Functional Characteristics of Enterococcus faecium SA5 and Its Potential in Conversion of Ginsenoside Rb1 in Ginseng (Enterococcus faecium SA5의 기능적 특성과 인삼 ginsenoside Rb1의 전환)

  • The fermentation of Panax ginseng can yield many compounds from ginsenosides that have a wide variety of biological functions. Lactic acid bacteria (LAB) strains are capable of converting ginsenosides. The purposes of this study were to: (1) characterize Enterococcus faecium SA5, an isolated LAB from Mongolian mare milk, (2) identify the existence of extracellular ${\beta}$-glucosidase activity in the milk, and (3) ascertain if the ${\beta}$-glucosidase has the capacity of converting ginsenoside in Korean ginseng. The results revealed that E. faecium SA5 was acid-resistant, bile salt-resistant, and has antibiotic activities against 4 pathogenic microorganisms (Salmonella typhimurium KCTC 3216, Listeria monocytogenes KCTC 3710, Bacillus cereus KCTC 1012, Staphylococcus aureus KCTC 1621). In addition, E. faecium SA5 had tolerance against some antibiotics such as colistin, gentamycin and neomycin. It was also found that E. faecium SA5 possessed bile salt hydrolase activity, which could lower blood cholesterol level. When incubated in 10% (w/v) skim milk as a yogurt starter, E. faecium SA5 caused to decrease pH of the medium as well as increase in viable cell counts. Using TLC and HPLC analysis on the samples incubated in MRS broth, our study confirmed that E. faecium SA5 can produce ${\beta}$-glucosidase, which was capable of converting ginsenoside $Rb_1$ into new ginsenosides $Rg_3-s$ and $Rg_3-r$. It was concluded that E. faecium SA5 possessed a potential of probiotic activity, which could be applied to yogurt manufacture as well as ginsenoside conversion in ginseng.

Characterization of Microbial Pathogen Bacillus thuringiensis Isolates from Soil Against Mosquito and Silkworm Larvae (II) (토양에서 분리한 살충성 Bacillus thuringiensis의 모기와 누에 유충에 대한 독성효과 (II))

  • Eight strains of Bacillus thuringiensis were isolated from soil in Korea and characterized. The isolates were named HL-24, HL-25, HL-33, HL-34, HL-35, HL-38, HL-39, HL-40. Strains HL-24 and HL-25 produced irregular parasporal crystals, HL-33 and HL-35 produced bipyramidal crystals, and others were round form in their cells. The biochemical characteristics of the eight isolates were only minor different in specific characteristics to the known serotypes of Bacillus thuringiensis. The HL-25, HL-33 and HL-34 strains showed resistances to cephalothin, colistin and penicillin G, and HL-39 and HL-40 strains were resistant to penicilin G. The strains of HL-24, HL-25, HL-33 and HL-34 were toxic to Bombyx mori lavae and HL-24, HL-25, HL-38, HL-39 and HL-40 strains killed Culex pipiens 3rd instar larvae. The HL-24 and H25 strains showed lethal activity against two kinds of the larvae, however lethality against mosquito larvae was low.

Emergence of Conjugative Multidrug-Resistant Pseudomonas aeruginosa (접합가능한다제내성녹농균의출현)

  • The emergence and spread of multidrug-resistant Pseudomonas aeruginosa (MRPA) have become a serious problem worldwide. The involvement of metallo-β-lactamases (MBLs) in inducing carbapenem resistance is particularly acute. However, unlike other members of the Enterobacteriaceae genus, new clones of P. aeruginosa are constantly emerging and rapidly replacing previously prevalent dominant clones. Therefore, this study aimed to perform antimicrobial resistance gene analysis, integron gene cassette analysis using DNA sequencing, and plasmid transfer analysis by conjugation to investigate the antimicrobial resistance dynamics of 18 P. aeruginosa strains isolated from various medical samples at a general hospital in Busan from September 2017 to September 2019. All 18 strains showed extensively drug-resistant (XDR) phenotype and were resistant to most antibiotics, except colistin (100%) but were susceptible to aztreonam (22.2%) and ceftazidime (16.6%). Approximately 66.7% of the strains had Class 1 integrons showing various antimicrobial resistances. Notably, IMP-6 ST235 (66.7%), VIM-2 ST357 (16.7%), and IMP-1 ST446(16.7%) were identified. The identification of IMP-1-producing ST446, previously unreported in Korea, is noteworthy considering the emergence and prevalence of another MRPA high-risk clone.

