Coping with large litters: the management of neonatal piglets and sow reproduction

  • As a result of intensive breeding, litter size has considerably increased in pig production over the last three decades. This has resulted in an increase in farrowing complications. Prolonged farrowing will shorten the window for suckling colostrum and reduce the chances for high-quality colostrum intake. Studies also agree that increasing litter sizes concomitantly resulted in decreased piglet birth weight and increased within-litter birth weight variations. Birth weight, however, is one of the critical factors affecting the prognosis of colostrum intake, and piglet growth, welfare, and survival. Litters of uneven birth weight distribution will suffer and lead to increased piglet mortality before weaning. The proper management is key to handle the situation. Feeding strategies before farrowing, management routines during parturition (e.g., drying and moving piglets to the udder and cross-fostering) and feeding an energy source to piglets after birth may be beneficial management tools with large litters. Insulin-like growth factor 1 (IGF-1)-driven recovery from energy losses during lactation appears critical for supporting follicle development, the viability of oocytes and embryos, and, eventually, litter uniformity. This paper explores certain management routines for neonatal piglets that can lead to the optimization of their colostrum intake and thereby their survival in large litters. In addition, this paper reviews the evidence concerning nutritional factors, particularly lactation feeding that may reduce the loss of sow body reserves, affecting the growth of the next oocyte generation. In conclusion, decreasing birth weight and compromised immunity are subjects warranting investigation in the search for novel management tools. Furthermore, to increase litter uniformity, more focus should be placed on nutritional factors that affect IGF-1-driven follicle development before ovulation.

Factors associated with farrowing assistance in hyperprolific sows

  • Objective: The present study was performed to determine risk factors associated with the frequency of farrowing assistance in hyperprolific sows in a tropical environment and to investigate the impacts of farrowing assistance on piglet colostrum consumption and sow colostrum yield. Methods: Farrowing data from 352 Landrace×Yorkshire crossbred sows and 5,554 piglets in five commercial swine herds in Thailand were investigated. The sows were classified according to parity numbers: 1 (n = 72), 2 to 4 (n = 128), 5 to 6 (n = 84), and ≥7 (n = 68) and the total number of piglets born per litter (TB): 10 to 13 (n = 90), 14 to 16 (n = 117), and ≥17 (n = 145). The incidence of farrowing assistance and associated parameters were investigated. Results: The TB and farrowing duration averaged 15.8±0.2 and 279.9±11.2 min, respectively. The percentage of sows that required farrowing assistance was 29.8% and varied among herds from 5.7% to 53.3% (p<0.001). The percentage of piglets born after birth assistance using manual intervention was 8.4%. Sows with parity numbers 1 and 2 to 4 had a lower frequency of farrowing assistance than sows with parity numbers ≥7 (p<0.01). The colostrum yield of sows that required farrowing assistance did not differ from sows that farrowed without assistance (5.3±0.2 and 5.1±0.1 kg; p = 0.288); however, the colostrum consumption of piglets born from sows that required farrowing assistance was lower than those born from sows that farrowed without assistance (302.2±15.7 and 354.2±5.6 g; p<0.001). Blood oxygen saturation of the piglets born after birth assistance tended to be lower than the piglets that farrowed without birth assistance (87.8%±1.3% vs 90.4%±0.4%; p = 0.054). Conclusion: The frequency of farrowing assistance in sows varied among herds and was influenced by parity number. The piglets born after receiving birth assistance should receive special care to improve their blood oxygen saturation and enhance colostrum intake.

