해조의 식용분말화에 관한 연구 (A Study on the Nutritive Value and Utilization of Powdered Seaweeds)

  • I. Subject of the study A study on the nutritive value and utilization of powdered seaweeds. II. Purpose and Importance of the study A. In Korea the shortage of food will be inevitable by the rapidly growing population. It will be very important study to develop a new food from the seaweeds which were not used hitherto for human consumption. B. The several kinds of seaweeds have been used by man in Korea mainly as side-dishes. However, a properly powdered seaweed will enable itself to be a good supplement or mixture to certain cereal flours. C. By adding the powdered seaweed to any cereals which have long been staple foods in this country the two fold benefits; saving of cereals and change of dietary pattern, will be secured. III. Objects and scope of the study A. Objects of the study The objects will come under four items. 1. To develop a powdered seaweed as a new food from the seaweeds which have been not used for human consumption. 2. To evaluate the nutritional quality of the products the analysis for chemical composition and animal feeding experiment will be conducted. 3. Experimental cocking and accepability test will be conducted for the powdered products to evaluate the value as food stuff. 4. Sanitary test and also economical analysis will be conducted for the powdered products. B. Scope of the study 1. Production of seaweed powders Sargassum fulvellum growing in eastern coast and Sargassum patens C.A. in southern coast were used as the material for the powders. These algae, which have been not used for human consumption, were pulverized through the processes of washing, drying, pulverization, etc. 2. Nutritional experiments a. Chemical composition Proximate components (water, protein, fat, cellulose, sugar, ash, salt), minerals (calcium, phosphorus, iron, iodine), vitamins (A, $B_1,\;B_2$ niacin, C) and amino acids were analyzed for the seaweed powders. b. Animal feeding experiment Weaning 160 rats (80 male and 80 female rats) were used as experimental animals, dividing them into 16 groups, 10 rats each group. Each group was fed for 12 weeks on cereal diet (Wheat flour, rice powder, barley powder, potato powder, corn flour) with the supplementary levels of 5%, 10%, 15%, 20% and 30% of the seaweed powder. After the feeding the growth, feed efficiency ratio, protain efficiency ratio and ,organs weights were checked and urine analysis, feces analysis and serum analysis were also conducted. 3. Experimental cooking and acceptability test a. Several basic studies were conducted to find the characteristics of the seaweed powder. b. 17 kinds of Korean dishes and 9 kinds of foreign dishes were prepared with cereal flours (wheat, rice, barley, potato, corn) with the supplementary levels of 5%, 10%, 15%, 20% and 30% of the seaweed powder. c. Acceptability test for the dishes was conducted according to plank's Form. 4. Sanitary test The heavy metals (Cd, Pb, As, Hg) in the seaweed powders were determined. 5. Economical analysis The retail price of the seaweed powder was compared with those of other cereals in the market. And also economical analysis was made from the nutritional point of view, calculating the body weight gained in grams per unit price of each feeding diet. IV. Results of the study and the suggestion for application A. Chemical composition 1. There is no any big difference in proximate components between powders of Sargassum fulvellum in eastern coast and Sargassum patens C.A. in southern coast. Seasonal difference is also not significant. Higher levels of protein, cellulose, ash and salt were found in the powders compared with common cereal foods. 2. The levels of calcium (Ca) and iron (Fe) in the powders were significantly higher than common cereal foods and also rich in iodine (I). Existence of vitamin A and vitamin C in the Powders is different point from cereal foods. Vitamin $B_1\;and\;B_2$ are also relatively rich in the powders.'Vitamin A in ·Sargassum fulvellum is high and the levels of some minerals and vitamins are seemed4 to be some influenced by seasons. 3. In the amino acid composition methionine, isoleucine, Iysine and valine are limiting amino acids. The protein qualities of Sargassum fulvellum and Sargassum patens C.A. are seemed to be .almost same and generally ·good. Seasonal difference in amino acid composition was found. B. Animal feeding experiment 1. The best growth was found at.10% supplemental level of the seaweed Powder and lower growth rate was shown at 30% level. 2. It was shown that 15% supplemental level of the Seaweed powder seems to fulfil, to some extent the mineral requirement of the animals. 3. No any changes were found in organs development except that, in kidney, there found decreasing in weight by increasing the supplemental level of the seaweed powder. 4. There is no any significant changes in nitrogen retention, serum cholesterol, serum calcium and urinary calcium in each supplemental level of the seaweed powder. 5. In animal feeding experiment it was concluded that $5%{\sim}15%$ levels supplementation of the seaweed powder are possible. C. Experimental cooking and acceptability test 1. The seaweed powder showed to be utilized more excellently in foreign cookings than in Korean cookings. Higher supplemental level of seaweed was passible in foreign cookings. 2. Hae-Jo-Kang and Jeon-Byung were more excellent than Song-Pyun, wheat cake, Soo-Je-Bee and wheat noodle. Hae-Je-Kang was excellent in its quality even as high as 5% supplemental level. 3. The higher levels of supplementation were used the more sticky cooking products were obtained. Song-Pyun and wheat cake were palatable and lustrous in 2% supplementation level. 4. In drop cookie the higher levels of supplementation, the more crisp product was obtained, compared with other cookies. 5. Corn cake, thin rice gruel, rice gruel and potato Jeon-Byung were more excellent in their quality than potato Man-Doo and potato noodle. Corn cake, thin rice gruel and rice gruel were excellent even as high as 5% supplementation level. 6. In several cooking Porducts some seaweed-oder was perceived in case of 3% or more levels of supplementation. This may be much diminished by the use of proper condiments. D. Sanitary test It seems that there is no any heavy metals (Cd, Pb, As, Hg) problem in these seaweed Powders in case these Powders are used as supplements to any cereal flours E. Economical analysis The price of the seaweed powder is lower than those of other cereals and that may be more lowered when mass production of the seaweed powder is made in future. The supplement of the seaweed powder to any cereals is also economical with the criterion of animal growth rate. F. It is recommended that these seaweed powders should be developed and used as supplement to any cereal flours or used as other food material. By doing so, both saving of cereals and improvement of individual's nutrition will greatly be achieved. It is also recommended that the feeding experiment for men would be conducted in future.

