A Study on the Antimicrobial Activity of Microcystis aeruginosa by Redox Reaction of Cu-Zn Alloy Metal Fiber (구리-아연 합금사의 산화-환원 반응을 통한 Microcystis aeruginosa의 사멸 특성에 관한 연구)

  • This study is focused on the antimicrobial activity of cyanobacteria Microcystis aeruginosa by the reduction and oxidation reaction of copper and zinc alloy metal fiber filter. Cu/Zn ion is easily makes radicals with molecular hydroperoxide. Especially, hydroperoxide radical shows strong toxicity to the strains. Plasma membrane causes conformational change when hydroperoxide radical binds to plasma membrane. Elution of copper ion from copper and zinc alloy metal fiber is detected in the cyanobacteria solution as 0.5 ppm, and that of zinc ion is 0 ppm respectively. Zinc ion is figured to form a hydroxide in the cyanobacteria solution and precipitated to form a sludge. The concentration of chlorophyll-a in the cyanobacteria solution was proved to be the index of antimicrobial level of Microcystis aeruginosa.

The Copper Adsorption onto Hwangto Suspension from Pankok-ri, Kosung-gun (경남 고성군 판곡리 황토 현탁액의 구리 흡착 특성)

  • Adsorption behavior of Cu onto Hwangto, from Pankok-ri, Kosung-gun, suspension was studied using Cu batch adsorftion experiment and computer program MINTEQA2 and FITEQL 3.2. The sorption of copper was investigated as a function of pH, copper concentration and $NaNO_3$ background concentration (0.01 and 0.1 M). The concentration of copper was analyzed using ICP-AES. The sorption of copper onto Hwangto suspension increased with increasing pH and copper concentration. The adsorption percentage of copper drastically increased from pH 5.5 to 6.5, and reached nearly 100% at pH 7.5. Because the amount of copper solution and the ionic strength of background electrolyte may not affect the sorption of copper onto Hwangto, the copper ion may be combined at the surface of Hwangto as an inner-sphere complex. Using the MINTEQA2 program, the speciation of copper was calculated as a function of pH and copper concentration. The concentration of $Cu^{2+}$ decreased and that of $Cu(OH)_2$ increased with increasing pH. The uptake of copper in the Hwangto suspension was simulated by FITEQL3.2 program using two sites-three pKas model, which is composed of silicate reaction site and Fe oxide reaction site. The copper absorption reaction constants were calculated in the case of 2~6 mL of copper solution. The Fe oxide reaction site rapidly adsorbs copper ion between pH 4.5~6.5. Silicate reaction site adsorbs little copper ion at low copper concentration but much at high copper concentration. The removal amount of copper by precipitation was negligible in comparison with that of adsorption. The Fe oxide reaction site may has higher adsorption affinity of copper ion than silicate reaction site.

  • To remove NO from flue gas, a direct decomposition method to $N_2$ and $O_2$ was investigated by using copper / zeolite catalyst. The copper ion-exchanged HY type zeolite has high activity on NO decomposition. The decomposition activity was increased with the increase of ion-exchange level, contacting time and reaction temperature in the range of 30$0^{\circ}C$ -50$0^{\circ}C$ , and decreased with the oxygen addition.

Formation of Crystalline Copper Thin Films by a Sputtering-assisted Magnetic Field System at Room Temperature

  • A sputtering-assisted magnetic field system was successfully developed for depositing crystalline Cu thin films at room temperature. This system employs a plasma source and an ion-beam gun with two magnetic field generators, which is covered with sputtering target and the ion-beam gun, simultaneously serving as sputtering plasma and a magnetic field generator. The formation of crystalline Cu thin films at room temperature was dominated by magnetic fields, which was revealed by preliminary experiments. This system can be employed for producing crystalline metal thin films at room temperature.

Oxidative Modification of Neurofilament-L by Copper-catalyzed Reaction

  • Neurofilament-L (NF-L) is a major element of neuronal cytoskeletons and known to be important for neuronal survival in vivo. Since oxidative stress might play a critical role in the pathogenesis of neurodegenerative diseases, we investigated the role of copper and peroxide in the modification of NF-L. When disassembled NF-L was incubated with copper ion and hydrogen peroxide, then the aggregation of protein was proportional to copper and hydrogen peroxide concentrations. Dityrosine crosslink formation was obtained in copper-mediated NF-L aggregates. The copper-mediated modification of NF-L was significantly inhibited by thiol antioxidants, N-acetylcysteine, glutathione, and thiourea. A thioflavin-T binding assay was performed to determine whether the copper/$H_2O_2$ system-induced in vitro aggregation of NF-L displays amyloid-like characteristics. The aggregate of NF-L displayed thioflavin T reactivity, which was reminiscent of amyloid. This study suggests that copper-mediated NF-L modification might be closely related to oxidative reactions which may play a critical role in neurodegenerative diseases.

