전해수 및 유기산처리에 의한 양상치에 오염된 Listeria monocytogenes의 생육저해 (Effect of Electrolyzed Water and Organic Acids on the Growth Inhibition of Listeria monocytogenes on Lettuce)

  • 본 연구는 신선편이 양상치를 대상으로 전해수와 유기산의 단독 또는 병용처리에 의한 위해미생물의 살균효과를 조사하였다. 산성전해수는 액상배지에서 L. monocytogenes를 60초 내에 완전히 사멸하였으나, 알칼리전해수는 약 1.7 log CFU/g 정도 감소하였다. 그러나 L. monocytogenes를 접종한 양상치에서 산성전해수는 현저하게 살균력이 감소하여 3분 침지시 약 2.5 CFU/g의 감소를 나타내었으나 알칼리전해수는 산성전해수와 거의 살균력의 차이가 없었다. 한편, L. monocytogenes에 오염된 양상치를 전해수와 유기산 단독 또는 병용처리 하였을 때 무처리구에 비하여 산성전해수와 알칼리전해수는 약 2 log CFU/g 정도의 감소를 나타냈으며, $1\%$ acetic acid, lactic acid, citric acid 및 ascorbic acid를 처리하였을 경우 각각 1.52.5 log CFU/g의 감소를 나타냈으나, 산성전해수와 알칼리전해수에 $1\%$ 유기산을 병용처리 하였을 경우 약 2.63.7 log CFU/g의 현저한 감소를 나타내었다. 특히 산성전해수와 알칼리전해수 모두 $1\%$ citric acid와 병용처리 하였을 때 가장 강한 살균력을 나타내었다. 양상치에 오염된 총균수, 효모 및 곰팡이의 경우도 L. monocytogenes에 오염된 양상치의 살균력과 비슷한 경향을 나타내었다. 또한, L. monocytogenes를 접종한 양상치를 전해수 처리한 후 온도별로 저장하면서 생육변화를 측정한 결과, 병용처리구에서 현저하게 생육이 저해되는 것으로 나타나 양상치를 세척시 알칼리전해수와 $1\%$ citric acid를 병용처리하면 총균수는 물론 L. monocytogenes에 의한 오염방지에 매우 효과적이었다. 한편, SEM 결과, 양상치에 오염된 L. monocytogenes 는 알칼리수와 $1\%$ citric acid의 병용처리구에서는 현저하게 세포막이 파괴되어 세포가 사멸하는 것으로 나타났다.

  • 효모의 구연산 생산성을 향상시키기 위하여 구연산 생성이 향상된 변이균주를 분리.사용하여 비교적 높은 당농도에서의 구연산발효 및 균체 순환식 반연속배양에 의한 구연산발효 실험을 실시하였다. 친균주인 Candida lipolytica S-109를 NTG처리하여 친주균에 비하여 구연산생성능이 30%정도 향상되고 fumarate, malate, isocitrate 생성이 억제된 변이균주 J-24를 분리하였다. 친균주 S-109와 변이균주 J-24를 10% glucose 함유 배지에서 6일간 배양하여 각각 56g/l, 72g/l의 구연산을 생성하였다. 배양액내의 구연산축적량을 향상시키기 위한 비교적 높은 glucose농도는 16%였으며 85g/l의 구연산을 생성하여 53% 수율을 보였다. P/C비, urea 및 yeast extract 농도를 각각 0.8-1.0 $\times$ $10^{-3}$, 0.15%, 0.25%로 할 때 93g/l의 구연산을 축적하여 58% 수율로 향상되었다. 효율적인 생산기를 연장하고 생산물 저해를 억제하며 유도기를 단축하기 위하여 균체순환식 반연속배양을 실시한 결과 그 생산성이 0.79g/l.h로서 회분식의 053g/l.h에 비하여 1.5배 높았다.

