Effect of Core Stabilization Intervention Program on Erector Spinae Contractile Properties and Isokinetic Muscle Function in Adults with Sedentary Lifestyle Patterns (코어안정화 운동이 좌식생활 패턴 성인의 척추기립근 수축 속성 및 체간 등속성 근기능에 미치는 영향)

  • Objective: The aim of this study was to investigate effect of core stabilization exercises on the erector spinae contractile properties and trunk isokinetic muscle function of middle age with low physical activity and sedentary lifestyle. Method: Twenty (female: n=10, male: n=10) middle-age subjects (age: 37.25 ± 6.08 years, height: 168.01 ± 6.84 cm, weight: 71.37 ± 11.75 kg) participated in this study. Tensiomyography was measured on the erector spinae, and the isokinetic trunk muscle function test was measured at an angular velocity of 60 °/s and 90 °/s. All subjects performed the core stabilization exercises for 60 min per day, 3 times a week, for 7 weeks. A paired t-test was performed with a significance level of 0.05. Results: Tensiomyography of the erector spinae revealed a significant post-exercise increase in the maximum radial displacement (p < .05) and velocity of contraction (p < .05), however, there wasn't a significant post-exercise change in the contraction time. Additionally, the isokinetic muscle function test of the trunk revealed a significant post-exercise increase in trunk extensor relative strength (p < .05) and strength ratio (p < .05). Conclusion: Our results indicated that core stabilization exercises reduced erector spinae muscle stiffness, increased the velocity of erector spinae contraction. Additionally, data showed the improvement in the trunk extensor strength help induce a more balanced development in trunk muscle.

  • The effects of Artemisia princeps var orientalis(mugwort), Circium japonicum var ussuriense (Unggungqui) on cadiovascular system in hyperlipidemic rats were investgated. Thirty rats devided into 5 experimental groups, were fed with the diet contained 1% chlesterol, 0.25% sodium cholate, 10% coconut oil and 5% lard by the same method of previous paper1). Contractile and relaxation responses in the isolated artria and thoracic aortae were measured and the morphological changes of the aortic endotherium from the rats were inspected. The responses of the right atrial to isoproterenol were significantly lower value in Ungungqui powder diet group(UP) and mugwort powder diet group(MP) than the control. The contraction force by injectin of phenylephrine and calcium in isolated thoracic aortae was significantly low value in the UP and the MP groups compaired to the control. The relaxation rate by acetylcholine in isolated thoracic aortae represented significantly higher value in UP than control. The morphological changes of endothelial cell suface was smallest in UP and the damage of endothelium by retarded in MP. Although Ungungqui and mugwort extract diet groups(UE, ME) were advanced, those were less than control.

The Preventive Effect of Fursulthiamine on Dietary Hypertension in Rats (Fursulthiamine이 흰쥐의 식이성 고혈압에 대한 예방효과)

  • Dietary hypertension induced by feeding cholesterol and vitamin D3 in rats for 7 weeks was prevented by the addition of fursulthiamine and diallyl disulfide in the diet. In the isolated preparation of the hypertentive rats, the contractile forces were potentiated by norepinephrine, serotonin and potassium chloride, as compared to that of the normotensive rats. However, the increased contractile forces of the hypertensive rats were inhibited in the aorta of the rats fed with fursulthiamine. Decreases in triglyceride contents were observed in the sera of the hypertensive rats fed with fursulthiamine. Therefore, it is suggested that dietary hypertension in rats may be prevented by supplement of fursulthiamine in the diet.

