Optimum Chilling Terms for Germination of the Dehisced Ginseng(Panax ginseng C. A. Meyer) Seed (개갑 인삼종자의 발아 적정 저온감응기간)

  • Experiments were conducted to study the optimum chilling period for breaking physiological dormancy of dehisced ginseng(Panax ginseng C. A. Meyer) seed. Embryo of ginseng seed is too small to be noticed with naked eyed on harvesting time. Embryo grew to half size of endosperm after seeds were stratified for 3 months. It takes 6 months for this embryo to reach the size enough for germination. And it grew faster indoors than outdoors. Dehisced ginseng seed with full-size embryo did not germinate at room temperature and required chilling treatment for 75days in outdoors and 90 days in cold chamber. While seed receiving sufficient chilling treatment were left to be in room temperature, the chilling effect decreased remarkably.

The Use of Multiple Tests in Predicting the Vigor of Soybean Seeds (대두 종자 발아세 검정방법 비교)

  • Five soybean varieties were used to measure seed vigor according to the artificially different deterioration. A vigor index derived from the product of percent germination x hypocotyllength was used as the parameter in comparison of other tests for seed vigor. There were the varietal differences in initial vigor. The warm germination test was the best measurement for seed vigor at the advanced stages of seed deterioration. All other vigor measurements, except ATP and GADA measurements, showed highly significant correlations with the vigor index. Hypocotyl length, conductivity index and cold germination measurements for seed vigor were appropriate for predicting seed quality.

  • The study was performed on the effects of crop load of pear (Pyrus pyrifolia Nakai) trees on shoot growth, reserved nutrients, and fruit production for two years, as well as regrowth of cuttings treated with various cold temperatures in 2012 and 2013. Crop load adjustments included three levels of fruiting; 54 (60%), 90 (100%), and 126 fruits per tree (140%). As increasing crop load from 60% to 140%, water sprout growth decreased, and one-year old shoot resulted in declined concentrations of total carbon, macro-nutrients, and free sugar. The 140% of crop load greatly increased fruit yield of approximately 35 tons per ha in both 2011 and 2012 but slightly reduced weight, size, and soluble solid contents in fruit. However, 140% of crop load increased annual production income to approximately over 10 million won compared to the 60% and 100% of crop load. Germination rates in the cuttings did not rapidly decrease up to $-20^{\circ}C$ in a growth chamber condition. The 140% of crop load decreased germination rates less than 70% of cuttings grown under $-30^{\circ}C$. Amounts of electrolyte leakage in the cuttings under various cold temperatures were not significantly different among the adjustment of crop load.

Changes in Seed Vigour of Sweet and Super Sweet Corn Hybrids as Affected by Storage Conditions (단옥수수와 초당옥수수의 저장조건에 따른 종자 활력변화)

  • An experiment was conducted to characterize the seed vigour of sweet (su) and super sweet (sh2) corn seeds stored at different temperatures and relative humidities (RH). Hybrid seeds of Early Sunglow ${\times}$ Golden Cross Bantam 70 (su) and Xtrasweet 82 ${\times}$ Fortune (sh2) were stored at different temperatures ($5\;and\;15^{\circ}C$) and RH(70 and 85%) for 10 months. Results of the experiment show that seed deterioration of super sweet corn was much faster than that of sweet corn under all storage conditions. Germination rate of sweet corn seeds at $25^{\circ}C$ and emergence rate in cold test showed similar patterns. Emergence rate of super sweet corn in cold test was significantly lower than the germination rate at $25^{\circ}C$. Germination rate of both sweet and super sweet corns was positively correlated to the emergence rate in cold test, but the correlation coefficient of super sweet corn was much lower compared to the sweet corn. This implies that the viability of super sweet corn seeds should be tested in the cold test to estimate field emergence rate. Seeds of sweet corn could be stored for 5 months under all storage conditions without significant seed deterioration, while those of super sweet corn should be stored at low temperature and RH. The emergence rate of sweet corn in cold test was not correlated to the leakage of total sugars, electrolytes or ${\alpha}-amylase$ activity, while that of super sweet com was positively correlated to the ${\alpha}-amylase$ activity, negatively correlated to the leakage of electrolytes, and was not correlated to the leakage of total sugars.

Effects of Gibberellin Treatment on the Dormancy-breaking and Germination Promotion of Carpinus laxiflora Seeds Stored for 16 Years (지베렐린 처리에 따른 서어나무 16년 저장 종자의 휴면타파 및 발아촉진 효과)

