Agrobacterium-mediated Transformation of Eleutherococcus sessiliflorus using Embryogenic Calli and the Regeneration of Plants (오갈피(Eleutherococcus sessiliflorus)의 배형성 세포를 이용한 고빈도 형질전환 및 재분화)

  • We have developed a reliable and high-frequency genetic transformation and regeneration system via somatic embryogensis of Eleutherococcus sessiliflorus. Embryogenic callus obtained from seed were co- cultivated with Agrobacterium tumefaciens strain EHA101/pIG121Hm harboring genes for intron-$\beta$-glucoronidase(GUS), kanamycin and hygromycin resistance. Following co-cultivation, two types of samples(fine embrogenic calli and early globular embryo clusters) were cultivated on Murashige and Skoog(MS) medium containing 1 mg/L2.4-D for 3day in dark. Transient expression of GUS gene was found to be higher in the early globular embryo clusters than in the embryogenic calli. Also, co-cultivated period affected expression of GUS gene; the best result was obtained when globular embryo clusters were co-cultivated with Agrobacterium for 3 days. Subsequently, this callus transferred to selective MS medium containing 1mg/L2.4-D, 50mg/L kanamycin or/and 30mg/L hygromycin and 300mg/L cefortaxime. These embryogenic calls were subcultured to the same selection medium at every 2 weeks intervals. Approximately 24.5% of the early globular embryos co-cultivated with Agrobacterium for 3days produced kanamycin or/and hygromycin-resistant calli. Transgenic somatic embryos were converted into plantlets in half strength MS medium supplemented with 3mg/L GA$_3$ kanamycin and were confirmed by GUS histochemical assay and polymerase chain reaction analysis. Genomic Southem blot hybridization confirmed the incorporation of NPT II gene into the host genome.

Antioxidative and Antiaging Activities and Component Analysis of Lespedeza cuneata G. Don Extracts Fermented with Lactobacillus pentosus

  • Lespedeza cuneata G. Don is a traditional herb that has been associated with multiple biological activities. In this study, we investigated the antioxidative/antiaging activities and performed an active component analysis of the non-fermented and fermented (using Lactobacillus pentosus) extracts of Lespedeza cuneata G. Don. The antioxidative activities of the fermented extract were higher than those of non-fermented extracts. The elastase inhibitory activity, inhibitory effects on UV-induced MMP-1 expression, and ability to promote type I procollagen synthesis were investigated in Hs68 human fibroblasts cells. These tests also revealed that the fermented extract had increased antiaging activities compared with the non-fermented extract. A component analysis of the ethyl acetate fractions of non-fermented and fermented extracts was performed using TLC, HPLC, and LC/ESI-MS/MS to observe changes in the components before and after fermentation. Six components that were different before and after fermentation were investigated. It was thought that kaempferol and quercetin were converted from kaempferol glucosides and quercetin glucosides, respectively, via bioconversion with the fermentation strain. These results indicate that the fermented extract of L. cuneata G. Don has potential for use as a natural cosmetic material with antioxidative and antiaging effects.

A Study on the Development of a Novel Pressure Sensor based on Nano Carbon Piezoresistive Composite by Using 3D Printing (3D 프린팅을 활용한 탄소 나노 튜브 전왜성 복합소재 기반 압력 센서 개발 연구)

  • This paper presents an ongoing study to develop a novel pressure sensor by means of a Nano Carbon Piezoresistive Composite (NCPC). The sensor was fabricated using the 3D printing process. We designed a miniaturized cantilever-type sensor electrode to improve the pressure sensing performance and utilized a 3D printer to build a small-sized body. The sensor electrode was made of 2 wt% MWCNT/epoxy piezoresistive nano-composite, and the sensor body was encapsulated with a pipe plug cap for easy installation to any pressure system. The piezoresistivity responses of the sensor were converted into stable voltage outputs by using a signal processing system, which is similar to a conventional foil strain gauge. We evaluated the pressure-sensing performances using a pressure calibrator in the lab environment. The 3D-printed cantilever electrode pressure sensor showed linear voltage outputs of up to 16,500 KPa, which is a 200% improvement in the pressure sensing range when compared with the bulk-type electrode used in our previous work.