  • The methods to separate and remove copper in the mixed solution ((399 ppm Cu, 208 ppm Fe, 15.3 g/L Ni, 2.1 g/L Co) with nickel, cobalt and iron were investigated. With hydroxide precipitation method, copper and iron ions were completely precipitated and removed from the solution at pH 7 while some nickel and cobalt also were precipitated. 99.75% copper could be precipitated and removed as copper sulfide from the solution with adding $Na_2S$ (1.25 w/v concentration) of 2 times equivalent of Cu at pH 1. Copper was selectively absorbed on TP 207 ion exchange resin at equilibrium pH 2.0 and could be eluted from copper-loaded resin using 5% $H_2SO_4$.

  • The active site for the formation of acrolein in propylene oxidation reaction over copper(Ⅱ)-exchanged zeolite Y has been studied. At the early stage of the reaction, the formation of hydrocarbons was observed. The formation of hydrocarbons decreased gradually during the course of reaction, apparently due to the poisoning the Bronsted acid sites. The formation of acrolein was quite low when the formation of hydrocarbons was proceeding. The formation of acrolein was depend on the copper ion content and this can be related to the availability of the copper ions inside the supercage. It seems that it is the copper ion not the Bronsted acid site which is primarily responsible for the formation of acrolein.

  • Recently SIMS has attracted interest as new technique to distinguish the primary and the secondary arc beads. A Cs+ primary ion beam was used to detect the $^{12}C^-$, $^{63}Cu^-$, $^{18}O^-$, $^{35}Cl^-$ secondary ions which are formed during depth profiles in the copper wires. In this work, we studied thermal diffusion in the copper wire which are occurred with supplying over-current. The results demonstrated that Carbon and Chloride are diffused in PVC-coated copper wire deeper than none PVC-coated. However Oxygen showed the reverse diffusion property.

  • Metal ion-induced and it’s regulatory genes were screened in virulent salmonella typhimurium UK1 and tested cross-regulation with various stresses. Using the techniqud of P22-MudJ(Km, lacZ)-directed lacZ operon fusion, LF40 cuiA::MudJ and Lf153 cuiD::MudJ which were induced by copper were selected. cuia and cuiD were determined anaerobic coper inducible and copper tolerance response gene, respectively. Also cuiA and cuiD locus were determined at 81 and 8min, respectively, on salmonella Genetic Map. The two regulators were identified as cuaR, and cudR, which controls cuiA and cuiD, respectively. cuaR, and cudR appeared as negative regulators because the expression of cuiA-lac-Z and cuiD-lacZ were increased. Copper adapted UK1 showed high resistance to H$_{2}$O$_{2}$, but cuiD did not. The product of the cudR locus was responsible for decreasing the tolerance to copper and H$_{2}$O$_{2}$. Furthemore cuiA and cuiD locus were found to be part of a regulon under the control of a trans-acting regulators, rpoS, oxyR and relA. Therefore, the results suggest CTR participate with oxidative stress on Salmonella.

Potentiometric Back Titration of Isoniazid in Pharmaceutical Dosage Forms Using Copper Based Mercury Film Electrode (구리수은막 전극에을 사용한 이소니아자이드의 전위차 역적정)

  • A simple, rapid potentiometric back titration of Isoniazid (INH) in the presence of Rifampicin (RIF) in tablets and syrups is described. The method is based on the oxidation of INH by a known excess of copper (II) ion and the back titration of unreacted copper (II) ion potentiometrically with ascorbic acid using a lab-made Copper Based Mercury Film Electrode (CBMFE). The titration conditions have been optimized for the determination of 1.0-10.0 mg of INH in pure and dosage forms. The precision and accuracy of the method have been assessed by the application of lack-of-fit test and other statistical methods. Interference was not caused by RIF and other excipients present in dosage forms. Application of the method for INH assay in tablets and syrups was validated by comparison of the results of proposed method with that of the British Pharmacopoeia (BP) method using F- and t- statistical tests of significance.
