Changes of Lectin from Viscum coloratum by Fermentation with Lactobacillus plantarum : Effect of pH and Temperature, Suger Specificity and Lymphocyte Stimulting Activity (유산균 발효에 의한 겨우사리 중의 렉틴 성분의 변화 : pH, 온도의 영향, 당 특이성, 림프구 자극분열효과)

  • Lectin from mistletoe(Viscum coloratum) fermented by Lactobacillus plantarum was compared with the lectin from unfermented mistletoe. Agglunating activity of fermented mistletoe was stable at pH 3.77~8.71, at temperature range of $0~40^{\circ}C$ and in the presence of 9 mental ions, which results are similar to unfermented one, but less stable at pH 2.03~3.00 and more stable at temperature $60~80^{\circ}C$ than lectin from unfermented one. Agglunating activity of lectin from mistletoe fermented for 1 or 2 days and from fraction number 42~54 was not inhibited by all sugars used except for lectin from fraction number 21~34. Mitogenic activity to murine lymphpocytes of lectin from mistletoe was decreased by fermentation process.

Seasonal Variation in Photosynthetic Characteristics and Chlorophyll Content of the Loranthus tanakae, Viscum album var. coloratum and its Hosts in Korea (계절에 따른 국내 자생 꼬리겨우살이, 겨우살이 및 기주목의 광합성 특성과 엽록소 함량)

  • We investigated seasonal variation in photosynthetic characteristics and chlorophyll content of the Loranthus tanakae, Viscum album var. coloratum and its hosts in Korea. The maximum photosynthesis and transpiration rate of L. tanakae were $9.36{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at $941{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PAR (photosynthetically active radiation) in June, $5.06{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at $1,596{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PAR in July, respectively. The maximum photosynthesis and transpiration rate of V. album var. coloratum were $6.51{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at $418{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PAR in Autumn, $3.91{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at $1,735{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PAR in Autumn, respectively. V. album var. coloratum was able to conduct photosynthesis in November whereas its host and L. tanakae were not able to conduct photosynthesis. Especially transpiration rate of L. tanakae were always higher than its host and V. album var. coloratum. The chlorophyll a+b contents of L. tanakae was $8.23mg{\cdot}g^{-1}$ in July, V. album var. coloratum was $10.27mg{\cdot}g^{-1}$ in June, and chlorophyll a/b ratio of L. tanakae was 1.7~3.7, V. album var. coloratum was 1.1~4.5, depend on season.

  • In general, studies of aerial parasitic plants known collectively as mistletoe have been carried out to investigate their ecological and agricultural characteristics. However, with the recently increased level of interest in medicinal resources, research on different types of Korean mistletoe has also increased. This study was carried out to review the work on the taxonomy and ecology of Korean mistletoe in preparation for the industrial use of these plants in the future. Mistletoe types are flowering plants belonging to Santalales, which exist in the form of parasites on the branches of trees or shrubs. In Korea, five taxa of four genera in two families of mistletoe exist: Viscum coloratum (Komarov) Nakai f. coloratum, Viscum coloratum (Komarov) Nakai f. rubroaurantiacum (Makino) Kitagawa and Korthalsella japonica (Thunb.) Engl. in Santalaceae, along with Loranthus tanakae Franch. et Sav. and Taxillus yadoriki (Sieb. ex Maxim.) Danser in Loranthaceae. As taxonomic studies of these species remain insufficient and given that the distribution ranges of these species are very wide, further observations pertaining to the morphological variations in each species are necessary. The distribution of mistletoes is known to be determined by the host specificity, the interval between the hosts, the environmental condition, the habits of the host plant, the eating characteristics of mediators in the area, and their habitat selection features.

Anti-oxidant, Anti-aging, and Whitening Effects of Viscum album var. coloratum In Vitro (한국산 겨우살이의 항산화, 항노화 및 미백 효과)

  • In this study, we investigated the anti-oxidant, anti-aging, and skin whitening effects of Korean mistletoe (Viscum album var. coloratum). The mistletoe fraction was composed of four types: hexane (HX), ethyl acetate (EA), butanol (BU), and methylene chloride (MC). In total phenol content assay, HX showed the highest phenol content among four fractions. In addition, EA significantly increased 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and catalase-like activities, and MC significantly increased superoxide dismutase (SOD)-like activity. When we compared IC50 value in the hyaluronidase and elastase inhibition assay, MC had the lowest IC50. In addition, we also performed tyrosinase inhibition assay to demonstrate the possibility of Korean mistletoe as a cosmetic component. HX showed the highest tyrosinase inhibition rate among the fractions.

