This study was to analyze the genetic diversity of Quercus gilva Blume growing in Jeju Island for developing a preservation strategy. We examined the genetic diversity and structure using 6 ISSR primers and investigated 67 polymorphic ISSR amplicons in 80 trees distributed among five populations. The average of proportion of polymorphic loci were 93%, the average level of Shannon's information index was 0.237, and Nei's genetic diversity was 0.156. According to the analysis of the molecular variance (AMOVA), $F_{st}$ was 0.169 indicating there was a genetic variation among five populations. 17% of the total variation was allocated among the five populations, while the other 83% of the total variation was in individual trees in each population. The result could be due to the uneven number of trees among the five populations. Based on these results, the preservation strategy could be developed, for examples, considering for designation as "forest genetic resources conservation area" about the habitat, monitoring continuously, fostering the growth of seedling, ex situ preservation of genetic resources, and comparing the differences of environmental and genetic characteristic with population in ex situ.

A comprehensive analysis of the population structure of the sandfish (Arctoscopus japonicas), the most abundant fishery resource in the East Sea of Korea, has not been carried out, despite its importance in Korea. The present study examined the genetic diversity and differences between five populations (two Japanese and three Korean populations) of A. japonicas captured in the East Sea using both the 401 bp sequence of mitochondrial DNA (mtDNA, cytochrome b) and five microsatellite DNA (msDNA) markers. The results of the analysis using the Cyt b sequence revealed 27 haplotypes. Based on msDNA variations, the estimated expected heterozygosity (HE) in each population ranged from 0.68 (Gampo, Korea) to 0.7765 (Erimo, Japan). Pairwise FST and AMOVA tests using both the Cyt b sequence and msDNA data pointed to significant differences between the Korean and Japanese populations (mtDNA; FST=0.2648, p<0.05, msDNA; FST=0.0814, p<0.05). These results were similar to the results of UPGMA, PCA, and structure analysis. In these analyses, the five populations were assigned to two groups (Korean populations and Japanese populations). These results shed light on the genetic diversity and relationships of A. japonicas and contribute to research on the evaluation, conservation, and utilization of Korean A. japonicas as genetic resources.
Mating system parameters were estimated in a natural population of Pinus koraiensis which was located at Gwongeumseong in Mt. Seorak, South Korea. The estimated parameters from allozyme were as follows: 0.882 of multilocus outcrossing rates($t_m$), 0.881 of singlelocus outcrossing rates($t_s$), 0.368 of correlated paternity($r_p$), and 2.7 of number of effective pollen contributors. The estimated parameters from cpSSR markers were as follows: 0.831 of average of outcrossing rates and 12.4 of the average number of effective pollen contributors. The average outcrossing rate from two genetic markers was 0.857, which was similar to those estimated in other conifer species. More number of potential pollen contributors was estimated from cpSSR marker analysis compared with that estimated from allozyme marker analysis. This result sugges$t_s$ that cpSSR markers may be more useful than allozyme markers for identifying potential pollen contributors in the analysis of mating system.
Tribolodon hakonensis(Cypriniformes; Leuciscinae) is anadromous; they are born in freshwater, migrate back to the ocean, then return to their home stream for spawning from mid-March to early-June. Here, five microsatellites were used to assess the level of gene flow among T. hakonensis populations from the Samcheok-Oship Stream, South Korea. The frequencies of dominant alleles across several loci differed between down-and upstream populations divided by several weirs, and pairwise multilocus $F_{ST}$ estimate was significantly high(0.083). However, there were no signs of any loss of genetic variation in the upstream population. Assignment tests of individuals in admixture model(K=2) to a set of baseline samples showed fairly correct assignment to each cluster; all of upstream individuals sere included in the first cluster, while the majority of downstream individuals(65%) comprise the second cluster. These results indicate reduced gene flow between up- and downstream populations but allowing passive downstream drift. It is likely that man-made structures might at least partially be a factor for creating and consolidating the current distribution patterns of genetic variation among T. hakonensis populations in the Samcheok-Oship Stream. This information will assist governing agencies in making informed decisions regarding conservation of anadromous fishes in Korean drainage systems.
The distribution status and habitat characteristics of the endangered species, Lethenteron reissneri were investigated in 2011~2012 in Korea. During the study period, L. reissneri were collected 60 stations among 755 sampling stations investigated. The total numbers of ammocoetes and adults were 208 and 435, respectively from eight stations of the Han River drainage system, four stations of Geum River drainage system, 32 stations of Seomjin and Yeongsan River drainage systems, and 16 stations of Nakdong River drainage system. The Han and Geum river drainage systems, the Sapkyo Stream and independent streams running toward the East Sea were estimated to have suffered sudden reductions of population density, whereas the Seomjin River drainage system showed increased population density compared with historic occurrence records. L. reissneri inhabited the middle-upper stream of clean water with well-developed rapids and lots of aquatic organisms. According to IUCN Red List categories and criteria L. reissneri was assessed to be vulnerable (VU) [B2ab (ii,iii,iv,v);D1] because of the rapid reduction of the occupying areas and the small number of mature individuals. Besides the distribution status and habitat characteristics we discussed the conservation strategies of L. reissneri.
