결명자의 아질산염 소거작용 (The Nitro-scavenging Effects by the Component of Cassiae Torae Semen)

  • 전통기호음료성분에 의하여 나타나는 기능특성을 조사하기 위한 연구의 일환으로 결명자의 메탄올 추출물을 유기용매에 의한 분획, silica gel column chromatography, thin layer chromatography에 의하여 여러가지 획분으로 분획하여 아질산염 소거작용을 조사한 결과, 아질산염 소거율은 ethyl acetate 획분>chloroform 획분>수용성 획분>ethyl ether 획분의 순으로 나타났으며, ethyl acetate 획분에서 분리한 compound D의 아질산염 소거효과는 매우 강력하여 ascorbic acid보다 10배 뛰어났다.

  • Oriental pear was used as treatment of asthma, control of blood pressure tonic medicine vasoaction, diabetes in oriental medicine. In this study, it was investigated that pear phenolic compound effects on cardiovascular system as blood pressure and renin and atrial natriuretic peptide(ANP) in plasma, Cardiac hypertrophy. The experiments were performed on Sprague Dawley rats, 2K1C hypertension model was prepared by constricting the left renal artery with a sliver clip. Animals were then divided into four groups, 5mg/Kg(PPC-A) 10mg/Kg(PPC-B) 15mg/Kg(PPC-c) and control group. Pear phenolic compound solution were supplied with them for 3weeks, a day's interval. The results are that The blood pressure was significantly decreased at 15days in PPC-A group than control group. The plasma ANP was increased in PPC-A and PPC-C group, and cardiac hypertrophy was significantly decrease in PPC-C group compared with control group.

Effect of various chromatographic terpenoid fractions of Luffa cylindrica seeds on in-vitro antimicrobial studies

  • The objective of the present investigation is to evaluate the antimicrobial potency of the terpenoid fractions isolated from Luffa cylindrica seeds against various pathogenic microbes. The seeds were powdered and extracted with methanol in soxhlet appratus based on phytochemical screening. Three terpenoid components were isolated by column chromatography and identified by thin layer chromatography and chemical analysis which were designated as ${LCSF_4}^*$, ${LCSF_6}^*$ & ${LCSF_8}^*$ respectively. Disc diffusion method was employed to determine the antimicrobial effectiveness of test compounds I, II and III $({LCSF_4}^*,\;{LCSF_6}^*\;&\;{LCSF_8}^*)$ against 6 microbial species viz., Staphylococcus (S.) aureus, Bacillus (B.) subtilis, Escherichia (E.) coli, Pseudomonas (P.) aeruginosa, Candida (C.) albicans and Aspergillus niger. The disc was saturated with $100{\mu}l$ of each compound, allowed to dry and introduced on the upper layer of seeded agar plate. The plates were incubated overnight at $37^{\circ}C$. Microbial growth was determined by measuring the zonal inhibition diameters. Compound I showed maximum potency against gram positive S. aureus (21 mm) in comparison with standard ciprofloxacin (38 mm), whereas the same compound was completely devoid of activity against both the fungi tested. Compound II was found to be highly sensitive against both the gram negative E. coli (20 mm) and P. aeruginosa (22 mm). Compound II was found to exhibit maximum potency against the fungi C. albicans (15 mm) and A. niger (20 mm). Compound III was found to be very effective against both the gram positive S. aureus (20 mm) and B. subtilis (15 mm) respectively.

