This study was made to evaluate the temperature effects on anaerobic digestion of swine manure. A laboratory single-stage, high-rate, anaerobic digester was operated at 10, 20 and 30 day's HRT at the temperature of 35$\circ$C or 55$\circ$C. The conclusions from this study are as follows: (1) COD and BOD reductions were similar in both the mesophilic and thermophilic digestions. (2) With thermophilic digestion, volatile reduction increased to 67%, as compared with 60% of mesophilic digestion. With thermophilic digestion, the pH increased to 8.5 as compared with 8.0 of mesophilic digestion. With thermophilic digestion, the concentration of volatile acid increased to 763 mg/l, as compared with 250 mg/l of mesophilic digestion. While the gas was produced by mesophilic digestion at 0.74m$^3$/kg of VS fed, it increased to 0.87 m$^3$/kg VS fed by thermophilic digestion. The refractory VS was about 25% of the infiuent VS.
In this study, two types of reactors were operated to examine the properties of methanol uptake under the high-rate denitrification process. In a sequencing batch reactor, the denitrifying activity was enriched up to 0.80 g-N/g-VSS-day for 72 days. Then, the enriched denitrifying sludge was transferred to a completely stirred tank reactor (CSTR). At the final phase on Day 46-50, the nitrogen removal efficiency was around 100% and the total nitrogen removal rate reached 0.097±0.003 kg-N/㎥-day. During the continuous process, the sludge settling index (SVI30) was stabilized as 118.3 mL/g with the biomass concentration of 1,607 mg/L. The continuous denitrifying process was accelerated by using a sequencing batch reactor (SBR) with a total nitrogen removal rate of 0.403±0.029 kg-N/㎥-day with a high biomass concentration of 8,433 mg-VSS/L. Because the reactor was open to ambient air with the dissolved oxygen range of 0.2-0.5 mg-O2/L, an increased organic carbon requirement of 5.58±0.70 COD/NO3--N was shown for the SBR in comparison to the value of 4.13±0.94 for the test of the same biomass in a completely anaerobic batch reactor. The molecular analysis based on the 16S rRNA gene showed that Methyloversatilis discipulorum and Hyphomicrobium zavarzinii were the responsible denitrifiers with the sole organic carbon source of methanol.

Background: Airway mucus hypersecretion plays an important role in the pathogenesis of asthma, and is associated with the induction of MUC5AC expression in airway secretion. The MUC5AC gene is highly polymorphic; however, there are few studies about the association between the polymorphisms of the MUC5AC gene and asthma susceptibility or asthma phenotypes. We have investigated the association of MUC5AC promoter polymorphisms with the risk of asthma and asthma phenotypes. Methods: We determined the genotypes of the MUC5AC promoter (-1274G>A) in 78 asthma patients and in 78 age, sex-matched control individuals in the Korean population. Genomic DNAs from blood were analyzed by PCR and RFLP (restriction fragment length polymorphism) determination. We examined $FEV_1$, total eosinophil count, serum IgE level, $PC_{20}$ and the presence of atopy (by a skin test) in asthma patients. Results: The mean age of the patients was $47.7{\pm}16.1$ years and 38.5% were men, and the mean $FEV_1$ was $84.4{\pm}22.3%$ of predicted in the asthma patients. The -1274G>A polymorphism of the MUC5AC promoter in asthma patients was not significantly different as compared with normal individuals (GG 57.7%, AG 34.6% and AA 7.7% in asthma patients vs. GG 56.4%, AG 38.5% and AA 5.1% in control subject, p = 0.752, Cod). Several clinical parameters in asthma patients such as $FEV_1$, total eosinophil count, serum IgE level, $PC_{20}$ and the presence of atopy, were not associated with the -1274G>A polymorphism of the MUC5AC promoter. Conclusion: The -1274G>A single nucleotide polymorphism (SNP) frequency of the MUC5AC promoter was not associated with asthma in a Korean population.
The runoff characteristics of non-point source pollutions in the municipal area of Jeonju were investigated and analyzed by using the SWMM (Storm Water Management Model). The flow rates and water qualities of runoff from two types of drainage conduits were measured respectively. One was a conventional combined sewer system and the other was a separated sewer system constructed recently From August to November in 2004, investigations on two rainfall events were performed and flow rate, pH, BOD, COD, SS, T-N and T-P were measured. These data were also used for model calibration. On the basis of the measured data and the simulation results by SWMM, it is reported that $80-90\%$ of pollution load is discharged in the early-stage storm runoff. Therefore, initial 10-30 mm of rainfall should be controlled effectively for the optimal treatment of non-point source pollution in urban area. Also, it was shown that the SWMM model was suitable for the management of non-point source pollution in the urban area and for the analysis of runoff characteristics of pollutant loads.