  • Chitosan is a polysaccharide with cationic amino groups in its structure and has useful properties as functional materials. Various end-use developments of chitosan are in progress. When the cotton fabric is pretreated with chitosan, the hand property of cotton fabric may be improved expecially for the summer apparel. In this study, as a cross-linking agent to introduce chitosan into cotton, BTCA(butane-1,2,3,4-tetracarboxylic acid) or CA(citric acid) was added in order to prevent detachment of chitosan by the cross-linking. During the cross-linking procedure, via the padding-drying-heat setting, amino groups of chitosan and hydroxyl groups of cotton, carboxyl groups of BTCA/CA are cross-linked by forming anhydrous cyclic rings. Since BTCA has four carboxyl groups, cross-linking by thermal treatment is easy, leading to the trials in wrinkle-recovery treatment of cotton fabrics. However, the high price of the BTCA reagent has been a shortcoming in the actual application for industrial use. Therefore, in this study, we tried the application of CA having three carboxyl groups, which is relatively low priced, as the substituting cross-linking agent. The hand of the treated fabrics were evaluated by measuring physical properties. In addition, based on the physical properties, three-dimensional images were introduced by using 3D CAD systems and results were compared.

TRPHs - 중금속 복합오염토양의 동시 처리를 위한 과황산 산화 - 구연산 세척 혼성공정 개발 (A Continuous Process of Persulfate Oxidation and Citric acid Washing for the Treatment of Complex-Contaminated Soil Containing Total Recoverable Petroleum Hydrocarbons and Heavy Metals)

  • A continuous process of persulfate oxidation and citric acid washing was investigated for ex-situ remediation of complex contaminated soil containing total recoverable petroleum hydrocarbons (TRPHs) and heavy metals (Cu, Pb, and Zn). The batch experiment results showed that TRPHs could be degraded by $Fe^{2+}$ activated persulfate oxidation and that heavy metals could be removed by washing with citric acid. For efficient remediation of the complex contaminated soil, two-stage and three-stage processes were evaluated. Removal efficiency of the two-stage process (persulfate oxidation - citric acid washing) was 83% for TRPHs and 49%, 53%, 24% for Cu, Zn, and Pb, respectively. To improve the removal efficiency, a three-stage process was also tested; case A) water washing - persulfate oxidation - citirc acid washing and case B) persulfate oxidation - citric acid washing (1) - citric acid washing (2). In case A, 63% of TRPHs, 73% of Cu, 60% of Zn, and 55% of Pb were removed, while the removal efficiencies of TRPHs, Cu, Pb, and Zn were 24%, 68%, 62%, and 59% in case B, respectively. The results indicated that case A was better than case B. The three-stage process was more effective than the two-stage process for the remediation of complex-contaminated soil in therms of overall removal efficiency.

구연산 HA임플란트 표면구조에 미치는 영향 (The Effects of Citric Acid on HA coated Implant Surface)

  • The present study was performed to evaluate the effect of citric acid on the change of implant surface microstructure according to application time. Implants with pure titanium machined surface, and HA coated surface were utilized. Pure titanium machined surface and HA coated surface were rubbed with pH 1 citric acid for 30s., 45s., 60s., 90s., and 120s. respectively. Then, the specimens were processed for scanning electron microscopic observation. The following results were obtained. 1. The specimens showed a few shallow grooves and ridges in pure titanium machined surface implants. The roughness of surfaces conditioned with pH 1 citric acid was slightly increased. 2. In HA-coated surfaces, round particles were deposited irregularly. The specimens were not significant differences within 45s. But, began to be changed from 60s. The roughness of surfaces was lessened and the surface dissolution was increased relative to the application time. In conclusion, pure titanium machined surface implants and HA coated surface implants can be treated with pH 1 citric acid for peri-implantitis treatment if the detoxification of these surfaces could be evaluated.

수종의 관주용액의 수산화칼슘제재에 대한 칼슘 용출효과 (Elution of calcium ion from calcium hydroxide products using various root canal irrigants)

