Morphological and Molecular Characterization of Heterodera schachtii and the Newly Recorded Cyst Nematode, H. trifolii Associated with Chinese Cabbage in Korea

  • The sugar beet cyst nematode, Heterodera schachtii is a well known pathogen on Chinese cabbage in the highland fields of Korea. However, a race of cyst forming nematode with close morphological resemblance to H. trifolii was recently isolated from the same Chinese cabbage fields. Morphological species differentiation between the two cyst nematodes is challenging, with only minor differences between them. Thus, this study described the newly intercepted H. trifolii population, and reviewed morphological and molecular characteristics conceivably essential in differentiating the two nematode species. A comparison of morphometric measurements of both infective juveniles and vulval cones of cysts showed significant differences between the two cyst nematodes. When total RNA and genomic DNA were extracted from a mixed field population, COI genes and ITS regions were clearly amplified with primers of the two Heterodera species, suggesting that Heterodera population collected from the Chinese cabbage field consisted of a mixture of two species. COI and ITS of H. trifolii were predominantly amplified from nucleotides prepared from H. trifolii monoxenic population whereas those of H. schachtii were strongly detected in H. schachtii monoxenic cultures. Thus, this study confirms the coexistence of the two species in some Chinese cabbage fields; and the presence of H. trifolii in Korea is reported here for the first time.

  • Modern volcanoes, Laoheishan and Huoshaoshan, have erupted during $1720\~1721$ in the Wudalianchi volcanic group, NE china. They comprise scoria and spatter cones that consist of potassium-rich phono-tephritic pyroclastic deposits and lavas, and include wide lava flow fields. The Laoheishan scoria cone is a polygenetic multiple volcano that overlaps earlier and later edifices with more complicated internal structures produced in greater scale and in earlier time than the Huoshaoshan. There is a funnel-shaped crater in the center of the later edifice of the Laoheishan scoria cone. The Huoshaoshan spatter cone is a monogenetic simple volcano with a central pit crater. The volcanic sequences indicate eruption processes that followed a repeated pattern that progressed through 5 stages of explosive and effusive eruption including lava fountains and Strombolian eruptions in the Laoheishan, and a recognizable pattern of 2 stages that started with Strombolian eruption and progressed through lava effusion in the Huoshaoshan.

One-cyclic Volcanic Processes at Udo Crater, Korea (우도(牛島) 분화구(噴火口)에서의 일윤회(一輪廻) 화산과정(火山過程))

  • Udo Island, some 3 km off the coast of Sungsan Peninsula at the eastern promontory of Cheju Island, occurs in such a regular pattern on the sequences which reprent an excellent example of an eruptive cycle. The island comprises a horseshoe-shaped tuff cone, a nested cinder cone on the crater floor, and a lava delta which extends over northwest from the moat between two cones. The volcanic sequences suggest volcanic processes that start with emergent Surtseyan eruption, progress through Strombolian eruption and end with lava effusion followed by reworking of smooth tephra on the tuff cone. Eruptive environment and hydrology of vent area in the Udo tuff cone are poorly constrained because the stratigraphic units under the tuff cone are unknown. It is thoughl, however, that the tuff cone could be mainly emergent because the present cone deposits show no evidence of marine reworking, and standing body of sea water could play a great role. The emergent volcano is characterized by distinctive steam-explosivity that results primarily from a bulk interaction between rapidly ascending magma and a highly mobile slurry. The sea water gets into the vent by flooding accross or through the top or breach of tephra cone. Udo tuff cone was constructed from Surtseyan eruption which went into with tephra finger jetting activities in the early stage, late interspersed with continuous uprush activities and proceeded to only continuous uprush activities in the last. When the enclosure of the vent by a long-lived tephra barrier would prevent the flooding and thus allow the vent to dry out, the Surtseyan eruption ceased to transmit into Strombolian activities, which constructed a cinder cone on the crater floor of the tuff cone. The Strombolian eruption ceased when magma in the conduit gradually became depleted in gas. In the case of Udo, the last magmatic activity was Hawaiian-type (and/or fountain) which accumulated basalt lava delta. And then the loose tephra of the tuff cone reworked over the moat lava and the northeastern flank.