An autotrophic denitrification reactor (ADR-l) and a heterotrophic denitrification reactor (HDR-2) were operated to remove nitrate and nitrite in an anoxic environment in raw sewage. The $NO_3$-N removal rate of ADR-l was shown to range from 52.8% to 78.7%, which was higher than the $NO_3$-N removal rate of HDR-2. Specific denitrification rates (SDNR) of ADR-l and HDR-2 were 3.0 to 4.0 and 1.1 to $1.2\;mgNO_3$-N/gVSS/h, respectively. From results of restriction fragment length polymorphism (RFLP) of the 16S rRNA gene, Aquaspirillum metamorphum, Alcaligenes defragrans, and Azoarcus sp. were $\beta$-Proteobacteria that are affiliated with denitritying bacteria in the ADR-l. Specifically, Thiobacillus denitrificans was detected as an autotrophic denitrification bacteria. In HDR-2, the $\beta$-Proteobacteria such as Denitritying-Fe-oxidizing bacteria, Alcaligenes defragrans, Acidovorax sp., Azoarcus denitrificans, and Aquaspirillum metamorphum were the main bacteria related to denitrifying bacteria. The $\beta$-and $\alpha$-Proteobacteria were the important bacterial groups in ADR-l, whereas the $\beta$-Proteobacteria were the main bacterial group in HDR-2 based on results of fluorescent in situ hybridization (FISH). The number of Thiobacillus denitrificans increased in ADR-l during the operation period but not in HRD-2. Overall, the data presented here demonstrate that many heterotrophic denitritying bacteria coexisted with autotrophic denitrifying bacteria such as Thiobacillus denitrificans for nitrate removal in ADR-l. On the other hand, only heterotrophic denitritying bacteria were identified as dominant bacterial groups in HDR-2. Our research may provide a foundation for the complete nitrate removal in raw sewage of low-COD concentration under anoxic condition without any external organic carbon or the requirement of post-treatment.
The free-living amoeba and Acanthamoeba sp. are widely distributed in fresh water, soil, air and dust in the world. We studied distribution of amoeba from low Nakdong River(Mulgum and Maeri) and removal efficiency in water treatment process of Busan metropolitan city. During this investigation, water quality showed pH $7.4{\sim}9.6({\pm}1.1)$, water temperature $2.0{\sim}29.0({\pm}17)^{\circ}C$, turbidity $4.8{\sim}27.4({\pm}11.0)$ NTU, chlorophyll-a $10.3{\sim}109.0({\pm}44.3)\;mg/m^{3}$, BOD $1.7\sim4.9({\pm}2.6)$ mg/L, COD $3.1\sim-6.9({\pm}5.0)$ mg/L and total coliform $17\sim920({\pm}200.5)$ MPN/100 mL. The free-living amoeba were detected highly than Acanthamoeba sp., 11 out of 22 in raw water samples were positive (50%) for Acanthamoeba sp. from February 2005 to December 2005. The seasonal characteristics of tree-living amoeba and Acanthamoeba sp. in raw water were mainly distributed through the spring to the early fall. When tree-living amoeba and Acanthamoeba sp. were passed through the water treatment of pilot-plant, approximately 80% was sure to be removed through pre-ozonation, sedimentation, send filtration. 100% was removed after post-ozonation process. All of the isolated amoebas from Nakdong River were Acanthamoeba sp. AC311 18S ribosomal RNA gene with 98% nucleotide sequence homology.
The purpose of this study is to explore the ecosystem service and benefit indicators of natural seaweed beds. Ecosystems of natural seaweed beds provide a wide range of services and benefits to human society including provisioning services, regulating services, supporting services, and cultural services. Indicators for each of the ecosystem services are chosen by marine plants ecologists and as follows. Ecosystem indicators of natural seaweed beds for provisioning services are well-being food(amount of seaweed harvested/amount of fish landed, fish biomass, area of natural seaweed beds, the number of species, contribution to the second production), raw materials(amount of biomass by breed, amount of aquaculture feed), genetic resources(amount of genetic material extracted, amount of genetic material contained by age and habitat), and medicinal resources(amount of medicinal material extracted). Ecosystem indicators of natural seaweed beds for regulating services are air purification(amount of fine dust/NOx or $SO_2$ captured), climate regulation(amount of $CO_2$ sequestered), waste treatment(amount of N, P stored, biochemical degradation capacity COD), and costal erosion prevention(length and change of natural coast line, amount of sediment prevented). Ecosystem indicators of natural seaweed beds for supporting services are lifecycle and maintenance(primary production, contribution to the second production) and gene pool protection(amount of compositional factors in ecosystem, introduced species). Ecosystem indicators of natural seaweed beds for cultural services are recreation and tourism(the number of visits of an area) and information for cognitive development(amount of time spent in education, research and individual learning about ecosystem of natural seaweed beds).
This study aimed to assess the impact of livestock excreta discharged from an Intensive Livestock Farming Area (ILFA) on river water quality during a rainfall event. The Bangcho River, which is one of the 7 tributaries in the Cheongmi River watershed, was the study site. The Cheongmi River watershed is the second largest area for livestock excreta discharge in Korea. Our results clearly showed that, during the rainfall event, the water quality of the Bangcho River was severely deteriorated due to the COD, $NH_4-N$, T-N, $PO_4-P$, T-P, and heavy metals (Cu, Zn, and Mn) in the run-off from nearby farmlands, where the soil comprised composted manure and unmanaged livestock excreta. In addition, stable isotope analysis revealed that most of nitrogen ($NH_4-N$ and $NO_3-N$) in the run-off was from the ammonium and nitrate in the livestock excreta. The values of ${\delta}^{15}N_{NH4}$ and ${\delta}^{15}N_{NO3}$ for the Bangcho River water sample, which was obtained from the downstream of mixing zone for run-off water, were lower than those for the run-off water. This indicates that there were other nitrogen sources upstream river in the river. It was assumed from ${\delta}^{15}N_{NH4}$ and ${\delta}^{15}N_{NO3}$ stable isotope analyses that these other nitrogen sources were naturally occurring soil nitrogen, nitrogen from chemical fertilizers, sewage, and livestock excreta. Therefore, the use of physicochemical characteristics and nitrogen stable isotopes in the water quality impact assessment enabled more effective analysis of nitrogen pollution from an ILFA during rainfall events.