Pinus pumila, which occurs in the northeast Asia, is found limitedly in Daecheongbong area of Mt. Seorak in the South Korea. This population was chosen to study spatial pattern, genetic diversity and spatial genetic structure. There were 48 polymorphic and 30 monomorphic I-SSR markers. A total of 65 individuals which distributed in the study site (40 m ${\times}$ 70 m) showed weakly aggregate distribution (Aggregate Index = 0.871). A total of 40 genets were observed from 65 individuals through I-SSR genotype comparison. Proportion of distinguishable genotype (G/N), genotype diversity (D) and genotype evenness (E) were 61.5%, 0.977 and 0.909, respectively. In spite of the small number and the limited distribution, Shannon's diversity index (I = 0.567) was relatively high as compared with those of other plant species. Spatial autocorrelation using Tanimoto's distance showed that the genetic patch was established within 12 m. Based on Mantel tests, there was relatively low correlation between genetic distance and geographic distance. Therefore, it seems the P. pumila population was formed by many parent trees in early stage. For ex situ genetic conservation of P. pumila, the sampling strategy is efficient at least above 12 m between individual trees.
This study was conducted to investigate the effect of cone protective net and the variation of cone and seed characteristics of a unique and rare dwarf stone pine (Pinus pumila Regel) population in Mt. Seorak which is in the southern peripheral of South Korea. Iron protective net (50 mesh, 25 ${\times}$ 25 cm) was effective in reducing birds and rodents damage to conelet, thereby it was possible to safely collect seed genetic resources. Early July was observed to be the most appropriate season to cover conelet with Iron protective net. P. pumila have ovoid-triangle shaped, wingless seeds and indehiscent cones, seeds dissemination is mainly by the hoarding behavior of nutcrackers and rodents. Statistical analysis showed that there were significant differences among individuals within population, and the mean characteristics of the species were 35.3 mm, 25.6 mm, 1.38, 6.6 g and 39.3 ea in the cone length, cone width, cone index, cone weight and in number of seeds per cone, respectively. Coefficients of variations in seed weight and number of seeds per cone were relatively high (21.7%, 21.5%, respectively) compared to other traits. Based on the correlation analysis between cone characteristics and seed characteristics, the length of cone and seed showed a significant positive correlation with the seed size and the seed weight, but the width and thickness of seed showed a negative correlation with the number of seeds per cone.
The conservation project to protect an endangered fish, Gobiobotia naktongensis was executed against declining the gene diversity of the fish after the construction of Youngju Dam in a sand-bed stream (Naeseongcheon Stream). We tried to move the populations of G. naktongensis from submerged planned sites to alternative habitats, bred artificially and augmented the juveniles to optimal habitat, and monitored the results of the restoration implementation. No entity of G. naktongensis was confirmed at the planned submerged sites despite attempting to capture more than 8 times and eventually the movement to alternative habitat could not be implemented. About 40 individuals of G. naktongensis were captured in the Naeseongcheon Stream and a total of 5,000 individuals were artificially spawned up. The population of juvenile inherited the genetic diversity from the brood stock. The bred juveniles were discharged at the selected optimal site that had a habit condition what was similar to their natural habitat. The micro-dispersion around the discharging area was found at the early stages of the augmentation. The re-capturing rate of discharged juveniles was reduced as time passed. The discharged juveniles seemed to adapt to the natural environment of Naeseongcheon Stream. The observation of their high abdominal distension and excrement demonstrated that the juveniles fed successfully in the discharging area. Therefore, securement of genetic homogeneity and enhancement of restoration population of G. naktongensis in upstream and downstream of Youngju Dam site from the artificial seed producing seemed to be primarily successful and long-term monitoring and analysis of the effect was expected to be necessary.

A Fairy Pitta is a bird known to breed only in mainland China, Taiwan, Japan and Korea and is listed as Vulnerable in the IUCN Red List. We carried out a DNA analysis to contribute to conserve the genetic diversity of Fairy Pitta. 32 samples were collected at Jeju Island, the Korean Peninsula and Taiwan from 2004 to 2013 and DNA was extracted from them and several sequences were amplified-it through PCR. And then we performed the population genetic analysis. We found there was a transversion between nucleotide sequences at CO1 gene, while there was no changes at Cyt-b gene. And we confirmed the polymorphism from two genes was caused from genetic drift not from selection. Through this analysis, the group within the Peninsula was found bigger than other two groups based on the analysis of CO1 gene, and the group from Taiwan was found bigger than other two groups through the analysis of Cyt-b gene. The population genetic structure of mitochondria gene of three group was showing CO1 gene had 5 haplotypes and Cyt-b gene had 6 haplotypes. Haplotype 2 in CO1 gene was found in three group and many individuals of samples had this haplotype. Like CO1 gene, haplotype 2 in Cyt-b gene was found in three group and was included in plenty of individuals. Other haplotypes were not overlaped and broke off among the three groups. To prevent from the extinction of Fairy Pitta and to obtain the genetic diversity, we need to compare with other regional group such as Japan, China and perform additional research in the non-breeding area.
Reproductive phenology and physiological responses of Epilobium hirsutum L. to moisture content and nutrient content of soil were analysed in order to obtain basic data for effective conservation and restoration. Epilobium hirsutum L. is a perennial plant. But Epilobium hirsutum L. grew reproductively in all moisture and nutrient gradients. Flower bud, flowers and peduncle were respectively ripened in earlier under highest moisture condition and highest nutrient condition. And, number of flowers and peduncle were more quickly increased under highest moisture condition and highest nutrient condition. Chlorophyll content was high under highest moisture condition and higher middle moisture condition. However, we found no significant difference of chlorophyll content regard to nutrient gradients. There was no difference in minimum chlorophyll fluorescence among all moisture and nutrient gradients. The photochemical efficiency values of PS II were 0.75 in all moisture gradients, and it was 0.78 in highest nutrient gradient. The chlorophyll content of Epilobium hirsutum L. increased as the moisture content increased, and the Fv/Fm value increased as the organic matter increased. Our results showed that high moisture and nutrient content of soil advance their breeding season and promote reproductive growth. It might be important basic informations for the maintenance of population and the management of habitat of Epilobium hirsutum L. an endangered plant species.