한국 남해안에 서식하는 백합과 5종의 근육 및 내장의 일반성분 조성 (Proximate Composition in the Muscle and Viscera of Five Veneridae Clams (Bivalvia) from Southern Coast of Korea)

  • 백합과 5종에 속하는 유용패류인 살조개, 바지락, 백합, 개조개 및 가무락조개의 체성분에 따른 유사 종간의 근육과 내장에 따른 일반성분 조성을 분석하였다. 분석 결과 근육과 내장의 일반성분의 경우 수분, 조단백질 및 탄수화물은 근육부위에서 내장부에 비해 전체적으로 높은 값을 나타났다. 이러한 결과는 조지방과 탄수화물을 제외한 다른 항목에서는 유의한 차이를 나타내지 않았다. 조지방의 경우 백합 내장에서 $3.67{\pm}0.57%$로 높게 나타났으며, 바지락 근육에서 가장 낮은 $2.50{\pm}1.40%$를 나타내었다. 탄수화물의 경우 백합의 내장에서 $3.61{\pm}0.45%$ 나타났으며, 바지락과 살조개 내장에서 $3.45{\pm}1.4%$$3.39{\pm}0.07%$로 높게 함유하고 있었다.

Prevalence and Characterization of Typical Aeromonas salmonicida Chum Salmon Isolates in Korea

  • Aeromonas salmonicida is an important fish pathogen commonly associated with furunculosis in salmonids. Typical A. salmonicida strains have the surface virulence A-layer protein, a major virulence determinant encoded by the vapA gene. In this study, 880 chum salmon Oncorhynchus keta were collected from the east coast of Korea during 2006-2011, including 560 wild adults and 320 artificially hatched fry pools, and the presence of typical A. salmonicida was examined by PCR using the typical A. salmonicida-specific vapA gene primers. The results demonstrated that 34.5% of the samples (304/880 samples) were PCR positive, implying that a typical A. salmonicida infection is highly prevalent among chum salmon in Korea. Twenty typical A. salmonicida isolates were recovered based on their brown pigmentation on Trypticase Soy Agar (TSA) plates, which indicates the existence of the A-layer protein. Further biochemical analyses with the four randomly selected typical A. salmonicida isolates revealed some variations in their amino acid decarboxylation and carbohydrate fermentation activity. A phylogenetic analysis based on the entire vapA gene sequence suggested that the A. salmonicida isolates from chum salmon were clustered with those isolated from Atlantic salmon in Europe. Further study is needed to resolve such an interesting relationship in detail.