약물-표적 단백질 연관관계 예측모델을 위한 쌍 기반 뉴럴네트워크 (Pairwise Neural Networks for Predicting Compound-Protein Interaction)

  • In-silico 기반의 약물-표적 단백질 연관관계 예측은 신약 탐색 단계에서 매우 중요하다. 그러나 기존의 예측모델은 입력 값이 고정적이며 표적 단백질의 특질 값이 가공된 데이터로 한정됨으로써 예측 모델의 확장성과 유연성이 부족하다. 본 논문에서는 약물-표적 단백질 연관관계를 예측하는 확장 가능한 형태의 머신러닝 모델을 소개한다. 확장 가능한 머신러닝 모델의 핵심 아이디어는 쌍기반의 뉴럴 네트워크로써, 약물과 단백질의 미가공 데이터를 사용하여 특질을 추출하고 특질 값을 각각의 뉴럴 네트워크 레이어에 입력한다. 이 방법은 추가적인 지식없이 자동적으로 약물과 단백질의 특질을 추출한다. 또한 쌍기반 레이어는 특질 값을 풍부한 저차원의 벡터로 향상 시킴으로써 입력 값의 차이로 인한 편향 학습을 방지한다. PubChem BioAssay(PCBA) 데이터 셋에 기반한 5-폴드 교차 검증법을 통하여 제안한 모델의 성능을 평가했으며, 이전의 모델보다 우월한 성능을 보였다.

Effective Thematic Words Extraction from a Book using Compound Noun Phrase Synthesis Method

  • Most of online bookstores are providing a user with the bibliographic book information rather than the concrete information such as thematic words and atmosphere. Especially, thematic words help a user to understand books and cast a wide net. In this paper, we propose an efficient extraction method of thematic words from book text by applying the compound noun and noun phrase synthetic method. The compound nouns represent the characteristics of a book in more detail than single nouns. The proposed method extracts the thematic word from book text by recognizing two types of noun phrases, such as a single noun and a compound noun combined with single nouns. The recognized single nouns, compound nouns, and noun phrases are calculated through TF-IDF weights and extracted as main words. In addition, this paper suggests a method to calculate the frequency of subject, object, and other roles separately, not just the sum of the frequencies of all nouns in the TF-IDF calculation method. Experiments is carried out in the field of economic management, and thematic word extraction verification is conducted through survey and book search. Thus, 9 out of the 10 experimental results used in this study indicate that the thematic word extracted by the proposed method is more effective in understanding the content. Also, it is confirmed that the thematic word extracted by the proposed method has a better book search result.

복합 치아종의 보존적 외과적 적출: 증례보고 (CONSERVATIVE SURGICAL REMOVAL OF COMPOUND ODONTOMA: CASE REPORTS)

  • 치아종은 외배엽성 상피세포와 중배엽성 세포의 과성장으로 형성되는 혼합종양이다. 대부분 정상 치아 발생 시기 동안 형성되며 어느 정도 크기까지만 자라므로 진성 종양이 아닌 과오종으로 여겨진다. 형태에 따라 복합 치아종과 복잡 치아종으로 구별된다. 복합 치아종은 주로 구강 전방부에서 발견되며 작은 치아와 비슷한 방사선 불투과성 구조물을 함유하고 있는 병소이다. 치아종의 치료는 대부분 보존적인 외과적 적출술이 추천되며 재발은 거의 없다. 본 증례는 유치 및 영구치의 맹출 장애를 일으킨 복합 치아종으로 진단된 세 어린이에서 병소의 외과적 적출술을 시행하고 약 1년간 관찰한 결과 자발적 맹출을 보여 이에 보고하는 바이다.

양기능성 커플링제 실란에 의한 실리카-천연고무 복합소재의 계면간 결합 고찰 (Observation of Interfacial Adhesion in Silica-NR Compound by Using Bifunctional Silane Coupling Agent)

