$^{13}C$ NMR Analysis for the Characterization of Heme Electronic/Molecular Structure in Horse Myoglobin Cyanide (Myoglobin 시안 단백질에 포함된 Heme에 대한 전자 및 분자구조 규명을 위한 $^{13}C$ NMR분석)

  • The reverse detection heteronuclear multiple quantum coherence, HMQC study of metcyano complex of horse myoglobin(MbCN) has provided the complete assignment of hyperfine shifted resonances of heme carbons attached with proton(s). The application of HMQC experiment to the paramagnetic low-spin MbCN gives clear $^1H$ and $^{13}C$ coherences for the paramagnetic amino acid residues as well as heme side chains, and can be extended to the low-spin paramagnetic hemoprotein derivative for the assignment of natural abundance $^{13}C$ resonances. This assignment strategy can avoid possible ambiguities that may result from the sole utilization of $^1H$ nuclear Overhauser effect for the assignment of heme $^1H$ signals resonating in the diamagnetic region. The resulting 2,4-vinyl ${\alpha}$-carbons and 7-propionate ${\beta}$-carbon follow anomalous anti-Curie behavior, and are indicative of incoplanarity with heme plane. Magnetic/electronic asymmetry of heme induced by proximal histidine(His) makes spread that the hyperfine shifted heme carbon resonances over the range of 250 ppm at $25^{\circ}C$. These heme carbon resonances would be the much more sensitive probe than those of proton resonances in analyzing the nature of heme electronic structure of myoglobin.

Blood Vessel Strain Imaging Using Linear Array Transducer (선형 트랜스듀서를 이용한 혈관 변형률 영상법)

  • The intrasvascular ultrasound (IVUS) imaging technique is used to diagnose cerebrovascular diseases such as stroke. Recently, elasticity imaging methods have been investigated to diagnose blood clots attached to blood vessel intima. However, the IVUS imaging technique is an invasive method that requires a transducer to be inserted into blood vessel. In this paper, strain images are obtained of blood clots attached to blood vessel intima with data acquired from outside the blood vessel using a linear array transducer. In order to measure the displacement of blood vessel accurately, experimental data are acquired by steering ultrasound beams so that they can intersect the blood vessel wall at right angles. The acquired rf data are demodulated to the baseband. The resulting complex baseband signals are then processed by an autocorrelation algorithm to compute the blood vessel movement and thereby produce strain image. This proposed method is verified by experiments on a plastic blood vessel mimicking phantom. The efficacy of the proposed method was verified using a home-made blood vessel mimicking phantom. The blood vessel mimicking phantom was constructed by making a 6 mm diameter hollow cylinder inside it to simulate a blood vessel and adhering 2 mm thick soft plaque to the inner wall of the hollow cylinder. The RF data were acquired using a clinical ultrasound scanner (Accuvix XQ, Medison, Seoul. Korea) with a 7.5 MHz linear array transducer by steering ultrasound beams in steps of $1^{\circ}$ from $-40^{\circ}$ to $40^{\circ}$ for a total of 81 angles. Experimental results show that the plaque region near the blood vessel wall is softer than background tissue. Although the imaging region is restricted due to the limited range of angles for which scan lines are perpendicular to the wall, the feasibility of strain imaging is demonstrated.

  • As long span and complex bridges are constructed often recently, safety estimation became a big issue. Various types of measuring instruments are installed in case of long span bridge. New wireless technologies for long span bridges such as sending information through a gateway at the field or sending it through cables by signal processing the sensing data are applied these days. However, The case of occurred accidents related to bridge in the world have been reported that serious accidents occur due to lack of real-time proactive, intelligent action based on recognition accidents. To solve this problem in this study, the idea of "communication among things", which is the basic method of RFID/USN technology, is applied to the bridge monitoring system. A sensor node module for USN based intelligent bridge system in which sensor are utilized on the bridge and communicates interactively to prevent accidents when it captures the alert signals and urgent events, sends RF wireless signal to the nearest traffic signal to block the traffic and prevent massive accidents, is designed and tested by performing TinyOS based middleware design and sensor test free Space trans-receiving distance.

The Effects of ATP - sensitive $K^+$ Channel on the Muscle Fatigue in Mouse Skeletal Muscle Cell (골격근 세포에서 ATP-의존성 $K^+$통로의 활성화가 근피로에 미치는 영향)

