Development of a new vitrification solution, VSL, and its application to the cryopreservation of gentian axillary buds

  • Vitrification methods are convenient for cryopreserving plant specimens, as the specimens are plunged directly into liquid nitrogen (LN) from ambient temperatures. However, tissues and species with poor survival are still not uncommon. The development of vitrification solutions with high survival that cover a range of materials is important. We attempted to develop new vitrification solutions using bromegrass cells and found that VSL, comprising 20% (w/v) glycerol, 30% (w/v) ethylene glycol, 5% (w/v) sucrose, 10% (w/v) DMSO and 10 mM $CaCl_2$, gave the highest survival following cryopreservation, as determined by fluorescein diacetate staining. However, the cryopreserved cells showed little regrowth, for unknown reasons. To check its applicability, VSL was used to cryopreserve gentian axillary buds and the performance was compared with those of conventional vitrification solutions. Excised gentian stem segments with axillary buds (shoot apices) were two-step precultured with sucrose to induce osmotic tolerance prior to cryopreservation. Gentian axillary buds cryopreserved using VSL following the appropriate preculturing approach exhibited 78% survival (determined by the regrowth capacity), which was comparable to PVS2 and PVS1 and far better than PVS3. VSL had a wider optimal incubation time (20-45 min) than PVS2 and was more suitable for cryopreserving gentian buds. The optimal duration of the first step of the preculture was 7-11 days, and preculturing with sucrose and glucose gave a much higher survival than fructose and maltose. VSL was able to vitrify during cooling to LN temperatures, as glass transition and devitrification points were detected in the warming profiles from differential scanning calorimetry. VSL and its derivative, VSL+, seem to have the potential to be good alternatives to PVS2 for the cryopreservation of some materials, as exemplified by gentian buds.

  • Laser cladding process accompanies phase transformations from melting (on heating) through solidifying (on cooling) at the same time within a small material volume and to final solid phase. The phase transformations are not reversible, but an irreversible thermodynamic process; they accompany either absorption or release of thermal energy (referred to latent heat) during transformation. Yet, most analyses on materials processed by laser as a heat source have been performed on models of neglecting the latent heat in the process and those did not Justify the simplification. With literatures on the laser material process, we have not place an answer to how little the assumption affects on analyses. This led us to our current study: the effects of latent heat on thermo-mechanical analysis. To this end, we developed a fairly accurate program accommodating an algorithm for enforcing the latent heat whenever necessary and ran it combining with ABAQUS$^{TM}$. The simulation techniques we used in this study were verified by directly comparing our prediction with experimental publications elsewhere; our numerical results agreed accurately with the experiments. On the effects of the latent heat, we performed two alternatives about considering the latent heat in analysis, and compared each other. As a result, we found that more accurate conclusions might come out when considering the latent heat in process analyses.s.

Estimation of Radioactive Inventory for a major component of Reactor in Decommissioning (해체시 원자로 주요 구성품에 대한 방사능 재고량 평가)

  • DORT and ORIGEN2 code were used for calculation of neutron flux and inventory in reactor pressure vessel(RPV) of Kori unit-1, To calculate neutron flux using DORT code, the reactor was divided into 94 mesh from the center of core to RPV and from 0 to 45 degree along the azimuth. The cross-sections of main nuclides were recalculated using neutron flux in the RPV region. The results showed that 95% of the total activity in RPV came from the nuclides of $^{55}$ Fe, $^{60}$ Co, $^{59}$ Ni and $^{63}$ Ni. And the total activity with cooling of more than 50 years after decommissioning was no more than 0.2% of at the time of shutdown. Considering the weight of RPV is 210 tons, the initial total activity of RPV reached 5.25${\times}$10$^{6}$ GBq. To verify results of ORIGEN2 calculation, comparison between calculated and measured value at RPV of Kori unit-1 was peformed. The comparison results showed a good agreement.

Effect of Microwave Irradiation on Exfoliation of Graphene Oxide (마이크로파 조사가 산화그래핀의 화학적 박리에 미치는 효과)

  • Graphene oxide has been synthesized by microwave-assisted exfoliation of graphite oxide prepared by modified Hummers method. Graphite was oxidized in a solution of $H_2O_2$ and $KMnO_4$ at $65{\sim}80^{\circ}C$, followed by 10 % $H_2O_2$ solution treatment at $80{\sim}90^{\circ}C$. The graphite oxide was exfoliated under microwave irradiation of 1 kW and was reduced to graphene effectively by hydrazine hydrate ($H_4N_2{\cdot}H_2O$) treatment. The exfoliation of graphene oxide was significantly affected by the microwave irradiation on (heating)/off (cooling) period. An on/off period of 10 s/20 s resulted in much more effective exfoliation than that of 5 s/10 s with the same total treatment time of 10 min. This can be explained by the higher exfoliation temperature of 10 s/20 s. Repetition of the graphite oxidation and exfoliation processes also enhanced the exfoliation of graphene oxide. The thickness of the final graphene products was estimated to be several layers. The D band peaks of the Raman spectra of the final graphene products were quite low, suggesting a high crystal quality.