  • In endodontic treatment, calcium hydroxide has been used as intracanal medicament. Although calcium hydroxide should be removed thoroughly before permanent root canal filling, no effective method for its removal has been reported. Because of irregularity of root canal walls, root curvatures and anatomic variations, it is insufficient to remove calcium hydroxide from the canal wall only by mechanical instrumentation Considering the chemical effects of irrigants on calcium hydroxide, $Ca^{++}$ dissolving effect from two calcium hydroxide products is investigated, using dis- tilled water, NaOCl, citric acid and EDTA. Vitapex$^{(R)}$ 0.1g and calcium hydroxide 0.03g were dissolved in distilled water, 5% NaOCl, 50% citric acid and 17% EDTA respectively, at 1, 3, 5, 10min. time interval. The solution was filtered using filter paper(pore size $5{\mu}m$) and $Ca^{++}$ concentration was determined by ion chromatography. The result were as follows : 1. Distilled water, NaOCl, citric acid and EDTA abstracted more $Ca^{++}$ from calcium hydroxide than Vitapex$^{(R)}$ except NaOCl 1, 5, 10 time interval. 2. EDTA and citric acid abstracted more $Ca^{++}$ from Vitapex$^{(R)}$ and calcium hydroxide than distilled water or NaOCl. The overall result support the view that water-based calcium hydroxide product is easily removed than oil-based calcium hydroxide product and EDTA, citric acid are more effective in $Ca^{++}$ elution than NaOCl or distilled water.

Study on the Effect of Surface Finishing Methods on Pitting Corrosion Behavior of 304 Stainless Steel Alloy

  • In this study the effect of different surface finishing techniques on the pitting corrosion behaviour of a commercial 304 stainless steel alloy was investigated. Surface finishing methods were divided into two categories, i.e. mechanical and chemical. Mechanical treatment methods include power tooling such as grinding, emery paper brushing, stainless steel wire brushing and stainless steel shot blasting. Chemical treatment methods include chemical passivation (phosphoric acid, citric acid, nitric acid) and electro-cleaning (phosphoric acid and citric acid). Potentiodynamic polarization experiments were carried out in 3.5 wt. % NaCl solution at room temp. (20 $^{\circ}C$). The results showed that chemical treatment methods improved the corrosion resistance of stainless steel 304, measured in terms of pitting potential ($E_{pit}$). Corrosion resistance of the specimens was increased in the order of; electro-cleaning > manual passivation > mechanical cleaning. Surface of electro-cleaned specimens was smoother than rest of the surface treatment methods. Chrome content in chemically treated specimens was higher than in mechanically treated specimens as shown by EDX analysis.

신맛(酸味)이 용혈성 빈혈흰쥐의 혈액성분에 미치는 효과 - 여성빈혈의 간호중재개발을 위한 동물실험 - (Effects of Sour Tastes on Blood Components in Anemic mice - Animal Experiment for Nursing Intervention of Female Anemia -)

  • Many women in the world have suffered from anemia produced by menstruation, pregnancy and delivery. In the theory of oriental medicine, a sour taste is believed to have a tonic effect on the blood. Thus this paper is to investigate the effect of sour tastes on the improvement in anemia-induced female mice, using citric acid and Fructus schizandrae(五味子). The method used in this experiment was the change of RBC, WBC, Hemoglobin, Hematocrit, Fe, and TIBC in the blood of female mice who were fed citric acid and Fructus schizandrae(五味子). The results obtained were as follows ; 1. Compared with the control group (anemia- induced group without treatment), the mean number of RBC in the blood of mice was significantly increased only in evaluating the change of sample group fed Fructus schizandrae (五味子) 500mg/kg. 2. Compared with the control group (anemia- induced group without treatment), the mean number of WBC in the blood of mice was not significantly larger in both sample groups fed citric acid and Fructus schizandrae(五味子). 3. Compared with the control group (anemia- induced group without treatment), the mean number of Hemoglobin in the blood of mice was significantly larger only in evaluating the change of the sample group fed Fructus schizandrae(五味子). 4. Compared with the control group (anemia- induced group without treatment), the mean percentage of Hematocrit in blood of mice was significantly increased only in evaluating the change of sample group fed Fructus schizandrae(五味子) 500mg/kg and sample group fed Fructus schizandrae(五味子) 250mg/kg for 3days. 5. Compared with control group (anemia- induced group without treatment), the mean volume of Fe in serum of mice was significantly increased only in evaluating the change of sample group fed Fructus schizandrae(五味子) 500mg/kg. 6. Compared with control group (anemia- induced group without treatment), the mean TIBC in serum of mice was significantly increased only in evaluating the change of sample group fed Fructus schizandrae(五味子) 500mg/kg and sample group fed Fructus schizandrae(五味子) 250mg/kg for 7days. According to these results, a sour taste is presumed to have a general tonic effect on anemia, but more study must be taken on the effects of citric acid in improving female anemia.