  • Objective : This study was undertaken to evaluate the effect of Sunghyangchungisan (SHCS) on the regulation of vascular tone and $Ca^{2+}$ metabolism in arterial tissues. Vascular rings isolated from rabbit carotid artery were myographed isometrically in isolated organ baths and the effect of SHCS on contractile activities, endothelial function and $Ca^{2+}$ metabolism were determined. Methods : In phentobarbital sodium-anesthetized rabbits, SHCS administered through ear vein (100 mg/Kg body wt.) or intragastric dwelling tube (300 mg/Kg body wt.) attenuated phenylephrine (PE, 10 ${\mu}g$/Kg, i.v.)-induced increases in both systolic and diastolic cartoid arterial blood pressure. Results : In experiments with isolated arterial strips, SHCS relaxed arterial rings which were pre-contracted by phenylephrine (PE, 1 ${\mu}M$). The responses to SHCS were partially dose-dependent at concentrations lower than 0.5 mg/ml. When SHCS was applied prior to the exposure to PE, it inhibited the PE-induced contraction by a similar magnitude which was comparable to the relaxation of pre-contracted arterial rings. Washout of SHCS after observing its relaxant effect resulted in a full recovery of PE-induced contractions, indicating that the action mechanism is reversible. The observation that SHCS did not change the $ED_{50)$ of PE oh its dose-response curve ruled out the possible interaction of SHCS with ${\alpha}$-receptors. The relaxant effect of SHCS was not affected by removal of endothelium or a nitric oxide synthase inhibitor, L-NAME. Methylene blue, an inhibitor of the soluble guanylate cyclase, did not affect the relaxant effect of SHCS. These results suggest that the action of SHCS is not mediated by the endothelium nor soluble guanylate cyclase. Constant cGMP production determined in arterial strips in the presence or absence of SHCS is consistent with this conclusion. When contraction was induced by additive application of $Ca^{2+}$ in arterial rings which were pre-depolarized by high $K^+$ in a $Ca^{2+}$-free solution, the relaxant effect of SHCS was attenuated by increasing the $Ca^{2+}$ concentration. SHCS, when applied to the arterial rings pre-contracted by PE and then relaxed by nifedipine, a $Ca^{2+}$ channel blocker, did not show additive relaxation. SHCS partially blocked $Ca^{2+}$ influx stimulated by PE and high $K^+$ which was determined by 5-min ^{45}Ca$ uptake, while it did not affect $Ca^{2+}$ efflux. Conclusions : From above results, it is suggested that SHCS relax PE-induced contraction of rabbit carotid artery in an endothelium independent manner, andinhibition of $Ca^{2+}$ influx may contribute to the underling mechanism.

Change of isometric contractile force and muscle activity applying ice and hot according to the time on biceps brachii muscle (온열 및 한랭의 적용시간에 따른 상완이두근의 등척성 수축력과 근활성도 변화)

  • This study has investigated the effect of isometric contractile force and muscle activity applying ice and hot according to the time from the biceps brachii muscle. In this study, 20 university students participants without musculoskeletal and neurological disorders. By applying a hot pack 5 min, 10 min, 20 min and 30 min respectively and ice pack for 5 min, 10 min, and 20 min respectively. After that measurement are skin temperature, contractile force and muscle activity. Skin temperature of the hot and cold 5 min applied that rapidly changing. Increasing the time it takes to apply a variance has been reduced(p<.001). Isometric contractile force was not statistically significant but highest when applying the hot pack 5 minutes and lowest when applying the hot pack 30 minutes(p<.001). Muscle activity and median frequency was highest when applying the hot pack 5 minutes. To analyze the above results, it was found that isometric contractile force and muscle activity changed according to the applying time. These result lead us to the conclusion that this study will be more evidence for changes in muscle contraction to apply hot pack and ice pack on clinic.

Effects of the Fractionation of Sophorae Radix Water Extract on the Langendorff hearts (고삼 유기용매 분획층이 적출 심장에 미치는 영향)

  • The water extract of Sophorae radix was tested for its preventive effects against cardiovascular anaphylaxis elicited in experimental animals. H₂O and ethyl acetate fractionation of Sophorae radix water extract improved anaphylactic cardiac dysfunction in passively sensitized isolated guinea hearts: improvement was noted in the maximal contractile force, post-challenge contractile force, post-challenge coronary flow and creatine kinase change elevation. These results suggest that H₂O and ethyl acute fractionation of Sophorae radix water extract possesses anti-anaphylactic effect in cardiovascular system.

