Polyvinylidene fluoride (PVDF) is a stable and biocompatible material that has been broadly used in biomedical applications. Due to its piezoelectric property, the electrospun nanofiber of PVDF has been used to culture electroactive cells, such as osteocytes and cardiomyocytes. Here, taking advantage of the piezoelectric property of PVDF, we have fabricated a PVDF nanofiber scaffolds using an electrospinning technique for differentiating human embryonic stem cells (hESCs) into neural precursors (NPs). Surface coating with a peptide derived from vitronectin enables hESCs to firmly adhere onto the nanofiber scaffolds and differentiate into NPs under dual-SMAD inhibition. Our nanofiber scaffolds supported the differentiation of hESCs into SOX1-positive NPs more significantly than Matrigel. The NPs generated on the nanofiber scaffolds could give rise to neurons, astrocytes, and oligodendrocyte precursors. Furthermore, comparative transcriptome analysis revealed the variable expressions of 27 genes in the nanofiber scaffold groups, several of which are highly related to the biological processes required for neural differentiation. These results suggest that a PVDF nanofiber scaffold coated with a vitronectin peptide can serve as a highly efficient and defined culture platform for the neural differentiation of hESCs.
We develop a ceramic composite separator prepared by coating $ZrO_2$ nanoparticles with a poly(vinylidene fluoride-co-hexafluoropropylene) (PVdF-HFP) copolymer on a polyvinyl alcohol (PVA) mechanical support prepared by electrospinning technique to improve thermal properties. The gurley number of the ceramic composite separator shows much lower value than that of a PE separator even though it possesses the polymeric coating layer with ceramic nanoparticles. In addition, the proposed sample shows higher electrolyte uptake than PE separator, leading to enhancing the ionic conductivity of the proposed sample and, by extension, the rate discharge properties of lithium ion batteries. Thermal stability of the ceramic composite separator is dramatically improved without any degradation in electrochemical performance compared to the performance of conventional PE separators.
Vascular tissue engineering has been accessed to mimic the natural composition of the blood vessel containing intima, media, and adventitia layers. We fabricated mechanically expanded PLLA/PLCL nanofibers using electrospinning and UTM. The pore size of the meshes was increased the gelatin immobilized AAc-PLLA/PLCL nanofibers ($203.30{\pm}49.62microns$) than PLLA/PLCL nanofibers ($59.99{\pm}8.66microns$) after mechanical expansion. To increase the cell adhesion and proliferation, we introduced carboxyl group, and gelatin was conjugated on them. The properties of the PLLA/PLCL nanofibers were analyzed with SEM, ATR-FTIR, TBO staining, and water contact angle measurement, general cell responses on the PLLA/PLCL nanofibers such as adhesion, proliferation, and infiltration were also investigated using smooth muscle cell (SMC). During the SMC culture, the initial viability of the cells was significantly increased on the gelatin immobilized AAc-PLLA/PLCL nanofibers, and infiltration of the cells was also enhanced on them. Therefore, gelatin immobilized AAc-PLLA/PLCL nanofibers and mechanically expanded meshes may be a good tool for vascular tissue engineering application.
The fabrication of gelatin nanofibers by electro spinning has been examined using the TFE/DW co-solvent system. It has been found that no beads-on-string structure was formed for the solution containing ionic salts. The resulting fibers exhibited a uniform diameter ranging from 110 to 125 nm. As the concentration of ionic salts increases, the beads become smaller and more spindle like, due to the increase of viscosity and conductivity. The addition of ionic salts induces a higher charge density on the surface of ejected jet during spinning, leading that higher elongation forces are applied to the jet. The higher enhancement of viscosity and conductivity was observed in gelatin solutions by the use of divalent salt. However, the concentration of ionic salts scarcely affected the variation of fiber diameter. While very low crystallinity was observed from XRD pattern for the sample containing no ionic salt, which increased with increasing the concentration of ionic salts.
We have fabricated a novel thermo-responsive nanofibrous surfaces by grafting PIPAAm by electron beam irradiation onto poly(3-hydroxybutyrate-co-3-hydroxyvalerate)(PHBV) nanofibrous mats. The electrospun PHBV nanofiber structures revealed randomly aligned fibers with average diameter of 400 nm. Increased atomic percent of nitrogen was observed on the PIPAAm-grafted PHBV mats after electron beam irradiation determined by ESCA. The amounts of PIPAAm-grafted onto PHBV films were $6.49{\mu}g/cm^2$ determined by ATR-FTIR. The PIPAAm-grafted surfaces exhibited decreasing contact angles by lowering the temperature from 37 to $20^{\circ}C$, while ungrafted PHBV surfaces had negligible contact angle change. This result indicates that PIPAAm surfaces, which are hydrophobic at the higher temperature, became markedly more hydrophilic in response to a temperature reduction due to spontaneous hydration of the surface-grafted PIPAAm. Thermo-responsive nanofibers showed good tissue compatibility. Cultured cells were well detached and recovered from the surfaces by changing culture temperature from 37 to $20^{\circ}C$.
Poly(${\varepsilon}$-caprolactone) (PCL) nanofibers and PCL/silica membranes were synthesized by sol-gel derived electrospinning and casting, respectively. Smooth PCL nanofibers were obtained from the precursor containing N,N-dimethylformamide (DMF). PCL/silica membranes were prepared by varying the tetraethyl orthosilicate (TEOS) contents from 0 to 40 vol% to investigate the effect of silica addition on mechanical properties and cytotoxicity of the membranes. Although the strength of the membranes decreased from 12 to 8 MPa with increasing the silica content, the strength remained almost constant 7 weeks after dipping in phosphate buffered saline solution (PBS). The strength reduction was attributed to the presence of a patterned surface pores and micro-pores present in the walls between pores. The crystal structure of the membranes was orthorhombic and the crystallite size decreased from 57 to 18 nm with increasing the silica content. From the agar overlay test, the PCL/silica membranes exhibited neither deformation and discoloration nor lysis of L-929 fibroblast cells.