  • Excitation-contraction coupling in skeletal muscle is process by which depolarization of the muscle fiber membrane, elicited by a nerve action potential, triggers the release of $Ca^{2+}$ from the sarcoplasmic reticulum(SR). The resulting rise in intracellular $Ca^{2+}$ concentration$([Ca^{2+}]_i)$ activates the troponin complex, thereby initiating the contraction of the muscle. The question remains as to what factors are involved in the inhibition of SR $Ca^{2+}$ release in fatigued muscle. The purpose of this study was determine whether ATP-sensitive $K^+(K_{ATP})$ channels are activated and contribute to decrease in $[Ca^{2+}]_i$ during fatigue development in the mouse skeletal muscle. To elucidate a role of $K_{ATP})$ in relation to ECC, I measured the modulation effects of $K_{ATP})$ channel blocker(glibenclamide) and opener(pinacidil) on $[Ca^{2+}]_i$ after fatiguing electrical field stimulation(FEFS). Intracellular $Ca^{2+}$ signals were recorded by conforcal laser microscopy(LSM 410) and monitored using the fluorescent $Ca^{2+}$-Sensitive indicator Fluo-3 AM. The results of this study were as followed: 1. The relative [Ca2'li after FEFS in the pre-glibenclamide-treated group was higher than the control. And relative $[Ca^{2+}]_i$ after FEFS in the pre-glibenclamide-treated group was lower than the control. 2. The relative $[Ca^{2+}]_i$ after FEFS for 3 min in the control, pre-glibenclamide-treated group and pre-pinacidil-treated group showed a similar pattern; the gradually significant decrease in $[Ca^{2+}]_i$. But, these decreasing pattern was most significant in the control. These findings suggest a tight relationship between $K_{ATP})$ and $Ca^{2+}$ in ECC during fatigue. Therefore, 1 thought that activation of $K_{ATP})$ channels may be one of mechanisms of the fatigue in skeletal muscle.

Incident Detection for Urban Arterial Road by Adopting Car Navigation Data (차량 궤적 데이터를 활용한 도심부 간선도로의 돌발상황 검지)

  • Traffic congestion cost is more likely to occur in the inner city than interregional road, and it accounts for about 63.39% of the whole. Therefore, it is important to mitigate traffic congestion of the inner city. Traffic congestion in the urban could be divided into Recurrent congestion and Non-recurrent congestion. Quick and accurate detection of Non-recurrent congestion is also important in order to relieve traffic congestion. The existing studies about incident detection have been variously conducted, however it was limited to Uninterrupted Traffic Flow Facilities such as freeway. Moreover study of incident detection on the interrupted Traffic Flow Facilities is still inadequate due to complex geometric structure such as traffic signals and intersections. Therefore, in this study, incident detection model was constructed using by Artificial Neural Network to aim at urban arterial road that is interrupted traffic flow facility. In the result of the reliability assessment, the detection rate were 46.15% and false alarm rate were 25.00%. These results have a meaning as a result of the initial study aimed at interrupted traffic flow. Furthermore, it demonstrates the possibility that Non-recurrent congestion can be detected by using car navigation data such as car navigator system device.

Analysis of the Necessary Mechanical Properties of Embroiderable Conductive Yarns for Measuring Pressure and Stretch Textile Sensor Electrodes (생체 신호 측정 압력 및 인장 직물 센서 전극용 자수가 가능한 전도사의 필요 물성 분석)

  • In this study, we investigated the necessary mechanical properties of conductive multifilament yarns for fabricating the electrodes of biosignal measurement pressure and stretch textile sensors using embroidery. When electrodes and circuits for smart wearable products are produced through the embroidery process using conductive multifilament yarns, unnecessary material loss is minimized, and complex electrode shapes or circuit designs can be produced without additional processes using a computer embroidering machine. However, because ordinary missionary threads cannot overcome the stress in the embroidery process and yarn cutting occurs, herein, we analyzed the S-S curve, thickness, and twist structure, which are three types of silver-coated multifilament yarns, and measured the stress in the thread of the embroidery simultaneously. Thus, the required mechanical properties of the yarns in the embroidery process were analyzed. In the actual sample production, cutting occurred in silver-coated multifilament rather than silver-coated polyamide/polyester, which showed the lowest S-S curve. In the embroidery process, the twist was unwound through repetitive vertical movement. Further, we fabricated a piezoresistive pressure/tension sensor to measure gauge factor, which is an index for measuring biological signals. We confirmed that the sensor can be applied to the fabrication of embroidery electrodes, which is an important process in the mass production of smart wearable products.

NLRP3 Inflammasome in Neuroinflammatory Disorders (NLRP3 인플라마좀 작용 기전 및 신경 질환에서의 역할)

  • Immune responses in the central nervous system (CNS) function as the host's defense system against pathogens and usually help with repair and regeneration. However, chronic and exaggerated neuroinflammation is detrimental and may create neuronal damage in many cases. The NOD-, LRR-, and pyrin domain―containing 3 (NLRP3) inflammasome, a kind of NOD-like receptor, is a cytosolic multiprotein complex that consists of sensors (NLRP3), adaptors (apoptosis-associated speck like protein containing a caspase recruitment domain, ASC) and effectors (caspase 1). It can detect a broad range of microbial pathogens along with foreign and host-derived danger signals, resulting in the assembly and activation of the NLRP3 inflammasome. Upon activation, NLRP3 inflammasome leads to caspase 1-dependent secretion of the pro-inflammatory cytokines IL-1β and IL-18, as well as to gasdermin D-mediated pyroptotic cell death. NLRP3 inflammasome is highly expressed in CNS-resident cell types, including microglia and astrocytes, and growing evidence suggests that NLRP3 inflammasome is a crucial player in the pathophysiology of several neuroinflammatory and psychiatric diseases, such as Alzheimer's disease, Parkinson's disease, multiple sclerosis, stroke, traumatic brain injury, amyotrophic lateral sclerosis, and major depressive disorder. Thus, this review describes the molecular mechanisms of NLRP3 inflammasome activation and its crucial roles in the pathogenesis of neurological disorders.