수치모델을 이용한 고성만 양식 참굴의 연구 -I. 먹이가 참굴의 성장에 미치는 영향- (Modeling Oyster Populations Dynamics -I. Effect of Available Food on Growth of the Pacific Oyster Crassostrea gigas in Goseong Bay, Korea-)

  • Effects of available food on growth of commercially cultured Pacific oysters, Crassostrea gigas in Goseong Bay on the south coast were studied using a numerical model. levels of total protein, carbohydrate and lipid in particulate organic matter in the water column as well as chlorophyll a concentration were determined for estimating total available food for oyster growth. Environmental parameters including water temperature, salinity and total suspended solid were also monitored for the model. Oyster growth was also monitored by means of measuring shell length and tissue wet weight increase on a monthly basis. Simulation results from the numerical model indicated that chlorophyll a is not a good representative of available food for the oysters in Goseong Bay. In contrast, available food in the water column measured by filtration of the organic particles and analyzed in terms of total lipids, carbohydrates and protein was well matched with simulated oyster growth in the Bay which is similar to observed growth. The model also suggested that oysters have relatively low retention efficiency of $50\%$ or less. This result indicates that oysters in the bay utilize only a part of food particle available in the water column, as reported in other studies.

청예용 호밀의 수확시기가 사초의 수량과 사료가치에 미치는 영향 (effect of Harvesting Daates on Forage Yield and Qality of Winter Rye)

  • This experiment was carried out to determine the effect of harvesting dates on the dry matter yield and quality of "Kool grazer" winter rye(Seca1e cereale L.) as a soilage crop at the Forage Experimental Field of College of Agrlc. and Life Sciences, SNU, Suweon. Four-day harvasting interval was allocated to measure the growth, yield and quality of rye from April 14 to May 18, 1987. Rye was rapidly grown as 3 cm/day and the growth was linearly recorded from the elongation to flowering stage. The dry matter and in vitro digestible dry matter(1VDDM) yields per ha were linearly increased up to 0.26 and 0.19 tonlday from April 14 to May 4 and April 14 to 29, respectively, but major breaks in yield increases occurred at the 50% heading stage, than yields increased. The highest crude protein yield was observed at the first heading stage of April 29, then decreased. Acid detergent fiber (ADF) and crude protein(CP) contents of rye at the heading stage of May 5 were 35 and 17%, respectively and the nutritive value of rye in terms of ADF and CP contents was evaluated as the I st grade hay, but neutral detergent fiber(NDF) content of rye was 47 % at the boot stage ofApril 22 and evaluated as the 2nd grade hay in terms of NDF content. The results indicate that the first heading stage or the end of April would be the most suitable harvesting time for early maturing winter rye as a soilage crop in terms of yield and quality in the middle northwestern coast rcgion of Korea. of Korea.

제주도산 아열대성 어류 18종의 일반성분 및 무기질 함량 (Proximate Composition and Mineral Contents of 18 Subtropical Fish Species from Jeju Island)

  • The proximate composition and mineral contents of 18 subtropical fish species caught off the coast of Jeju Island, Korea were determined. The mean moisture content was highest (78.0%) in the eight species of lean fish, lowest (72.6%) in the four species of medium-fat fish, and intermediate (75.4%) in the six species of low-fat fish. The lipid content was in the order of medium-fat fish > low-fat fish > lean fish. The protein content ranged from 17.7% to 18.9%. The mean macro-mineral contents in the 18 species were 290.2 mg/100 g edible portion K, 146.6 mg P, 113.5 mg Na, 67.2 mg Ca and 38.3 mg Mg. The mean value of micro-mineral contents were 2.1 mg Fe, 1.7 mg Cu and 1.5 mg Zn. These results suggest that subtropical fish from Jeju Island are good sources of protein, lipid and minerals.

Molecular Cloning of the 3'-Terminal Region of Garlic Potyviruses and Immunological Detection of Their Coat Proteins

  • cDNAs complementary to the 3'-terminal regions of two potyvirus genomes were cloned and sequenced. The clone G7 contains one open reading frame (ORF) of 1,338 nucleotides and a 3' untranslated region (3'-UTR) of 403 nucleotides at the 3'-end excluding the 3'end poly(A) tail. The putative viral coat protein (CP) shows 55%-92% amino acid sequence homology to those of Allium potyviruses. The genome size of the virus was analyzed to be about 9.0 kb by Northern blot analysis. Five cDNA clones were screened out using GPV2 oligonucleotide as a probe. One of these clones, DEA72, which has a longest cDNA insert, contains one ORF of 1,459 nucleotides and a 3'-UTR of 590 nucleotides at the 3'-end excluding the 3'-end poly(A) tail. The putative viral CP shows 57%-88% amino acid sequence homologies to those of Allium potyviruses. The genome size of the virus was analyzed to be about 9.6 kb by Northern blot analysis. The results of immunoblot and Northern blot analyses suggest that almost all of the tested garlic plants showing mosaic or streak symptoms are infected with DEA72-potyvirus in variable degrees but rarely infected with G7-potyvirus in variable degrees but rarely infected with DEA72-potyvirus in variable degrees but rarely infected with G7-potyvirus. Immunoelectron microscopy using anti-DEA72 CP antibody shows that this potyvirus is about 750 nm long and flexuous rod shaped.