Characteristics of Culture Conditions for the Production of Crude Biosurfactant by Bacillus subtilis JK-1 (Bacillus subtilis JK-1의 생물계면활성제 생산을 위한 배양 특성)

  • Optimal culture conditions were characterized for production of crude biosurfactant of Bacillus subtilis JK-1. During incubation of B. subtilis JK-1, the bacterial growth pattern, changes of the surface tension at variable temperatures, pH and NaCl concentrations in bacterial culture medium were studied. The strain was able to grow and produce biosurfactant at $15-45^{\circ}C$, in the pH range of 6-10, and at 0-10% (w/v) NaCl. In case, culture broth pH was gradually changed to neutral or weak alkaline. Optimal culture conditions for crude biosurfactant production were at $35^{\circ}C$ and pH 7.0 after 48 h incubation and the surface tension of biosurfactant was 24.0 mN/m. Besides, as the concentration of NaCl was increased from 0 to 10% (w/v), the growth was decreased, pH of the culture broth was converted from weak alkaline to acidic, and the surface tension rised.

  • The temperature dependency and the stability of enzyme systems for $11{\alpha}-hydroxyla­tion$ of progesterone were investigated using Aspergillus phoenicis. Though A. phoenicis conserves high enzyme activities for lactose hydrolysis even at high temperatures, the bioconversion reaction of progesterone by this strain was found to be very temperature sensitive. The compositions of reaction mediums of inside and outside of cells were analyzed using sonication technique. At early stage of reaction, the concentration of $11{\alpha}-hydroxyprogesterone$ of cell inside was higher than that of outside. But as the reaction proceeded further, the $11{\alpha}-hydroxyprogesterone$ existing inside of cells being converted into another products, its concentration was lower within the cells that in the bulk medium. Even in the reaction mediums containing organic solvents, A. phoenicis was founded to be able to metabolite, so that $11{\alpha}-hydroxyprogesterone$ can be produced continuously from fixed bed reactions packed with immobilized A phoenicis in vivo.

  • The present process of grading leather quality by the rare eyes is not reliable. Because inconsistency of grading due to eyes strain for long time can cause incorrect result of grading. Therefore it is necessary to automate the process of grading quality of leather based on objective standard for it. In this paper, leather automatic classification system consists of the process obtaining the information of leather and the process grading the quality of leather from the information. Leather is graded by its information such as texture density, types and distribution of defects. This paper proposes the algorithm which sorts out leather information like texture density and defects from the gray-level images obtained by digital camera. The density information is sorted out by the distribution value of Fourier spectrum which comes out after original image is converted to the image in frequency domain. And the defect information is obtained by the statistics of pixels which is relevant to Window using searching Window after sort out boundary lines from preprocessed images. The information for entire leather is used as standard of grading leather quality, and the proposed algorithm is practically applied to machine vision system.

Bioconversion of Ginsenoside Rb1 to Compound K using Leuconostoc lactis DC201

  • Ginseng (Panax ginseng) is frequently used in Asian countries as a traditional medicine. The major components of ginseng are ginsenosides. Among these, ginsenoside compound K has been reported to prevent the formation of malignancy and metastasis of cancer by blocking the formation of tumor and suppressing the invasion of cancer cells. In this study, ginsenoside $Rb_1$ was converted into compound K, via secreted ${\beta}$-glucosidase enzyme from the Leuconostoc lactis DC201 isolated, which was extracted from Kimchi. The strain DC201 was suspended and cultured in MRS broth at $37^{\circ}C$. Subsequently, the residue from the cultured broth supernatant was precipitated with EtOH and then dissolved in 20 mM sodium phosphate buffer (pH 6.0) to obtain an enzyme liquid. Meanwhile, the crude enzyme solution was mixed with ginsenoside $Rb_1$ at a ratio of 1:4 (v/v).The reaction was carried out at $30^{\circ}C$ and 190 rpm for 72 hours, and then analyzed by TLC and HPLC. The result showed that ginsenoside Rb1 was transformed into compound K after 72 hours post reaction.

