Investigation of Microbial Contamination and Working Environment in University Foodservices (대학급식소 작업시설과 환경의 미생물 오염도 분석 및 작업환경 실태조사)

  • The purpose of this study was to identity the probability of cross-contamination from the environment. For this, we examined foodservices at 20 universities/colleges for microbiological analysis of their working facilities and environment as well as their preventive equipment against cross-contamination. Seventy percent of the 20 foodservices were found to maintain one unified working area, which suggests high probability of contamination of food/utensils/equipment in the cooking area by pre-preparation or dish washing. According to the microbiological analysis, the hygiene acceptance ratio of working facilities in the clean zone was 70%, which was higher than the average 45% hygiene acceptance ratio of working facilities in the contamination operating zone. There was a significant difference in the total plate count (P<0.001) and coliform count (P<0.01), which demonstrates that work tables in the clean zone were in a good state compared to those in the contamination operating zone. In the contamination operating zone, refrigerator shelves had a high probability of cross-contamination. Regarding the floor surface and airborne microbes, cooking areas which should be maintained as clean zones had higher cross-contamination probability than those in the contamination operating zone. So corrective actions such as cleaning and sanitizing, keeping dry floors, lowered temperature and humidity, shoe disinfecting facilities, and checking concentrations, are necessary to manage floor surfaces and airborne microbes in the cooking area.

A Estimation of Contamination Degree by the Measurement of Phase Difference on the Outdoor Insulator (옥외절연물의 오손도 파악을 위한 위상차 측정)

  • In this paper, a phase difference of liveline insulator was measured and analyzed to estimate the contamination degree. The probability of accident rises according to the increase of the contamination. So the estimation of contamination degree is very important in design and maintenance of outdoor insulator. There are many methods to measure the contamination degree, but those methods use the spacial sense to measure the contamination degree. So, we have no information about the contamination from the liveline insulator, but there is a measurement of phase difference to estimate the contamination degree. The phase difference is varied with the increase of ESDD, so the contamination degree could be estimated by using those characteristics. The experiment is performed to analyze the interrelationship between contamination degree and phase difference. The variable condition is relative humidity that increases from 30 to 100 % in a chamber. We use the DAS program to measure and analyze the data. From the result, the variation of phase difference is different by the contamination degree, so we can confirm the possibility of estimating the contamination degree by the phase difference.

Risk Assessment for Salmonellosis in Chicken in South Korea: The Effect of Salmonella Concentration in Chicken at Retail

  • Salmonellosis caused by chicken consumption has been a critical issue in food safety worldwide, including in Korea. The probability of illness from consumption of chicken was simulated in study, based on the recipe of Dakgalbi, a commonly eaten chicken dish in Korea. Additionally, the processing stage at slaughterhouses to decrease Salmonella concentration in broilers was modeled to explore its effect on the likelihood of illness. A Monte Carlo simulation model was created using @RISK. Prevalence of Salmonella in chickens at the retail stage was found to be predominantly important in determining the probability of illness. Other than the prevalence, cooking temperature was found to have the largest impact on the probability of illness. The results also demonstrated that, although chlorination is a powerful tool for decreasing the Salmonella concentration in chicken, this effect did not last long and was negated by the following stages. This study analyzes the effects of variables of the retail-to-table pathway on the likelihood of salmonellosis in broiler consumption, and also evaluates the processing step used to decrease the contamination level of Salmonella in broilers at slaughterhouses. According to the results, we suggest that methods to decrease the contamination level of Salmonella such as chlorination had little effect on decreasing the probability of illness. Overall, these results suggest that preventing contamination of broiler with Salmonella must be a top priority and that methods to reduce the concentration of Salmonella in broilers at slaughterhouses hardly contribute to safe consumption of Salmonella-contaminated chicken.

  • Satellite contamination from back-scattered molecules has long been analyzed using the BGK theory or the DSMC technique which are rather inefficient in that they are complicated or take a long time in the analysis. This study presents a new technique of estimating the back-scattering contamination, which is very simple and easy to use like the view factor method and also very accurate. This method, called the modified view factor method, is equivalent to the DSMC in so far as the molecular thermal velocity is much smaller than the satellite velocity and the mean free path much longer than the satellite.

The Contamination Characteristics of BTEX and TPH Components in Silty Soils with the Oil Leakage Event from Point Source (점오염원 형태의 유류누출 사건에 의한 실트질 토양층에서 BTEX와 TPH 성분의 오염도 연구)

  • The contamination characteristics of BTEX and TPH components in silty soils with the oil leakage event from point source were studied. The over ratios of three soil pollution standard for TPH component were $1.5{\sim}1.7$ times higher than that of BTEX component. The mean and maximum values of BTEX and TPH components with sample points were B-zone > A-zone > C-zone, and the highest concentrations were measured at $1{\sim}2m$ depth below surface. BTEX and TPH components were increased with linear distance in zone within 120 m and 80 m from point source. For the zone more than 120 m, BTEX and TPH concentrations were under soil pollution standard. The cutoff values of indicator kriging using BTEX and TPH components were defined as confirmative limit, warn- ing limit and counterplan limit. The variograms of indicator-transformed data were selected linear model. The contamination ranges of BTEX and TPH components using confirmative limit and warning limit were estimated similar, but the contamination range of those using counterplan limit was much reduced. The maximum contamination probabilities were estimated by probability maps usinB confirmative limit, warning limit and counterplan limit. The maximum contamination probabilities with three soil pollution standard were estimated 26%, 26% and 13% for BTEX component, and 44%, 38% and 26% for TPH component.