  • This study assessed the vitality of Carpinus laxiflora seeds stored for more than 15 years in order to discover optimal pre-treatment conditions for removingthe causes of seed dormancy and improving revitalization rate. Seeds were collected in October 2000 and stored at -18℃ for 16 years. Experiments to assess the revitalization ability of the seeds were performed under the following conditions: a controlled environment; in warm stratification (30 days at 23℃); in cold stratification (30 days at 4℃ andfor 30, 45, 60, and 120 days); gibberellin (GA3) treatment (24 hours per day in GA3 solutions of 100, 500, 1000, 2000, and 3000 mg·L-1); and both cold stratification and GA3 treatment (30 days at 4℃, then in a GA3 solution of 100, 500, and 1000 mg·L-1 for 2 hours). The average germination percentage(GP) of untreated seeds was 2%, and the average GP of warm-stratification seeds was 10%. Cold-stratification seeds had the highest GP at 81% for the 45-day process, while the 120-day cold-stratification seeds had the lowest GP at 67.3%. The average GP of seeds treated with GA3 ranged from 77% (100 mg·L-1) to 99% (1000 mg·L-1), indicatingsignificant differences between the treatment concentrations. The treatment effect of GA3 was highest at 500 mg·L-1 and 1000 mg·L-1, and lowest at 100 mg·L-1. The average GP of seeds treated with GA3 following cold stratification was 68%, which was lower than the cold stratification-only (73.2%) and GA3-only (88.4%) treatments. A comprehensive comparison of the seed germination characteristics according to the four treatments determined that a GA3 500 mg·L-1 pre-treatment, with the highest average GP, was ideally suited to the revitalization of long-term stored C. laxiflora seeds. Consequently, C. laxiflora stored at -18℃ for 16 years indicated strong vitality and could be regenerated by proper pre-treatment.

The studies on affinity between Miscanthus sinensis and other plants (Miscanthus sinensis와 타 식물과의 친근성에 관한 연구)

  • Althought allelopathy has been cleary established for Pinus densiflora by Lee-Monsi, the auther suspected that Miscanthus sinensis which grows vigrously in such Korean pine forests might also exhibit the phenomenon. A total of 33 species having an affinity in the Miscanthus group were recorded in the filed. Out of these, five species; Patrinia scabiosaefolia, Lespedeza crytobotrya. Oenothera odorata, Raphanus sativus var. acanthiformis, and Zoysia japonica were considered the "A" group. Five other species which have no affinity to Misanthus sinensis, Amaranthus patulus, Solanum nigrum, Capsella bursa-pastoris var. triangularis, Chemopodium album var. centrorubrum and Alopeculus amurensis were considered the "B" group. Extracts of Miscanthus sinensis were applied to determine relative germination and growth. It was found the growth in the "B" group was promoted by a cold water extract of Miscanthus stems and leaves, but inhibited remarkably by an extract obtained by passing cold water through Miscanthus root. As expected, growth in the "A" group was promoted by both experimental treatments.romoted by both experimental treatments.

  • This research was carried out to promote the seed germination of Styrax japonica, by several pretreatments and to examine its seedling growth and seed component variations according to time and pretreatment. The results are as follows ; 1. Styrax japonica seeds are usually germinated after more than one year cold stratification. However, when the seeds were stratificated for 5 months after dipped in the solution of 70% $H_2SO_4$ for 30 minutes and also treated with $GA_3$(3000ppm), they showed 55% germination. This demonstrated that the seeds could be germinated in one year when pretreated. 2. The growth of one-year-old seedlings after pretreatment was the same as that of two-year-old seedlings when compared 160 days after seeding. 3. Before seeding, one-year-old, pretreated seeds of Styrax japonica showed increased moisture content but decreased crude fat, carbohydrate and exude ash, which were similar to the components of two-year-old seeds.

Physiological and Ecological Studies on the Low Temperature Damages of Rice (Oryza sativia L.) (수도의 저온장해에 관한 생리 생태학적 연구)