Identification of Botrytis cinerea, the Cause of Post-Harvest Gray Mold on Broccoli in Korea

  • In this study, we identified the causative agent of post-harvest gray mold on broccoli that was stored on a farmers' cooperative in Pyeongchang, Gangwon Province, South Korea, in September 2016. The incidence of gray mold on broccoli was 10-30% after 3-5 weeks of storage at $3^{\circ}C$. Symptoms included brownish curd and gray-to-dark mycelia with abundant conidia on the infected broccoli curds. The fungus was isolated from infected fruit and cultured on potato dextrose agar. To identify the fungus, we examined the morphological characteristics and sequenced the rDNA of the fungus and confirmed its pathogenicity according to Koch's postulates. The results of the morphological examination, pathogenicity test, and sequencing of the 5.8S rDNA of the internal transcribed spacer regions (ITS1 and ITS4) and three nuclear protein-coding genes, G3PDH, HSP60, and RPB2, revealed that the causal agent of the post-harvest gray mold on broccoli was Botrytis cinerea. To our knowledge, this is the first report of post-harvest gray mold on broccoli in Korea.

Comparison of Extractive Nitrogenous Constituents between the Diploid and the Triploid of Oyster Crassostrea gigas Whole Body

  • In order to investigate the composition of extractive nitrogenous components in the diploid and the triploid oysters, Crassostrea gigas, cultured at the south coast of Korea, the whole edible part (whole body) was analyzed into extractive nitrogen, free amino acids, oligopeptides, ATP and its related compounds, quaternary ammonium bases, and guanidino compounds using specimens collected from April to May of 1992. The major free amino acids in the diploid and the triploid were taurine, proline, alanine, glycine, glutamic acid hypotaurine, glutamine, arginine, aspartic acid, and $\beta-alanine$. There was no conspicuous difference in the constituents of free amino acids between the diploid and the triploid. A lot of hypotaurine was detected in the diploid and the triploid of oyster and the contents of them were 107 mg and 123 mg/100g, respectively. The compounds, glycinebetaine, homarine and trigonelline were found in both the diploid and the triploid. Among them, glycinebetaine was the most prominent in all the samples. The amount of protein, glycogen, extractive nitrogen, oligopeptides, ATP and its related compounds, and free amino acids in the triploid was higher than that of the diploid (p<0.10)

비식용해조의 사료효과 증진에 관한 연구(제1보, 유난의 사료가치 시험) (A STUDY ON THE FOOD EFFECT OF NON-EDIBLE MARINE ALGAE(Part 1: Test on the Food Value for Chicken))

  • This research has been dealt with the nutritional component analysis of two kinds of non-edible marine algae, Sargassum herneri (Turner) C. Ag. and Zostera marina Linne which grow abundantly around the southern coast of Korea, These marine algae were mixed in several kinds of samples rates with the combined feed for poultry sold in the market. These were given to 35 chicken in seven test divisions respectively. We have experimented with 35 chicken grown up for two weeks after hatchout. The combined food for poultry obtained from the market was set up as control divisions. The experiments were as follows: 1) The average weight increase in each test division during feeding (Table 5, Fig.2). 2) Food conversion rate and food efficiency in each test division (Table 7). 3) The comparison of digestive rate of crude protein during feeding (Table 8), The results were as follows: 1) The weight increasing rate of the test animal stock fed the food containing $5\~10\%$ of Sargassum horneri (Turner) C. Ag. powder was higher than the rate of those fed only market food for poultry. 2) The stock given food containing $10\%$ Zostera marine powder showed lower growth than the control divison. 3) No apparent trouble owing to salt component involved in the marine algae was found. 4) The stock given food containing sodium glutaminate and Sargassum horneri (Turner) C. Ag. had better result than that without sodium glutaminate.
