Rediscovery of a Method for Preparation of Traditional Grape Tea (전통적인 포도차 제조방법의 재현에 관한 연구)

  • In the Joseon Dynasty, various fruit teas were popularized to promote health, with spread of Donguibogam and Hyangyag gugeupbang. As interest in fruit tea has recently increased, studies on its manufacture have become necessary. We used response surface analysis for rediscovery and commercialization of grape tea. Major materials of traditional grape tea are grape juice, pear juice, ginger juice, and honey, and the sugar contents of these materials were 12.3, 14.1, 3.3 and 75 $^{\circ}Brix$, respectively. When sensory examinations were conducted with subjects aged 40-60 years, the difference between dilution ratios of 100% and 150% was not significant, but tea diluted by 150% showed somewhat higher scores than did tea diluted by 100%. Ginger taste and sweetness were found to have the greatest effect on overall acceptance. Regression analysis on color, flavor, taste, and overall acceptance values, with reference to ginger juice and honey as independent variables, revealed that the $R^2$ values were 0.8411, 0.6717, 0.9499, and 0.9015, respectively. Contour maps were superimposed to obtain an optimal combination of ingredients for traditional grape tea, and the indicated levels of ginger juice and honey were 0.46-0.69% and 3.85-5.20%, in combination with grape juice, pear juice, and water concentrations of 28%, 9% and 60% (all w/w), respectively. Thus, it is now possible to prepare traditional grape tea.

Quality Characteristics of Pan Bread Added with Black Garlic Flour (흑마늘 가루를 첨가한 제빵의 품질 특성)

  • This study examines physiochemical characteristics of functional bread with black garlic flour added such as its content, texture, sensory test and quality properties. The loaf volume and weight of black garlic flour-added pan bread showed that for the control, the loaf volume was the greatest, and the more the black garlic flour content increased, the smaller the loaf volume became. The change of crust chromaticity showed that the L value decreased significantly, and crust chromaticity thickened, and as the black garlic flour content increased, a value and b value decreased. The characteristics of texture showed that the hardness and chewiness decreased as the black garlic flour content increased, and gumminess increased significantly while there was no significant difference in cohesiveness. The flavor, taste and texture of pan bread with black garlic flour content didn't showed a significant difference with the control. Consequently, the black garlic flour-added wheat flour was no significant difference with the control up to 3% black garlic flour-added wheat flour, which can be used as a product.

Effect of p-Phenylenediamine to Rat Skin (p-Phenylenediamine이 흰쥐의 피부 조직에 미치는 영향)

  • Red brownish p-pheylenediamine (PPD) has been widely used hair dye for women. The dye was known to cause systemic anaphylaxis, dermatitis and bladder cancer. But the effect of PPD toxicity with oxygen free radical has not been studied. This study investigated the degree of skin injury by PPD. PPD ($2.5\%$ PPD in $2\%\;NH_{4}OH$) was applied to the rat skin ($25 mg/16.5\;cm^2$) 3 or 5 times every other day. Histopathological findings demonstrated the proliferation of epithelial cells and the increased keratinization by PPD. The activities of glucose 6-phosphatase (G6Pase) was decreased and acid phosphatase (ACP) was increased in PPD-applied rat skin. Groups in which PPD was applied 5 times were more damaged than groups applied 3 times. To examine the relationship between tissue damage and oxygen free radicals, effect of PPD on xanthine oxidase (XO) activity was measured and XO activity was more significantly increased in the group treated with PPD 5 times than 3 times. However, reduced glutathione (GSH) content, and the activities of catalase (CAT), super-oxide dismutase (SOD) and glutathione S -transferase (GST) were more decreased in PPD-applied groups than in controls. Even though the activities of XOD was not changed in the group treated with PPD 3 times, the decreased activities of oxygen free radical system and the damaged skin tissue were observed. This result might be caused by the production of toxic PPD metabolites in rat skin. In conclusion, topical PPD application led to skin injury in a dose-dependent manner, probably due to the generation rate of oxygen free radical.

Effect of Red Ginseng-Chungkukjang Extracts on Lipid Profiles of Serum in Alcohol Administered Diabetes-Induced Rats (알코올을 투여한 당뇨 흰쥐의 혈당과 혈청지질에 미치는 홍삼 청국장 추출물의 영향)

  • To evaluate red ginseng-chungkukjang extracts (RC) on levels of blood sugar and serum lipids in diabetes rats fed with ethanol, SD rats were supplemented 2 mL of $20\%$ ethanol solution with or without RC by gastric intubation for 2 weeks after streptozotocin (STZ) injection and then body weight gains, food efficiency ratio (FER), water intake, urine volume, organ weight, levels of blood sugar and serum lipids were determined. Water intake and urine volume were not restored in STZ-treated rats by RC supplementation. On the other hand, decreased body weight gam and FER were restored in diabetes rats by RC supplementation. Furthermore, levels of blood sugar, serum triglyceride, total cholesterol and LDL-cholesterol were significantly decreased by RC supplementation. The rate of mortality in diabetes rats was significantly inhibited by RC supplementation. These results suggest that inhibited rate of mortality in diabetes rats by supplementation of ethanol with RC was considered to be due to improvement of blood sugar and serum lipids levels by components of RC.