  • 본 실험은 홀스타인 젖소의 초유에 함유된 lactoferrin, IgA, $IgG_1,\;IgG_2$의 함량과 병원성미생물의 항균효과, 유산균증식효과를 알아보기 위하여 수행하였다. 홀스타인종 젖소 초유의 lactoferrin, IgA, $IgG_1,\;IgG_2$의 함량은 여름철의 경우 분만 후 첫째 날에 각각 0.30 mg/mL, 0.37 mg/mL, 4.00 mg/mL, 0.37 mg/mL, 겨울철의 경우 분만 후 첫째 날에 각각 1.16 mg/mL, 2.60 mg/mL, 13.35 mg/mL, 1.30 mg/mL로 측정되었다. 무열처리한 초유와 $65^{\circ}C$에서 30분간 열처리한 초유에서 배양한 병원성 미생물의 변화는 E. coli 균주에 대하여 초유의 항균활성이 나타났다. 초유 첫째 날, 둘째 날, 세째 날의 유산균수 성장촉진 효과는 B. longum, L. acidophilus S. themophilus의 상업용 혼합균주와 L. acidophilus, L. casei, L. bulgaricus, L. lactis subsp. cremoris에서 모두 정상유보다 균수가 증가하였다. 초유 첫째 날, 둘째 날, 세째 날의 pH는 배양 15시간 후 B. longum, L. acidophilus S. themophilus의 상업용 혼합균주와 L. acidophilus, L. casei, L. bulgaricus, L. lactis subsp. cremoris에서 각각 4.97-5.22 및 5.96-6.47로 나타났으나, 정상유에서는 각각 4.89, 6.50-6.81이었다. 초유 첫째 날, 둘째 날, 세째 날의 산도는 B. longum, L. acidophilus S. themophilus의 상업용 혼합균주와 L. acidophilus, L. casei, L. bulgaricus, L. lactis subsp. cremoris에서 각각 0.75-0.88% 및 0.29-0.48%로 나타났으나, 정상유에서는 각각 0.70%, 0.20-0.25% 이었다.

  • 초유 및 성숙유의 지방질 조성을 TLC-FID 방법으로 분석, 정량하였던 바 분리된 지방질 성분 중 triglyceride 수준이 초유에서보다 성숙유에서 유의하게 높았으며, 기타의 지방질 성분들은 초유 및 성숙유간에 유의한 차이가 나타나지 않았다.

Isolation and Characterization of an immunomodulatory Protein from Bovine Colostrum

  • A colostral protein that augments natural killer (NK) cell activity was isolated from bovine colostrum. This protein, designated matemal immunity enhancing factor (MIEF), increased NK cell-mediated cytotxicity against human tumor targets wheb added to cultures of resting peripheral blood lymphocytes. The NK cell stimulatory activity of the MIEF was demonstrated at the concentrations as low as 0.1-0.01 $\mu$g/ml. Purified MIEF showed an apparent molecular weight of 22,000 in SDS-polyacrylamide gel electrophoresis. The unusual biochemical characteristics of the MIEF distinguish it from other cytokines. The MIEF was soluble at a cold tgemperature, and precipitated by raising the temperature. This themal precipitability was reversible, and dependent on the concentration, pH and ionic strength. Maximal precipitation was observed at neutral pH, and higher ionic strength.

Effect of Andrographis paniculata supplementation during the transition period on colostrum yield, immunoglobulin G, and postpartum complications in multiparous sows during tropical summer

  • Objective: This study evaluated the effect of Andrographis paniculata (A. paniculata) supplementation in sow diets before and after farrowing on the sow and piglets' performances during early postpartum period and on sows' backfat and longissimus muscle losses during lactation. Methods: Seventy Landrace×Yorkshire sows and their offspring (1,186 piglets) were distributed into three groups: control (n = 31), treatment-250 (n = 18), and treatment-1000 (n = 21). From 110.2±0.7 days of gestation until farrowing (5.8 days) and throughout the lactation period (25.2 days), sows in the control group were given the conventional lactation diet, while sows in the treatment-250 and treatment-1000 groups received supplements of 250 ppm and 1,000 ppm of A. paniculata, respectively. Results: In sows with parity 3-5, piglets from the treatment-1000 group had higher colostrum intake than the control and treatment-250 groups (p<0.05), but not in sows with parity 6-9. Colostrum immunoglobulin G (IgG) increased in treated sows versus controls for parity 6-9 (p<0.05), but was consistent for parity 3-5. Piglet performance until day 3 postnatal was similar across groups (p>0.05). Treatment-250 sows had higher feed intake post-farrowing than treatment-1000 sows (p<0.05). Longissimus loss was less in both treatment groups than control (p<0.05), but backfat loss was similar across groups (p>0.05). Post-partum complications were consistent across groups (p>0.05). Farrowing duration and piglet birth intervals in sows with parity 6-9 were prolonged in the treatment-1000 group. Conclusion: Supplementing with 1,000 ppm A. paniculata for 5.8 days pre-farrowing and 25.2 days post-farrowing enhanced sow colostrum IgG and piglet colostrum intake, while also reducing longissimus loss in sows. However, for sows of parity 6-9, this supplementation led to prolonged farrowing, increased intervals between piglet births, increased stillbirth, and reduced piglet birth weight. These effects should be considered when using A. paniculata supplementation.