Germination and Seedling Induction of Viscum album var. coloratum (Kom.) Ohwi after Artificial Inoculation on the Branch of Host Plants (겨우살이의 종자의 기주목 접종 및 유묘 활착기술)

  • Berries of Korean mistletoe (Viscum album var. coloratum [Kom.] Ohwi) contained one seed, which have, in general, one or two embryos but very rarely three embryos. Mucilaginous substances in berries may help them adhere to the branches of host trees. It was observed that seeds need more than one and half years to develop into normal and healthy seedlings from the time of inoculation. Many factors such as adhesion of berry, thickness of host branch, orientation of haustorial root, etc. influenced the successful development of mistletoe plants. Through the application of six-year observation results on the germination of seeds and growth of seedlings, about 80% of germination rate for mistletoe seeds and 61% of survival ratio for germinated seeds, which is more than 23 times higher in natural conditions, were obtained after inoculation of seeds on the one-year-old branches of Malus pumila var. dulcissima and Quercus mongolica trees. The technological aspects of the success can be applied to other host plants and provide a critical clue to an artificial propagation system, for this medicinally valuable genus. This is the first successful report on artificial inoculation and plant development of Korean mistletoe.

Isolation and Characterization of $\beta$-Galactoside Specific Lectin from Korean Mistletoe (Viscum album var. coloratum with Lactose-BSA-Sepharose 4B and Changes of Lectin Conformation

  • Lectins and its A- and B-chains from Korean mistletoe (Viscum album var. coloratum) were isolated by affinity chromatography on the Sepharose 4B modified by lactose-BSA conjugate synthesized by reductive amination of ligand (lactose) to .epsilon.-amino groups of lysine residues of spacer (BSA) after reduction by $NaCNBH_3$. The lactose-BSA conjugate was coupled to Sepharose 4B activated by cyanogen bromide. The molecular weight determined by SDS-PAGE were a 31 kD of A-chain and a 35kD of B-chain. Amino acid analysis and N-terminal sequencing were performed. The effects of pH, temperature and guanidine chloride on the conformation of the lectin were investigated by measuring its intrinsic fluorescence and compared with its hemagglutinating activities. Blue shift was detected on the acidic pH and there was a close relationship between activities and conformation of the lectin. Under denaturing conditions, the tryptophan emission profile of lectin showed typical denaturaiional red shift which also correspond to the conformations and activity of lectin.

Bleeding Time Prolongation Effect of Methanol Extract of Viscum album var. coloratum

  • The methanol extract of Viscum album var. coloratum, Korean mistletoe, showed potent prolongation effects on the bleeding time in rats in vivo, and whole blood clotting time and plasma recalcification time in rats ex vivo. The prolongation effect on the bleeding time of Korean mistletoe is comparable to that of Viscum album L., European mistletoe, 185.6% and 176.5%, respectively. However, the water extracts of the both plants did not show any prolongation effects. Platelet activating factor (PAF) receptor binding assay was carried out to elucidate the action mechanisms of the extracts, and both of the methanol extracts did not show any inhibitory activity. The $LD_{50}$ of the methanol extracts of both mistletoes are more than 2 g/kg. These results suggest that the mehtanol extract of Korean mistletoe might be a potential candidate to develop new drug to improve microcirculation.

  • A lectin (agglutinin, VCA) from Korean mistletoe (Viscum album L. coloratum) was isolated by affinity chromatograpy on a asialofetuin-Sepharose 4B. The molecular weights of A- and B-chains of VCA were different from those of VAAS. The VCA recognized the antibody of VAAs in the Western blot analysis and ELLA system. We also investigated the synergistic effects of the components in mistletoe by dividing the extract into different molecular weight fractions.

Korean Mistletoe (Viscum album Coloratum) Extract Induces Eel (Anguilla japonica) Non-specific Immunity

  • Background: The immunomodulatory effects of Korean mistletoe (Viscum album Coloratum) on the innate immune responses of eel (Anguilla japonica) were studied. Methods: Mistletoe, Freund’s complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control was injected into eel peritoneal cavities. Results: Nitroblue tetrazolium (NBT)-positive cells in the head kidney of eel were significantly augmented by the second day post-injection of mistletoe. Reactive oxygen intermediates (ROI) were more produced in mistletoe-injected fish kidney leucocytes than in FCA-injected ones. The level of lysozyme activity in the serum of fish 2 days after injection with mistletoe was also significantly higher than that in the serum of the control fish. The optimal concentration of mistletoe in inducing the highest serum lysozyme activity was revealed to 500${\mu}$g/200 g of fish. In phagocytic activity assay, mistletoe-sensitized eel kidney phagocytes captured more zymosan than did the control fish. Conclusion: Korean mistletoe appeared to be a good activator of the non-specific immune responses of eel.

Flounder were inoculated with mistletoe, Freunds complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control into their peritoneal cavities. Reactive oxygen intermediate (ROI) products were more enhanced in mistletoe-injected fish kidney phagocytes than in FCA-injected ones. The level of lysozyme activity detected in the serum of fish 4 d after injection with mistletoe was also significantly higher than that found in the serum of the control fish. The appropriate concentration of mistletoe in eliciting the highest level of serum lysozyme activity was 500 $\mu$m/300 g of fish. In phagocytic activity assays, mistletoe-sen-sitized flounder kidney phagocytes captured more yeasts than those of the control fish. Korean mistletoe appeared to be a good activator of the non-specific immune responses of Japanese flounder.