A Survey for Needs and Preference of Food and Nutrition information on Mass Media for Korean Female Adults (대중매체 식품영양정보에 대한 성인 여성의 요구도 조사)

  • Objectives: This study was conducted to examine the preferences and needs of typical Korean females adults for food and nutrition information provided by the mass media. Methods: A total of 343 females (77 in their 20s, 85 in their 30s, 88 in their 40s and 93 in their 50s) residing in the Seoul/Gyeonggido area was surveyed on general characteristics, main sources of food and nutrition information and needs for sources and contents of nutrition information. Results: The survey showed that typical Korean females obtained knowledge of food and nutrition mainly through the Internet (30.4%) and broadcasting (29.0%). Typical Korean females were interested in 'dietary management for weight control' (21.9%), 'the prevention and treatment of disease' (20.0%), 'food safety' (16.8%), 'proper dietary habits' (14.6%), 'cookery' (11.8%), 'functional foods' (9.6%), 'restaurant details' (3.5%) and 'life-cycle-specific dietary guideline' (1.6%). Needs for food and nutrition program forms on TV were 'educational programs' (34.3%), 'documentaries' (20.8), 'expert lecture-style' (13.0%), 'entertainment programs' (11.9%), 'expert conversation' (11.4%), 'news-style' (4.6%) and 'public campaign advertisements' (4.0%). On the Internet, 38.6% of the respondents preferred to get information provided by food and nutrition-related institutions (38.6%) while 26.1% preferred webtoons for nutritional information. The favored forms in mobile applications were 'monitoring their diets' (29.5%), 'data-based texts information' (21.4%), 'experts feedback' (20.6%), 'communities' (15.1%) and 'games' (13.1%). The rates of the preference to obtain information from experts such as nutritionists and dietitians and doctorsor dietitian turned reporters increased markedly with older ages. Conclusions: Since the mass media is a main source of food and nutrition information for the general public, the effectiveness and accuracy of the information provided should be enhanced by taking the needs of the public into account. The quality of information should be improved by involving more nutrition experts.

A Study on Traditional Kimchi made with Heating (가열하여 담그는 전통 김치)

  • During the Joseon period, kimchi was mostly made by heating the ingredients. Since salt was pricey at the time, in order to save salt and time, people used a method that involved destroying the cell wall by heating the vegetables. However, this method is no longer passed down. Thus, in this paper, we re-discovered how kimchi was made through heating while analyzing the recipes for kimchi during the Joseon period. There were 27 kinds of kimchi made through blanching. To keep the vegetables from becoming soggy, 2 kinds of kimchi were made by putting the ingredients in potassium aluminum sulfate water and 3 kinds were made through blanching the ingredients in limewater. There were 7 kinds of kimchi made by heating in vinegar, 5 kinds by boiling the ingredients, 6 kinds by stir-frying the ingredients, 2 kinds by stir-frying the ingredients with salt, and 3 kinds by steaming the ingredients. In order to eradicate unwanted germs, leaving only Lactobacillus, 25 kinds of kimchi were made by draining the boiled mixture. A total of 17 kinds of kimchi were made by heating the kimchi pot with compost including that of horses. For elders with weak teeth and poor digestion, 7 kinds of kimchi were made after heating, including 3 kinds of sukkkakdugi (cubed radish kimchi made with boiled radish). 3 kinds of chaekimchi (julienned kimchi) and 3 kinds of chaekkakdugi (kimchi with julienned radish) for elders existed as well.

Antioxidant Activity of Fermented Wild Grass Extracts (산야초 발효액의 항산화 활성)

  • Wild grass is edible, and it grows in the mountains or field areas. Wild grass has diverse biological effects, such as antiobesity, anti-cancer, antioxidant activities and immune stimulation. Currently, many studies are aimed at enhancing the efficacy of medicinal foods on biological activity using a bioconversion technology, including the fermentation process. In this study, the quality characteristics and antioxidative activity of the fermented wild grass was investigated. The antioxidant activity of fermented wild grass was assessed by various radical scavenging assays using DPPH(2,2-diphenyl-1-picrylhydrazyl), FRAP(ferric ion reducing antioxidant power), reducing power, and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)). Moisture contents of the fermented wild grass is $49.6{\pm}0.06%$. Contents of crude ash, crude protein, and crude fat were $0.65{\pm}0.01$, $0.65{\pm}0.04$, and $3.3{\pm}0.59%$, respectively. Moreover, fermented wild grass showed that the hunter's color values were 80.36(lightnees), 11.47(redness), and 44.53(yellowness), respectively. Total phenolic contents of the fermented wild grass was $1,185{\pm}159{\mu}g$ GAE(gallic acid equivalent)/g. The antioxidative activities of the fermented wild grass were significantly increased in a dose dependent manner. In addition, fermented wild grass did not show any cytotoxicity up to 500 ${\mu}g/m{\ell}$. However, the anti-adipogenic effect of the fermented wild grass extract was barely detectable. This antioxidant potential is partly due to the phenolic compounds that are present in the fermented wild grass extracts.

