  • In order to screen cold tolerant soybean lines, germination at various temperature, and emergence and seedling height at 1$0^{\circ}C$ were investigated. Since the most conspicuous varietal difference of the germination speed was observed at 1$0^{\circ}C$, the germination test at 1$0^{\circ}C$ would be effective in screening cold tolerant lines.

  • Rose seed shows low germination percentages (about 20%) because it has a high amount of substances that inhibit germination in the pericarp. We investigated the effect of orientation of achene, the day after pollination (DAP), and cold storage with or without half-cut in achene for the germination percentage in R. rugosa. Germination percentages of intact or half-cut achenes were investigated in a 16-hour photoperiod at $25^{\circ}C$ room on basal MS medium for two weeks. In germination percentage, maximum 100% was measured within one week when half-cut achenes were cultured on an orientation that the embryos facing to the light. Half-cut achenes at 90 DAP were germinated 100% regardless of cold storage. Various LED lights (red, blue, yellow, green, and white) were illuminated over the half-cut achenes to gain the effect of light color. Germination percentage of R. rugosa seeds under blue LED reached the greatest with 90% within one week of culture and these seedlings were the best with a steady growth rate. It is concluded that half-cut achenes would be an effective method to improve seed germination in R. rugosa without stratification or scarification. This system could be applied to breeding studies in rose cultivars.

  • Pollen-derived embryos cultured on the hormone-free medium showed a low germination frequency (12.5%) and poor growth response after germination. The greatest frequency of germination (81.3%) was obtained from the embryos cultured on medium with 0.3mg/L GA$_3$.The greatest frequency of generation (81.3) was obtained from embryos cultured on medium with 0.3mg/L GA$_3$. The embryos precultured for 20 days on medium with 0.3mg/L GA$_3$were transferred to the medium with various combination of hormones such as IAA, kinetin, zeatin, 6-benzylaminopurin (BA) and Gh$_3$. The germination frequency of cotyledonary stage embryos showed above 72% on media with all of the hormonal combinations, but the embryos germinated on medium with 2mg/L BA or 0.1mg/L kinetin and 0.3mg/L GA$_3$ developed more vigorously into plantlets than those of other hormonal combinations. Torpedo-stage embryos cultured on medium with 0.3 mg/L Gh$_3$ were pretreated for 8 weeks at 2-week intervals at 4$^{\circ}C$, The germination frequency of the cold-preheated embryos increased with the increment of pretreatment period from 2 to 8 weeks. The greatest frequency of germination (73.3%) was obtained from the embryos pretreated for 8 weeks at 4$^{\circ}C$. The chromosomes of the root-tip cells of W plane grown for 40 days after germination were observed. Most of the regenerated plants were haploid (55.8%) or diploid (315%), but triploid (1.3%), tetraploid (5.2%), or aneuploid (6.5%) were also detected among them.

  • This research was conducted to develop a seed propagation protocol for the use of Broussonetia kazinoki Siebold ex Siebold & Zucc, which native to East Asia. Light and temperature conditions, GA3 treatment, and low-temperature stratification were performed to evaluate the germination characteristics. The germination percentage at 4, 15/6, 20/10 and 25/15℃ was 0, 42. 5, 44. 4 and 91.7%, respectively. Thus, the optimal germination temperature of the B. kazinoki seeds was 25/15℃. Although germination was delayed by relatively low temperature conditions, GA3 treatment allowed for increased germination rates even under the low temperature conditions. Cold stratification reduced time to germination. Relatively low germination percentages at 20/10℃ and 15/6℃ indicated that the seeds had been under the state of conditional dormancy. Through this study, B. kazinoki seeds have conditional dormancy because they can be germinated through GA3 treatment even under relatively low temperature conditions and it is judged that it will support the commercial propagation practice in the future.

Studies and on relationship between Amylase activities winter hardiness of germinating seeds in winterwheat varieties (소맥품종에 있어서 발아종자의 Amylase 활력과 내한성에 관한 연구)

  • The studies were conducted to know the relationship between $\beta$-amlyase activities and hardiness for the germinated seedlings of, winter wheat varieties which were classified with eye estimated cold resistance in field as, susceptible, moderate and resistant. These varieties were tested in continued five days from germination in four replicated split plot design. For the measurement of $\beta$-amylase, improved A. K. Balls method (2) was employed. Result obtained will be summarized as follows. 1. Tested varieties showed highly significant differences in $\beta$-amylase activity, while no differences were obtained between dates after germination. 2. Winter hardy varieties, Yukseung #3, Chin Kwang and Suwon#85 showed higher amylase activities than the moderate hardy varieties, Jukdalma, Kangdosinryuk and Norin #4, while lower activities were measured in susceptible varieties, Norin #6, Kangdo and Norin#12. 3. With measurement of $\beta$-amylase activity, rurther detail classification to cold resistance is seemed available than eye-estimating in the field condition. 4. In accordance with testing dates, amylase activities were not so clear on 1st, 2nd, 4th and 5th days from germination, while clear differences were found on 3rd day from germination. 5. Amylase activity obtained on 3rd day after germination is considered easy and effective method to estimate cold resistance of wheat varieties with a classification standard.

