단일모드 광섬유와 결합기를 이용하여 간결한 구조의 광섬유 공초점 간섭 현미경을 구성하였으며, 위상 변위법을 응용한 표면 검색 방법을 제안하여 통신용의 반도체 레이저와 같이 비교적 긴 파장의 광원과, 낮은 NA의 대물렌즈를 사용하더라도 정밀한 시료 표면 검색이 가능함을 보였다. 이때 시료 표면의 높낮이는 시료로부터 반사된 빛의 위상으로부터 결정되며, 종래의 공초점 현미경에 비하여 주사시간을 크게 단축할 수 있었다. 끝으로 종래의 방법에 비해 제안된 방법은 시료의 반사율 변화에 덜 민감함을 확인할 수 있었다.
Mixing between two or more reagents is one of important processes in biochemical microfluidics. In efficient micromixer design, it is essential to analyze flow pattern and evaluate mixing efficiency with good precision. In this work, mixing efficiency for Y-channel micromixer is measured by fluorescence intensity using LIF(Laser Induced Fluorescence) Confocal Microscope. The Y-channel micromixers are fabricated with polydimethylsiloxane(PDMS) and those are bonded to glass plate through Plasma bonding. Nile Blue A is injected into the micromixer as a fluorescence dye for measuring of fluorescence intensity by He/Ne laser. For visualization of the flow pattern, dynamic image capturing is carried out using CAM scope. For the comparison with computer simulation, modified SIMPLE algorithm for incompressible flow equation is solved for the same geometry as in the experiment. Throughout the experiments and computer simulation, accurate mixing efficiency evaluation process for a PDMS Y-channel micromixer is established.
본 연구에서는 레버 증폭 구조를 이용한 플렉서 기반의 나노 스테이지를 설계, 제작하였다. 제작된 나노 스테이지를 샘플 스테이지로 채용한 공초점현미경을 개발하였다. 2차원 이미징의 구현을 위해서, 기존의 공초점현미경은 레이저를 이미징 평면상에 스캐닝하는 방식으로 구현된다. 본 연구에서는 백 나노미터의 구동정밀도를 가지는 나노 스테이지 위에 놓인 샘플을 2차원으로 스캐닝하면서 2차원 이미지를 구현할 수 있는 공초점현미경을 개발하였다. 플렉서 기반의 나노 스테이지는 이중 판스프링, 변위증폭 레버, PZT 엑추에이터 그리고 변위 센서로 구성 되어 있다. 스테이지의 구동 성능 해석을 위해 상용 유한요소 해석 프로그램을 이용하였다. 현미경에 사용되는 광원은 적색광 레이저이며, 레이저는 여러 광학요소를 거쳐 샘플스테이지의 샘플에 입사되고, 반사된 빛은 광센서인 PMT(Photo Multiplying Tube)로 계측되게 된다. 계측된 빛의 크기를 이용하여서 2차원 이미징을 구현하였다. 개발된 공초점 현미경으로 생쥐 귀의 피부조직을 관찰하여 현미경의 이미징 성능을 검증하였다. 설계된 샘플 스테이지는 기존의 공초점 현미경의 기계적인 Beam 스캐너를 대신함으로써 현미경의 광경로 및 전체시스템을 간소화 하였다.
최근 여러 가지 비침습적인 방법으로 피부조직의 생검 또는 조직학적 처리과정 없이 피부의 실시간 이미지를 이용한 연구가 가능해졌다. Confocal scanning laser microscope (CSLM) 이미지는 830 nm의 근적외선 파장을 사용하는 비침습적 방법으로 피부의 내부 구조를 이미지화할 수 있으며, CSLM을 이용한 연구를 통해 비침습적인 피부 측정법으로 색소 침착 부위의 기저층과 진피 유두 사이에서 melanin cap, papillary ring과 같은 지표가 뚜렷하게 관찰된다고 보고되었다. 본 연구에서는 scanning probe image processor (SPIP)소프트웨어를 사용하여 CSLM 디지털 이미지 정량 분석을 처음 시도하였으며 본 연구결과는 정성적 피부조직 이미지 데이터를 정량화함으로써 향후 CSLM 활용범위를 높이게 될 것으로 기대하고 있다.

The number of the living endothelial cells and the shape of those are very import clinical parameters for the evaluation of the quality of cornea. In this paper, we developed the automated endothelial cell counting and shape analysis algorithm for a confocal microscope. Since, the endothelial images from the confocal microscope has a non-uniform illumination and low contrast between cell boundaries and cell bodies, it is very difficult to segment the cells from the endothelial images. To cope with these difficulties, we proposed the new two stage image processing algorithm. At first stage algorithm, we used a high-pass filter and histogram equalization to compensate the non-uniform brightness pattern and a morphological filter and a watershed method are applied to detect the boundary of cells. From this stage, we could count the number of cells in an endothelial image. At second stage algorithm, we used a Voronoi diagram method to classify the shape of cells. This cell shape analysis and the percent of hexagonal cells are very sensitive in detecting the early endothelium damage. To evaluate the performance of the proposed system, we p개cessed seven endothelial images obtained using a confocal microscope. The proposed system correctly counted 95.5% cells and classified 92.0% of hexagonal cell shapes. This result is better than any others in this research area.
A Confocal Laser Scanning Microscope (CLSM) is applied to determine three-dimensional fiber orientation states in injection-molded short fiber composites. Since the CLSM optically sections the composites, more than two planes either on or below the surface of composites can be obtained. Therefore, three dimensional fiber orientation states are determined without destruction. To predict the orientation states, velocity and temperature fields are calculated by using a hybrid FEM/FDM method. The change of orientation state during packing stage is also considered by employing a compressible Hele-Shaw model. The predicted orientation states show good agreement with measured ones. However, some differences are found at the end of cavity. They may result from other effects, which are not considered in the numerical analysis.
The purpose of this study was to evaluate the effect of multiple application of all-in-one dentin adhesive system on microtensile bond strength using confocal laser scanning microscope and microtensile bond strength test. Flat occlusal dentin surfaces were prepared using low-speed diamond saw. In group I, Scotchbond Multipurpose (SM) was applied by manufacturer's recommendation. In group II, after Adper Prompt L-Pop was applied for 15s and light cured for 10s. the second coat was re-applied and light-cured. In group III, after light-curing the second layer. the third coat was re-applied and light-cured. Specimens bonded with a resin-composite were sectioned into resin-dentin stick for measuring the adhesive layer thickness by confocal laser scanning microscope and evaluating micro-tensile bond strength. The adhesive layers of three-step dentin adhesive system. 3 coats of Adper Prompt L-Pop had significantly thicker than SM. 2 coats of Adper Prompt L-Pop (p < 0.05). However. there was no significant differences in bond strengths between SM and 3 coats of Adper Prompt L-Pop (p > 0.05). And SM. 3 coats of Adper Prompt L-Pop had significantly higher than 2 coats of Adper Prompt L-Pop in bond strengths (p < 0.05).
