In this study, we analyzed the attachment amount and the surface properties, such as shape and attachment aspect of silver nanoparticles on the surface of salt and sucrose. In addition, we investigated the antimicrobial activity of silver nanoparticles by measuring total colony counts and total coliforms in sewage according to the time and the amount of silver nanoparticles. As a result, it is found that silver nanoparticles are attached on the surface of salt and sucrose from the SEM images and there is no microorganisms on the surface of salt and sucrose. Silver nanoparticles on salt were rod shape but silver nanoparticles on sucrose were round shape. Also, the content of silver was 0.735 mg/g on salt and 0.885 mg/g on sucrose. In all experiments, total colony counts and total coliforms sharply declined initially, and it decreased gradually as change the time. When injection amount of nano silver sucrose and salt increased, the width of decline was greater. The amount of total colony counts and total coliforms of nano silver salt was much lower and the disinfection efficiency was higher comparing with nano silver sucrose. That means the case of nano silver salt is much better at the antimicrobial activity.

Purpose: This study was aimed to investigate the colony counts by different disinfecting processes and duration of the operation for humidifiers in an intensive care unit (ICU) in Busan. Method: The four steps of disinfecting process were taken with four different humidifiers according to the use of disinfectant (1:100 $Deconex^{(R)}$ 50FF/nothing) and refilling water (tap water/distilled water). First, the colony counts using water sample from wick was conducted at 0 hr, 24 hr, and 48 hr respectively after turning on the humidifiers. Next, the colony counts using waterdrop from the outlet was done at 0 hr, 24 hr, 48 hr, and 72 hr respectively after turning on the humidifiers. Result: No colony was counted after disinfecting with 1:100 $Deconex^{(R)}$ 50FF until 72 hr after turning on the humidifiers. However, without disinfecting with $Deconex^{(R)}$ 50FF, the colony count was increased in the humidifiers from 24 hr after turning on the humidifiers. The result was the same regardless of the refilling water. Conclusion: According to the results, the study found that disinfection of humidifiers in the ICUs with 1:100 $Deconex^{(R)}$ 50FF every 72 hr is effective to remove general bacteria.

Three media, i. e., MOD-SD, M98-5 and M98-5 supplemented with chicken fecal extract were tested as isolation media for anaerobic bacteria present in the duodenum, jeju-ileum and cecum of chicken. The results showed that the mean colony counts of medium M98-5 were similar with those of MOD-SD medium in all intestinal samples at the incubation periods of 2, 6 and 10 days. Supplementation with chicken fecal extract of M98-5 medium significantly increased (p < 0.05) the colony counts of bacteria from the duodenum, jeju-ileum and cecum. The colony counts at 6-day incubation were similar with those at 10-day incubation, but were much higher than the counts at 2-day incubation. The major types of bacteria found in the duodenum and jeju-ileum of chicken were tentatively identified as Lactobacillus, Streptococcus and E. coli. In the cecum, ten tentatively identified groups of bacteria, namely, Streptococcus, Staphylococcus, Lactobacillus, E. coli, anaerobic coccus, Eubacterium, Propionibacterium, Clostridium, Fusobacterium and Bacteroides were isolated. Anaerobes were found to comprise nearly the entire microbial population of the cecum. Predominating in all sections of the intestine were homofermentative lactobacilli. The main Lactotacillus species in chicken intestine were L. acidophilus, L. fermentum and L. brevis.
This study characterized water quality of groundwater and purified water used for drinking water in 30 schools in Gyeongsangbukdo Province. The results of the study showed that 43% of groundwater and 45% of purified water were not suitable for drinking. Among them, microbial contaminations were the most serious problem. In raw groundwater, the exceeding rates of total colony counts was the highest in August (53.6%). Purified water samples showed higher rate of exceeding drinking water standard for total colony counts while showing lower rate of exceeding drinking water standard for total coliforms and fecal coliforms in March and August. Overall, proper managements for microbial contaminations are required for both groundwater and purified water. Furthermore, special attention should toward students not to drink water when groundwater or purified water exceeds the drinking water quality standard.
To evaluate the effect of disinfectants for Alginate impression, several commercially available disinfectants were tested to Alginate model contaminated by Methicillin resistant Staphylococcus aureus. Alginate specimens were artificially contaminated by Staphylococcus aureus and soaking in four commercially available disinfectants for 1, 5, 10 minutes and colonies were counted for each case. Significant reduction of bacterial colony counts was noted after soaking in 0.5% and 0.05% sodium hypochlorite and after soaking in 2% glutaraldehyde, but not after soaking in Hypochlorus add. Commercially available sodium hypochlorite and glutaraldehyde were found to be effective disinfectants for Alginate impression. But hypochlorus add was not adequate disinfectant.
