Efficient Labeling of Porcine Hematopoietic Cells by Fluorescence-Conjugated Nanoparticles

  • Nanotechnology is currently receiving considerable attention in various fields of biotechnology. The uptake of nanoparticles by cells for labeling and tracking is a critical process for many biomedical therapeutic applications. However, nanoparticle labeling of porcine hematopoietic cells has not been demonstrated so far. In the present study, silica-coated nanoparticles conjugated with rhodamine B isothiocyanate (SR-RITC) were used to investigate the uptake of nanoparticles by porcine hematopoietic cells. Flow cytometric and confocal microscopic analyses reveled that the cells were efficiently internalized by the silica-coated nanoparticles. Furthermore, biocompatibility tests demonstrated that the SR nanoparticles were not cytotoxic, and they had no impact on proliferation. Our study demonstrates that silica-coated nanoparticles are taken up very rapidly and with high efficiency into porcine hematopoietic cells, with no apparent deleterious effects. Therefore, silica-coated nanoparticles appear to be a promising tool for tracking porcine hematopoietic cells.

A Study on the Life Enhancement of TiN Coated Drill (TiN 박막을 코팅한 드릴의 수명향상에 관한 연구)

  • Recently, various film coated insert tools have been used in order to improve tool life by several different vapor deposition or chemical vapor deposition. Especially, TiN coated drills have been broadly studied because of improving drill performance in terms of drill life, work quality and its brilliant color. Nevertheless, because of the poor adhesion between TiN film and drill, it was difficult to attain the better drill performance. Therefore, to improve adhesion of TiN films, we sputtered titanium as interlayer prior to TiN deposition on drill by PECVD(Plasma Enhanced Chemical Vapor Deposition). The results indicate that Ti/TiN coated drills achieve about 2.6 times life improvement, while TiN coated drills only 2 times. Wear characteristics of tested drills were examined using SEM, and the results were correlated with drill life and roughness of drilled holes.

A study on the Cutting Force Variation Comparison between Low CBN and Coated Low CBN Tools in Turning of SCM440 (Low CBN 코팅공구의 SCM440 선삭시 절삭력변화에 관한 연구)

  • In recent years, high hardness steel is used for most of the material in many areas including aircraft, nuclear power, space exploration and automotive parts. Low CBN tools are widely used in industrial field which can effectively process high hardness steel of HRC 45 or harder. The results of this study demonstrated, when high hardness steel, SCM440 is turned with Low CBN tools coated with TiN and TiAlN coatings respectively, that both the thrust force and cutting force tends to increase with more increase in cutting force than thrust force, as the feed rate increases at constant cutting speed. In addition, the size of the cutting force and thrust force does not change with the increased cutting speed at the same feed rate, but the tool life is reduced if the cutting speed is increased to shorten the machining time. Therefore, it is recommended to limit the cutting speed at 250 m/min maximum or less. Furthermore, comparing the cutting force of the three tools at the same cutting condition, Tin coating tool showed the smallest cutting force and Low CBN was the next, and the TiAlN coating tools showed the largest cutting force.

  • Inconel 718 alloy has been applied to high temperature, high load and corrosion resistant environments due to its superior properties. However, This alloy is a difficult-to-cut nickel-based superalloy and the chipping or notch wear is mainly generated on the cutting edge of the tool. In this study, the machinability of Inconel 718 is investigated to improve tool life under various cutting conditions with TiCN-based coated ball-end mills. The cutting conditions can be suggested to improve both the tool life and machined surface quality in Inconel 718 high speed machining.

  • The ideal surface roughness is obtained by tool geometry and feed rate in face milling. however actual surface roughness is affected by various factors such as cutting conditions. vibration and used tool. To improve the quality and productivity of the machining parts, lots of research on the evaluation of tool life and control of surface roughness has been required. Therefore, the width of flank wear, cutting force, and surface roughness are monitored to analyse the characteristics of surface roughness. This experimental investigation is mainly focused on the characteristics of surface roughness in multi-insert milling using TiN coated tool.

  • Recently, a wide range of industrial production area has a competitive advantage through cost reduction. Moreover with the development of industrial technology, base material and cutting tool help the machining technology. But most of the machining enterprises have not hold the R&D facilities and human resources. This mainly disturbs the industrial development and th increase of production efficiency. Especially in the interrupted machining process, it showed different behavior with continuous machining process. So it needs to research and develop the tool life and tool wear mechanism analysis.

  • Nickel-based heat resisting alloys are commonly used for high temperature application such as in aircraft engines and gas turbines. In this paper, the machinability of Nickel-based heat resisting alloys was investigated with respect to the wear and the chip formation by tool type and cutting condition. Relationship between three types of tool and chip formation was experimentally investigated. Among the three types of tool tested, coated tools(CVD, PVD) ara available for the difficult-to-cut-materials such as Nickel-based heat resisting alloys and etc.

  • In this paper present a new technique (strain-telemetering)for detection of coated tool failure in face milling processes. In the cutter body the strain signals received fro the transmitter is transformed in to frequency modulation(FM) signals in face milling processes. A receiver which is place near by the Vertical milling machine receives the FM signals, then the signals will be sent to a computer which determines whether th tool is failure. And machined surface of workpiece is detected by the SEM. In this paper, A on-line monitoring of the tool failure detection system based on the strain -telemetering apparatus has bee represented.

  • Generally, automotive cylinder head is manufactured by FCD45 material that is hard to cutting materials, and it needs the accurate machining tool fir higher output of engine. This paper is concerned with a study on TiN coated 6 edges reamer for improving machining accuracy of cylinder head guide pin. The reamer shape is changed from 4 edges to 6 edges for the improvement of machining accuracy. Also, TiN coating is applied to the improvement of surface roughness of cylinder head guide pin and tool life of 6 edges reamer. It is noted that 6 edges reamer are effective in controlling the dimensional accuracy and surface roughness as well as increasing tool life.
