  • Direct colony hybridization on agarose gel plate was established for the identification of recombinant plasmids. The hybridization of the probe to nucleic acids on dried gel without transferring to solid supports was more effective and simpler than hybridization of such probes to materials immobilized on filters such as nitrocellulose or nylon. D-cycloserine in overlaying agamse was essential for releasing the nucleic acids from colony.

  • Purpose: The purpose of this study is to evaluate the efficacy of mesenchymal stem cell (MSC) isolation by the magnetic-activated cell sorting (MACS) method in tendon tissue-derived cells compared to the colony picking method for isolation of MSCs by picking colony-forming cells. Materials and Methods: Human tendon-derived cells were isolated by enzyme digestion using normal tendon tissues from three donors. We used the magnetic kit and well-known MSC markers (CD90 or CD105) to isolate MSCs in tendon-derived cells using MACS. Cloning cylinders were used to isolate colony-forming cells having MSC characteristics in tendon-derived cells. Colony-forming unit-fibroblast (CFU-F) assay was used to evaluate the self-renewal capacity of cells isolated using the colony picking method or MACS. For comparison of differentiation potentials into osteogenic or adipogenic lineage between two groups, alizarin red S and oil red O staining were performed at 14 days after induction of differentiation in vitro. Results: Flow cytometry results showed that early passage tendon-derived cells expressed CD44 in 99.13%, CD90 in 56.51%, and CD105 in 86.19%. In the CFU-F assay, CD90+ or CD105+ cells isolated with MACS showed larger colony formation in size than cells isolated using the colony picking method. We also observed that CD90+ or CD105+ cells were constantly differentiated into both osteogenic and adipogenic lineages in cells from all donors, whereas cells isolated using the colony picking method were heterogeneous in differentiation potentials to the osteogenic and adipogenic lineages. Conclusion: CD90+ or CD105+ cells isolated using MACS showed superior MSC characteristics in the self-renewal and multi-differentiation capacities compared with cells isolated using the colony picking method.

  • We investigated possible effect of different concentration of sugar solution and addition of antiseptic in the solution on oviposition and colony development of Bombus ignitus and B. terrestris. The rates of oviposition, colony foundation and progeny-queen production of B. ignitus were 1.2-3.0 fold higher in the 40$\%$ sugar solution than those of the 50$\%$ sugar solution. The rates of oviposition, colony foundation and progenyqueen production were 1.1-2.6 fold higher in the 40$\%$ sugar solution added in 0.3$\%$ sorbic acid as antiseptic than those of the 40$\%$ sugar solution. Further, the death rate within one month was 1.7 fold lower in the 40$\%$ sugar solution added in 0.3$\%$ sorbic acid than that of 40$\%$ sugar solution alone. In the comparison of the colony development tested using imported sugar solution, the Beehappy???, the 40$\%$ sugar solution added to antiseptic and the 40$\%$ sugar solution without antiseptic, the 40$\%$ sugar solution added to antiseptic was about equal to the Beehappy??? in colony development of B. terrestris. Further, the number of adults produced was 1.2-3.0 fold higher in the 40$\%$ sugar solution added to antiseptic than that of the Beehappy???. Therefore the 40$\%$ sugar solution was more effective than the 50$\%$ sugar solution, and the 40$\%$ sugar solution added to antiseptic was the most effective in colony development and mass rearing of bumblebee.

  • The purpose of this study is to understand spatio-temporal changes of active fungal biomass and water in Tricholoma matsutake soil colonies during the mushroom fruiting season. The active fungal biomass was estimated by analyzing ergosterol content at four different points within four replicated locations in a single circular T. matsutake colony at Ssanggok valley in the Sogri Mt. National Park in Korea during 2003 to 2005. The four points were the ahead of the colony, the front edge of the colony and 20 cm and 40 cm back from the front edge of the colony. Ergosterol content was 0.0 to 0.7 ${\mu}g$ per gram dried soil at the ahead, 2.5 to 4.8 ${\mu}g$ at the front edge, 0.5 to 1.8 ${\mu}g$ at the 20 cm back and 0.3 to 0.8 ${\mu}g$ at the 40 cm back. The ergosterol content was very high at the front edge where the T. matsutake hyphae were most active. However, ergosterol content did not significantly change during the fruiting season, September to October. Soil water contents were lower at the front edge and 20 cm back from the front edge of the colony than at the ahead and 40 cm back during the fruiting season. Soil water content ranged from 12 to 19% at the ahead, 10 to 11% at the edge, 9 to 11% at the 20 cm back and 11 to 15% at the 40 cm back. Our results suggest that the active front edge of the T. matsutake soil colony could be managed in terms of water relation and T. matsutake ectomycorrhizal root development.

  • As Sacbrood virus (SBV), a causative agent of larval death and colony collapse in Apis cerana honey bee, is prevalent and poses one of the most significant threats to the Korean apiculture, development of methods to counter this viral disease is urgently needed. In this study we tested some SBV controlling methods, such as requeen, shook swam, adding Apis mellifera and spraying yogurt to SBV inoculated colony. Colony size measured by number of sealed brood and adult were evaluated every 15 days until two months while instances of recurrence were recorded up to five months after applying treatment methods. We also test the effects of yogurt on healthy and SBV-infected larvae at both in vitro reared larvae and colony level. Our result showed that all SBV controlling methods had similar success rates with respect to elimination of SBV clinical symptom up to 30 days post treatment. Mix-species and spraying yogurt method had similar pattern of sealed brood and adult number and higher than that of other SBV controlling methods up to 45 days post treatment. These two groups also showed the lower percentage of SBV recurrence (50% and 66.7%) at 120 days post treatment than other group that mostly colony had clinical symptom. Result on in vitro reared larvae challenged with yogurt showed that yogurt have neither harmless on healthy larvae nor remedial effect on SBV infected larvae. However, at colony level, colony in group received yogurt treatment removed significantly more SBV-infected larvae, SBV-killed larvae, and even healthy larvae in comparison to the control, suggested that yogurt could trigger the hygienic behavior of nurse bee. Our results recommended that it is practical in beekeeping by adding A. mellifera and spraying yogurt to control SBV in A. cerana colony.

  • Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a pleiotropic hematopoietic growth factor and an activator of lmature myeloid cells and recombinant GM-CSF is increasingly under clinical studies for the treatment of various diseases including cancer, infectious diseases and hematopoietic diseases. We constructed a reconbinant mouse GM-CSF expression plasmid with pelB leader sequence and His. Tag under T7 promoter control, and showed that the construct produced a 20 kDa recombinant protein in 8M urea. We also showed that the 20 kDa recombinant protein prepared in 8M urea sitmulated colony formation in vitro, indicating that the recombinant mGM-CSF can be renatured to its native form to show the colony stimulating activity.

  • The Ant Colony System (ACS) is a variant of Ant colony optimization algorithm which is well-known in Traveling Salesman Problem. This paper proposed a hybrid method based on genetic algorithm (GA) and ant colony system (ACS), called GACS, to solve traffic routing problem. In the GACS, we use genetic algorithm to optimize the ACS parameters that aims to attain the shortest trips and time through new functions to help the ants to update global and local pheromones. Our experiments are performed by the GACS framework which is developed from VANETsim with the ability of real map loading from open street map project, and updating traffic light in real-time. The obtained results show that our framework acquired higher performance than A-Star and classical ACS algorithms in terms of length of the best global tour and the time for trip.