Effects of $GA_3$ on Seed Germination and Seedling Survival Rate of Acanthopanax senticosus Maxim. (가시오갈피의 $GA_3$처리에 따른 종자발아와 유묘생존)

  • This experiment was carried out to investigate the effects of $GA_{3}$ and cold stratification as presown treatments on seed germination, seedling emergence and final survival rate of Acanthopanax senticosus Maxim. Seeds collected 145 days after-ripening period followed by 10 days of cold stratification was effective in promoting germination. Dehisced seeds treated with 500 ppm of $GA_{3}$ for 3 days was also effective in promoting germination. However, seedling emergence rate remained low in both treatment. Seedling emergence rate was higher for seeds germinated in Heungnong-Bio and Klasman-Bio than in vermiculite, perlite, vermiculite-perlite mixture, or sand. After 40 days of cold stratification, seedling emergence was significantly higher in the 500 ppm $GA_{3}$ treatment than nontreatment for both dehisced and non-dehisced seeds. However, for dehisced seeds, $GA_{3}$ treatment before sowing resulted in decreased final seedling survival rate.

Dormancy-breaking and Germination of Chelidonium majus L. subsp. asiaticum H. Hara Seeds by Stratification and Gibberellins

  • The demand for Chelidonium majus L. subsp. asiaticum H. Hara is expected to increase due to its pharmacological properties such as antibacterial, antioxidant, and anti-inflammatory effects. However, an effective propagation system for this species has not yet been established. This study was conducted to analyze the seed dormancy and germination characteristics of C. majus L. subsp. asiaticum H. Hara native to Korea and establish a mass propagation system. The dormancy type was primarily classified by analyzing the general information of the collected seeds. The seed dormancy breaking was investigated by comparing the effects of cold stratification (0, 2, 4, 8, 10, or 12 weeks) with warm stratification (S, summer temperature, 25/15℃) and intermediate temperature stratification (A, autumn temperature, 15/10℃) of alternating temperature stratification (S12-A4 or S12-A8 weeks). After dormancy break, 500 mg/L GA3 and GA4+7 treatment replaced cold stratification and improved seed germination. The results of this study are expected to provide basic data for future seed propagation and mass propagation by analyzing the dormancy and germination characteristics of C. majus L. subsp. asiaticum H. Hara seeds.

Inhibition of Incidence of Fungi in Cold Storage Room by Acetic Acid (Acetic acid에 의한 저온저장고 내의 균발생 억제 효과)

  • Postharvest diseases developed on harvested products result in a great economic loss. The objective of this research was to develop a chemical treatment to control major postharvest pathogens including Botrytis cinerea and Penicillium expansum in the cold storage room. Acetic acid ($2.4mg{\cdot}L^{-1}$, $4.8mg{\cdot}L^{-1}$) inhibits spore germination and mycelial growth of B. cinerea and P. expansum on PDA at room temperature ($25^{\circ}C$) and low temperature ($2^{\circ}C$). Fumigation of cold storage room with $SO_2$ ($5g{\cdot}m^{-3}$) gas or gaseous acetic acid ($4.8g{\cdot}m^{-3}$) prior to operation greatly reduced population of fungi in the cold storage room.

Influence of Rainfall on Germination of Malting Barley at Harvesting Season (수확기의 강우가 맥주보리 발아에 미치는 영향)

  • In order to find out the effect of rainfall at harvesting season on germination of malting barley, the seeds sampled from Cheju island and southern part of Korea were examined. The germination rate of seeds from Cheju island where rainfall was frequent at harvesting season, ranged from 46 to 71%, even though disease infection of the seeds was not that serious as would be expected. High sugar content of seed was resulted from the degradation of carbohydrates during the harvest season. From TZ test the rainfall - affected seeds were found to be highly viable but in the state of secondary dormancy. Results of cold germination test showed that the seeds were recovered from the secondary dormancy. Therefore, it was recommended that the seeds affected by the rainfall at harvesting season should be utilized after 12 months when the dormancy period terminated. The germination percent of the seeds was significantly enhanced by prechilling and / or 1 ppm gibberellic acid treatment. Different seed lots showed different rate of germination and the degree of dormancy.

  • This experiment was conducted to investigate the effects of temperature, light, seed harvesting time, storage temperature, and period on its germination of Astragalus membranaceus. Optimum germination temperature was ${20{\sim}25^{\circ}C}$ and it was found to be light insensitive seed. The germination rates were not significantly different between 1-year and 2-year old plants, and seed harvesting times. Seeds stored at such cold temperatures as ${-4^{\circ}C\;and\;-20^{\circ}C}$ for 15 months showed over 80% in germination rate at the 6th day after placement. On the other hand, low germination rate of 60% at the 11th day was observed from the seeds stored at $4^{\circ}$ and room temperature.