  • Probabilistic exposure assessment has been recognized as an important tool in microbial risk assessment, because of obtained the desired results to characterize of variability and uncertainty associated with the microbial hazards. In addition, it will be provided much more actuality information than the point-estimate approaches. In this study, we present methodology using mathematical probability distribution in exposure assessment and estimating of contamination level of Listeria monocytogenes in meat and meat products as a case study. The result of estimation contaminatin level was mean ($50^{th}$ percentile) -4.08 Log CFU/g minimum ($5^{th}$ percentile) -4.88 Log CFU/g, maximum ($95^{th}$ percentile) -3.56 Log CFU/g.

Choosing Optimal STR Markers for Quality Assurance of Distributed Biomaterials in Biobanking

  • The quality assurance (QA) is of utmost importance in biobanks when archived biomaterials are distributed to biomedical researchers. For sample authentication and cross-contamination detection, the two fundamental elements of QA, STR genotyping is usually utilized. However, the incorporated number of STR markers is highly redundant for biobanking purposes, resulting in time and cost inefficiency. An index to measure the cross-contamination detection capability of an STR marker, the mixture probability (MP), was developed. MP as well as other forensic parameters for STR markers was validated using STR genotyping data on 2328 normal Koreans with the commercial AmpFlSTR kit. For Koreans, 7 STR marker (D2S1338, FGA, D18S51, D8S1179, D13S317, D21S11, vWA) set was sufficient to provide discrimination power of ${\sim}10^{-10}$ and cross-contamination detection probability of ${sim}1$. Interestingly, similar marker sets were obtained from African Americans, Caucasian Americans, and Hispanic Americans under the same level of discrimination power. Only a small subset of commonly used STR markers is sufficient for QA purposes in biobanks. A procedure for selecting optimal STR markers is outlined using STR genotyping results from normal Korean population.

Investigation of Pollution of Polycyclic Aromatic Hydrocarbons and Heavy Metals in Soil near Railway Rails (철도레일 부근 토양의 다환방향족 탄화수소 및 중금속 오염도 조사)

  • Trains have been a major means of transport in Korea during these past decades. However, train facilities such as stations and repair shops are contaminated with organic and inorganic substances. There is a high probability of train facility contamination with polyaromatic hydrocarbons (PAHs). This study evaluated the PAH and heavy metal contamination of soil near railroads in the Kyungpook area. A total of 18 soil samples were collected from the railroads and analyzed for 16 PAHs and 6 heavy metal species. The contamination level of the top soil was found to be slightly higher than that of the subsoil for contamination with PAHs. The ratio of carcinogenic PAH concentration to the total PAH concentration was relatively high, with a maximum of 0.9. The toxicity equivalent (TEQ) of the PAHs were 500.6 ng/kg in the topsoil and 355.5 ng/kg in the subsoil. The ratio of low molecular PAHs (LPAHs) to high molecular PAHs (LPAHs) ranged from 6.7 to 29.5; this shows that contamination is primarily due to combustion of fuel rather than due to petroleum. The ratio of phenanthrene to anthracene and the ratio of fluoranthene to pyrene also show that contamination occurred due to combustion for transportation. The heavy metal contamination level was lower than the Korean standard, but higher than the background concentration; this indicates that the soil was affected by the operation of the railways.

Using GIS Modeling to Assess the Distribution and Spatial Probability of Soil Contamination of Geologic Origin in Korea (GIS 모델링을 이용한 국내 지질 기원 토양오염의 분포 현황과 공간적 개연성 연구)

  • Soil contaminants measured and managed in Korea include those of geologic origin such as arsenic, cadmium, copper, lead, zinc, nickel, mercury, and fluoride. This study identifies the distribution of these contaminants using GIS modeling to analyze the spatial probability of soil contamination originating from geology. The modeling found that cadmium, copper, lead, nickel, and mercury often exceed the regulated standard by <1%. Concentrations of arsenic and zinc greatly exceeded the standard in the vicinity of mines and industrial complexes: mining and industry seemed to have substantial effects on the concentrations of these metals. Although fluoride was sampled at the lowest number of points, its frequency of exceeding the standard was the highest. No obvious source of artificial contamination has been identified, and fluoride's distribution characteristics showed continuity over a wide area, suggesting a strong correlation between geological characteristics and fluoride concentration. The highest frequencies of fluoride exceeding the standard were in Jurassic granite (40.00%) and Precambrian banded gneiss (34.12%). As these rocks contributed to the formation of soil through their weathering, high fluoride concentrations can be expected in soil in areas where these rocks are distributed.

Estimation of Shelf Life Distribution of Seasoned Soybean Sprouts Using the Probability of Bacillus cereus Contamination and Growth

  • Growth of Bacillus cereus was assessed during the storage of seasoned soybean sprouts at 0,5, 10, and $15^{\circ}C$. No lag time in its growth curve was observed and thus the specific growth rate of B. cereus in the exponential growth phase was estimated for bootstrapped microbial count data. The distribution of the specific growth rate could be explained by the BetaGeneral distribution function, and temperature dependence was described by the Ratkowsky square root model. The temperature dependence of the growth could be successfully incorporated into the differential equation of microbial growth to predict the B. cereus count on the seasoned soybean sprouts under fluctuating temperature conditions. Safe shelf lives with different probabilities to reach $10^5\;CFU/g$ were presented at four different temperatures, considering the variation in initial contamination and specific growth rate by the Monte Carlo method and 2-step bootstrapping, respectively. Safe shelf lives defined as the time with a probability of less than 0.1% of reaching the critical limit, were 13.4, 5.2, 3.6, and 2.8 days at 0, 5, 10, and $15^{\circ}C$, respectively.