  • 양기능성 실란 커플링제인 bis(triethoxysilylpropyl)tetrasulfide(TESPT)가 실리카/천연고무 복합소재 내에서 실리카의 hydroxy기와 고무 계면간에 화학적 결합반응을 하여 실리카-실란-고무간의 3차원 사슬구조를 형성한 것을 열분해 가스 크로마토그래피 질량분석기(Py-GC/MS)와 주사전자현미경(SEM)을 통해 관찰하였다. TESPT의 첨가로 배합물 내의 황 함유율이 증가하여 스코치 시간($t_{10}$)은 감소하고, TESPT 내 sulfur donor 역할을 하는 황이 활성화제 및 촉진제와의 복합반응으로 가교반응이 지속적으로 이루어짐에 따라서 가교속도지수(CRI)의 값이 감소하였다. 또한 TESPT가 첨가된 컴파운드는 첨가되지 않은 컴파운드에 비해 가공성 및 기계적 물성이 향상되었다. 결과적으로, 실리카로 충전된 천연고무 복합소재에 실란 커플링제(TESPT)가 첨가되어 화학반응으로 실리카-실란-고무간의 3차원 사슬구조가 형성됨을 관찰하였고 이에 따라 가교밀도가 증가하여 복합소재의 물성 증가에 기여함을 보여주었다.

Compound IKD-8344, a Selective Growth Inhibitor Against the Mycelial Form of Candida albicans, Isolated from Streptomyces sp. A6792

  • In the course of screening for selective growth inhibitors against the mycelial form of Candida albicans, we isolated a Streptomyces sp. A6792 from soils. The inhibitor was isolated from the above bacterium and identified through several spectral analyses with UV and mass spectrophotometries, and various NMR. The compound was determined to be a macrocyclic dilactone antibiotic, IKD-8344 (molecular weight: 844, molecular formula: $C_{48}H_{76}O_{12}$). The compound selectively inhibited the growth of mycelial form of C. albicans with an MIC of 6.25 ${\mu}g/ml$. It also exhibited strong inhibitory effect preferentially on the mycelial form of various Candida spp. including C. krusei, C. tropicalis, and C. lusitaniae, with MICs ranging from 1.56 to 25 ${\mu}g$/ml. Furthermore, the compound showed no significant toxicity against SPF ICR mice up to 60 mg/kg. These results suggest that IKD-8344 is a useful lead compound for the development of novel antifungal agents, based on the preferential growth inhibition against Candida spp.

  • Pseudomonas sp. DJ77 의 chromosomal DNA로 부터 6-8kb XhoI 절편 상에 존재하는 phenanthrene 분해에 관련된 유전자군을 vector pBLUESCRIPT SK(+)에 클로닝하였다. 이렇게 얻은 재조합 plasmid인 pHENX7을 가지고 있는 JM101 균주는 3-methylcatechol을 노란색의 meta-cleavage 화합물로 전환학 수 있었다. 그러자 삽입된 절편의 방향이 반대가 되도록 제조한 pHENX7R 은 extradiol dioxygenase 활성을 나타내지 않기 때문에 전사방향을 알 수 있었다. pHENX7과 이의 유도체들을 지니는 JM101 균주에서 PhnC(24kDa), PhnD(31 kDa), PhnE(34 kDA), PhnF(15 kDa) 의 4 polypeptide 를 확인학 수 있었고 개개의 유전자의 위치와 범위를 알 수 있었다. 유전자 순서는 phnC-phnD-phnE-phnF-phnG 이었으며, phnD, phnE, phnG 는 각기 gluthione S-transferase, meta-cleavage compound hydrolase, extradiol dioxygenase, metacleavage compound dehydrogenase 의 유전자이었다.

Insulin secretory activity and mechanism of compound K

  • Purpose: Panaxadiols are more potent than panaxatriols as far as insulin secretory activity is concerned. In this study, we examined insulin secretory activity and mechanism of compound K (CK), a major intestinal bacterial metabolite of ginsenosides. Method: Insulin secretory activity of CK was examined using pancreatic beta cells and in Oral Glucose Tolerance Test assay. In addition, insulin secretory mechanism was studied in terms of calcium dependent or independent pathways. Results: In vitro, CK enhanced the insulin secretion concentration-dependently when compared to glucose-stimulated control cells. Insulin secretory mechanism of CK seems to block ATP sensitive K channels, which was confirmed by diazoxide (K channel opener) but, insulin resistance ameliorating activity of CK can't be ruled out. In vivo, CK showed hypoglycemic effect in OGTT.