Application of Saccharified Acorn-starch for Biomass and Lipid Accumulation of Microalgae (당화된 도토리의 전분이 미세조류 바이오매스 증식과 바이오오일 함량에 미치는 영향)

  • The growth of the algae strain Chlorella vulgaris under mixotrophic conditions in the presence of saccharified acorn-starch (acorn-glucose) was evaluated with the objective of increasing biomass growth and triacylglycerols (TAGs) content. The results indicated that 81.3% of starch was converted to glucose in acorns. C.vulgaris algal strains grown with acorn-glucose produced higher biomass and TAGs content than with autotrophic growth. The highest biomass production and TAGs content with 3 g/L acorn-glucose were 12.44 g/L and 32.9%, respectively. Biomass production with 3 g/L acorn-glucose was 16.4 fold higher than under autotrophic growth condition. These findings suggested that 3 g/L acorn-glucose is economic and efficient for biomass production/productivity and TAGs content of microalgae. This study provides a feasible way to reduce the cost of bioenergy production from microalgae.

Spectral Element Formulation for Analysis of Lamb Wave Propagation on a Plate Induced by Surface Bonded PZT Transducers (표면 부착형 PZT소자에 의해 유발된 판 구조물의 램파 전달 해석을 위한 스펙트럼 요소 정식화)

  • This paper presents spectral element formulation which approximates Lamb wave propagation by PZT transducers bonded on a thin plate. A two layer beam model under 2-D plane strain condition is introduced to simulate high-frequency dynamic responses induced by a piezoelectric (PZT) layer rigidly bonded on a base plate. Mindlin-Herrmann and Timoshenko beam theories are employed to represent the first symmetric and anti-symmetric Lamb wave modes on a base plate, respectively. The Euler-Bernoulli beam theory and 1-D linear piezoelectricity are used to model the electro-mechanical behavior of a PZT layer. The equations of motions of a two layer beam model are derived through Hamilton's principle. The necessary boundary conditions associated with the electro-mechanical properties of a PZT layer are formulated in the context of dual functions of a PZT layer as an actuator and a sensor. General spectral shape functions of response field and the associated boundary conditions are obtained through equations of motions converted into frequency domain. Detailed spectrum element formulation for composing the dynamic stiffness matrix of a two layer beam model is presented as well. The validity of the proposed spectral element is demonstrated through numerical examples.

Experimental Study on the Drawbar Pull and Structural Safety of an Onion Harvester Attached to a Tractor (트랙터 부착형 양파수확기의 작업 속도에 따른 견인 부하와 구조 안정성에 관한 실험적 연구)

  • Recently, due to labor shortages in rural areas within South Korea, the demand for upland-field machinery is growing. In addition, there is a lack of development of systematic performance testing of upland-field machinery. Thus, this study examined structural safety and drawbar pull based on soil properties, as a first step for systematic performance testing on the test bed. First, the properties of soil samples from 10 spots within the experimental site were examined. Second, the strain was measured and converted into stress on 8 points of an onion harvester that are likely to fail. More specifically, the chosen parts are linked to the power, along with the drawbar pull, using a 6-component load cell equipped between the tractor and the onion harvester. The water content of the soil ranged between 5.7%-7.5%, and the strength between 250-1171 kPa. The test soil was subsequently classified into loam soil based on the size distribution ratio of the sieved soil. The onion harvester can be considered as structurally safe based on the derived safety factor and the drawbar pull of 115-1194 kgf, according to the working speed based on agricultural fieldwork.