Isolation and Purification of Berberine in Cortex Phellodendri by Centrifugal Partition Chromatography (Centrifugal Partition Chromatography에 의한 황백으로부터 Berberine의 분리 및 정제)

  • Cortex Phellodendri (CP) is derived from the dried bark of Phellodendron amurense. It has been widely used as a drug in traditional Korea medicine for treating diarrhea, jaundice, swelling pains in the knees and feet, urinary tract infections, and infections of the body surface. Many analytical methods have been used to study oriental herbal medicines, such as thin-layer chromatography, column liquid chromatography, and high performance liquid chromatography (HPLC). In this study, preparative centrifugal partition chromatography (CPC) was successfully carried out in order to separate pure compounds from a CP methanol extract. The optimum two-phase CPC solvent system was composed of n-butanol: acetic acid: water (4:1:5 v/v/v). The flow rate of the mobile phase was 3 mL/min in ascending mode with rotation at 1,000 rpm. The CPC-separated fraction and purification procedures were carried out by preparatory HPLC. The $^1H$ NMR spectrum revealed that the resonances at ${\delta}$ 4.10 and 4.20 ppm corresponded to three protons ($-OCH_3$), whereas those at ${\delta}$ 6.10 ppm corresponded to two protons ($-OCH_2O-$). Further, two aromatic protons (H-11 and H-12) conveys a doublet-doublet pattern. The H-11 doublet and H-12 doublet appear at ${\delta}$ 7.98 and 8.11, respectively. The $^{13}C$ NMR. spectrum showed a tetrasubstituted with a methylenedioxy group at C2 and C3, and two methoxy groups at C9 and C10. The chemical structure of the berberine was identified by $^1H$, $^{13}C$-nuclear magnetic resonance and electrospray ionization-mass spectroscopy spectral data analysis.

Characteristics of Alcohol Fermentation in Citrus Hydrolysate by Different Kinds of Sugar (첨가당의 종류에 따른 감귤 가수분해물의 알코올발효 특성)

  • This study investigated the alcohol fermentation characteristics of citrus hydrolysate by adding various sugars(sucrose, honey, fructose and fructooligosaccharide). As a result, the alcohol content was shown to be similar among all the sugars. Fructose, glucose, sucrose and maltose were detected as a major free sugar. In particular, the contents of fructose and glucose were shown to be higher in sucrose addition, lactic, citric and malic acids were detected as major organic acids of citrus wine. When the sensory characteristics of citrus wines were compared, flavor was shown to have a sensory score of 5.1 in sucrose addition, showing the most preference. However, no significant difference in preference was found among the sugars. Color, taste and overall acceptability were shown to have the most preference in sucrose addition, and then in honey, fructose, and fructooligosaccharide in order. Therefore, a further study on the improvement of quality and sensory preference using aging process and complex sugars is required.

Effect of Beni-Koji with Chitosan-Ascorbate on Serum Lipid Profile of Rats Fed a High Fat Diet (Chitosan-Ascorbate 함유 홍국이 고지방 식이 흰쥐의 혈청지질에 미치는 영향)

  • Dietary effect of soyfiber beni-koji (SBK) with chitosan-ascorbate (CA) on the level of serum lipids in rats fed a high fat diet was investigated The experimental groups which were divided into high fat diet control group(HC-group), $2\%\;SBK+0.1\%$ CA mixture diet group(CA1-group), $2\%\;SBK+0.15\%$ CA mixture diet group(CA2-group), and $2\%\;SBK+0.2\%$ CA mixture diet group (CA2-group) were fed for 4 weeks. Weight gains in CA2- and CA3-group were $5.3\%\;and\;9.5\%$ lower than that of HC-group, respectively, while there was no significant difference in feed intakes, feed efficiency ratio and organs weight Level of serum triglyceride in $C_3-group\;was\;21\%$ lower than that of HC-group. Level of serum total cholesterol and LDL-cholesterol in CA2- and CA3-group were $22.1\~22.7\%\;and\;58.6\~64.3\%$ lower than those of HC-group, respectively. Atherogenic index decreased with the higher level of CA. Level of lipid peroxide in CA3-group was $24\%$ lower than that of HC-group, while there was no significant difference in GSH(Glutathione-S-transferase) content O type activities of XOD(xanthine oxidase) in the treated groups were lower, especially the activity in CA3-group was $51.6\%$ lower than that of HC-group. Also, O/T ratio of XOD was lower, showing $21.7\~23.5\%$ in treated groups and $34.0\%$ in HC-group(p<0.05). GST activities were 332.52 units in HC-group and $350.28\~355.63$ units in the treated groups, but there were no significant differences among them.
