberine(II), Jatrorrhizine(III) and coptisine(IV), and one aporphine alkaloid, magnoflorine(V). Although the isolations of the compounds I, II, IIII, IV and V from different sources were reported, this is the first report that Berberis amurensis contained the compounds. When the contents of compound I(palmatine) and II(berberine) were quantified and compared with those of other plant parts, cortex contained higher palmatine and berberine than any other part of the plant.

A Study on the Extraction Efficiency of Berberine in Coptidis Rhizoma (황련 중 berberine의 추출효율에 관한 연구)

  • In this paper, optimum extraction condition for berberine which is the indan companent of Coptidis rhizoma was examined at various conditions in the aspect of temperature, solvent and time followed by quantitation with reversed phase HPLC. The efficiency for extraction has been shown the best at $60^{\circ}C$ with 2~3 hours reflux time in methanol or acetic acid. Any significant effect can not be shown from the addition of SLS to extraction solvent.

Effects of Protoberberine Compounds on Serotonin Content in P815 Cells (Protoberberine 화합물이 P815 세포중의 serotonin 함량에 미치는 영향)

  • The effects of protoberberine compounds on serotonin biosynthesis in P815 cells were investigated. Protoberberine compounds such as berberine, palmatine and coralyne decreased serotonin content dose-dependently, but coptisine did not. The $IC_{50}$ values of berberine, palmatine and coralyne were $3.0\;{\mu}M,\;16.5\;{\mu}M\;and\;14.5\;{\mu}M$, respectively. Protoberberine compounds at concentrations up to $20\;{\mu}M$ were not cytotoxic towards P815 cells. The activity of tryptophan hydroxylase, a ratelimiting enzyme in serotonin biosynthesis, was inhibited by the exposure of berberine, palmatine and coralyne in P815 cells (14.9-19.3% inhibition at $2-15\;{\mu}M$), but that of aromatic L-amino acid decarboxylase was not. These results suggest that the inhibition of tryptophan hydroxylase activity by berberine, palmatine and coralyne might partially contribute to the decrease in serotonin content in P815 cells.

Quantitative and Qualitative Analysis of Alkaloids in Coptis chinensis (Coptidis Rhizoma) by LC-DAD and LC-ESI/MS

  • The quality control of natural products is principal key to guarantee the Good Manufacturing Practices (GMP) and Good Clinical Practices (GCP) for the functional food, pharmaceuticals and cosmeceuticals in the industry. In this study, we examined the quantitative analysis of berberine as marker substance of Coptidis Rhizoma by high performance liquid chromatography-photodiode array detector (HPLC-DAD). The HPLC method was validated and met all the requirements for the quality control analysis recommended by FDA and ICH. The berberine was separated on a Xterra $C_{18}$ column ($5{\mu}m$, $4.6{\times}250mm$) using mobile phase consisting of distilled water and acetonitrile with $KH_2PO_4$ (3.4 g) and $Na_2SO_4$ (1.7 g). Calibration curve of berberine has been estimated (y = 42293.47x-41589 with the correlation coefficient 0.9999). The amount of berberine was calculated as 4.25%. And berberastine, palmatine, columbamine, jatrorrhizine, epiberberine, berberine and coptisine in the Coptidis Rhizoma were identified by high performance liquid chromatography - electrospray ionization-mass spectrometer (HPLC-ESI-MS) method.

Corydalis Tuber Induces Apoptosis in MCF-7 Cells, Via Inhibition of Bcl-2 and $Bcl-_{XL}$ Expression (현호색 메탄올 추출물이 Bcl-2와 $Bcl-_{XL}$ 발현 억제를 통해 유방암 세포의 자멸사에 미치는 영향)

  • 목적 : 본 연구는 현호색의 메탄올 추출물(Corydalis Tuber Extract:CTE)이 인간 유방암 세포인 MCF-7 세포의 자멸사에 미치는 영향을 알아보고자 하였다. 방법 : 현호색 메탄올 추출물로 인간유방암에서 유래된 MCF-7 세포를 처리하였다. CTE의 세포자멸사 유도 효과는 MTT, FACS, TUNEL, DNA laddering and immunoblot assay를 통하여 측정하였다. 결과: 본 연구에서 현호색 메탄올 추출물은 MCF-7 세포의 활성을 감소시켰다. CTE로 처리한 MCF-7 세포는 용량에 따라 세포 자멸사 과정이 증가했다. CTE에 노출하였을 때 Bcl-2와 $Bcl-_{XL}$의 발현을 억제하고, 미토콘드리아로부터 세포질로 cytochrome-c를 방출하며, caspase와 PARP의 절단을 유발하여 세포자멸사가 증가했다. 현호색 메탄올 추출물에 존재하는 성분중에서 coptisine이 세포 활성도를 효과적으로 감소시킨 것으로 사료된다. 결론: 실험결과 현호색 메탄올 추출물이 유방암의 임상 치료에 있어 가능성 있는 치료 방법이 될 수 있을 것으로 사료된다.

HPLC Separation of Isoquinoline Alkaloids for Quality Control of Corydalis species

  • A simple and rapid analytical method was developed for the determination of eight isoquinoline alkaloids in Corydalis species. Eight isoquinoline alkaloids, including 2 aporphine alkaloids (isocorydine and glaucine) and 6 protoberberine alkaloids (coptisine, palmatine, berberine, canadine, corydaline, and tetrahydrocoptisine) were used as chemical markers which have a various biological activity and determined for quality control of Corydalis (C.) species (C. ternata, C. yanhusuo, and C. decumbens). To evaluate the quality of these herbal medicines, LC chromatographic separation of alkaloids were preferentially investigated on reversed-phase C18 column with pH variation and composition of mobile phase. In addition, the separation of these alkaloids in herbal extracts was found to be significantly affected on mobile phase composition using gradient elution. Especially for C. yanhusuo extract, berberine was seriously interfered with other alkaloid extracted from sample matrix when mobile phase composition was not optimized. As results, these compounds were successfully separated within 28 min using 10 mM ammonium acetate containing 0.2% triethylamine (adjusted at pH 5.0) as a mobile phase with gradient elution. On the basis of optimized HPLC conditions, 23 different Corydalis species samples were analyzed for the determination of alkaloid levels. In addition, principal component analysis (PCA) combined with the chromatographic data could be successfully classified the different geographic origin samples.