  • Experiments were conducted to investigate rice varietal response to low water and air temperatures at different growth stages from 1975 to 1980 in a phytotron in Suweon and in a cold water nursery in Chooncheon. Germination ability, seedling growth, sterility of laspikelets, panicle exertion, discoloration of leaves, and delay of heading of recently developed indica/japonica cross(I/J), japonica, and indica varieties at low air temperature or cold water were compared to those at normal temperature or natural conditions. The results are summarized as follows: 1. Practically acceptable germination rate of 70% was obtained in 10 days after initiation of germination test at 15\circ_C for japonica varieties, but 15 days for IxJ varieties. Varietal differences in germination ability at suboptimal temperature was greatest at 16\circ_C for 6 days. 2. Cold injury of rice seedlings was most severe at the 3.0-and 3.5-leaf stage and it was reduced as growth stage advanced. A significant positive correlation was observed between cold injury at 3-leaf stage and 6-leaf stage. 3. At day/night temperatures of 15/10\circ_C seedlings of both japonica and I/J varieties were dead in 42 days. At 20/15\circ_C japonica varieties produced tillers actively, but tillering of I/J varieties was retarded a little. At 25/15\circ_C, both japonica and I/J varieties produced tillers most actively. Increase in plant height was proportional to the increase in all varieties. 4. In I/J varieties the number of differentiated panicle rachis branches and spikelets was reduced at a day-night temperature of 20-15\circ_C compared to 25-20 or 30-25\circ_C, but not in japonica varieties although panicle exertion was retarded at 20-15\circ_C. The number of spikelets was not correlated with the number of primary rachis branches, but positively correlated with that of secondary rachis branches. 5. Heading of rice varieties treated with 15\circ_C air temperature at meiotic stage was delayed compared to that at tillering stage by 1-3 days and heading was delayed as duration of low temperature treatment increased. 6. At cold water treatment of 17\circ_C from tillering to heading stage, heading of japonica, I/J, and cold tolerant indica varieties was delayed 2-6, 3-9, and 4-5 days, respectively, Growth stage sensitive to delay of heading delay at water treatment were tillering stage, meiotic stage, and booting tage in that order, delay of heading was greater in indica corssed japonica(Suweon 264), japonica(Suweon 235), and cold tolerant indica(Lengkwang) varieties in that order. Delay of heading due to cold water treatment was positively correlated with culm length reduction and spikelet sterility. 7. Elongation of culms and exertion of panicles of rice varieties treated with low air temperature 17\circ_C. Culm length reduction rate of tall varieties was lower than that of short statured varieties at low temperature. Panicle exertion was most severaly retarded with low temperature treatment at heading stage. Generally, retardation of panicle exertion of 1/1 varieties was more severe than that of japonica varieties at low temperature. There was a positive correlation between panicle exertion and culm length at low temperature. 8. The number of panicles was increased with cold water treatment at tillering stage, but reduced at meiotic stage. As time of cold water treatment was conducted at earlier growth stage, culm length was shorter and panicle exertion poorer. 9. Sterility of all rice varieties was negligible at 17\circ_C for three days but 30.3-85.2% of strility was observed for nine-day treatment at 17\circ_C. Among the tested varieties, sterility of Suweon 264 and Milyang 42 was highest and that of Suweon 290 and Suweon 287 was lowest. The most sensitive growth stage to low temperature induced sterility was from 15 to 5 days before heading. There was positive correlation between sterility of rice plants treated with low temperature at meiotic and heading stage. 10. Percentage of spikelet sterility was greatest at cold water treatment at meiotic stage (auricle distance -15~-10cm) and it was higher in 1/1 (Suweon 264, Joseng tongil), japonica (Nongbaek, Towada), and cold tolerance indica(Lengkwang) varieties in the order. Level of cold water and position of young-ear affected on the sterility of varieties at meiotic stage; percentage of spikelet sterility of variety, Lengkwang, of which young-ear was located above the cold water level was high, but that of short statured variety, Suweon 264, of which young-ear was located in the cold water was lower. 11. Percentage of ripened grains was not reducted at 15\circ_C air temperature for three days at full heading stage in all varieties. However, at six-day low temperature treatment Suweon 287, Suweon 264 showed percentage of ripended grains lower than 60%, but at nine-day low temperature treatment all varieties showed percentage of ripened grains lower than 60%. Low temperature treatment of 17\circ_C from 10 days after heading for 20 days did not affect on the ripening of all varieties. 12. Uptake of nitrogen, phosphorous, potassium, calcium, and magnesium in whole plants was higher at average air temperature of 25\circ_C, but concentration of the elements was lower compared to those at 19\circ_C. However, both total uptake and concentration of manganese were higher at 19\circ_C compared to 25\circ_C. 13. Higher application of nitrogen, phosphorus, silicate, and compost increased yield of rice due to increased number of panicles and spike let fertility in cold water irrigated paddy.

  • Allelopathic activity of the volatiles from leaves of Perilla frutescens an Artemisia asiatica was determined on the basis of bioassay, which tested germination and seedling growth of radish, rice, mung bean and lettuce. Seedling growth was more inhibited by phytotoxic volatiles than germination. Volatile components collected by headspace cold trapping-Tenax GC adsorption were analyzed by GC-MS. Fifteen volatile components in P. frutescens and 15 components in A. asiatica were identified. By steam distillation-extraction, 4 flavor components in P. frutescens and 10 components in A. asiatica were identified. The inhibitory activity of the fractions, obtained by steam distillation-extraction, was determined by virtue of bioassay on radish. Volatile allelochemicals of the most active fraction, neutral fraction, isolated from P. frutescens contained 9 components. In A. asiatica, 24 volatile allelochemicals were identified.

  • In order to determine the mechanism of cold tolerance in crops, changes in biochemical factors related with the biological reduction of molecular oxygen upon cold shock treatment were analyzed at an early stage of Brassica germination. As the cold shocked seedlings were recovered under the normal growth condition for 24 hours, the peroxidase activities in cold sensitive rape(B. napus) and cold tolerant 'Sandongchae'(B. campestris) were considerably increased by 33% and 87% in root fraction and, 84% and 206% in hypocotyl, respectively. The content of superoxide($H_2O_2$) in hypocotyl fraction was dramatically accumulated until 8 hours after recovery and then gradually decreased. The extent of superoxide accumulation was severer in B. napus than B. campestris. At 24 hours after cold shock, $H_2O_2$ content was decreased to the nearly control level in B. campestris but still remained by 38%, in E. napus. Even though $H_2O_2$ content in hypocotyl fraction was decreased only 2% in B. napus during cold shock, while in B. campestris it was severely decreased about 15%. On the other hand, the cold shock at 3 days after Uniconazole treatment was more effective in increase of peroxidase activity than each separate treatment.