Biomass and Net Primary Productivity of Pinus densiflora Natural Ecosystem in Kangwondo, Korea (강원도산(江原道産) 소나무천연림생태계(天然林生態系)의 Biomass 및 Net Primary Production에 관(關)한 연구(硏究))

  • Dimension analysis was used to estimate biomass and net primary production(NPP) in a 36-year-old Japanese red pine (Pines densiflora S. et Z.) natural forest in Kangwondo. Best estimation was made by the equation model of $Wt=aD^bH^c$ where Wt is weight in kg, D is DBH in cm, and H is total tree height in m. Total aboveground biomass was estimated at 198.82 t/ha. Organic matter was distributed in the Japanese red pine stand as follows ; bolewood 68.8%, live branches 16.5%, bolebark 5.4%, foliage 4.6%, dead branches 3.7%, and cones 0.6%. Net primary production was estimated at 15.87 t/㏊/yr and was distributed: bolewood 44.5%, live branches 30.9%, foliage 14.1%, current twig 7.1%, and bolebark 3.3%. Leaf efficiency was estimated at 1.876. The power equation for biomass and IVPP yielded similar results as the results for Pines densiflora in Japan.

N-Acetyl-D-Glucosamine Kinase Promotes the Axonal Growth of Developing Neurons

  • N-acetyl-D-glucosamine kinase (NAGK) plays an enzyme activity-independent, non-canonical role in the dendritogenesis of hippocampal neurons in culture. In this study, we investigated its role in axonal development. We found NAGK was distributed throughout neurons until developmental stage 3 (axonal outgrowth), and that its axonal expression remarkably decreased during stage 4 (dendritic outgrowth) and became negligible in stage 5 (mature). Immunocytochemistry (ICC) showed colocalization of NAGK with tubulin in hippocampal neurons and with Golgi in somata, dendrites, and nascent axons. A proximity ligation assay (PLA) for NAGK and Golgi marker protein followed by ICC for tubulin or dynein light chain roadblock type 1 (DYNLRB1) in stage 3 neurons showed NAGK-Golgi complex colocalized with DYNLRB1 at the tips of microtubule (MT) fibers in axonal growth cones and in somatodendritic areas. PLAs for NAGK-dynein combined with tubulin or Golgi ICC showed similar signal patterns, indicating a three way interaction between NAGK, dynein, and Golgi in growing axons. In addition, overexpression of the NAGK gene and of kinase mutant NAGK genes increased axonal lengths, and knockdown of NAGK by small hairpin (sh) RNA reduced axonal lengths; suggesting a structural role for NAGK in axonal growth. Finally, transfection of 'DYNLRB1 (74-96)', a small peptide derived from DYNLRB1's C-terminal, which binds with NAGK, resulted in neurons with shorter axons in culture. The authors suggest a NAGK-dynein-Golgi tripartite interaction in growing axons is instrumental during early axonal development.

A Literature Study on the Korean Acupuncture for Headache and Vertigo (두통과 현훈 치료관련 침구문헌 비교연구)

  • Background : The varieties of manifestations referred to headache and vertigo have been long complicated physicians in the efficient diagnosis and treatment of these diseases. Objectives : Establish a distinctive and efficient acupuncture method for the treatment of headache and dizziness based in literature research. Method : We reviewed four Korean medical literature, "治腫指南Guide to Swollen Sore Treatment", "東醫寶鑑Treasured Mirror of Eastern Medicine", "鍼灸經驗方Experiential Prescriptions of Acupuncture and Moxibustion", and "舍岩鍼法Essential Rhymes on Acupuncture and Moxibustion by Master Sa-am", and analyzed the therapeutic characteristics in the treatment of headache and vertigo. Result : 1. "治腫指南Guide to Swollen Sore Treatment" inserted needle in GV20百會 and waited until the skin erupts and heat burns, then applied in those regions affected. 2. "東醫寶鑑Treasured Mirror of Eastern Medicine" mostly applied moxibustion on headache, however, needling acupoints such as GV20百會, GV24神庭, GV23上星, GV22顖會 were also considered important. 3. "鍼灸經驗方Experiential Prescriptions of Acupuncture and Moxibustion" used moxa cones on headache in order to disperse heat and direct Gi downward. 4. "校勘 舍岩道人鍼法Essential Rhymes on Acupuncture and Moxibustion by Master Sa-am" defined the cause of dizziness, wood excavates earth and earth kills water. Conclusions : After previous study on eye, oral, glottal, labial, dental diseases and stroke, we could also find various efficient methods according to headache and vertigo, through literature research of Korean medical classics. This study will concurrently result in establishing distinctive therapeutic method characteristic of Korea.