Effect of C1 Esterase Inhibitor on the Cardiac Dysfunction Following Ischemia and Reperfusion in the Isolated Perfused Rat Heart

  • Complement-mediated neutrophil activation has been hypothesized to be an important mechanism of reperfusion injury. It has been proposed that C1 esterase inhibitor (C1 INH) may prevent the complement- dependent activation of polymorphonuclear leukocytes (PMNs) that occurs within postischemic myocardium. Therefore, The effect of C1 INH was examined in neutrophil dependent isolated perfused rat heart model of ischemia (I) (20 min) and reperfusion (R) (45 min). Administration of C1 INH (5 mg/Kg) to I/R hearts in the presence of PMNs $(100{\times}10^6)$ and homologous plasma improved coronary flow and preserved cardiac contractile function (p<0.001) in comparison to those I/R hearts receiving only vehicle. In addition, C1 INH significantly (p<0.001) reduced PMN accumulation in the ischemic myocardium as evidenced by an attenuation in myeloperoxidase activity. These findings demonstrate the C1 INH is a potent and effective cardioprotective agent inhibits leukocyte-endothelial interaction and preserves cardiac contractile function and coronary perfusion following myocardial ischemia and reperfusion.

  • The rates of deterioration of contractile forces of isolated hearts from ginseng component treated rats were determined. Rat papillary muscles were also used to study the influence of ginseng on the mechanical performance of heart. Rats weighing 200-300g were administered orally with ginseng ethanol extract (100mg/kg/day), ginseng total saponin (50mg/kg/day) and ginsenoside Rbl (5mg/kg/ day) for a week respectively. The isolated hearts from rats were perfused with Krebs-Henseleit solution by Langendorff perfusion apparatus. The force-velocity relation was clearly seen with the load-generator equipped isotonic shortening recording apparatus. The control group was only able to maintain 60% of their initial contractile forces after 120 minutes of perfusion, whereas ginseng ethanol extract treated group was able to sustain nearly their initial strength even after 120 minutes of perfusion. The similar effects were seen in the hearts treated with total ginseng saponin and ginsenoside Rb$_{1}$. Ginseng ethanol extract did alter mechanical performance of rat ventricular myocardium. It increased both maximum velocity(Vmax) of isotonic shortening and isometric force (P$_{0}$) and showed increased velocity of shortening significantly (P<0,05) at any one afterload.d.

Influence of Dichloroisoproterenol and Panax Ginseng on the action of some drugs. (Dichloroisoproterenol및 인삼(人蔘)의 이삼약물(二三藥物)의 작용(作用)에 미치는 영향(影響))