  • Prior to the experimental and production stages of goods, the strengths should be evaluated in the design stage. The introduction of commercial codes at the design stage gives benefits such as cost and time economies in the production and strength evaluation. In this study, structural analysis and fatigue analysis are carried out using ANSYS modeling of the 3D geometry of the wheel. In a comparison of dish-type and spoke-type wheels, it is shown that the deformation and maximum equivalent stress for the dish-type wheels are lower than those for spoke-type wheels. Nevertheless, spoke-type wheels are often used because they are light and have exhibit excellent cooling performance. Furthermore, according to the results of life analysis, aluminum wheels show improved resistance to fatigue compared to steel wheels.

  • Connectors are very important components that transmit electric signals to different parts. It must maintain intensity of the connector to prevent defects from impact and maintain contact to transmit electric signals. Most of the external parts of the connector, which act as the main framework, are formed by injection molding. However, bend deformation occurs for injection molded products due to the residual stress left inside the product after product molding. When the bend deformation is large, it does not come into complete contact when being assembled with other parts, which leads to connector contact intensity not being properly maintained. In result, the main role of the connector, which is to transmit electric signals, cannot be performed. In order to address this problem, this study conducted bend deformation cause analysis through bend deformation analysis to predict and prevent bend deformation of housings and wafers, which are injection molded products of pressure welded connectors that are normally applied in compact mobile and display products. Bend deformation analysis was carried out by checking the charging time, pressure distribution and temperature distribution through wire to board connector wafer and housing injection molding analysis. Based on the results of the bend deformation analysis results, the cause of the bend deformation was analyzed through deformation resulting from disproportional cooling, deformation resulting from disproportional contraction, and deformation resulting from ingredient orientation. In result, it was judged that the effects for bend deformation were biggest due to disproportional contraction for both the pressure welded connector wafer and housing.

Environmental Control of Plant Production Factory Using Programmable Logic Controller and Computer (PLC와 컴퓨터를 이용한 식물생산공장의 환경제어)

  • This study was conducted to develop a system and an control algorithm for control the environment of a plant factory. The greenhouse control system for environmental control was largely composed of a computer and a PLC. The screen of control program was composed of a greenhouse figure which was included machinery and equipments for greenhouse, the graph of environmental factors of inside greenhouse and the image of greenhouse. In order to reduce temperature change, the operation time of ventilation window was changed by 3 stage according to difference between a target and present temperature. When is heating, a temperature variation was shown to be $16.7{\pm}0.8^{\circ}C$. When is cooling, a temperature variation was shown to be $23.1{\pm}0.6^{\circ}C$. When is humidifing, a humidity variation was shown to be $39.3{\pm}1.6^{\circ}C$ %RH. An environmental control system and a control algorithm were proved that it was shown a good performance in a control accuracy. So a computer control system should be adapted to a control system of a greenhouse and a plant factory.

  • In this study, We analyzed change of sea surface temperature due to cooling water around nuclear power station. Study area is around of Uljin nuclear power station, which is relatively large power station. There are many problems in monitering environmental change around of nuclear power station, because area is relatively large. We used Landsat 5, 7 Imagery which are useful in temperature analysis and can be easily obtained. After we georeferenced Landsat Imagery, radiance and sea surface temperature were calculated. As a result, As we compared sea surface temperature of surrounding area of nuclear power station with same area located 3 km east, there are $2.049^{\circ}C$ temperature difference.

  • Polymer electrolyte membrane fuel cell (PEMFC) is very interesting power source due to high power density, simple construction and operation at low temperature. But it has problems such as high cost, improvement of performance, effect of temperature and initial start at low temperature. These problems can be approached to be solved by using experiment and mathematical method which are general principles for analysis and optimization of control system for heat and hydrogen detecting management. In this paper, insulation vessel and control system for stable operation of fuel cell at low temperature were developed for experiment. The constant temperature capability and the heating time at sub-zero temperatures with insulation control system were studied by using a heating bar of 60W class. PEMFC stack which was made by 4 cells with $50\;mc^2$ active area in each cell is a thermal source. Times which take to reach constant temperature by the state of insulation vacuum were measured at variable environment temperatures. The test was performed at two conditions: heating mode and cooling mode. Constant temperature capability was better at lower environment temperature and vacuum pressure. The results of this experiment could be used as basis data about stable operation of fuel cell stack in low temperature zone.

  • Annual dose on the containment building wall of the interim storage facility at normal condition was calculated to estimate the dose rate transition of the facility of PWR spent nuclear fuel. In this study, source term was generated by ORIGEN-ARP with 4.5 wt% initial enrichment, 45,000 MWd/MTU burnup and 10 years cooling time. Modeling of the storage facility and the containment building and radiation shielding evaluations were conducted by MCNP code depending on the distance between the wall and the facility in the building. In the case of the centralized storage system, the distance required for the annual dose rate limit from 10CFR72 was estimated to be 50 m.