Development of Voltammetric Nanobio-incorporated Analytical Method for Protein Biomarker Specific to Early Diagnosis of Lung Cancer (폐암 조기 진단을 위한 단백질 바이오마커 측정용 전압-전류법 기반의 나노바이오 분석법 개발)

  • In this article, a portable and cost-effective voltammetric biosensor with nanoparticles was developed for the measurements of heterogeneous nuclear ribonucleoprotein A1 protein (hnRNP A1) biomarker which can potentially be used for lung cancer diagnosis. Gold nanoparticles were first electrodeposited onto screen printed carbon electrode (SPCE) followed by immobilizing a single stranded DNA aptamer specific to hnRNP A1 onto the electrode surface. Ethanolamine was also used when immobilizing DNA aptamer on the surface to prevent signals from non-specific adsorption events. Sequential injection of hnRNP A1 biomarker and anti-hnRNP A1 conjugated with alkaline phosphatase (ALP) onto the aptamer chip surface allows to form the sandwich complex of DNA aptamer/hnRNP A1/ALP-anti-hnRNP A1 on the electrode surface which further reacted with 4-aminophenyl phosphate (APP). The electrocatalytic reaction of the enzyme, ALP, and the substrate, APP, resulting in the oxidative current response changes at -0.05 and -0.17 V (vs. Ag/AgCl) against the hnRNP A1 concentration was measured using cyclic and differential pulse voltammetry, respectively. The Au nanoparticles-integrated voltammetric biosensor was applied to analyze human normal serum solutions possibly suggesting potential applicability for lung cancer diagnosis.

Development of Underwater Positioning System using Asynchronous Sensors Fusion for Underwater Construction Structures (비동기식 센서 융합을 이용한 수중 구조물 부착형 수중 위치 인식 시스템 개발)

  • An underwater positioning method that can be applied to structures for underwater construction is being developed at the Korea Institute of Ocean Science and Technology. The method uses an extended Kalman filter (EKF) based on an inertial navigation system for precise and continuous position estimation. The observation matrix was configured to be variable in order to apply asynchronous measured sensor data in the correction step of the EKF. A Doppler velocity logger (DVL) can acquire signals only when attached to the bottom of an underwater structure, and it is difficult to install and recover. Therefore, a complex sensor device for underwater structure attachment was developed without a DVL in consideration of an underwater construction environment, installation location, system operation convenience, etc.. Its performance was verified through a water tank test. The results are the measured underwater position using an ultra-short baseline, the estimated position using only a position vector, and the estimated position using position/velocity vectors. The results were compared and evaluated using the circular error probability (CEP). As a result, the CEP of the USBL alone was 0.02 m, the CEP of the position estimation with only the position vector corrected was 3.76 m, and the CEP of the position estimation with the position and velocity vectors corrected was 0.06 m. Through this research, it was confirmed that stable underwater positioning can be carried out using asynchronous sensors without a DVL.

Insect Juvenile Hormone Antagonists as Eco-friendly Insecticides (친환경 살충제로서의 곤충 유충호르몬 길항제)

  • Because of their specificity to target insects and relatively low toxicity to non-target organisms, insect growth regulators (IGRs) have been regarded as attractive alternatives to chemical insecticides. Commercially available IGRs are classified into juvenile hormone agonists (JHAs), ecdysone agonists (EAs), and chitin synthesis inhibitors (CSIs) according to their mode of action. Recently, JH-mediated interaction of methoprene-tolerant (Met), which is JH receptor, and its binding partners have been replicated in vitro using yeast cells transformed with the Met and FISC/CYC genes of A. aegypti. Using this in vitro yeast two-hybrid β-galactosidase assay, juvenile hormone antagonists (JHANs) have been identified from various sources including chemical libraries, plants, and microorganisms. As juvenile hormone (JH) is an insect specific hormone and regulates development, reproduction, diapause and other physiological processes, JHANs fatally disrupt the endocrine signals, which result in abnormal development and larval death. These results suggested that JHANs could be efficiently applied as IGR insecticides with a broad insecticidal spectrum. This review discuses JH signaling pathway mediated by Met and future prospects of JHANs as environmentally benign IGR insecticides.