Antibiotic resistance pattern of avian pathogenic Escherichia coli isolated from chickens (닭에서 분리된 조류 병원성 대장균의 항생제 내성 양상)

  • Avian pathogenic Escherichia coli (APEC) is an important bacterial pathogen of chickens and causes colibacillosis such as airsacculitis, perihepatitis, omphalitis, peritonitis, salpingitis, and pericarditis. As the transfer of antibiotic resistance from animal to humans can be possible, surveillance on antibiotic resistance of APEC is very important. A total 34 APEC isolates from diseased chickens during the period from 2007 to 2009 were obtained. The susceptibility of the isolates to 13 antibiotics was determined by disc diffusion assay. Resistance to erythromycin was found in 97.1% of APEC isolated, followed by resistance to tetracycline (85.3%), doxycycline (82.3%), ampicillin (73.5%), sulfisoxazole (67.6%), enrofloxacin (67.6%), ciprofloxacin (64.7%), norfloxacin (61.7%) trimethoprim/sulfamethoxazole (52.9%), gentamycin (26.5%), amoxicillin (8.8%), colistin (5.9%), and amikacin (2.9%). The blaTEM genes were detected in 25 (100%) of the 25 ampicillin-resistant APEC isolates. Among the 29 tetracycline-resistant APEC isolates, tetA and tetB genes were detected in 18 (62.1%) and 9 (31%) isolates, respectively. Twenty six (76.5%) isolates were multiresistant to at least 6 antibiotics and seven (20.1%) isolates were multiresistant to at least 10 antibiotics. This results indicated that multiple antibiotic-resistant APEC is widespread in chicken flocks in Korea.

Antimicrobial Effect of Grapefruit Seed Extract on Vibrio parahaemolyticus Isolated from the Southern Adjacent Sea of Korea (남해안연안에 분포되어 있는 Vibrio Parahaemolyticus의 성상 및 Grapefruit Seed Extract 처리에 의한 향균효과)

  • To investigate the distribution of Vibrio parahaemopyticus in the southern 4 coastal areas, Vibrio parahaemolyticus was isolated from seawater, shellfish and sediment from May to October in 1991, and antimicrobial effect of grapefruit seed extract (GFSE) on the growth of isolated strains were examined. In the 120 sample from 4 coastal areas, 16 strains of Vibrio parahaemolyticus were isolated and identified. The distribution serotype of isolated strains was 10 types of monovalent k-antiserum. Especially k-5 and k-28 were highly distribyted with 3 and 4 strains. 31.3% of isolated strains showed positive on Kanagawa phenomenon test. All isolated Vibrio parahaemolyticus were sensitive to chloramphenicol and gentamycin, 5 and 6 strains were resistant to streptomycin and colistin, respectively. Isolated strains were compared with geographical, month and sample. The distribution of 16 isolated Vibrio parahamolyticus was high at Hadong with 50%(8 strains), on July with 43.8%(7 strains) and from seawater with 37.5%(6 strains) respectively. Minimal inhibitory level of GFSE to Vibrio parahaemolyticus was 50 ppm. With 100 ppm treatment of GFSE, the destroy of cell membrane function, outflow of cell ingredients and ghost morphology of cell were investigated.

  • To investigate the epidemiological trait of intestinal diseases of animals caused by thermophilic Campylobacter spp., isolation of etiological agent was carried out. Isolated Campylobacter spp. were biotyped, serotyped and the susceptibility of the isolates to antimicrobial agents were examined. Th results were as follows. 1. Isolation rates of Campylobacter spp. from 649 fecal materials of 208 cattle, 300 pigs and 141 chickens were 25.5%, 23.7% and 38.3%, respectively. 2. The majority of the 130 isolates of C jejuni was classified as biotype I(50.6%) and biotype II (34.6%). Most of the 46 isolates of C coli were biotype I (71.7%). 3. Isolated C jejuni strains showed 14 different serotype, and serotype 4, 26, 36 were most frequent. Isolated C coli strains showed 5 different serotype and serotype 31 and 21 were relatively common. 4. Isolated Campylobacter spp. were highly susceptible to nalidixic acid, amikacin, gentamicin, colistin and chlorampehnocol.