  • This paper outlines research to study two aspects of Cryptooridium. First, specific antigenic determinants were identified and followed through the growth cycle of C. parvum to investigate antigenic sharing of molecular epitopes among the different life cycle stages. Secondly, the importance of passive immune protective mechanisms in cryptosporidial infection was assessed by following the course of infection in neonatal mice which have been subjected to treatments using either monoclonal antibodies (mAbs) or hyperimmune bovine colostrum.

Immunomodulatory Effects of Human Colostrum and Milk

  • The immune system is not fully developed in human neonates and infants; breastfeeding is important in this stage as the bioactive components of human breast milk are known to have anti-microbial, anti-inflammatory, and immunomodulatory effects, and can therefore contribute to an infant's immunity against allergies, asthma, autoimmune diseases, and inflammatory bowel disease. Herein, the positive effect on the immune system by human colostrum and milk are reviewed.

Evaluation and Characterization of Milk-derived Microvescicle Isolated from Bovine Colostrum

  • Extracellular microvesicles are membranous nano-sized cellular organelles secreted by a variety of cells under normal and pathological conditions and heterogeneous in size ranging from 30 nm to $1{\mu}m$. They carry functional microRNAs that can influence immunity and development. For a particular application of microvesicles, choice of isolation method is particularly important; however, their isolation methods from colostrum in particular have not been described clearly. In this work, differential ultracentrifugation as a conventional method, ultracentrifugation with some modification such as additional precipitations, ultrafiltration, sucrose gradient separation and ExoQuick$^{TM}$ as a commercial reagent were compared. The goal was to compare mainly microvesicular total microRNA yield, distribution and purity among the methods then select the best isolation method for bovine colostrum microvesicles based largely on microRNA yield with the view of applying the vesicles in work where vesicular microRNA cargo is the target bioactive component. Highest yields for vesicular microRNA were obtained using conventional methods and among them, subsequent ultracentrifugation with 100,000 g and 135,000 g conventional method 2 was selected as it had the highest RNA to protein ratio indicating that it pelleted the least protein in relation to RNA an important factor for in vivo applications to assess microvesicle functionalities without risk of contaminating non-vesicular biomaterial. Microvesicles isolated using conventional method 2 were successfully internalized by cells in vitro showing their potential to deliver their cargo into cells in vitro and in vivo in case of functional studies.

Calf Rearing Systems in Smallholder Dairy Farming Areas of Zimbabwe : A Diadnostic Study of the Nharira-Lancashire Area

  • A formal survey was carried out in Nharira-Lancashire areas located in Chivhu to assess the calf rearing systems practiced in smallholder dairy farming areas of Zimbabwe. A total of 47 farmers, collectively owning 305 cows and 194 calves of various breeds, participated in the survey. All the farmers allowed their calves to suckle their dams all day to obtain colostrum. The colostrums intake period was significantly (p < 0.05) shorter (5.2 vs 4.1 days) in the small scale commercial area (SSCA) compared to communal area (CA). Milk was first sold to the Nharira-Lancashire Milk Centre a day after the colostrum intake period ended. Most of the CA (91.3%) and SSCA (77.8%) farmers penned their cows and calves together at night during the colostrum intake period. Thereafter the calves were penned separate from their dams. After colostrum intake, two types of calf suckling systems were practised; twice a day suckling and twice a day then changed to once a day suckling. In both systems, suckling was allowed for 30 minutes after the cows had been hand milked. There was no significant (p < 0.05) difference in the mean weaning age of calves between the CA and SSCA (5.8 vs 5.4 months). The most common weaning method was through separation of the calves from the dams. The limitaitions to calf production in Chivhu were the prohibitively high costs of calf meals, poor feed resources during the dry season, a general lack of knowledge on calf rearing diseases and inappropriate calf housing.