A Study on the Afforestation of Fire-Resistant Forest through an Application of Forest Fire Investigation (산림 화재감식 응용을 통한 내화림 조성에 관한 연구)

  • In this study, species resistant to forest fire were extracted from high trees through an investigation of combustion mechanism. Here, the average flameless ignition temperatures of living leaves, branches and barks of oak were respectively, $365^{\circ}C$, $440^{\circ}C$ and $435^{\circ}C$, and the average flameless ignition temperatures of living leaves, branches, barks and cones of pine tree were respectively, $320^{\circ}C$, $405^{\circ}C$, $435^{\circ}C$ and $363^{\circ}C$. It shows that generally, pine tree has a lower flameless ignition temperature than oak and thus, has a relatively high risk of flameless ignition. When comparing risk of fire depending on ignition characteristics, Quercus serrata had a low risk of stem fire, and Quercus acutissima and Quercus serrata had a low risk of crown fire, as well. When analysing risk of fire depending on a duration of flame, also, Quercus dentata had a low risk of stem fire, and Quercus variabilis Blume had a low risk of crown fire too. Lastly, when comparing risk of fire, according to heat release rates, Quercus acutissima had a low risk of stem fire, and Quercus acutissima and Quercus dentata had a low risk of crown fire, as well. In conclusion, it was observed that as fire-resistant species, Quercus serrata and Quercus dentata have a low risk of ignition, Quercus variabilis Blume has a short duration of flame after discharging ignition when a forest fire occurs, and Quercus acutissima, Pinus Rigida and Antipathes japonica Brook have low heat release rates.

Endodontic treatment of mandibular molar with root dilaceration using Reciproc single-file system

  • Biomechanical preparation of root canals with accentuated curvature is challenging. New rotatory systems, such as Reciproc, require a shorter period of time to prepare curved canals, and became a viable alternative for endodontic treatment of teeth with root dilaceration. Thus, this study aimed to report a clinical case of endodontic therapy of root with accentuated dilaceration using Reciproc single-file system. Mandibular right second molar was diagnosed as asymptomatic irreversible pulpitis. Pulp chamber access was performed, and glide path was created with #10 K-file (Dentsply Maillefer) and PathFile #13, #16 and #19 (Dentsply Maillefer) up to the temporary working length. The working length measured corresponded to 20 mm in the mesio-buccal and mesio-lingual canals, and 22 mm in the distal canal. The R25 file (VDW GmbH) was used in all the canals for instrumentation and final preparation, followed by filling with Reciproc gutta-percha cones (VDW GmbH) and AH Plus sealer (Dentsply Maillefer), using thermal compaction technique. The case has been receiving follow-up for 6 mon and no painful symptomatology or periapical lesions have been found. Despite the difficulties, the treatment could be performed in a shorter period of time than the conventional methods.

Runoff Characteristics of a Small Catchment in Eoseungsaeng-oreum, Jeju Island (제주도 스코리아콘의 유출 특성 - 어승생오름 소유역을 사례로 -)

  • In order to examine the runoff characteristics of scoria cones in Jeju Island, hydrological observations were conducted in the experimental basin (5.1 ha) of Eoseungsaeng-oreum which has been predominantly covered with Carpinus laxiflora and Quercus serrata. Although runoff has continuously occurred during the observed period, the baseflow gradually increased from April and decreased from October. The peak flow approximately corresponded to every rainfall events except for the rainfall events which has slight total precipitation and no previous precipitation. The experimental basin shows flash runoff response and short lag time; the mean lag time is 35.8 minutes. Although the runoff ratio of quick flow is proportional to total precipitation, the increasing rate is low and the maximum runoff ratio is 24.7%. In addition, the runoff ratio is less than 1% in 68.3% of the rainfall events, suggesting that the portion of quick flow to total precipitation is low. The rainfall events with relatively long event time demonstrated a secondary peak generated by translatory flow. The runoff characteristics seem to be related to local impermeable beds in the experimental basin.

The effect of ankle Kinesio taping on range of motion and agility during exercise in university students

  • Objective: The purpose of this study was to examine the effects of ankle Kinesio taping on range of motion and agility during exercise in university students. Design: Cross-sectional study. Methods: Thirty subjects were randomly allocated to two groups: taping group (n=15) and non-taping group (n=15). All groups underwent the same exercise program including stretching for 30 minutes. The exercise program proceeded in the following order: five minutes of stretching, a 20-minutes exercise program, and additional five minutes of stretching. Of the eight exercise methods suggested by Purcell et al, seven were chosen (lateral shuffle, forward and backward running, agility ladder, figure-of-8, forward jogging while jumping over cones, wall jumps and zigzags); $90^{\circ}$ cuts with lateral shuffle were omitted. The range of motion of ankle dorsiflexion and plantarflexion was measured using the goniometer. Agility was measured using the side hop test. Results: For ankle range of motion, the taping group showed significant differences in dorsiflexion and plantarflexion on both sides (p<0.05). The non-taping group showed significant differences only in left plantarflexion (p<0.05). There was a significant difference in dorsiflexion on both sides between the taping group and the non-taping group (p<0.05). All groups showed significant differences in agility on the left and right ankle (p<0.05). There was a significant difference in left ankles between the taping group and the non-taping group (p<0.05). Conclusions: Kinesio taping increased range of motion and agility during exercise in university students. Additional research on Kinesio taping for improving range of motion and agility is needed.