  • this study was performed for the purpose of surveying the distribution and characteristics of the Vibrio species in the diseased farm flounders (Paralichthys olivaceus) during the winter season in Korea. Samples were collected along the southern coast in the Sinam, Keoje Island, and Teosu areas from November '90 to Feburary '91. Nine species of Vibrio were identified as V. tubiashii (85 strains), V. damsela (78 strains), V. anguillarum (21 strains), V. compbelli (9 strains), V. fluvialis (9 strain), V. costicola (1 strain), V. alginolyticus (1 strain), V. gazogenes (1 strain), V. marinus (1 strain), and unidentified 3 groups of bacteria (154 strains) which may have related with V. tubiashii were isolated. Eleven kinds of discs (BBL Co.) were used for antibiotic susceptibility test and 116 strains were selected for the experiment. Most bacteria were powerfully inhibited their growth by 4 antibiotics such as cephalothin, chloramphenicol, colistin, and tetracycline, but they were poorly inhibited by 4 antibiotics such as carbenicillin, kanamycin, penicillin G, and streptomycin.

  • The present study was conducted to investigate isolation and antimicrobial drug susceptibility of Pasteurella spp from pneumonic calves and cows (pneumonic lungs) in Gyeongnam south area from January to November 2000. Pasteurella multocida and P haemolytica were isolated 136 (53.8%) and 22(8.7%), respectively, from 158 of 253 pneumonic calves of 1 to 12 months of age and from pneumonic lungs and simultaneous isolation rate of P mutocida and P haemolytica was 4 (1.6%). Seasonal isolation rate of Pasteurella spp in calves and pneumonic lungs varied from 3 to 65.2%, and it was higher in Fall. The majority of biochemical and cultural properties of P multocida and P haemolytica isolated from calves and pneumonic lung were identical to those of the reference strains employed. Pneumonic lesions of the examined lungs were histologically identified as purulent bronchopneumonia and fibrinous pneumonia. All isolates were resistant to penicillin, amikacin, lincomicin, colistin, cloxacilling, sulfonamide, kanamycin, sulfamethoxasoletrimetoprim, and gentamicin(>80%), but some of them were susceptible to amoxicillin, norfloxacin, cephalexin, cefazolin, orbifloxacin, enrofloxacin, and ciprofloxacin(>60%).

Antibiotic resistance and plasmid profile of salmonella spp isolated from swine in Kyoungbuk province (경북지방 돼지유래 salmonella 속균의 약제내성과 plasmid profile)

  • This study was conducted to investigate the antibiotic resistance and plasmid profiles of 58 salmonella spp isolated from mesenteric lymphnodes of slaughter pigs in Kyoungbuk province during the period from September 1997 to June 1998. The results obtained are as follow that all isolates were susceptible to amikacin, gentamicin, ciprofloxacin and enrofloxacin, and the majority of isolates were highly susceptible to norfloxacin, colistin, nalidixic acid and apramycin while they were moderately susceptible to kanamycin, cephalothin, chloramphenicol, ampicillin, trimethoprim and penicillin. The majority of isolates were over 90% resistant rates to lincomycin, sulfadimethoxine, vancomycin, methicillin and erythromycin. The plasmid profiles of 58 salmonella spp are developed 1 to 4 fractions, 0.9 to 29.5 Kb molecular range sizes and U strains (45.5%) were showed plasmid profiles by agarose gel electrophoresis. 5 derby harbored 29.5 Kb and 7 Kb, and S schwarzengrund had 14 Kb and 0.9 Kb harboring sizes. Four of 10 S agona and 2 of 4 S typhimurium were harbored 3.1 Kb and n.5 Kb, respectively. Thirty-five untypable strains are developed variable size fractions its showed small size plasmid profile less than 6 Kb and 22 (62.8%) of them had no detectable plasmids.

Isolation and identification of Moraxella cuniculi from a rabbit with keratoconjunctivitis

  • A Gram-negative, catalase- and oxidase-positive, coccus-shaped bacterium was isolated from a rabbit with keratoconjunctivitis. Colonies of the isolate were round, smooth, and exhibited hemolytic activity on 5% sheep blood agar. Scanning electron microscopy revealed 0.4 to $0.5{\mu}m$ diameter oval cocci. Partial 16S rRNA gene (1446 bp) sequence analysis demonstrated the isolate had significant homology with the Moraxella cuniculi CCUG2154 strain isolated from a rabbit in Germany in 1973. Our isolate was designated as APQAB1701. Antibiotic susceptibility tests demonstrated that APQAB1701 was sensitive to 24 antibiotics; 3 of the antibiotics (nalidixic acid, spectinomycin, and colistin) had minimal inhibitory concentrations ${\geq}32{\mu}g/mL$ against the isolate.