와연형태(窩緣形態)에 따른 와연누출(邊緣漏出)에 관(關)한 실험적(實驗的) 연구(硏究) (A STUDY ON THE MARGINAL LEAKAGE OF RESTORATIONS WITH DIFFERENT CAVOSURFACE MARGINS)

  • The purpose of this study was to evaluate the marginal leakage of glass ionomer cement with different cavosurface margins. 192 class V cavities were prepared on freshly extracted non-carious teeth and glass ionomer cement were inserted according to the manufacturer's instructions. Cavity preparations for this investigation were performed in four groups. The experimental specimens were made by packing the glass ionomer cement (Fuji Ionomer Type II G-C Co. Japan) into the prepared 192 cavities of four groups with different modes: Group I. - The 48 cavities with $90^{\circ}$ butt-joint cavosurface preparation and restored with glass ionomer cement. Group II. - The 48 cavities with butt-joint preparation modified by $135^{\circ}$ beveling the cavosurface in the dentin and restored with glass ionomer cement. Group III. - The 48 cavities with butt-joint preparation modified by cutting a chamfer in the dentin and restored with glass ionomer cement. Group IV. - The same 48 cavities as group I, and overfilled with glass ionomer cement beyond the cavosurface angle. And four groups above described divided into three subgroups by means of conditioning the cavity walls: Control group. - Glass ionomer cement filled in the prepared 64 cavities after being cleaned with a stream of tap water. Phosphoric acid treatment group. - Glass ionomer cement filled in the prepared 64 cavities after being conditioned with a 50% phosphoric acid. Citric acid treatment group. - Glass ionomer cement filled in the prepared 64 cavities after being conditioned with a 50% citric acid. All 192 specimens were immersed in the 2.0% basic fuchsin solution and subjected to thermal stress at one-minute intervals ($4{\pm}2^{\circ}C$ to $60{\pm}2^{\circ}C$) for 70 minutes before exposure to the dye. The specimens were sectioned ecclesiologically through the center of the restorations for different periods of immersion time, 24 hours, 7 days, 14 days 30 days. The sections were examined under a stereoscopic microscope. The results were as follows: 1. The degree of marginal leakage in group II and III was greater than that in group I and IV. 2. The degree of marginal leakage in phosphoric acid treatment group was similar with that in control group. 3. The degree of marginal leakage in citric acid treatment group was less than that in control group. 4. In all groups, the degree of marginal leakage in phosphoric acid treatment group was greater than that in citric acid treatment group. 5. There is no statistical difference of the degree of marginal leakage according to the immersion time in the dye solution.

Evaluating Commercial Spray Applications of Lactic Acid, Hot Water, and Acidified Sodium Chlorite for the Reduction of Escherichia coli on Beef Carcasses

  • This study examined the effects of lactic acid spray, hot water spray, or their combined treatment, as well as the effects of acidified sodium chlorite (ASC), for the decontamination of Escherichia coli on beef carcass surfaces using a commercial intervention system. With this system, the effects of 2 or 4% lactic acid (v/v), hot water ($89{\pm}1^{\circ}C$), or their combined treatment, were examined in terms of reducing inoculated E. coli. ASC (266 ppm), which was adjusted to pH 2.5 using acetic acid or citric acid, was applied using a hand-held spray system. When the beef carcasses were treated with 2 or 4% lactic acid for 10.4 s, less than 1 log reductions of inoculated E. coli were observed. A hot water spray treatment for 9.8 s resulted in a 2.1 log reduction of inoculated E. coli. However, when the hot water was followed with either 2 or 4% lactic acid, no difference in E. coli reduction was found between the hot water alone or the combined treatment with lactic acid. When ASC was adjusted to pH 2.5 with acetic acid and citric acid, 3.8 and 4.1 log reductions of E. coli were observed, respectively. Overall, the lactic acid spray treatment was least effective, and the ASC treatment was most effective, for the E. coli decontamination of beef carcasses. Therefore, these data suggest that ASC would be a more effective intervention against E. coli than most of the methods currently being used. However, more research is required to evaluate the effects of ASC on other organisms, as well as to identify application methods that will not affect meat quality.