  • Dichloroisoproterenol(DCI) i; one of the well known ${\beta}$-adrenergic receptor blocking agents. According to Moran and Perkins, DCI has sympathomimetic like action in relatively low concentrations. Fleming and Hawkins confirmed that DCI acts upon the receptors concerned with positive chronotropic and inotropic actions in the heart. Vogins reported that DCI, in concentration of $5{\times}10^{-8}$ to $5{\times}10^{-6}g/ml$, had properties of sympathomimetic amine causing positive inotropic and chronotropic actions in normal rat atria. And James and Nadeau found that DCI had not only adrenergic blocking effect in moderate and higher concentrations, but it also blocked the effect on the sinus node by vagal stimulation and of directly administered acetylcholine in higher concentrations. As stated above by many authors, DCI has complicated actions according to its concentrations. Our aim at the present experiments was to study the effects of DCI to the action of ouabain and acetylcholine upon the excised rabbit atria, as well as to the action of barium chloride and acetylcholine upon the excised rabbit intestine. In addition, Pan ax Ginseng is widely used as tonics in oriental nations, its pharmacological action, however, has not been clearly established. So we atempted to investigate the effects of the water extract of Panax Ginseng to the action of ouabain and DCI upon both atria and intestine. The results obtained were as follows. 1) DCI has a negative inotropic effect on the excised rabbit atria at concentration of $10^{-5}$ and a positive inotropic effect at concentration of $10^{-6}$. 2) DCI (at concentration of $10^{-6}$) potentiates the positive inotropic effect of ouabain upon the excised rabbit atria. 3) DCI antagonizes the action of acetylcholine upon the excised rabbit atria. 4) The water extract of Panax Ginseng, at concentration of $10^{-3}$, decreases the contractile force of rabbit atria, and tends to slightly increase that of rabbit atria at $10^{-4}$. 5) The water extract of Panax Ginseng exhibits a synergistic action with ouabain on the contractile force of rabbit atria. 6) DCI, in concentrations of $10^{-7}{\sim}10^{-6}$, depresses the tone and amplitude of contraction of the excised rabbit intestine. The depression of the intestinal tone markedly appears in pretreatment with reserpine 2mg/kg 24 hours. 7) DCI antagonizes the contractile effect of barium chloride on the excised rabbit atria. 8) DCI has no significant influence on the action of acetylcholine upon the excised rabbit intestine. 9) The series of those evidences indicates that DCI has a sympathomimetic-like action and more over a relaxing action directly on the excised rabbit intestine. 10) The water extract of Panax Ginseng in concentrations of $10^{-4}{\sim}10^{-3}$, has transient depression of the intestinal tone, but later gradually recovers its normal motility: 11) The water extract of Panax Ginseng has a synergistic action with ouabain on the intestinal contractility.

  • Objectives : It has been reported that Cordyceps militaris Mycelia(CMM) has an effect on deficiency allergic asthma(虛喘) clinically. The aim of this study was to determine an appropriate oriental treatment and the doses of CMM to treat asthma. Methods : In order to study the effect of herbal acupuncture solution of the CMM on allergic asthma, the mouse were pretreated by CMM herbal acupuncture at BL13, LU4 before antigen sensitization. 2 days later Mice were actively sensitized with a subcutaneous injection of ovalbumin(OA) and 13 days later they were provoked with OA aerosols. IL-4, lymphocyte, macrophage in bronchoalveolar lavage fluid(BALF), IgE in serum, WBC, RBC, HGB in blood, and in vitro isometric contractile responses of the isolated tracheal smooth muscle(TSM) to acetylcholine$(ACh,\;0.1-1000\;{\mu}M)$, KCl were measured. Results : Contractile responses of TSM to ACh were significantly increased in CMM herbal acupuncture 1 group $(Ach\;1000\;{\mu}M)$, CMM herbal acupuncture 2 group $(ACh\;1,\;10\;{\mu}M)$, CMM herbal acupuncture 3 group $(Ach\;0.3,\;1,\;30,\;300\;{\mu}M)$. The sensitivity of TSM to ACh was significantly decreased in CMM herbal acupuncture 3 group. The maximal contractile response of TSM to ACh was significantly decreased in CMM herbal acupuncture 1, 3 group. The maximal contractile response of TSM to KCl was significantly decreased in CMM herbal acupuncture 1, 2, 3 group. The counts of lymphocytes in BALF was significantly increased in CMM herbal acupuncture 3 group. The counts of macrophages in BALF was significantly decreased in CMM herbal acupuncture 3 group. Interleukin-4 level in BALF was significantly increased in CMM herbal acupuncture 1,3 group. and it was increased in CMM herbal acupuncture 2 group, but there was no significance. Serum IgE level was significantly decreased in CMM herbal acupuncture 1, 2, 3 group. The counts of WBC in blood was significantly increased in CMM herbal acupuncture 1, 3 group Conclusion : Based on the above results it is assumed that CMM herbal acupuncture at BL13, LU4 can help the treatment of deficiency allergic Asthm.