Antibiotics susceptibilities of Staphylococci were tested to get useful data for prevention food contamination and treatment of food poisoning. Tested were colony counts, isolation of Staphylococci, identification for the Staphylococcus aureus and resistant patterns to antimcrobial agents. The colonies and Staphylococci isolated from the atmosphere of 37 home kitchens in the morning (07:00-08:00) on Nov. 19, 1977 in Busan. The findings are as follows: 1. The average of colony counts was shown to be 9.6 ranging from 47 to 1 at mean temperature of 18$\circ$C (the highest 21$\circ$C, the lowest 15$\circ$C) 2. Out of total 37 kitchens, 18 Staphylococci were isolated from 15 kitchens with 40.5 per cent, and 4 Staphylococcus aureus were identified with 22.2 per cent from 18 Staphylococci. 3. Isolated Staphylococci were resistant to penicillin(100%), dihydrostreptomycin(93.3%), tetracycline (93.3%) and oleandomycin(66.6%), while the strains were sensitive to sulfonamide(88.6%) and colistin(83.3%). 4. Staphylococcus aureus identified were resistant to dihydrostreptomycin, penicillin and tetracycline, while the strains were sensitive to sulfonamide and colistin.
This study was investigated to evaluate the distribution of airborne microorganisms at underground shops in Seomyun and Nampodong, Pusan. The number of bacterial colonies on the nutrient agar plate plates were calculated by the open petri dish method for 30 minutes in indoor air of underground shops at every seasons in a year. There was no statistically significant difference between Seomyun and Nampodong in mean colony counts. Isolation rates of Gram positive rods was highest, and Gram positive cocci and Gram negative rods were followed. In Nampodong underground shops, Enterobacteriaceae strains was isolated. Mean colony counts according to seasons was higher at summer and autumn in Seomyun, and spring and winter in Nampodong. In near future, a study on the distribution of bacteria causing respiratory infection should be followed.
A survey was conducted to measure concentration of airborne microbe in a hospital using RSC air sampler during October~November 1991.The result was as follows: 1) In an agar strip GK-A media for total counts of microbial particles. The highest count were 1384 CFU/m$^3$ in the main lobby, followed by 912 CFU/m$^3$, in the obstetric room, 688 CFU/m$^3$ in 1CU. By gram staining, the distribution for organisms in the air were shown 74.1% in gram possitive cocci followed by 16.8%, in gram possitive bacilli 6.7% in gram negative bacilli and 4.7% in yeast, but low organism was detected in recovery room with 194 CFU/m$^3$. 2) In agar strip S media for Staphylococci the count at the main lobby was detected in the recovery room with 92 CFU/m$^3$, Tests of coagulase were negative Staphylococci with 78%, and positive Staphylococci with 22%. The Staphylococci were highly resistance to penicillin, ampicillin and sensitive to amikacin, cefazolin, gentamycin and chloramphenicol. 3) In agar strip C media for coliform bacteria the colony counts at the main lobby was 139 CFU/m$^3$ and treatment room was 190 CFU/m$^3$, most frequently isolated microorganisms were non fermentative bacilli. 4) In agar strip HS media for yeast and molds. Most frequently colony counts 17~76 CFU/m$^3$, 0.5% lactophenol cotton blue stains were shown unidentified 77.2%, 8.1%, in Penicillium 8.1% in Aspergillus, and 3.8% in mucor.
This research was performed to investigate the dynamics of microbial community by RBC (Rotating Biological Contactor) using Rhodococcus sp. EL-GT and activated sludge. Cell counts revealed by DAPI were compared with culturable bacterial counts from nutrient agar. Colony counts on nutrient agar gave values 20~25% and 1~15% of cell counts (DAPI). The cell counts for the dynamics of bacterial community were determined by combination of in situ hybridization with fluorescently-labelled oligonyucleotide probes and epifluorescence microscopy. Around 90~80% of total cells visualized DAPI were also detected by the bacteria probe EUB 338. For both reactors proteobacteria belonging to the gamma subclass were dominant in the first stage (1 and 2 stage) and proteobacteria belonging to the gamma subclass were dominant in the last stage (3 and 4 stage).
The use of tissue conditioners has come into favor for preprosthetic treatment and the treatment of Denture stomatitis, but the major problem in the use of the tissue conditioner is the growth of C.albicans. To compare the growth of C.albicans according to the type and the wearing period of the tissue conditioner, three commercial tissue conditioners were relined to intraoral plates which were delivered to 14 smoking, 12 nonsmoking healthy men. Cultures were made from the conditioner surface at 2, 7, 14, 21 days after after intraoral placement. The frequency of positive culture and colony counts of C.albicans were measured by imprint culture technique. The following results were achieved : 1. The frequency of positive cluture had increased significantly for all materials used. 2. The frequency of positive culture had incresed significantly around 2,14 days for day for COE-Comfort, SR-Ivoseal. 3. There were no significant difference in the colony counts of C.albicans among the mate rials used. 4. There were an over-all increase with the wearing period of tissue conditioners and significant increase around 2, 7, 14 days for COE-Comfort, and 2, 14 days for SR-Ivoseal, viscogel in colony counts. 5. Smoking had no effect on the growth of C.albicans. That is, there were no difference among the materials used in the growth of C.albicans. In the clinical application of tissue conditioners, we should avoid a long term use of it, but in inevitable cases, special disinfection procedures should be considered.
